Gene Expression Patterns in Different Wool Densities of Rex Rabbit Using cDNA Microarray

This study was carried out to identify gene expression patterns in different wool densities of rex rabbit. The mid-dorsal skin samples from 8 rex rabbits were studied. They were divided into two groups according to different wool densities. Total RNA was isolated and labeled by reverse transcription reaction with Cy5-dCTP and Cy3-dCTP for cDNA probe. The cDNA probe was hybridized with cDNA microarrays containing 14 601 rabbit’s genes. The differently expressed genes were analysed with the Gene Ontology （GO） classification and the pathway analysis. Hierarchical clustering was performed to clarify genes in association with different wool densities. The 2 657 differentially expressed genes were identified. Among them, 1 103 genes were functionally known genes, 687 genes were up-regulated and 419 were down-regulated. GO analysis indicated that these altered genes were associated with metabolism, signal transduction, cell cycle, cell adhesion, cell proliferation, cell division, apoptosis, and other processes. KEGG analysis showed that 95 signal pathways associated with up-regulated genes and 87 signal pathways associated with down-regulated genes had changed significantly （P0.05）. Some important differentially expressed genes in different wool densities of rex rabbit were identified, such as MMP2, TGF-β1, TGF-β2, IGF-1, ITGB1, RPS6KB1, BMP2, ActRIIB, CDK2, and CCNA2. Hierarchical clustering analysis separated the differentially expressed genes into 5 main characteristic groups. Data from cDNA microarray experiments clearly distinguished between group A and group B. Some important genes were identified, which might be useful in further study on wool density markers of rex rabbit.

Effect of Humulus scandens for Traditional Fiber Sources on Digestion,Diarrhea,and Performance of Growing Rabbits

This work was to determine which concentration of a mixture of fresh Humulus scandens （HS） and fresh alfalfa could be included in diets for rabbits. The fresh forage of three experimental diets is composed of 100% alfalfa, 50% alfalfa ＋ 50% HS, 100% HS, respectively. 30 rabbits [（1.42 ± 0.056） kg] were used to determine nutrient digestibility. A feeding trial was carried out using 90 weaning rabbits [35 d of age; （512±6.5） g]. Increasing content of HS in the diet did not affect digestibility of DM （dry matter）, CF （crude fiber）, CP （crude protein）, GE （gross energy）, and EE （ether extract）. But diarrhea decreased markedly （P〈0.01） compared with the diets that had no HS. ADF1 （average daily feed intake） increased by 4.3% （P 〈 0.05） and 3.6% （P 〈 0.05） when rabbits fed the diet with 100% HS of fresh forage compared with the diets containing 0 and 50% HS, respectively. Feed efficiency improved significantly （P 〈 0.05） by 2.07 and 5.30% when rabbits fed 0 and 50% HS of fresh forage, respectively, compared with 100% HS. There was no effect on ADG （average daily weight gain）. In conclusion, the mixture （50：50） of HS and alfalfa constitutes an alternative source of fiber for fattening rabbits and prevents diarrhea effectively.

Genetic Diversity of Ustilaginoidea virens in Heilongjiang Province

In recent years, the occurrence of rice false smut has become increasingly serious and the area of damage has expanded year by year, so the false smut has become one of the diseases which have seriously affected rice yield and quality in Heilongjiang Province. However, the genetic diversity research on false smut in Heilongjiang Province was relatively weak. In the current investigation, seven polymorphic SSR markers were taken to analyze the genetic diversity among 89 strains of the pathogens of rice false smut collected from five main rice growing areas in Heilongjiang Province, China. The results showed that the amplified bands of each pair of primers were between 2 and 7. A total of 43 bands were obtained, and each primer was expanded for 4.8 bands. The genetic similarity coefficient(GSC) between strains by using SSR molecular marker analysis showed that the GSC of the strain of Ustilaginoidea virens was 0.613-0.955, with an average of 0.741. According to the results cluster analysis, when the genetic distance was 0.73, 89 strains were divided into three groups, and five strains of rice smut strains were distributed in three populations, and strains in Harbin City were mainly distributed in group Ⅲ. In Mudanjiang City, the strains were mainly distributed in groups Ⅰ and Ⅲ, and the strains in Suihua City were mainly distributed in groups Ⅰ and Ⅱ, and the strains in Jiamusi City were evenly distributed in groups Ⅰ, Ⅱ and Ⅲ, and the strains in Qiqihar City were mainly distributed in groups Ⅱ and Ⅲ. The results showed that the genetic background of Ustilaginoidea virens in Heilongjiang Province was relatively low in complexity, and the overall inheritance of the strain was relatively stable.

试验旨在研究不同纤维源饲粮对家兔生长性能、屠宰性能、免疫器官发育及抗氧化能力的影响。试验选取96只体重相近、健康的35日龄伊拉肉兔,随机分为4组,每组8个重复,每个重复3只。4个试验组分别使用大豆皮、稻草、花生秧和苜蓿为唯一纤维源配制全价饲粮。预试期7 d,正式试验期30 d。结果显示,大豆皮组、花生秧组和苜蓿组的末重和平均日增重均显著高于稻草组(P<0.05),稻草组、花生秧组和苜蓿组的平均日采食量显著高于大豆皮组(P<0.05)。大豆皮组、花生秧组和苜蓿组的宰前活重显著高于稻草组(P<0.05),花生秧组和苜蓿组的胴体重、半净膛重和全净膛重均显著高于稻草组(P<0.05)。苜蓿组的肝脏指数显著高于其他组(P<0.05),圆小囊指数显著高于大豆皮组和花生秧组(P<0.05)。大豆皮组的血清谷胱甘肽过氧化物酶(GSH-Px)活性显著高于其他组(P<0.05)。研究表明,苜蓿作为家兔饲粮的纤维源综合表现较好,花生秧仅次于苜蓿,优于大豆皮和稻草。

Asperuloside Promotes Apoptosis of Cervical Cancer Cells through Endoplasmic Reticulum Stress-Mitochondrial Pathway

Objective: To investigate the effects of asperuloside on cervical cancer based on endoplasmic reticulum(ER) stress and mitochondrial pathway. Methods: Different doses(12.5–800 μg/mL) of asperuloside were used to treat cervical cancer cell lines Hela and CaSki to calculate the half maximal inhibitory concentration(IC_(50)) of asperuloside. The cell proliferation was analyzed by clone formation assay. Cell apoptosis, intracellular reactive oxygen species(ROS) and mitochondrial membrane potential were determined by flow cytometry. The protein expressions of cleaved-caspase-3, Bcl-2, Bax, Cyt-c, cleaved-caspase-4 and glucose-regulated protein 78(GRP78) were analyzed by Western blot. And the inhibitor of ER stress, 4-phenyl butyric acid(4-PBA) was used to treat cervical cancer cells to further verify the role of ER stress in the apoptosis of cervical cancer cells induced by asperuloside. Results: Asperuloside of 325, 650, and 1300 μg/mL significantly inhibited the proliferation and promoted apoptosis of Hela and CaSki cells(P<0.01). All doses of asperuloside significantly increased intracellular ROS levels, reduced mitochondrial membrane potential, significantly reduced Bcl-2 protein expression level, and increased Bax, Cyt-c, GRP78 and cleaved-caspase-4 expressions(P<0.01). In addition, 10 mmol/L 4-PBA treatment significantly promoted cell proliferation and reduced apoptosis(P<0.05), and 650 μg/mL asperuloside could reverse 4-PBA-induced increased cell proliferation, decreased apoptosis and cleaved-caspase-3,-4 and GRP78 protein expressions(P<0.05). Conclusion: Our study revealed the role of asperuloside in cervical cancer, suggesting that asperuloside promotes apoptosis of cervical cancer cells through ER stress-mitochondrial pathway.

Isorhamnetin Downregulates MMP2 and MMP9 to Inhibit Development of Rheumatoid Arthritis through SRC/ERK/CREB Pathway

Objective To investigate the effect of isorhamnetin on the pathology of rheumatoid arthritis(RA).Methods Tumor necrosis factor(TNF)-α-induced fibroblast-like synoviocytes(FLS)was exposed to additional isorhamnetin(10,20 and 40µmol/L).Overexpression vectors for matrix metalloproteinase-2(MMP2)or MMP9 or SRC were transfected to explore their roles in isorhamnetin-mediated RA-FLS function.RA-FLS viability,migration,and invasion were evaluated.Moreover,a collagen-induced arthritis(CIA)rat model was established.Rats were randomly divided to sham,CIA,low-,medium-,and high-dosage groups using a random number table(n=5 in each group)and administed with normal saline or additional isorhamnetin[2,10,and 20 mg/(kg·day)]for 4 weeks,respectively.Arthritis index was calculated and synovial tissue inflammation was determined in CIA rats.The levels of MMP2,MMP9,TNF-α,interleukin-6(IL-6),and IL-1β,as well as the phosphorylation levels of SRC,extracellular regulated kinase(ERK),and cyclic adenosine monophosphate response element-binding(CREB),were detected in RA-FLS and synovial tissue.Molecular docking was also used to analyze the binding of isorhamnetin to SRC.Results In in vitro studies,isorhamnetin inhibited RA-FLS viability,migration and invasion(P<0.05).Isorhamnetin downregulated the levels of MMP2,MMP9,TNF-α,IL-6,and IL-1βin RA-FLS(P<0.05).The overexpression of either MMP2 or MMP9 reversed isorhamnetin-inhibited RA-FLS migration and invasion,as well as the levels of TNF-α,IL-6,and IL-1β(P<0.05).Furthermore,isorhamnetin bound to SRC and reduced the phosphorylation of SRC,ERK,and CREB(P<0.05).SRC overexpression reversed the inhibitory effect of isorhamnetin on RA-FLS viability,migration and invasion,as well as the negative regulation of MMP2 and MMP9(P<0.05).In in vivo studies,isorhamnetin decreased arthritis index scores(P<0.05)and alleviated synovial inflammation.Isorhamnetin reduced the levels of MMP2,MMP9,TNF-α,IL-6,and IL-1β,as well as the phosphorylation of SRC,ERK,and CREB in synovial tissue(P<0.05).Notably,the inhibit

The outbreak of COVID-19 has drawn great attention around the world.SARS-CoV-2 is a highly infectious virus with occult transmission by many mutations and a long incubation period.In particular,the emergence of asymptomatic infections has made the epidemic even more severe.Therefore,early diagnosis and timely management of suspected cases are essential measures to control the spread of the virus.Developing simple,portable,and accurate diagnostic techniques for SARS-CoV-2 is the key to epidemic prevention.The advantages of point-of-care testing technology make it play an increasingly important role in viral detection and screening.This review summarizes the point-of-care testing platforms developed by nucleic acid detection,immunological detection,and nanomaterial-based biosensors detection.Furthermore,this paper provides a prospect for designing future highly accurate,cheap,and convenient SARS-CoV-2 diagnostic technology.

Two Nonmonotone Proximal Gradient Methods for Nonsmooth Optimization over the Stiefel Manifold

We propose two nonmonotone retraction-based proximal gradient methods for solving a class of nonconvex nonsmooth optimization problems over the Stiefel manifold. The proposed methods are equipped with the descent direction obtained by a proximal mapping restricted in tangent space of the manifold and the BarzilaiBorwein stepsizes determined by two recent iteration points and the corresponding descent directions. By employing, respectively, the Grippo-Lampariello-Lucidi nonmonotone line search strategy and the Dai-Fletcher nonmonotone line search strategy, our proposed methods are proved to be globally convergent. Analysis on the iteration complexity of NPGL for obtaining an?-stationary solution is provided. Numerical results on the sparse principle component analysis problems demonstrate the efficiency of our methods.

On a Flexible Loan Repayment Method Depending on Borrower's Asset with an Early Termination Clause

Aiming at the problem that the asset’s fluctuation influences the borrower’s repayment ability,a loan with a new and flexible repayment method is designed,which depends on the asset value of the borrower.The repayment method can reduce the loan default probability,but it causes the uncertainty of the pay off time.Because the repayment term is related to the regular repayment amount in this method,a boundary for the regular repayment amount is set up in order to avoid too long repayment term.This will balance the benefit of borrowers and lenders and improve the applicability of this method.By establishing a mathematical model of the residual value of the loan,this model can be transformed into an initial-boundary problem of a partial differential equation.The analytic solution and the expected time to pay off the loan are obtained.Finally,numerical analysis are shown.

Fixed-Time Anti-Disturbance Average-Tracking for Multi-Agent Systems Without Velocity Measurements

In this paper,the authors tackle a fixed-time average-tracking problem for multi-agent systems which do not require velocity measurements and are affected by external mismatched and matched disturbances.The objective requires the considered agents with second-order dynamics to follow the average value of several time-varying reference signals,each of which is acquired by only one agent.First of all,the average value is estimated by using a novel distributed average estimator in a fixed time.Second,an original observer is created to provide estimated values in a fixed time for external mismatched and matched disturbances as well as unmeasured velocities of agents.Then,the authors propose a novel fixed-time anti-disturbance average-tracking protocol with the aid of the observer and a sliding mode surface.In the end,the feasibility of theoretical analysis is illustrated by some simulation experiments.
