Overweight and obesity in young adults with patellofemoral pain:Impact on functional capacity and strength

Purpose:This study aimed to(a)investigate the proportion of overweight/obesity in a cohort of young adults with patellofemoral pain(PFP)and(b)explore the association of body mass index(BmI),body fat,and lean mass with functional capacity and hip and knee strength in people with ***:We included a mixed-sex sample of young adults(18-35 years old)with PFP(n=100).measurements for BmI,percentage of body fat,and lean mass(assessed by bioelectrical impedance)were *** capacity was assessed by the Anterior Knee Pain Scale,plank test,and single-leg hop *** of the knee extensors,knee flexors,and hip abductors was evaluated isometrically using an isokinetic *** proportion of overweight/obesity was calculated based on *** association between BmI,body fat,and lean mass and functional capacity and strength was investigated using partial correlations,followed by hierarchical regression analysis,adjusted for covariates(sex,bilateral pain,and current pain level).Results:A total of 38%of our cohort had their BmI categorized as overweight/*** BmI was associated with poor functional capacity(ΔR^(2)=0.06-0.12,pmass was associated with poor functional capacity(ΔR^(2)=0.04-0.13,p≤0.032)and reduced strength(ΔR^(2)=0.29-0.31,pmI,body fat,and lean mass should be considered in the assessment and management of young people with PFP because it may be detrimental to function and strength.

Background:We compared body mass index(BmI),body fat,and skeletal muscle mass between(1) a mixed-sex nonathletic cohort of people with patellofemoral pain(PFP) and pain-free people,and(2) a nonathletic cohort of people with PFP and pain-free people subgrouped by sex(i.e.,men and women with PFP ***-free men and women).methods:This cross-sectional study included 114 people with PFP(71 women,43 men) and 54 pain-free controls(32 women,22 men).All participants attended a single testing session to assess body composition measures,which included BmI,percentage of body fat(%BFBioimpedance),and skeletal muscle mass(both assessed by bioelectrical impedance analysis),and percentage of body fat(%BFskinfoid)(assessed by skinfold caliper analysis).A one-way univariate analysis of covariance(age and physical activity levels as covariates) was used to compare body composition measures between groups(i.e.,PFP ***-free group;women with PFP ***-free women;men with PFP ***-free men).Results:Women with PFP presented significantly higher BmI,%BFBiompedance,and %BFSkinfold,and lower skeletal muscle mass compared to pain-free women(p≤0.04;effect size:-0.47 to 0.85).men with PFP and men and women combined had no differences in BmI,%BFBioimpedance,%BFSkinfold,and skeletal muscle mass compared to their respective pain-free groups(p> 0.05).Conclusion:Our findings indicate that BmI and body composition measures should be considered as part of the evaluation and management of people with PFP,especially in women,who have demonstrated higher BmI and body fat and lower skeletal muscle mass compared to pain-free *** studies should not assess body composition measures in a mixed-sex population without distinguishing men participants from women participants.

Discrepancies between the responses to skin prick test to food and respiratory antigens in two subtypes of patients with irritable bowel syndrome

AIm:To compare the response to skin prick tests (SPTs) to food antigens (FAs) and inhalant allergens (IAs) in patients with two subtypes of irritable bowel syndrome (IBS) and healthy controls. mETHODS:We compared the results of SPTs for IAs and FAs in 87 volunteers divided into three groups:diarrhea predominant IBS (D-IBS) GroupⅠ(n = 19), constipation predominant IBS (C-IBS) Group Ⅱ (n = 17), and normal controls Group Ⅲ (n = 51). RESULTS:Of the 285 tests (171 for FAs and 114 for IAs) performed in GroupⅠwe obtained 45 (26.3%) positive responses for FA and 23 (20.1%) for IA. Of the 153 tests for FA in Group Ⅱ, we obtained 66 (20.1%) positive responses, and of the 102 tests for IA, we obtained 20 (19.6%) positive responses. Of the 459 tests for FA performed in Group Ⅲ, we obtained 39 (84%) positive responses, and of the 306 for IA, we obtained 52 (16.9%) positive responses. The numbers of positive responses were not significantly different between the three groups, but in the D-IBS group, the number of SPTFA responses differed significantly from those for the other two groups (P mall number of cases studied, the higher reactivity to FAs in GroupⅠcompared to Groups Ⅱ and Ⅲ adds new information, and suggests the presence of a possible alteration in intestinal epithelial function.

ANP impairs the dose-dependent stimulatory effect of ANG II or AVP on H^(+)-ATPase subcellular vesicle trafficking

The effect of angiotensin II (ANG II) or arginine vasopressin (AVP) alone or plus atrial natriuretic peptide (ANP) on H+-ATPase subcellular vesicle trafficking was investigated in mDCK cells following intracellular pH (pHi) acidification by exposure to20 mmNH4Cl for 2 min in a Na+-free solution containing Schering 28080, conditions under which H+-AT-Pase is the only cell mechanism for pHi recovery. Using the acridine orange fluorescent probe (5mm) and confocal microscopy, the vesicle movement was quantified by determining, for each experimental group, the mean slope of the line indicating the changes in apical/basolateral fluorescence density ratio over time during the first 5.30 min of the pHi recovery period. Under the control conditions, the mean slope was 0.079 ± 0.0033 min-1 (14) and it increased significantly with ANG II [10-12 and 10-7 m, respectively to 0.322 ± 0.038 min-1 (13) and 0.578 ± 0.061 min-1 (12)] or AVP [10-12 and 10-6 m, respectively to 0.301 ± 0.018 min-1 (12) and 0.687 ± 0.049 min-1 (11)]. However, in presence of ANP (10-6 m, decreases cytosolic free calcium), dimethyl-BAPTA/Am (5 × 10-5 m, chelates intracellular calcium) or colchicine (10-5 m, 2-h preincubation;inhibits microtubule-dependent vesicular trafficking) alone or plus ANG II or AVP the mean slopes were similar to the control values, indicating that such agents blocked the stimulatory effect of ANG II or AVP on vesicle trafficking. The results suggest that the pathway responsible for the increase in cytosolic free calcium and the microtu-bule-dependent vesicular trafficking are involved in this hormonal stimulating effect. Whether cytosolic free calcium reduction represents an important direct mechanism for ANP impairs the dose-dependent stimulatory effect of ANG II or AVP on H+-ATPase subcellular vesicle trafficking, or is a side effect of other signaling pathways which will require additional studies.

乳腺癌是女性最常见的恶性肿瘤之一,在新增癌症病例中占23%。乳腺癌在女性癌症患者中的致死率位居第二位,仅次于肺癌。天然化合物与合成化合物相比具有低毒的特点,是许多疾病的治疗药物生产的最初来源。木香烯内酯是一种倍半萜内酯,由多种植物化学成分组成,通过细胞毒素作用对多种癌细胞发挥抗癌效应;但木香烯内酯在乳腺癌治疗中的可能作用尚不明确。本研究旨在探索天然化合物对mCF7细胞增殖的影响,研究ROS在由天然化合物诱导的乳腺癌细胞凋亡中潜在的治疗作用。我们首先检测了木香烯内酯对乳腺癌mCF7细胞增殖的影响,并证明其具有高效的抗癌能力。我们发现,木香烯内酯的IC50值为50μm,是强有力的细胞增殖抑制剂。利用荧光探针钙绿化素AV/PI活体实验,我们发现木香烯内酯对mCF细胞的增殖抑制和凋亡诱导效应,降低了 mCF-7细胞在剂量依赖下的生存能力。为了研究木香烯内酯诱导的细胞凋亡,我们采用了流式细胞检测技术。结果显示,在剂量依赖下,木香烯内酯诱导乳腺癌mCF7细胞凋亡。由于ROS的过度表达会诱导氧化压力,导致细胞功能丧失和细胞凋亡。因此,我们利用荧光染色的ROS追踪素—双氯荧光乙酸乙酯来检测细胞内ROS水平,并发现木香烯内酯显著提高ROS水平。有趣的是,细胞被一种特殊的ROS抑制剂乙酰半胱氨酸预处理之后,ROS介导的细胞凋亡效应被消除。所以在本研究中,我们证明木香烯内酯通过提高ROS水平,进而诱导细胞凋亡。然后我们研究了木香烯内酯作为外源诱导剂对乳腺癌mCF7细胞凋亡的影响。通过流式细胞检测技术和蛋白质免疫印记分析了 DNA成分和细胞凋亡相关的蛋白质表达水平。在乳腺癌mCF7细胞中,我们检测了木香烯内酯对凋亡外源途径(死亡受体途径)激活及对凋亡效应蛋白质Caspase-3和PARP的活化作用。结果显示,在25um和50um木香烯内酯处理的细胞中,蛋白质Caspase-3分裂为一个17kDa和一个20kDa的片段,蛋白质PARP也被活化剪切。在此基础上,我们进一步检测了木香烯内酯乳腺癌mCF7细胞凋亡过程中对凋亡内源途径(线粒体途径)的激活效应。我们检测了线粒体凋亡途径中抗凋亡蛋白质BcL2和促凋亡蛋白Bax的表达水平,并证明木香烯内酯可下调BcL2蛋白表达和上调Bax蛋白表达,从而证明木香烯内酯通过活化两种凋亡途径从而诱导乳腺癌mCF7细胞产生凋亡。我们的研究结果表明,木香烯内酯对乳腺癌细胞具有增殖抑制和凋亡促进效应,为人类癌症治疗提供了一种可能的新策略。

Quantification of Flavonoids in Brazilian Orange Peels and Industrial Orange Juice Processing Wastes

The flavonoid content in orange peels of different Brazilian citrus varieties such as bahia, lima, lima-of-persian, morcote, pera, ponkan, seleta, cravo, kinkan and pomelo was assessed. Industry processing juice wastes such as bagasse, bagasse residues, animal feeding bagasse, pulp WEUE and CORE-wash were also analyzed. The HPLC analysis indicates that the most abundant flavonoids found in these Brazilian citrus peels are hesperidin and naringin. The solvents used are selective for flavonoid extraction, and depending on their polarity, glycoside or aglycone flavonoids are extracted. The use of multivariate analysis shows that DmSO is the best solvent to extract glycosides flavanones while hexane displays high selectivity in the extraction of polymethoxylated flavones. The flavonoids present in the orange wastes, obtained at different stages of the industrial processing, are qualitative and quantitatively different. The identification and quantification of the flavonoid composition in each Brazilian citrus variety were evaluated and allowed the selection of the best solvent for the extraction of each specific class of flavonoids. These compounds were found to be more abundant in the fruit peels than in their juices, revealing their great industrial potential. The residual portion of the processing juices is also rich in flavonoids, depending on the processing step.

The relationships between climate conditions and wood density in tropical forests are still poorly *** quantify spatial dependence of wood density in the state of minas Gerais(mG,Brazil),map spatial distribution of density,and correlate density with climate variables,we extracted data from the Forest Inventory of minas Gerais for 1988 trees scaled throughout the territory and measured wood density of discs removed from the *** variables were extracted from the database of the Ecological-Economic Zoning of minas *** spatial analysis,tree densities were measured at 44 georeferenced sampling *** data were subjected to exploratory analysis,variography,cross-validation,model selection,and ordinary *** relationships between wood density and environmental variables were calculated using dispersion matrices,linear correlation,and *** density proved to be highly spatially dependent,reaching a correlation of 96%,and was highly continuous over a distance of 228 *** distribution of wood density followed a continuous gradient of 514-659 kg m^(−3),enabling corre-lation with environment *** was correlated with mean annual precipitation(−0.57),temperature(0.63),and evapotranspiration(0.83).Geostatistical methods proved useful in predicting wood density in native tropical forests with different climate *** results confirmed the sensitivity of wood density to climate change,which could affect future carbon stock in forests.

Crystal-oriented wrinkles with origami-type junctions in few-layer hexagonal boron nitride

Understanding layer interplay is the key to utilizing layered heterostructures formed by the stacking of different two-dimensional materials for device applications. Boron nitride has been demonstrated to be an ideal substrate on which to build graphene devices with improved mobilities. Here we present studies on the morphology and optical response of annealed few-layer hexagonal boron nitride flakes deposited on a silicon substrate that reveal the formation of linear wrinkles along well-defined crystallographic directions. The wrinkles formed a network of primarily threefold and occasionally fourfold origami-type junctions throughout the sample, and all threefold junctions and wrinkles formed along the armchair crystallographic direction. Furthermore, molecular dynamics simulations yielded, through spontaneous symmetry breaking, wrinkle junction morphologies that are consistent with both the experimental results and the proposed origami-folding model. Our findings indicate that this morphology may be a general feature of several two-dimensional materials under proper stress-strain conditions, resulting in direct consequences in device strain engineering.
