Impact of vitamin D supplementation on the outcome of tuberculosis treatment： a systematic review and meta-analysis of randomized controlled trials

Background Vitamin D supplementation is believed to be beneficial in the treatment of patients with tuberculosis （TB）,however,results from clinical trials have been *** We performed a systematic literature search across MEDLINE,EMBASE,the Cochrane Central Register of Controlled Trials,Springer,EBSCO,ProQuest,HighWire Press,and Web of Science,published as of December *** individually inspected citations and extracted data *** estimated pooled risk ratios （RR） and 95％ confidence intervals （CI） using random-effect *** also assessed risk of bias using the Jadad scale and the quality of the evidence using *** included all randomized controlled trials comparing vitamin D with or without standard TB therapy or *** A total of five studies were analyzed in our meta analysis covering 841 newly-diagnosed TB *** receiving vitamin D supplementation had a 39％ reduced risk of sputum smear or culture positive after six weeks of antiTB treatment than those in the control group,although this is not statistically significant （pooled RR 0.61,95％ CI 0.24 to 1.56,P=0.30）.Apart from an increased serum vitamin D level in the supplement group after eight weeks of treatment there was no evidence of any additional adverse effects related to vitamin *** The meta analysis results indicate that vitamin D supplementation does not seem to have any beneficial effect in the treatment of *** rigorous randomized controlled trials are needed to explore whether the supplementation of vitamin D could shorten treatment duration and to confirm whether the polymorphisms of vitamin D receptor have any potentially beneficial effect.

Relatively poor in vitro rooting has limited the large-scale commercial production of tree *** this study,on the basis of transcriptome sequencing,differentially expressed genes and the associated metabolic pathways were identified in tree peony roots at different stages of root formation under sandy loam cultivation.A total of 31.63 Gb raw data were generated and 120,188 unigenes(mean length of 911.98 bp)were annotated according to six databases(NR,NT,GO,KEGG,COG,and Swiss-Prot).Analyses of the ungerminated root primordium period,induced root primordium period,and root formation period detected 8,232,6,907,and 10,687 differentially expressed genes related to 133,132,and 133 metabolic pathways,*** significantly differentially expressed genes(Unigene13430_All and ***1_All)were associated with the auxin *** full-length Unigene13430_All coding sequence(843 bp)encoded 280 amino acids,whereas the full-length ***1_All coding sequence(1,470 bp)encoded 489 amino ***13430_All and ***1_All were identified as IAA gene family members and were respectively named PsIAA27 and *** qRT-PCR analysis and functional verification indicated that the expressions of PsARF19 and PsIAA27 in tree peony seedlings,cuttings and grafted seedlings were significant ***19 promoted root development,it might be a regulatory gene related to the formation of tree peony roots,while PsIAA27 inhibited lateral root development,and it might be involved in controlling auxin sensitivity during root *** results of this study may form the basis of future investigations on the mechanism mediating peony root *** transcriptome data will be an excellent resource for researchers interested in characterizing the rooting-related tree peony genes.

MicroRNAs play a pivotal role in the interaction between pathogens and host cells,participating as host defense mechanisms,as well as exploited by pathogens to subvert host cellular *** miR-127 has been reported to be involved in the control of innate immunity while the underlying mechanisms for miR-127 regulation of antibacterial innate immunity remain largely ***,we report microRNA regulations upon infection of mammalian cells with the common pathogen,Staphylococcus aureus,and its vital role in the bacteria clearance and pulmonary *** expression of miR-127 was induced by Staphylococcal infection in a time-and dose-dependent ***-127 up-regulates proinflammatory cytokines and antibacterial molecules in macrophages,leading to the enhanced clearance of ***,mice treated with miR-127 mimic displayed the alleviated bacterial pneumonia with reduced bacterial ***,our data revealed that miR-127 up-regulates the phosphorylation and ubiquitination of transcription factor STAT3 through inhibiting the expression of *** with STAT3 inhibitor or over-expression of A20 largely abolished the effect of miR-127 on the production of anti-bacterial *** thus for the first time reveal the unappreciated role for miR-127 in mediating antibacterial innate immune responses though targeting A20/STAT3 axis.

The EGF receptor tyrosine kinase inhibitors(EGFR-TKI) including Gefitinib have become a standard therapy in patients with non-small cell lung cancer(NSCLC).Unfortunately,acquired resistance eventually limits the clinical effects and application of gefitinib.P-elemene has been used in the clinic as one of the national II novel antitumor drugs in *** this study,we found that P-elemene significantly enhanced the susceptibility of lung cancers to gefitinib,as revealed by the decreased proliferation,migration,and invasion of A549 and H1299 ***,our data showed that the combination of P-elemene and gefitinib reversed the epithelial-mesenchymal transition(EMT) of lung cancer cells and reduced their stem-like ***,we demonstrated that the co-operative effect of p-elemene and gefitinib was largely dependent on their regulation of the epigenetic modifier *** ly,we showed that P-elemene and gefitinib synergistically inhibited the development of xenografts tumors in *** the adverse side-effect of chemotherapeutics and the increasing emergence of chemo-resistant cancers,our data suggest that the development of P-elemene and gefitinib might be a novel promising treatment for lung cancers.

Rheumatoid arthritis is a chronic autoinflammatory disease. Interlenkin-1b(IL-1b) is a key factor of cartilage destruction in rheumatic disease, but the regulation of IL-1b in rheumatoid arthritis is limited by the absence of suitable mouse models of the disease. Here, we show that ASC, a component of the inflammasome complex promotes IL-1b secretion and modulates synovial inflammation in the K/Bx N serum transfer induced arthritis models. That it crucially relies on the ASC inflammasome and IL-1b receptor signaling, but that regulation is unknown. In the macrophages, when we lacking PLZF has decreased the protein expression level of the inflammasome adaptor ASC, in contrast the pro IL-1b is not altered. As a result, PLZF deficiency in the macrophages significantly reduced ASC inflammasome-mediated caspase-1 activation, pyroptosis and IL-1 b secretion by number of NLRP3 stimuli. Importantly, decreased ASC inflammasome activation limited the pathology of rheumatoid arthritis was found in the PLZF deficiency mice. These results reveal PLZF as a novel regulator of ASC inflammasome mediated capase-1 activation and IL-1b secretion, and subsequently promote rheumatoid arthritis associated inflammasome disease and cartilage destruction.

Dysregulation of immune signaling has been associated with the aging process,but the cellular and molecular basis for this remains *** specifically,little is known about how telomere shortening impacts host defensive response and hence shapes disease *** we employed late generation Terc knockout(G3 Terc) mice to investigate the role of telomere dysfunction in host innate immunity and in inflammation-induced tissue *** the overall capability to eradicate pathogens appeared to be enhanced in mice with dysfunctional telomeres,a dramatic increase in disease vulnerability was seen in these animals upon staphylococcal infection,as manifested by exaggerated lung inflammation and increased ***,telomere dysfunction led to mitochondrial abnormalities,metabolic imbalance,and hence hyper-activation of NLRP3-mediated inflammasome in Terc macrophages relative to WT control cells following bacterial *** inhibition of IL-1 R pathway or alleviation of mitochondrial oxidant stress significantly reduced telomere-related ***,we found that one of the key self-limiting regulator of inflammatory signaling,tumor necrosis factor-associated interactive protein 3(TNFAIP3),controlled by peroxisome proliferator-activated receptor-gamma coactivator(PGC)-1α and estrogen-related receptor(ERR)α,was impaired under telomere dysfunction and contributed to inflammation propagation and disease ***,we herein provide the first evidences to connect telomere to metabolism,immunity and inflammation,which we believe would shed a new light on age-related pathologies.

Currently,acute lung injury(ALI) and acute respiratory distress syndrome(ARDS) featured with severe hypoxemia,airway dysfunction,and extensive inflammation lack effective *** activation of macrophages with mitochondria dismiss was proved to be associated with pathogenesis of *** from adipose-derived mesenchymal stem cells(AdMSC-Exos) have been distinguished by their low immunogenicity,lack of tumorigenicity,and high clinical safety,but their role in treating ALI and the mechanism involved need to be *** this study,we showed that AdMSC-Exos provided profound protection against LPS-induced ALI in mice,accompanied by macrophages shifting from M1 proinflammatory to M2 anti-inflammatory ***,our data indicated that AdMSC-Exos deliver mitochondrial component to recipient macrophages,which were metabolically disturbed during the development of ***,transfering of AdMSC-Exos significantly improved macrophages mitochondrial integrity,mitochondrial membrane potential(MMP) and oxidative phosphorylation level,leading to the resumption of metabolic and immune homeostasis of airway macrophages and mitigating lung inflammatory ***,our study reveals the potential role for AdMSC-Exos in protecting ALI,and suggests that macrophage-centered and mitochondria-targeted strategy might become a promising treatment for certain inflammatory lung diseases.
