Top priority current path between SiC particles during ultra-high temperature flash sintering: Presence of PyC “bridges”

Flash sintering(FS)is a novel technique for rapidly densifying silicon carbide(SiC)ceramics.This work achieved a rapid sintering of SiC ceramics by the utilization of ultra-high temperature flash sintering within 60 s.Pyrolysis carbon(PyC)“bridges”were constructed between SiC particles through the carbonisation of phenolic resin,providing a large number of current channels.The incubation time of the flash sintering process was significantly reduced,and the sintering difference between the centre and the edge regions of the ceramics was minimized,with an average particle size of the centre region and edge region being 12.31 and 9.02μm,respectively.The results showed that the porosity of the SiC ceramics after the flash sintering was reduced to 14.79% with PyC“bridges”introduced,and the Vickers hardness reached 19.62 GPa.PyC“bridges”gradually evolved from amorphous eddy current carbon to oriented graphite carbon,indicating that the ultra-high temperature environment in which the sample was located during the flash sintering was successfully constructed.Ultra-high temperature flash sintering of SiC is expected to be applied to the local repair of matrix damage in SiC ceramic matrix composites.

Ceramic fibrous aerogels are highly desirable for thermal management materials due to their high porosity,excellent elasticity,thermal conductivity,and good thermal resistance.However,the fabrication of nanofibrous aerogel with super-elasticity and good shape retention at the same time has remained challenging.To meet the requirements,a novel anisotropy nanofibrous-granular aerogel with a quasi-layered multi-arch-like and hierarchical-cellular structure is designed and prepared by vacuum-filtration-assisted freeze-drying and sintering.The quasi-layered multi-arch and flexible nanofibers endowed the aerogels with excellent mechanical robustness(ultimate stress up to 60 kPa with strain 60%)and super-elasticity with recoverable compression strain up to 60%.The introduced SiO_(2) aerogel nanoparticles and nanofibers are assembled into an arch-like structure and become the connection point of adjacent nanofibers,which endows low thermal conductivity(0.024 mW/(m·K))of composite aerogel.This novel strategy provides a fresh perspective for the preparation of nanofibrous aerogel with robust mechanical in thermal insulation and other fields.

ADP-ribosylation factor 6(Arf6),a small G-protein of the Ras superfamily,plays pivotal roles in multiple cellular events,including exocytosis,endocytosis,actin remodeling,plasma membrane reorganization and vesicular transport.Arf6 regulates the progression of cancer through the activation of cell motility and invasion.Aberrant Arf6 activation is a potential therapeutic target.This review aims to understand the comprehensive function of Arf6 for future cancer therapy.The Arf6 GEFs,protein structure,and roles in cancer have been summarized.Comprehending the mechanism underlying Arf6-mediated cancer cell growth and survival is essential.The structural features of Arf6 and its efforts are discussed and may be contributed to the discovery of future novel protein-protein interaction inhibitors.In addition,Arf6 inhibitors and mechanism of action are listed in the table.This review further emphasizes the crucial roles in drug resistance and attempts to offer an outlook of Arf6 in cancer therapy.

MoO_(x) nanoclusters on Mo-doped TiO_(2) nanosheets with enhanced singlet oxygen generation and sulfide conversion abilities for photocatalytic aerobic oxidative desulfurization

Photocatalytic aerobic oxidation desulfurization(PAODS)is a promising and sustainable alternative to conventional,energyintensive desulfurization techniques for petroleum products.However,its development is greatly plagued by the low capability in generating highly reactive oxygen species and sluggish kinetics of sulfide oxidation of reported photocatalysts.Here we report a class of MoO_(x)nanocluster decorated on ultrathin Mo-doped TiO_(2)nanosheet(MoO_(x)/MoTiO)catalyst for efficiently facilitating the photocatalytic aerobic oxidation of sulfides.We demonstrate that MoO_(x)/MoTiO can not only promote the generation of highly reactive singlet oxygen(^(1)O_(2))but also enhance the aerobic conversion of sulfides,which leads to a record dibenzothiophene oxidation activity of 3.90 mmol g^(-1)h^(-1).The multiple experimental characterizations and density functional theory calculations collectively reveal that the doped-Mo sites can interact with the photogenerated excitons,enabling directly energy transfer generation of^(1)O_(2)through a new exciton modulation mechanism,and the coordination unsaturated MoO_(x)clusters play the role of co-catalyst to enhance the separation of charge carriers,and effectively catalyze the reaction between sulfides and1O_(2)to form sulfones.

The fall armyworm,Spodoptera frugiperda(S.frugiperda),represents the most resistant insect species and poses serious threat to grain yield.Chlorantraniliprole(CHL),which targets the ryanodine receptors(RyRs)in insects,has demonstrated the efficacy in controlling S.frugiperda.Nevertheless,this has led to emerging resistance in several countries.To counter this resistance,a viable approach involves the development of novel compounds that bind against RyRs via distinct binding sites or modes.In this study,a series of 22 novel anthranilic diamide derivatives was designed and synthesized,and their insecticidal activities were evaluated.Most of these derivatives showed moderate to good insecticidal activity against S.frugiperda and Mythimna separata.Time-lapse fluorescence measurements of endoplasmic reticulum luminal calcium revealed that most derivatives elicited cellular responses similar as CHL when assessed on HEK293 cells expressing S.frugiperda ryanodine receptors(SfRyRs).The mode of action of compound 13a was studied and verified on the isolated neurons by calcium imaging technique.Finally,molecular docking analysis was employed to predict the binding mechanism of compound 13a against SfRyRs.Overall,these novel diamide derivatives hold promise as a valuable resource for guiding the future design of insecticidal compounds targeting RyRs.

Reprogramming of energy metabolism is one of the basic characteristics of cancer and has been proved to be an important cancer treatment strategy.Isocitrate dehydrogenases(IDHs)are a class of key proteins in energy metabolism,including IDH1,IDH2,and IDH3,which are involved in the oxidative decarboxylation of isocitrate to yield a-ketoglutarate(a-KG).Mutants of IDH1 or IDH2 can produce D-2-hydroxyglutarate(D-2HG)with a-KG as the substrate,and then mediate the occurrence and development of cancer.At present,no IDH3 mutation has been reported.The results of pan-cancer research showed that IDH1 has a higher mutation frequency and involves more cancer types than IDH2,implying IDH1 as a promising anti-cancer target.Therefore,in this review,we summarized the regulatory mechanisms of IDH1 on cancer from four aspects:metabolic reprogramming,epigenetics,immune microenvironment,and phenotypic changes,which will provide guidance for the understanding of IDH1 and exploring leading-edge targeted treatment strategies.In addition,we also reviewed available IDH1 inhibitors so far.The detailed clinical trial results and diverse structures of preclinical candidates illustrated here will provide a deep insight into the research for the treatment of IDH1-related cancers.

Resilience mediates the association between alexithymia and stress in Chinese medical students during the COVID-19 pandemic

Background Evidence indicates that medical students have had high rates of mental health problems,especially during the COVID-19 pandemic,which could be affected by alexithymia—a marked dysfunction in emotional awareness,social attachment and interpersonal relationships—and stress.However,psychological resilience might relieve alexithymia and stress levels.Aims This study aimed to investigate the role of resilience in alexithymia and stress in medical students.Methods A total of 470 medical students completed online and offline surveys,including the Toronto Alexithymia Scale-20(TAS-20),the Connor-Davidson Resilience Scale(CD-RISC)and the College Student Stress Questionnaire(CSSQ).The data of five participants were excluded because of a lack of integrity.Mann-Whitney U test or Kruskal-Wallis test was used to compare group differences in the CD-RISC scores among categorical variables.Spearman correlation analysis was employed to evaluate the associations between resilience and alexithymia and between resilience and stress.Mediation analysis was used to test the mediating effect of resilience between alexithymia and stress.Results Of the medical students considered in the analysis,382(81.28%)were female and 88(18.72%)were male.There was a significant negative correlation between the TAS-20 scores and the total and subtotal CD-RISC scores(p<0.001).The CSSQ scores also significantly negatively correlated with the total and subtotal CD-RISC scores(p<0.001).Resilience mediated the relationship between alexithymia and stress(total effect=1.0447,p<0.001).The indirect effect of alexithymia significantly impacted stress through resilience(effect=0.1670,95%CI:0.069 to 0.281).Conclusions Our findings suggest that resilience might effectively reduce alexithymia and stress.They also contributed to a better understanding of the mediating effects of resilience on alexithymia and stress during the COVID-19 pandemic.The evidence from these results encourages universities to focus on improving students’resilience.

Ibrutinib is a first-line treatment drug for B-cell malignancies.However,resistance to ibrutinib has been reported due to BTKC481Smutation.Although PROTAC strategy is expected to overcome this clinical resistance,it has limitations such as large molecular weight and moderate bioactivity,which restrict its potential clinical application.Herein,we report a new type of potent BTKC481S-targeting PROTAC degrader.Through design,computer-assisted optimization and SAR studies,we have developed a representative BTKC481Sdegrader L6 with a much smaller molecular weight and improved solubility.Notably,L6 demonstrates better BTK degrading activity and lower IC50value in ibrutinib-resistant cell line than the first-generation BTK degrader P13I.Optimization strategy of L6 provides a general approach in the development of PROTACs targeting BTK and other proteins for future study.

To the Editor:Proteolysis Targeting Chimera(PROTAC)is an emerging approach to selectively degrading target proteins by utilizing endogenous proteasome.Since PROTACs can degrade target proteins without high affinity,it is natural to speculate that this technology can be used to identify the targets of natural products。Although a recent study reported the employment of PROTACs to explore the unknown non-kinase target of a multi-kinase inhibitor sorafenib,whether PROTACs can be used to find the potential targets of natural products remains unexplored.
