Introduction: Malaria eradication campaigns all over the world are largely based on parasite and vector control. Vector identification, whether morphological or molecular, is an essential component of vector control. this study analyzed the possible causes of indeterminate polymerase chain reaction (PCR) results for mosquito species in Western part of Burkina Faso. Methodology: From July 2021 to November 2021, mosquitoes were collected during the period of high malaria transmission in the village of Séguéré, Houet province, Burkina Faso, and morphologically identified. After DNA extraction, samples were amplified by sine 200× PCR to identify species of the Anopheles gambiae complex. Indeterminate samples were then selected for further analysis. the parameters studied were: DNA dilution, the effect of protocol adjusting, and the type of protocol used. Results: A total of 130 “indeterminate” DNAs diluted 1:10 were analyzed. After dilution, the mean amount was 14.73 ± 3.59 ng/μL and absorbance 1.71 ± 0.1. PCR chain reaction yielded 94.62% (123/130) anopheline species in SINE PCR, 5.38% (7/130) “negative”. A significant difference between SINE PCR before dilution and after dilution was observed (P tification tests carried out using other protocols gave no positive results. From these results, we note that the adaptation of the protocol significantly reduced the polymerase amplification results of the species. Conclusion: It is therefore necessary to respect the amplification protocols. However, the persistence of “indeterminate” results suggests that further studies should be carried out to shed more light on the subject.

AIM: to investigate the effects of dextrans of various molecular weights (Mw) during a 12 h cold storage time-course on energetics, histology and mucosal infiltration of fluorescein isothiocyanate (FItC)-dextran. MEtHoDS: Rodent intestines were isolated and received a standard University of Wisconsin vascular flush followed by intraluminal administration of a nutrientrich preservation solution containing dextrans of varying Mw: Group D1, 73 kdal; Group D2, 276 kdal; Group D3, 534 kdal; Group D4, 1185 kdal; Group D5, 2400 kdal. RESULtS: Using FItC-labeled dextrans, fluorescent micrographs demonstrated varying degrees of mucosal infiltration; lower Mw (groups D1-D3: 73-534 kdal) dextrans penetrated the mucosa as early as 2 h, whereas the largest dextran (D5: 2400 kdal) remained captive within the lumen and exhibited no permeability even after 12 h. After 12 h, median injury grades ranged from 6.5 to 7.5 in groups D1-D4 (73-1185 kdal) representing injury of the regenerative cryptal regions and submucosa; this was in contrast to group D5 (2400 kdal) which exhibited villus denudation (with intact crypts) corresponding to a median injury grade of 4 (P of tissue energetics reflected a strong positive correlation between Mw and adenosine triphosphate (r 2 = 0.809), total adenylates (r 2 = 0.865) and energy charge (r 2 = 0.667).CoNCLUSIoN: our data indicate that dextrans of Mw > 2400 kdal act as true impermeant agents during 12 h ischemic storage when incorporated into an intraluminal preservation solution.

one of the most promising tools for the control of fungal plant diseases is spray-induced gene silencing(SIGS).In SIGS,small interfering RNA(siRNA)or double-stranded RNA(dsRNA)targeting essential or virulence-related pathogen genes are exogenously applied to plants and postharvest products to trigger RNA interference(RNAi)of the targeted genes,inhibiting fungal growth and ***,SIGS is limited by the unstable nature of RNA under environmental *** use of layered double hydroxide or clay particles as carriers to deliver biologically active dsRNA,a formulation termed BioClay^(tM),can enhance RNA durability on plants,prolonging its activity against ***,we demonstrate that dsRNA delivered as BioClay can prolong protection against Botrytis cinerea,a major plant fungal pathogen,on tomato leaves and fruit and on mature chickpea *** increased the protection window from 1 to 3 weeks on tomato leaves and from 5 to 10 days on tomato fruits,when compared with naked *** flowering chickpea plants,BioClay provided prolonged protection for up to 4 weeks,covering the critical period of poding,whereas naked dsRNA provided limited *** research represents a major step forward for the adoption of SIGS as an eco-friendly alternative to traditional fungicides.
