MicroRNAs(MiRNAs)carried by exosomes play pivotal roles in the crosstalk between cell components in the tumor microenvironment.Our study aimed at identifying the expression profile of exosomal miRNAs(exo-miRNAs)in the serum of multiple myeloma(MM)patients and investigating the regulation networks and their potential functions by integrated bioinformatics analysis.Exosomes in serum from 19 newly diagnosed MM patients and 9 healthy donors were isolated and the miRNA profile was investigated by small RNA sequencing.Differential expression of exo-miRNAs was calculated and target genes of miRNAs were predicted.CytoHubba was applied to identify the hub miRNAs and core target genes.The LASSO Cox regression model was used to develop the prognostic model,and the ESTIMATE immune score was calculated to investigate the correlation between the model and immune status in MM patients.The top six hub differentially expressed serum exo-miRNAs were identified.513 target genes of the six hub exo-miRNAs were confirmed to be differentially expressed in MM cells in the Zhan Myeloma microarray dataset.Functional enrichment analysis indicated that these target genes were mainly involved in mRNA splicing,cellular response to stress,and deubiquitination.13 core exo-miRNA target genes were applied to create a novel prognostic signature to provide risk stratification for MM patients,which is associated with the immune microenvironment of MM patients.Our study comprehensively investigated the exo-miRNA profiles in MM patients.A novel prognostic signature was constructed to facilitate the risk stratification of MM patients with distinct outcomes.

收集1个假性醛固酮减少症Ⅱ型(pseudohypoaldosteronism type II, PHA2)家系患者临床资料、化验检查及基因突变结果。先证者, 1岁7个月, 发现血钾6.69 mmol/L(参考范围3.5～5.3 mmol/L, 下同)、血压110/68 mmHg(90 mL·min-1·(1.73 m2)-1]、血肾素浓度0.30 μIU/mL(4.2～45.6 μIU/mL);其母亲、外祖父也有肾功能正常情况下高血钾、高血压、高血氯、代谢性酸中毒、低肾素。3例患者基因结果显示, 无赖氨酸激酶4(WNK4)基因第7号外显子存在1个杂合错义变异(c.1685A>G, p.E562G);氢氯噻嗪治疗均有效。通过文献复习, 将本文E562G家系与其他WNK4变异型进行了比较, 提示WNK4突变型的临床表型异质性。对于不明原因的高钾血症, 尤其合并高血压, 要警惕PHA2, 尽早进行基因筛查, 避免PHA2漏诊误诊。

目的从"肠-脑轴"角度探讨氟桂利嗪所致药源性帕金森综合征(DIP)的发病机制。方法30只雄性C57BL/6小鼠按随机数字表法分为空白对照组与氟桂利嗪组,每组15只。空白对照组小鼠给予0.1 mL 50%聚乙二醇400+50%生理盐水混合液灌胃,1次/d、持续2周;氟桂利嗪组小鼠给予浓度6 mg/mL氟桂利嗪+50%聚乙二醇400+50%生理盐水混合液灌胃,每日剂量30 mg/kg,1次/d、持续2周。2组小鼠均于开始给药后第1、3、5、10、14天时记录体质量,于第14天时进行转棒实验以评估运动功能,并留取小鼠粪便行16s RNA测序以观察肠道菌群情况,给予伊文思蓝溶液灌胃检测肠道转运功能,随后处死小鼠,取其脑组织、肠组织并制备成匀浆或冰冻切片,应用Western blotting实验检测黑质中多巴胺能神经元表达情况,应用RT-qPCR法检测黑质中炎性因子表达情况,应用免疫荧光染色检测结肠上皮组织中紧密连接蛋白ZO-1、Claudin-5表达情况。结果与空白对照组比较,开始给药后第1、3、5、7、10、14天时氟桂利嗪组小鼠的体质量比(与给药前体质量的比值)均明显更低。与空白对照组比较,氟桂利嗪组小鼠的转棒停留时间明显缩短,跌落时转棒转速明显降低,差异均有统计学意义(P0.05)。与空白对照组比较,氟桂利嗪组小鼠黑质中促炎因子白细胞介素-6、肿瘤坏死因子-α水平明显升高(1.00±0.00 vs.2.79±0.83;1.00±0.00 vs.3.39±1.37),肠道依文思蓝推进率明显降低(80.67%±4.51%vs.50.67%±6.03%),结肠上皮组织中ZO-1、Claudin-5表达明显降低(27.01±1.41 vs.16.32±2.83;37.00±2.80 vs.24.52±2.12),差异均有统计学意义(P<0.05)。空白对照组与氟桂利嗪组小鼠共同存在576种肠道菌群,空白对照组单独存在744种,氟桂利嗪组单独存在634种。基于加权UniFrac距离的主成分分析测量β多样性,2组小鼠的肠道菌群明显分离(PCoA1为39.88%,PCoA2为30.69%)。与空白对照组比较,氟桂利嗪组小鼠肠道菌群结构在门水平上以厚壁菌门、疣微菌门为优势菌群,在科水平上以拟杆菌科、毛螺球菌科、阿克曼氏菌科最常见。与空白对照组比较,氟桂利嗪组小鼠肠道菌群中阿克曼菌属、乳酸杆菌属的相对丰度明显降低,差异均有统计学意义(P<0.05)。结论氟桂利嗪可能通过引起肠道菌群结构紊乱,并基于"肠-脑轴"诱发神经炎症,从而导致DIP发病。

A fundamental challenge for cells is how to coordinate various biochemical reactions in space and time. To achieve spatiotemporal control, cells have developed organelles that are surrounded by lipid bilayer membranes. Further, membraneless compartmentalization, a process induced by dynamic physical association of biomolecules through phase transition offers another efficient mechanism for intracellular organization. While our understanding of phase separation was predominantly dependent on yeast and animal models, recent findings have provided compelling evidence for emerging roles of phase separation in plants. In this review, we first provide an overview of the current knowledge of phase separation, including its definition, biophysical principles, molecular features and regulatory mechanisms. Then we summarize plant-specific phase separation phenomena and describe their functions in plant biological processes in great detail. Moreover, we propose that phase separation is an evolutionarily conserved and efficient mechanism for cellular compartmentalization which allows for distinct metabolic processes and signaling pathways, and is especially beneficial for the sessile lifestyle of plants to quickly and efficiently respond to the changing environment.

The origin of the shallow decay segment in γ-ray bursts' (GRB) early light curves remains a mystery, especially those cases with a long-lived plateau followed by an abrupt falloff. In this paper, we propose to understand the origins of the long-lived plateau by considering the solidification of newborn quark stars with latent heat released as energy injection to the GRB afterglow, and we suggest that an abrupt falloff would naturally appear after the plateau due to the energy injection cutoff. We estimated the total latent heat released during the phase transition of quark stars from liquid to solid states to be on the order of ～ 1051 ergs, which is comparable to the emission energy in the shallow decay segment. We also estimated the time scale of radiating the latent heat through thermal photon emission, and found that the time scale agrees with the observations. Based on our estimation, we analyzed the process of energy injection to GRB afterglow. We will show that the steady latent heat of quark star phase transition will continuously inject into the GRB afterglow in a form similar to that of a Poynting-flux-dominated outflow and naturally produce the shallow decay phase and the abrupt falloff after the plateau. We conclude that the latent heat of quark star phase transition is an important contribution to the shallow decay radiation in some GRB afterglows, and explains the long-lived plateau followed by an abrupt falloff, if pulsar-like stars are really (solid) quark stars.

Spatial changes and driving factors of lake water quality in Inner Mongolia, China

Lakes play important roles in sustaining the ecosystem and economic development in Inner Mongolia Autonomous Region of China,but the spatial patterns and driving mechanisms of water quality in lakes so far remain unclear.This study aimed to identify the spatial changes in water quality and the driving factors of seven lakes(Juyanhai Lake,Ulansuhai Lake,Hongjiannao Lake,Daihai Lake,Chagannaoer Lake,Hulun Lake,and Wulannuoer Lake)across the longitudinal axis(from the west to the east)of Inner Mongolia.Large-scale research was conducted using the comprehensive trophic level index(TLI(Σ)),multivariate statistics,and spatial analysis methods.The results showed that most lakes in Inner Mongolia were weakly alkaline.Total dissolved solids and salinity of lake water showed obvious zonation characteristics.Nitrogen and phosphorus were identified as the main pollutants in lakes,with high average concentrations of total nitrogen and total phosphorus being of 4.05 and 0.21 mg/L,respectively.The values of TLI(Σ)ranged from 49.14 to 71.77,indicating varying degrees of lake eutrophication,and phosphorus was the main driver of lake eutrophication.The lakes of Inner Mongolia could be categorized into lakes to the west of Daihai Lake and lakes to the east of Daihai Lake in terms of salinity and TLI(Σ).The salinity levels of lakes to the west of Daihai Lake exceeded those of lakes to the east of Daihai Lake,whereas the opposite trend was observed for lake trophic level.The intensity and mode of anthropogenic activities were the driving factors of the spatial patterns of lake water quality.It is recommended to control the impact of anthropogenic activities on the water quality of lakes in Inner Mongolia to improve lake ecological environment.These findings provide a more thorough understanding of the driving mechanism of the spatial patterns of water quality in lakes of Inner Mongolia,which can be used to develop strategies for lake ecosystem protection and water resources management in this region.

Short Text Mining Framework with Specific Design for Operation and Maintenance of Power Equipment

In order to recover the value of short texts in the operation and maintenance of power equipment,a short text mining framework with specific design is proposed.First,the process of the short text mining framework is summarized,in which the functions of all the processing modules are introduced.Then,according to the characteristics of short texts in the operation and maintenance of power equipment,the specific design for each module is proposed,which adapts the short text mining framework to a practical application.Finally,based on the framework with the specific designed modules,two examples in terms of defect texts are given to illustrate the application of short text mining in the operation and maintenance of power equipment.The results of the examples show that the short text mining framework is suitable for operation and maintenance tasks for power equipment,and the specific design for each module is beneficial for the improvement of the application effect.

Dynamic Modeling of a Roller Chain Drive System Considering the Flexibility of Input Shaft

Roller chain drives are widely used in various high-speed, high-load and power transmission applications, but their complex dynamic behavior is not well researched. Most studies were only focused on the analysis of the vibration of chain tight span, and in these models, many factors are neglected. In this paper, a mathematical model is developed to calculate the dynamic response of a roller chain drive working at constant or variable speed condition. In the model, the complete chain transmission with two sprockets and the necessary tight and slack spans is used. The effect of the flexibility of input shaft on dynamic response of the chain system is taken into account, as well as the elastic deformation in the chain, the inertial forces, the gravity and the torque on driven shaft. The nonlinear equations of movement are derived from using Lagrange equations and solved numerically. Given the center distance and the two initial position angles of teeth on driving and driven sprockets corresponding to the first seating roller on each side of the tight span, dynamics of any roller chain drive with two sprockets and two spans can be analyzed by the procedure. Finally, a numerical example is given and the validity of the procedure developed is demonstrated by analyzing the dynamic behavior of a typical roller chain drive. The model can well simulate the transverse and longitudinal vibration of the chain spans and the torsional vibration of the sprockets. This study can provide an effective method for the analysis of the dynamic characteristics of all the chain drive systems.
