Programmed cell death induced by modified clay in controlling Prorocentrum donghaiense bloom

Modified clay(MC),an effective material used for the emergency elimination of algal blooms,can rapidly reduce the biomass of harmful algal blooms(HABs)via flocculation.After that,MC can still control bloom population through indirect effects such as oxidative stress,whichwas initially proposed to be related to programmedcell death(PCD)at molecular level.To further study theMC induced cell death in residual bloom organisms,especially identifying PCD process,we studied the physiological state of the residual Prorocentrum donghaiense.The experimental results showed that flocculation changed the physiological state of the residual cells,as evidenced by growth inhibition and increased reactive oxygen species production.Moreover,this research provides biochemical and ultrastructural evidence showing that MC induces PCD in P.donghaiense.Nuclear changes were observed,and increased caspase-like activity,externalization of phosphatidylserine and DNA fragmentation were detected in MC-treated groups and quantified.And the mitochondrial apoptosis pathway was activated in both MC-treated groups.Besides,the features of MC-induced PCD in a unicellular organism were summarized and its concentration dependent manner was proved.All our preliminary results elucidate the mechanism through which MC can further control HABs by inducing PCD and suggest a promising application of PCD in bloom control.

The analysis of endogenous glycoproteins and glycopeptides in human body fluids is of great importance for screening and discovering disease biomarkers with clinical significance.However,the presence of interfering substances makes the direct quantitative detection of low-abundance glycoproteins and glycopeptides in human body fluids one of the great challenges in analytical chemistry.Magnetic solid phase extraction(MSPE)has the advantages of easy preparation,low cost and good magnetic responsiveness.Magnetic adsorbents are the core of MSPE technology,and magnetic adsorbents based on different functional materials are widely used in the quantitative analysis of glycoproteins and glycopeptides in human body fluids,making it possible to analyze glycoproteins and glycopeptides with low abundance as well as multiple types,which provides a technical platform for screening and evaluating glycoproteins and glycopeptides in body fluids as disease biomarkers.In this paper,we focus on the recent advances in the application of MSPE technology and magnetic adsorbents for the separation and enrichment of glycoproteins and glycopeptides in human body fluids,and the future trends and application prospects in this field are also presented.

Design and mechanism insight on SiC quantum dots sensitized inverse opal TiO_(2) with superior photocatalytic activities under sunlight

The combination of SiC quantum dots sensitized inverse opal TiO_(2) photocatalyst is designed in this work and then applied in wastewater purification under simulated sunlight.From various spectroscopic techniques,it is found that electrons transfer directionally from SiC quantum dots to inverse opal TiO_(2),and the energy difference between their conduction/valence bands can reduce the recombination rate of photogenerated carriers and provide a pathway with low interfacial resistance for charge transfer inside the composite.As a result,a typical type-II mechanism is proved to dominate the photoinduced charge transfer process.Meanwhile,the composite achieves excellent photocatalytic performances(the highest apparent kinetic constant of 0.037 min^(-1)),which is 6.2 times(0.006 min^(-1))and 2.1 times(0.018 min^(-1))of the bare inverse opal TiO_(2) and commercial P25 photocatalysts.Therefore,the stability and non-toxicity of SiC quantum dots sensitized inverse opal TiO_(2) composite enables it with great potential in practical photocatalytic applications.

Natural gas hydrate(NGH)has recently received more attention as a cleaner alternative energy source that not only reduces carbon emissions caused by the use of conventional fossil fuels but also plays a key role in global climate change.Furthermore,hydrate-based technologies,particularly hydrate-based carbon capture and storage,have enormous promise for decreasing global carbon emissions,and porous media play an important role in all hydrate-based technologies.Accordingly,this paper reviews the recent applications of porous media in the field of methane hydrate(MH)formation and analyzes the influence of porous media systems on MH phase equilibria and formation kinetics.This is because the efficiency of hydrate-based technologies is determined mainly by the phase equilibrium and formation kinetics of hydrates.The influence of the nature of the media on MH formation in porous media systems is comprehensively summarized to understand how porous media can efficiently enhance the kinetics of hydrate formation.Promoters are necessary for rapid hydrate formation,and the effect of various promoters on MH formation was also evaluated.Based on the aforementioned overview and understanding,the mechanisms for MH formation in various porous media systems are proposed.Finally,the future perspectives and challenges of hydrate-based technologies in tackling global climate change were discussed.This review provides a fundamental understanding of the application and development of porous media in rapid hydrate formation,a fair evaluation of the performance of various porous media systems,and critical insights into major research foci.

Role of helicity of a-helical antimicrobia peptides to improve specificity

A major barrier to the use of antimicrobial peptides as antibiotics is the toxicity or ability to lyse eukaryotic cells. In this study, a 26-residue amphipathic a-helical antimicrobial peptide A12L/A20L （Ac-KWKSFLKTFKSLK KTVLHTLLKAISS-amide） was used as the framework to design a series of D- and L-diastereomeric peptides and study the relationships of helicity and biological activi- ties of o-helical antimicrobial peptides. Peptide helicity was measured by circular dichroism spectroscopy and demonstrated to correlate with the hydrophobicity of peptides and the numbers of D-amino acid substitu- tions. Therapeutic index was used to evaluate the selectivity of peptides against prokaryotic cells. By introducing D-amino acids to replace the original L-amino acids on the non-polar face or the polar face of the helix, the hemolytic activity of peptide analogs have been significantly reduced. Compared to the parent peptide, the therapeutic indices were improved of 44-fold and 22-fold against Gram-negative and Gram- positive bacteria, respectively. In addition, D- and L-diastereomeric peptides exhibited lower interaction with zwitterionic eukaryotic membrane and showed the significant membrane damaging effect to bacterial cells. helicity was proved to play a crucial role on peptide specificity and biological activities. By simply replacing the hydrophobic or the hydrophilic amino acid residues on the non-polar or the polar face of these amphipathic derivatives of the parent peptide with D-amino acids, we demonstrated that this method could have excellent potential for the rational design of antimicrobial pep- tides with enhanced specificity.

Shoot rot of Zizania latifolia and the first record of its pathogen Pantoea ananatis in China

The aquatic grass Zizania latifolia grows symbiotically with the fungus Ustilago esculenta producing swollen structures called Jiaobai,widely cultivated in China.A new disease of Z.latifolia was found in Zhejiang Province,China.Initial lesions appeared on the leaf sheaths or sometimes on the leaves near the leaf sheaths.The lesions extended along the axis of the leaf shoots and formed long brown to dark brown streaks from the leaf sheath to the leaf,causing sheath rot and death of entire leaves on young plants.The pathogen was isolated and identified as the bacterium Pantoea ananatis,based on 16 S ribosomal RNA(r RNA)gene sequencing,multilocus sequence analysis(atp D(β-subunit of ATP synthase F1),gyr B(DNA gyrase subunit B),inf B(translation initiation factor 2),and rpo B(β-subunit of RNA polymerase)genes),and pathogenicity tests.Ultrastructural observations using scanning electron microscopy revealed that the bacterial cells colonized the vascular tissues in leaf sheaths,forming biofilms on the inner surface of vessel walls,and extended between vessel elements via the perforated plates.To achieve efficient detection and diagnosis of P.ananatis,species-specific primer pairs were designed and validated by testing closely related and unrelated species and diseased tissues of Z.latifolia.This is the first report of bacterial sheath rot disease of Z.latifolia caused by P.ananatis in China.

The ongoing global pandemic of coronavirus disease 2019(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS‐CoV‐2),has caused devastating impacts on the public health and the global economy.Rapid viral antigenic evolution has led to the continual generation of new variants.Of special note is the recently expanding Omicron subvariants that are capable of immune evasion from most of the existing neutralizing antibodies(nAbs).This has posed new challenges for the prevention and treatment of COVID-19.Therefore,exploring broad-spectrum antiviral agents to combat the emerging variants is imperative.In sharp contrast to the massive accumulation of mutations within the SARS-CoV-2 receptor-binding domain(RBD),the S2 fusion subunit has remained highly conserved among variants.hence,S2-based therapeutics may provide effective cross-protection against new SARS-CoV-2 variants.here,we summarize the most recently developed broad-spectrum fusion inhibitors(e.g.,nAbs,peptides,proteins,and small-molecule compounds)and candidate vaccines targeting the conserved elements in SARS-CoV-2 S2 subunit.The main focus includes all the targetable S2 elements,namely,the fusion peptide,stem helix,and heptad repeats 1 and 2(HR1-HR2)bundle.Moreover,we provide a detailed summary of the characteristics and action-mechanisms for each class of cross-reactive fusion inhibitors,which should guide and promote future design of S2-based inhibitors and vaccines against new coronaviruses.

我国《民法典》规定的继续性合同的解除制度呈现出多元化发展趋势。为了避免不定期继续性合同的永久约束、保障当事人的自主决定权和人格自由发展,不定期继续性合同当事人有预告解除权,即不需要解除原因,只要在合理期限之前通知对方,就可以解除合同。继续性合同强调当事人之间的信赖,在信赖关系被打破或其他原因致维持合同对当事人不可承受时,无论是定期继续性合同的当事人,还是不定期继续性合同的当事人,都可以基于重大原因立即解除合同。《民法典》在个别有名继续性合同中承认当事人的任意解除权,即当事人解除合同既不需要解除原因,也不需要提前通知。任意解除权是完全的解除自由,需要充分的正当性理由。在传统的继续性合同解除制度之外,《民法典》在物业服务合同中规定了定期物业服务合同的业主预告解除权,这是继续性合同解除制度的新发展。然而,定期继续性合同预告解除制度的合理性基础并不充分,而且对契约严守制度的冲击太大,要限制其扩张。

Nano-and microphytoplankton community characteristics in brown tide bloom-prone waters of the Qinhuangdao coast,Bohai Sea, China

Since 2009, the newly recorded species Aureococcus anophagefferens has successively induced brown tides in the Qinhuangdao coastal waters of the Bohai Sea. here, we focus on the phytoplankton community structure in this region and try to point out the biological background characteristics of the outbreaks of successive brown tides. We analyzed species composition,cell abundance, diversity indices and size-fractionated chlorophyll a(Chl-a) concentration monthly from March 2013 to January2014. The phytoplankton community, as observed by microscopy, underwent succession from nano-celled chained diatoms to dinoflagellates in the spring and summer, and then to micro-celled diatoms and nano-celled chained diatoms as the most prominent groups in the subsequent autumn and winter. Canonical correspondence analysis revealed that changes in the abundance of diatoms were related to nutrient availability, especially of nitrogen, while water temperature was the crucial factor influencing the abundance of dinoflagellates and A. anophagefferens. Considering the contribution of nano-celled chained species to the micro-sized Chl-a,small phytoplankton(he majority of total Chl-a and were generally abundant during our study. Abundant and diverse small phytoplankton seemed to serve as a background or seedbank for the formation of blooms of A. anophagefferens and other small-celled algae.
