Assimilating FY-4A AGRI Radiances with a Channel-Sensitive Cloud Detection Scheme for the Analysis and Forecasting of Multiple Typhoons

This paper presents an attempt at assimilating clear-sky FY-4A Advanced Geosynchronous Radiation Imager(AGRI)radiances from two water vapor channels for the prediction of three landfalling typhoon events over the West Pacific Ocean using the 3DVar data assimilation(DA)method along with the WRF model.A channel-sensitive cloud detection scheme based on the particle filter(PF)algorithm is developed and examined against a cloud detection scheme using the multivariate and minimum residual(MMR)algorithm and another traditional cloud mask–dependent cloud detection scheme.Results show that both channel-sensitive cloud detection schemes are effective,while the PF scheme is able to reserve more pixels than the MMR scheme for the same channel.In general,the added value of AGRI radiances is confirmed when comparing with the control experiment without AGRI radiances.Moreover,it is found that the analysis fields of the PF experiment are mostly improved in terms of better depicting the typhoon,including the temperature,moisture,and dynamical conditions.The typhoon track forecast skill is improved with AGRI radiance DA,which could be explained by better simulating the upper trough.The impact of assimilating AGRI radiances on typhoon intensity forecasts is small.On the other hand,improved rainfall forecasts from AGRI DA experiments are found along with reduced errors for both the thermodynamic and moisture fields,albeit the improvements are limited.

Bioactive compounds derived from herbal medicinal plants modulate various therapeutic targets and signaling pathways associated with cardiovascular diseases(CVDs),the world’s primary cause of death.Ginkgo biloba,a well-known traditional Chinese medicine with notable cardiovascular actions,has been used as a cardio-and cerebrovascular therapeutic drug and nutraceutical in Asian countries for centuries.Preclinical studies have shown that ginkgolide B,a bioactive component in Ginkgo biloba,can ameliorate atherosclerosis in cultured vascular cells and disease models.Of clinical relevance,several clinical trials are ongoing or being completed to examine the efficacy and safety of ginkgolide B-related drug preparations in the prevention of cerebrovascular diseases,such as ischemia stroke.Here,we present a comprehensive review of the pharmacological activities,pharmacokinetic characteristics,and mechanisms of action of ginkgolide B in atherosclerosis prevention and therapy.We highlight new molecular targets of ginkgolide B,including nicotinamide adenine dinucleotide phosphate oxidases(NADPH oxidase),lectin-like oxidized LDL receptor-1(LOX-1),sirtuin 1(SIRT1),platelet-activating factor(PAF),proprotein convertase subtilisin/kexin type 9(PCSK9)and others.Finally,we provide an overview and discussion of the therapeutic potential of ginkgolide B and highlight the future perspective of developing ginkgolide B as an effective therapeutic agent for treating atherosclerosis.

Resistance index and browning mechanism of apple peel under high temperature stress

Apples are one of the most important economic crops worldwide.Because of global warming and an aggravation of environmental,abnormally high temperatures occur frequently in fruit-growing season and seriously affect normal fruit growth and reduce fruit quality and yield.We took five-year-old Ruixue’(Qinfu 1×Pink Lady;CNA20151469.1) fruits as test materials,and the ambient temperature during fruit development was monitored.The results showed that during the fruit-growing season,especially during the rapid growth stage (July to August),the maximum daily temperature exceeded 30℃ and lasted for more than 40 days.To determine the effects of high temperature stress on the apple fruit resistance,we treated expanding,veraison,and maturity-period fruits at different temperatures.It was found that the fruits of the expanding period showed strong resistance to high temperature stress,whereas during veraison and maturity,fruit resistance to high temperature stress decreased,and the fruit peel browning phenotype appeared.Meanwhile,the content of malonaldehyde (MDA),hydrogen peroxide (H_(2)O_(2)),and superoxide anion (O._(2)^(-)) in the peel gradually increased with increasing temperature.The content of total phenols,flavanol,and flavonoids in the peel decreased substantially at 45℃.Moreover,it was found that polyphenol oxidase gene (MdPPO1) was most sensitive to high temperature stress in apple.Furthermore,transient and stable MdPPO1 overexpression significantly promoted peel browning.The transgenic materials were more sensitive to high temperatures,and browning was more severe compared to non-genetically modified organism (WT).Stable MdPPO1 knockout calli obtained via clustered regularly interspersed short palindromic repeats (CRISPR/Cas9) gene knockout technology reduced the browning phenotype,and the resultant fruits were not sensitive to the effects of high temperature stress.Thus,MdPPO1 expression may be a key factor of high temperature-related changes observed in the browning phenotype that provides a scientific theoreti

An alfalfa MYB-like transcriptional factor MsMYBH positively regulates alfalfa seedling drought resistance and undergoes MsWAV3-mediated degradation

Drought is a major threat to alfalfa(Medicago sativa L.)production.The discovery of important alfalfa genes regulating drought response will facilitate breeding for drought-resistant alfalfa cultivars.Here,we report a genome-wide association study of drought resistance in alfalfa.We identified and functionally characterized an MYB-like transcription factor gene(MsMYBH),which increases the drought resistance in alfalfa.Compared with the wild-types,the biomass and forage quality were enhanced in MsMYBH overexpressed plants.Combined RNA-seq,proteomics and chromatin immunoprecipitation analysis showed that MsMYBH can directly bind to the promoters of MsMCP1,MsMCP2,MsPRX1A and MsCARCAB to improve their expression.The outcomes of such interactions include better water balance,high photosynthetic efficiency and scavenge excess H_(2)O_(2)in response to drought.Furthermore,an E3 ubiquitin ligase(MsWAV3)was found to induce MsMYBH degradation under long-term drought,via the 26S proteasome pathway.Furthermore,variable-number tandem repeats in MsMYBH promoter were characterized among a collection of germplasms,and the variation is associated with promoter activity.Collectively,our findings shed light on the functions of MsMYBH and provide a pivotal gene that could be leveraged for breeding drought-resistant alfalfa.This discovery also offers new insights into the mechanisms of drought resistance in alfalfa.

A distributed architecture of optical line terminal(OLT)equipment is proposed for response to national bandwidth acceleration requirements and for future smooth evolution to 50G passive optical network(PON).This architecture moves the forwarding function of the control board to each service board to improve the switching capacity and performance of the system.The traditional control boards of centralized architecture OLT equipment have exchange and traffic processing function,and every service board is only controlled by the control board.In the distributed architecture of OLT equipment,control boards still maintain exchange function,whereas service boards have traffic processing functions.This method separates the exchange and traffic processing functions,which improves reliability.This paper also presents optical access network equipment combined with telemetry technology that provides reliable guarantee for intelligent analysis and data mining.Compared with the traditional push mode of network management,the data collected by network management combined with telemetry technology and artificial intelligence(AI)analysis can be used for network planning,assurance of the bandwidth and accurate operation.

Belowground Bud Bank Is Insensitive to Short-Term Nutrient Addition in the Meadow Steppe of Inner Mongolia

Human activities and industrialization have signiﬁcantly increased soil nutrients,such as nitrogen(N)and phos-phorus(P),profoundly impacting the composition and structure of plant community,as well as the ecosystem functions,especially in nutrient-limited ecosystems.However,as the key propagule pool of perennial grasslands,how belowground bud bank and its relationship with aboveground vegetation respond to short-term changes in soil nutrients was still unclear.In this study,we conducted a short-term(2021–2022)soil fertilization experiment with N addition(10 g N m^(-2) yr^(-1))and P addition(5 g N m^(-2) yr^(-1))in the meadow steppe of Inner Mongolia,China,to explore the responses of belowground bud bank,aboveground shoot population and their relationships(represented by the ratio of bud to shoot density-meristem limitation index(MLI))for the whole community and three plant functional groups(perennial rhizomatous grasses-PR,perennial bunchgrasses-PB,and perennial forbs-PF)to nutrient addition.The short-term nutrient addition had no signiﬁcant inﬂuences on belowground bud density,aboveground shoot density,and MLI of the whole plant community.Plant functional groups showed different responses to soil fertilization.Speciﬁcally,N addition signiﬁcantly increased the bud density and shoot density of PR,especially in combination with P addition.N addition reduced the shoot density of PF but had no inﬂuence on its bud density and MLI.Nutrient addition had signiﬁcant effects on the three indicators of PB.Our study indicates that the belowground bud bank and its relationship with aboveground vegetation in temperate meadow steppe are insensitive to short-term soil fertilization,but plant functional groups exhibit speciﬁc responses in terms of population regeneration,which implies that plant community composition and ecosystem functions will be changed under the ongoing global change.

Mutation of the MAPK7 gene was related to human scoliosis.Mapk7 regulated the development of limb bones and skulls in mice.However,the role of MAPK7 in vertebral development is still unclear.In this study,we constructed Col2a1-cre;Mapk7 f/f transgenic mouse model to delete Mapk7 in cartilage,which displayed kyphosis and osteopenia.Mechanistically,Mapk7 loss decreased MEF2C expression and thus activated PTEN to oppose PI3K/AKT signaling in vertebral growth plate chondrocytes,which impaired chondrocyte hypertrophy and attenuated vertebral ossification.In vivo,systemic pharmacological activation of AKT rescued impaired chondrocyte hypertrophy and alleviated mouse vertebral defects caused by Mapk7 deficiency.Our study firstly clarified the mechanism by which MAPK7 was involved in vertebral development,which might contribute to understanding the pathology of spinal deformity and provide a basis for the treatment of developmental disorders of the spine.

Hypoglycemic activities of flowers of Xanthoceras sorbifolia and identification of anti-oxidant components by off-line UPLC-QTOF-MS/MS-free radical scavenging detection

Objective: To identify phytochemical constituents present in the extract of flowers of Xanthoceras sorbifolia and evaluate their anti-oxidant and anti-hyperglycemic capacities.Methods: The AlCl3colorimetric method and Prussian Blue assay were used to determine the contents of total flavonoids and total phenolic acids in extraction layers, and the bioactive layers was screened through anti-oxidative activity in vitro. The Waters ACQUITY UPLC system and a Waters ACQUITY UPLC BEH C18column(2.0 mm × 150 mm, 5 μm) were used to identify the ingredients. And anti-oxidative ingredients were screened by off-line UPLC-QTOF-MS/MS-free radical scavenging. The ameliorative role of it was further evaluated in a high-fat, streptozotocin-induced type 2 diabetic rat model and the study was carried out on NADPH oxidase(PDB ID: 2CDU) by molecular docking.Results: Combined with the results of activity screening in vitro, the anti-oxidative part was identified as the ethyl acetate layer. A total of 24 chemical constituents were identified by liquid chromatographymass spectrometry in the ethyl acetate layer and 13 main anti-oxidative active constituents were preliminarily screened out through off-line UPLC-QTOF-MS/MS-free radical scavenging. In vivo experiments showed that flowers of X. sorbifolia could significantly reduce the blood glucose level of diabetic mice and alleviate liver cell damage. Based on the results of docking analysis related to the identified phytocompounds and oxidase which involved in type 2 diabetes, quercetin 3-O-rutinoside, kaempferol-3-O-rhamnoside, isorhamnetin-3-O-glucoside, and isoquercitrin showed a better inhibitory profile.Conclusion: The ethyl acetate layer was rich in flavonoids and phenolic acids and had significant anti-oxidant activity, which could prevent hyperglycemia. This observed activity profile suggested X. sorbifolia flowers as a promising new source of tea to develop alternative natural anti-diabetic products with a high safety margin.

More common RNAemia in the early stage of severe SARS-CoV-2 BF.7.14 infections in pediatric patients

The risk factors of severe infections in children during the severe acute respiratory syndrome coronavirus 2(SARS‐CoV‐2)outbreak in Beijing remain elusive.SARS‐CoV‐2‐positive children admitted to the intensive care unit(ICU)with collected plasma specimens were enrolled and screened for common pathogens using capillary electrophoresis‐based multiplex PCR from December 12,2022,to January 24,2023.The SARS‐CoV‐2 subvariants were identified using next‐generation sequencing.Plasma was positive for two(positive;P),one(suspicious;S),or no(negative;N)SARS‐CoV‐2 genes were classified as plasmatic RNA‐positive(RNAemia;P+S)or without RNAemia(N).Clinical and laboratory data of the enrolled cases were then collected and analyzed.The 34 enrolled children included 26 males and 24 younger than three years.All were negative for other respiratory pathogens.BF.7.14(18/29)was the predominant subvariant.Viral loads in respiratory specimens,hours from symptom onset to the first respiratory specimen collection(time‐variable),with comorbidities and BF.7.14 and BA.5.2 distributions were significantly different in P vs.N and RNAemia vs.without RNAemia group.Among most cases,the T lymphocyte ratios decreased,while the cytokine level and the B lymphocyte ratio increased.The time variables were 2.22±2.05 and 4.00±2.49 days in BF.7.14 and BA.5.2 infections,respectively.In conclusion,SARS‐CoV‐2 was more likely to cause severe infections among males aged≤3 years old with comorbidities during the SARS‐CoV‐2 outbreak in Beijing,while RNAemia is more common in children at the early stage of severe BF.7.14 infections,and most had high cytokine levels and B‐cell activation.

Simulation-Based Investigation of Crack Formation in Forged MB15 Magnesium Alloy

In the process of riveting an MB15 forging die,cracks were discovered emerging along the longitudinal direction and near the riveting hole.Through fracture analysis,microscopic observation,energy spectrum analysis,metallographic examination,and hardness test,the properties and causes of the cracks are discussed.The results indicate that the cracking type is intergranular brittle cracking,occurring during the forging stage.Furthermore,the recrystallization at the crack site is found to be incomplete,which is attributed to the low deformation temperature.
