Malus x domestica microRNA MdmiR285N is a potential key regulator of plant immunity,as it has been predicted to target 35 RNA transcripts coding for different disease resistance proteins involved in plant defense to *** this study,the promoter region of MdmiR285N was isolated from the apple genome and analyzed in silico to detect potential regulatory regions controlling its transcription.A complex network of putative regulatory elements involved in plant growth and development,and in response to different hormones and stress conditions,was *** of theβ-Glucoronidase(GUs)reporter gene driven by the promoter of MdmiR285N was examined in transgenic apple,demonstrating that MdmiR285N was expressed during the vegetative growth ***,in transgenic Arabidopsis thaliana,spatial and temporal patterns of GUs expression revealed that MdmiR285N was differentially regulated during seed germination,vegetative phase change,and reproductive *** elucidate the role of MdmiR285N in plant immunity,MdmiR285N expression in wild-type apple plants and GUs activity in transgenic apple and Arabidopsis thaliana plants were monitored in response to Erwinia amylovora and Pseudomonas syringae *** DC3000.A significant decrease of MdmiR285N levels and GUs expression was observed during host-pathogen ***,these data suggest that MdmiR285N is involved in the biotic stress response,plant growth,and reproductive development.

Genotoxicity testing for radon exposure： Dolichopoda （Orthoptera, Rhaphidophoridae） as potential bio-indicator of confined environments

Radon represents the major source of natural radioactivity in confined environments. Despite the clear evidence of a direct association between residential exposure and human lung cancer provided by case-control studies, results relating indoor exposure and genotoxic/mutagenic effect induction are still contradictory. The present study attempts to estimate the genotoxic effects induced by exposure to radioactive radon in wild cricket populations sampled from caves where varying concentrations of radon are present. Cave crickets are also tested as possible bio-indicator organisms of genotoxic potential of contaminated residential and confined environments. six caves in Central Italy are considered covering a broad spectrum of radon radioactivity concentration （221-26,000 Bq/m3）. Dolichopoda specimens were sampled from each cave; both haemocytes and brain cells taken from individuals were tested for responsiveness to DNA damage induced by radon through the Comet assay. specimens from the least radioactive cave, housed in controlled conditions for 60 days before analysis, were used as control group. statistically significant increase of DNA damage was found in all groups of individuals from each cave, for both cell types. Very low values of all Comet parameters were found in control group individuals, which gave indications of a good responsiveness of the organism to the variable environmental levels of radioactive contamination. Results indicate that cave crickets represent a reliable tool for the detection of genotoxic potential induced by radioactive contamination of confined environments and can be proposed as a possible bio-indicator system for air （-radioactive） pollution related to indoor exposure [Current Zoology 60 （2）： 299-307, 2014].

Missing metastases as a model to challenge current therapeutic algorithms in colorectal liver metastases

In oncosurgical approach to colorectal liver metastases, surgery remains considered as the only potentially curative option, while chemotherapy alone represents a strictly palliative treatment. However, missing metastases, defined as metastases disappearing after chemotherapy, represent a unique model to evaluate the curative potential of chemotherapy and to challenge current therapeutic algorithms. We reviewed recent series on missing colorectal liver metastases to evaluate incidence of this phenomenon, predictive factors and rates of cure defined by complete pathologic response in resected missing metastases and sustained clinical response when they were left unresected. According to the progresses in the efficacy of chemotherapeutic regimen, the incidence of missing liver metastases regularly increases these last years. Main predictive factors are small tumor size, low marker level, duration of chemotherapy, and use of intra-arterial chemotherapy. Initial series showed low rates of complete pathologic response in resected missing metastases and high recurrence rates when unresected. However, recent reports describe complete pathologic responses and sustained clinical responses reaching 50%, suggesting that chemotherapy could be curative in some cases. Accordingly, in case of missing colorectal liver metastases, the classical recommendation to resect initial tumor sites might have become partially obsolete. Furthermore, the curative effect of chemotherapy in selected cases could lead to a change of paradigm in patients with unresectable liver-only metastases, using intensive first-line chemotherapy to intentionally induce missing metastases, followed by adjuvant surgery on remnant chemoresistant tumors and close surveillance of initial sites that have been left unresected.
