In vitro selected peptides bind with thymidylate synthase mRNA and inhibit its translation

Thymidylate synthase (TS),an essential enzyme for catalyzing the biosynthesisof thymidylate,is a critical therapeutic target in cancer *** studies have shown that TSfunctions as an RNA-binding protein by interacting with two different sequences on its ownmRNA,thus,repressing translational *** this study,peptides binding TS RNA with highaffinity were isolated using mRNA display from a large peptide library (>10^(13) differentsequences).The randomized library was subjected up to twelve rounds of in vitro selection *** the amino acid composition of the selected peptides (12.h round,R12. withthose from the initial random library (round zero,R0),the basic and aromatic residues in theselected peptides were enriched significantly,suggesting that these peptide regions might beimportant in the peptide-TS mRNA *** the amino acids at each random positionbased on their physicochemical properties and comparing the distributions with those of the initialrandom pool,an obvious basic charge characteristic was found at positions 1,12.17 and 18,suggestingthat basic side chains participate in RNA *** structure prediction showed that theselected peptides of R12.pool represented a helical propensity compared with R0 pool,and the regionswere rich in basic *** electrophoretic gel mobility shift and in vitro translation assaysshowed that the peptides selected using mRNA display could bind TS RNA specifically and inhibit thetranslation of TS *** results suggested that the identified peptides could be used as new TSinhibitors and developed to a novel class of anticancer agents.

Distribution of organochlorine compounds in water, porewater and sediments in Xiamen Harbour

The Concentrations of 18 organochlorine pesticides and 12.polychlorinated biphenyls (PCBs) in water, porewater and sediments from the Xiamen Harbour were analyzed quantitatively. It shows that, the levels of all organochlorine insecticides varied from 6. 60 to 19 .6 ng/dm3 (in water), 11. 9-78. 5 ng/dm3(in porewater) and not detected (ND) - 0. 58 ng/g dry weight (in sediments), and those of total DDTs were in the range ND - 0. 06 ng/g dry weight. Concentrations of PCBs and in- secticides in porewater were higher than in surface water, due to the higher affinity of these hydrophobic compound for sedimentary phase than to water. It implies there would be a potential flux of pollutants from sediments to overlying water. Compared with the concentrations of these pollutants measured in the Xiamen Harbour in 1993, it is one to several magnitude order lower, which suggests their inputs have been decreased in recent years and possible degradation of these compounds. The levels of these pollutants were also compared with other estuaries.

Genetic source,migration and accumulation of helium under deep thermal fluid activities:A case study of Ledong diapir area in Yinggehai Basin,South china Sea

Based on the geochemical parameters and analytical data,the heat conservation equation,mass balance law,Rayleigh fractionation model and other methods were used to quantify the in-situ yield and external flux of crust-derived helium,and the initial He concentration and thermal driving mechanism of mantle-derived helium,in the Ledong Diapir area,the Yinggehai Basin,in order to understand the genetic source,migration and accumulation mechanisms of helium under deep thermal fluid *** average content of mantle-derived He is only 0.0014%,the ^(3)He/^(4)He value is(0.002.2.190)×10^(−6),and the R/Ra value ranges from 0.01 to 1.52.indicating the contribution of mantle-derived He is 0.09%–19.84%,while the proportion of crust-derived helium can reach over 80%.Quantitative analysis indicates that the crust-derived helium is dominated by external input,followed by in-situ production,in the Ledong diapir *** crust-derived helium exhibits an in-situ 4 He yield rate of(7.66–7.95)×10^(−13)cm^(3)/(a·g),an in-situ 4 He yield of(4.10–4.2.)×10^(−4)cm^(3)/g,and an external 4 He influx of(5.84–9.06)×10^(−2.cm^(3)/*** results may be related to atmospheric recharge into formation fluid and deep rock-water *** ratio of initial mole volume of 3 He to enthalpy(W)is(0.004–0.018)×10^(−11) cm^(3)/J,and the heat contribution from the deep mantle(X_(M))accounts for 7.63%–36.18%,indicating that deep hot fluid activities drive the migration of mantle-derived 3 *** primary helium migration depends on advection,while the secondary migration is controlled by hydrothermal degassing and gas-liquid *** deep to shallow layers,the CO_(2.3) He value rises from 1.34×10^(9)to 486×10^(9),indicating large amount of CO_(2.has *** the influence of deep thermal fluid,helium migration and accumulation mechanisms include:deep heat driven diffusion,advection release,vertical hydrothermal degassing,shallow lateral migration,accumulation in traps far from faults,

Formation of apoptotic bodies is a typical character ofaPoptotic cell death, but how the processes are controlledis not known. In this study, we compared two apoptosisinducing systems in vascular endothelial cells (VEC). Wefound that the formation of aPoptotic bodies during apop-tosis induced by rattlesnake venom, which is an unique andspecific aPoptosis inducer to vascular endotheliaI cells, wasmuch faster than that induced by deprivation of survivalfactors (aFGF and serum). When we blocked the synthesisof mRNAs in cells treated with rattlesnake venom by DRB(5, 6- dichloro- 1 -β- D- rib ofur anosylb enzimidazole ), an in-hibitor of transcription, the formation of aPoptotic bodieswas dramatically inhibited. We examined the expressionof Psa gene and found that its expression was much higherin apoptosis induced by rattlesnake venom than that inaPoptosis induced by deprivation of aFGF and serum. Ourresults suggest that gene expression is important and P53gene may play a major role in inducing the formation ofapoptotic bodies in VEC.

Comparative Analysis of ARIMA and LSTM Model-Based Anomaly Detection for Unannotated Structural Health Monitoring Data in an Immersed Tunnel

Structural Health Monitoring(SHM)systems have become a crucial tool for the operational management of long *** immersed tunnels exposed to both traffic loads and the effects of the marine environment,efficiently identifying abnormal conditions from the extensive unannotated SHM data presents a significant *** study proposed amodel-based approach for anomaly detection and conducted validation and comparative analysis of two distinct temporal predictive models using SHM data from a real immersed ***,a dynamic predictive model-based anomaly detectionmethod is proposed,which utilizes a rolling time window for modeling to achieve dynamic *** the assumption of temporal data similarity,an interval prediction value deviation was employed to determine the abnormality of the ***,dynamic predictive models were constructed based on the Autoregressive Integrated Moving Average(ARIMA)and Long Short-Term Memory(LSTM)*** hyperparameters of these models were optimized and selected using monitoring data from the immersed tunnel,yielding viable static and dynamic predictive ***,the models were applied within the same segment of SHM data,to validate the effectiveness of the anomaly detection approach based on dynamic predictive modeling.A detailed comparative analysis discusses the discrepancies in temporal anomaly detection between the ARIMA-and LSTM-based *** results demonstrated that the dynamic predictive modelbased anomaly detection approach was effective for dealing with unannotated SHM *** a comparison between ARIMA and LSTM,it was found that ARIMA demonstrated higher modeling efficiency,rendering it suitable for short-term *** contrast,the LSTM model exhibited greater capacity to capture long-term performance trends and enhanced early warning capabilities,thereby resulting in superior overall performance.

In 2.2.,the Pakistan witnessed the hottest spring and wettest summer in *** devastating floods inundated a large portion of Pakistan and caused enormous ***,the primary water source and its contributions to these unprecedented floods remain *** on the reservoir inflow measurements,Multi-Source Weighted-Ensemble Precipitation(MSWEP),the fifth generation ECMWF atmospheric reanalysis(ERA5)products,this study quantified the contributions of monsoon precipitation,antecedent snow-melts,and orographic precipitation enhancement to floods in *** found that the Indus experienced at least four inflow up-rushes,which was mainly supplied by precipitation and snowmelt;In upper Indus,abnormally high temperature continued to influence the whole summer and lead to large amounts of snowmelts which not only was a key water supply to the flood but also provided favorable soil moisture conditions for the latter *** July,the snowmelt has higher contributions than the precipitation to the streamflow of Indus River,with contribution value of more than 60%.Moreover,the snowmelt could still supply 2.%-40%water to the lower Indus in July and August;The leading driver of 2.2. mega-floods over the southern Pakistan in July and August was dominated by the precipitation,where terrain disturbance induced precipitation account to approximately 33%over the southern *** results help to understand the mechanisms of flood formation,and to better predict future flood risks over complex terrain regions.

Three-dimensional structure of a low salinity tongue in the southern Taiwan Strait observed in the summer of 2.05

Cruise observations with CTD （conductivity-temperature-depth） profiler were carried out in the southern Taiwan Strait in the summer of 2.05. Using the cruise data, two-dimensional maps of salinity and temperature distributions at depths of 5, 10, 15, 2., and 30 m were generated. The maps show a low salinity tongue sandwiched by low temperature and high salinity waters on the shallow water side and high temperature and high salinity waters on the deep water side. The further analysis indicates that the low salinity water has a nature of river-diluted water. A possible source of the diluted water is the Zhujiang （Pearl） Estuary. Meanwhile, the summer monsoon is judged as a possible driving force for this northeastward jet-like current. The coastal upwelling and the South china Sea Warm Current confine the low salinity water to flow along the central line of the strait. Previous investigations and a numerical model are used to verify that the upstream of the low salinity current is the Zhujiang Estuary. Thus, the low salinity tongue is produced by four major elements： Zhujinag Estuary diluted water, monsoon wind driving, coastal upwelling and South china Sea Warm Current modifications.

Cathode interfacial materials(CIMs)stand as critical elemental in organic solar cells(OSCs),which can align energy levels,and foster ohmic contacts between the cathode and active layer of the ***,the lagging advancement in CIMs has concurrently engendered the oversight of theoretical inquiries pertaining to the impact of molecular structure on their *** into this realm,we present two propeller-shaped isomers,4,4',4''-(benzo[1,2.b:3,4-b':5,6-b'']trithiophene-2.5,8-triyl)tris(2.(3-(dimethylamino)propyl)-1H-benzo[de]isoquinoline-1,3(2.)-dione)(3ONIN)and 6,6',6''-(benzo[1,2.b:3,4-b':5,6-b'']trithiophene-2.5,8-triyl)tris(2.(3-(dimethylamino)propyl)-1H-benzo[de]isoquinoline-1,3(2.)-dione)(3PNIN),distinguished by their molecular planarity,as a promising foundation for crafting highly efficient *** study illuminates the superiority of 3PNIN with more plane structure,exemplified by its enhanced molar extinction coefficient,deeper lowest unoccupied molecular orbital(LUMO)and highest occupied molecular orbital(HOMO)energy levels,intensified self-doping effect,heightened electron mobility,and elevated conductivity,in comparison to its counterpart,*** a result,3PNIN and 3ONIN-treated OSC devices yield efficiencies of 17.73%and 16.82.,*** finding serves as a compelling validation of the critical role played by molecular planarity in influencing CIM performance.
