Background:Circular RNAs(circRNAs)have been recognized as significant regulators of pulmonary hypertension(PH);however,the differential expression and function of circRNAs in different vascular cells under hypoxia remain ***,we identified co-differentially expressed circRNAs and determined their putative roles in the proliferation of pulmonary artery smooth muscle cells(PASMCs),pulmonary microvascular endothelial cells(PMECs),and pericytes(PCs)under ***:Whole transcriptome sequencing was performed to analyze the differential expression of circRNAs in three different vascular cell *** analysis was used to predict their putative biological *** real-time polymerase chain reaction,Cell Counting Kit-8,and EdU Cell Proliferation assays were carried out to determine the role of circular postmeiotic segregation 1(circPMS1)as well as its potential sponge mechanism in PASMCs,PMECs,and ***:PASMCs,PMECs,and PCs exhibited 16,99,and 31 differentially expressed circRNAs under hypoxia,***1 was upregulated in PASMCs,PMECs,and PCs under hypoxia and enhanced the proliferation of vascular ***1may upregulate DEP domain containing 1(DEPDC1)and RNA polymerase II subunit D expression by targeting microRNA-432-5p(miR-432-5p)in PASMCs,upregulate MAX interactor 1(MXI1)expression by targeting miR-433-3p in PMECs,and upregulate zinc finger AN1-type containing 5(ZFAND5)expression by targeting miR-3613-5p in ***:Our results suggest that circPMS1 promotes cell proliferation through the miR-432-5p/DEPDC1 or miR-432-5p/POL2D axis in PASMCs,through the miR-433-3p/MXI1 axis in PMECs,and through the miR-3613-5p/ZFAND5 axis in PCs,which provides putative targets for the early diagnosis and treatment of PH.

重构GRAPES(Global/Regional Assimilation and Prediction System)全球、区域一体化变分同化系统中的极小化控制变量,提升中、小尺度同化分析能力,为中国气象局业务区域数值预报系统CMA-MESO提供千米尺度适用的同化方案。新方案用纬向风速(u)和经向风速(v)替代原有流函数和势函数作为新的风场控制变量,采用温度和地面气压(T,ps)替代原有非平衡无量纲气压作为新的质量场控制变量,同时不再考虑准地转平衡约束,而是采用连续方程弱约束保证分析平衡。背景误差参数统计和数值试验结果表明,采用重构后的极小化控制变量,观测信息传播更加局地,分析结构更加合理,避免了原方案在中、小尺度应用时存在的虚假相关问题。连续方程弱约束的引入,限制了同化分析中辐合、辐散的不合理增长,帮助新方案在分析更加局地的同时保证分析平衡。为期1个月的连续同化循环和预报试验结果表明,新方案可以减小风场和质量场分析误差,CMAMESO系统地面降水和10 m风场的预报评分显著提升。

On August 7,2023,Mangshi City,Dehong Prefecture experienced a local heavy rainstorm,and the geological disaster caused by the heavy rainfall caused casualties and property *** on the real-time observation data of automatic stations,Doppler weather radar detection and meteorological risk warning products,the disaster situation,social impact,forecast and early warning service,causes of heavy precipitation and forecast and early warning inspection were summarized and *** results show that the heavy rainfall was prominent locally,lasted for a long time and accumulated a large amount of *** were biases in model products,and it was difficult for forecasters to make subjective corrections in complex *** analysis ideas and focus points of heavy rainfall forecast,the improvement ideas and technical schemes of forecast deviation,and the improvement ideas and suggestions of services were *** provides a reference for the forecast and early warning of severe weather in the future.

Innovative 3D Microfluidic Intestinal Organoid Model for Assessing Cadmium Bioavailability in Food: Implications for Enhanced Exposure Risk Assessment

Given the severe toxicity and widespread presence of cadmium (Cd) in staple foods such as rice， accurate dietary exposure assessments are imperative for public health. In vitro bioavailability is commonly used to adjust dietary exposure levels of risk factors; however， traditional planar Transwell models have limitations， such as cell dedifferentiation and lack of key intestinal components， necessitating a more physiologically relevant in vitro platform. This study introduces an innovative three-dimensional (3D) intestinal organoid model using a microfluidic chip to evaluate Cd bioavailability in food. Caco-2 cells were cultured on the chip to mimic small intestinal villi's 3D structure， mucus production， and absorption functions. The model's physiological relevance was thoroughly characterized， demonstrating the formation of a confluent epithelial monolayer with well-developed tight junctions (ZO-1)， high microvilli density (F-actin)， and significant mucus secretion (Alcian blue staining)， closely resembling the physiological intestinal epithelium. Fluorescent particle tracking confirmed its ability to simulate intestinal transport and diffusion. The Cd bioavailability in rice measured by the 3D intestinal organoid model (9.07% ± 0.21%) was comparable to the mouse model (12.82% ± 3.42%) but significantly lower than the Caco-2 monolayer model (26.97% ± 1.11%). This 3D intestinal organoid model provides a novel and reliable strategy for in vitro assessment of heavy metal bioavailability in food， with important implications for food safety and risk assessment.

The GECAM series of satellites utilizes LaBr_(3)(Ce),LaBr_(3)(Ce,Sr),and NaI(Tl)crystals as sensitive materials for gamma-ray detectors(GRDs).To investigate the nonlinearity in the detection of low-energy gamma rays and address the errors in the calibration of the E-C relationship,comprehensive tests and comparative studies of the three aforementioned crystals were conducted using Compton electrons,radioactive sources,and mono-energetic *** nonlinearity test results of the Compton electrons and X-rays demonstrated substantial differences,with all three crystals presenting a higher nonlinearity for X/-rays than for Compton *** the LaBr_(3)(Ce)and LaBr_(3)(Ce,Sr)crystals having higher absolute light yields,they exhibited a noticeable nonlinear decrease in the light yield,especially at energies below 400 *** NaI(Tl)crystal demonstrated an"excess"light output in the 6-200 keV range,reaching a maximum"excess"of 9.2%at 30 keV in the X-ray testing and up to 15.5%at 14 keV during Compton electron testing,indicating a significant advantage in the detection of low-energy gamma ***,we explored the underlying causes of the observed nonlinearity in these *** study not only elucidates the detector responses of GECAM,but also initiates a comprehensive investigation of the nonlinearity of domestically produced lanthanum bromide and sodium iodide crystals.

当前提高城市经济韧性需要重点关注进口产品带来的深层影响。通过匹配285个城市数据与海关数据,构建计量模型实证检验进口产品规模对城市经济韧性的影响效应、传导机制及空间溢出。研究结果表明:扩大进口产品规模对城市经济韧性有显著提升效应;异质性分析表明,进口产品规模对城市经济韧性的影响在非资源型、大规模城市维度下更为显著;机制检验表明,进口产品规模能通过促进产业结构升级、提升产业集聚和增强技术创新能力提高城市经济系统恢复和再发展能力,进而增强城市经济韧性;进一步地,通过构建空间杜宾模型发现进口产品规模对城市经济韧性有正向空间溢出效应。为此,提出建议,建设韧性城市需重视进口产品对产业结构、产业集聚和技术创新的推动作用,以开放增强城市经济韧性。

Interorganelle contacts and communications are increasingly recognized to play a vital role in cellular function and *** particular,the mitochondria–endoplasmic reticulum(ER)membrane contact site(MAM)is known to regulate ion and lipid transfer,as well as signaling and organelle ***,the regulatory mechanisms of MAM formation and their function are still ***,we identify mitochondrial Lon protease(LonP1),a highly conserved mitochondrial matrix protease,as a new MAM tethering *** removal of LonP1 substantially reduces MAM formation and causes mitochondrial ***,deletion of LonP1 in the cardiomyocytes of mouse heart impairs MAM integrity and mitochondrial fusion and activates the unfolded protein response within the ER(UPR^(ER)).Consequently,cardiac-specific LonP1 deficiency causes aberrant metabolic reprogramming and pathological heart *** findings demonstrate that LonP1 is a novel MAM-localized protein orchestrating MAM integrity,mitochondrial dynamics,and UPR^(ER),offering exciting new insights into the potential therapeutic strategy for heart failure.

Patients with refractory immune thrombocytopenia(ITP)frequently encounter substantial bleeding risks and demonstrate limited responsiveness to existing *** cord-derived mesenchymal stem cells(UC-MSCs)present a promising alternative,capitalizing on their low immunogenicity and potent immunomodulatory effects for treating diverse autoimmune *** prospective phase I trial enrolled eighteen eligible patients to explore the safety and efficacy of UC-MSCs in treating refractory *** research design included administering UC-MSCs at escalating doses of 0.5×10^(6)cells/kg,1.0×10^(6)cells/kg,and 2.0×10^(6)cells/kg weekly for four consecutive weeks across three cohorts during the dose-escalation phase,followed by a dose of 2.0×10^(6)cells/kg weekly for the dose-expansion *** events,platelet counts,and changes in peripheral blood immunity were monitored and recorded throughout the administration and follow-up ***,12(with an addition of three patients in the 2.0×10^(6)cells/kg group due to dose-limiting toxicity)and six patients were enrolled in the dose-escalation and dose-expansion phase,*** patients(13/18,72.2%)experienced one or more treatment emergent adverse *** adverse events occurred in four patients(4/18,22.2%),including gastrointestinal hemorrhage(2/4),profuse menstruation(1/4),and acute myocardial infarction(1/4).The response rates were 41.7%in the dose-escalation phase(5/12,two received 1.0×10^(6)cells/kg per week,and three received 2.0×10^(6)cells/kg per week)and 50.0%(3/6)in the dose-expansion *** overall response rate was 44.4%(8/18)among all enrolled *** sum up,UC-MSCs are effective and well tolerated in treating refractory ITP(*** ID:NCT04014166).
