Long non-coding RNAs as promising targets for controlling disease vector mosquitoes

Hematophagous female mosquitoes are important vectors of numerous devastating human diseases, posing a major public health threat. Effective prevention and control of mosquito-borne diseases rely considerably on progress in understanding the molecular mechanisms of various life activities, and accordingly, the molecules that regulate the various life activities of mosquitoes are potential targets for implementing future vector control strategies. Many long non-coding RNAs (lncRNAs) have been identified in mosquitoes and significant progress has been made in determining their functions. Here, we present a comprehensive overview of the research advances on mosquito lncRNAs, including their molecular identification, function, and interaction with other non-coding RNAs, as well as their synergistic regulatory roles in mosquito life activities. We also highlight the potential roles of competitive endogenous RNAs in mosquito growth and development, as well as in insecticide resistance and virus-host interactions. Insights into the biological functions and mechanisms of lncRNAs in mosquito life activities, viral replication, pathogenesis, and transmission will contribute to the development of novel drugs and safe vaccines. Thousands of long non-coding RNA (lncRNAs) have been identified from different species of mosquitoes based on high-throughput transcriptome sequencing. Let-7 microRNA targets BmCentrin to modulate the development and functionality of the middle silk gland in the silkworm, Bombyx mori

The silk gland of the silkworm Bombyx mori serves as a valuable model for investigating the morphological structure and physiological functions of organs. Previous studies have demonstrated the notable regulatory role of let-7 microRNA in the silk gland, but its specific molecular mechanism remains to be elucidated across different segments of this organ. In this study, we further investigated the functional mechanism of let-7 in the middle silk gland (MSG). The MSG of a let-7 knockout strain was analyzed using a combined proteomic and metabolomic technique, revealing the enrichment of differential proteins and metabolites in the DNA synthesis and energy metabolism pathways. BmCentrin was identified as a novel target gene of let-7 in the MSG, and its downregulation inhibited the proliferation of BmN4-SID1 cells, which is exactly opposite to the role of let-7 in these cells. CRISPR/Cas9 genome editing and transgenic technologies were employed to manipulate BmCentrin in the MSG. Knockout of BmCentrin led to severe MSG atrophy, whereas the overexpression of BmCentrin resulted in beaded MSG. Further measurements of these knockout or overexpression strains revealed significant changes in the expression levels of sericin protein genes, the weight of the cocoon and the mechanical properties of the silk. Investigating the biological role of BmCentrin in the silk gland offers valuable insights for elucidating the molecular mechanisms by which let-7 controls silk gland development and silk protein synthesis in the silkworm.

Mathematical Model of Hydrodynamic Torque Converter and Analytic Description of Streamline

The mathematical model of a 3-element centripetal-turbine hydrodynamic torque converter and analytic description of fluid flow inside the hydrodynamic torque converter are investigated. A new torus coordinate system is proposed so as to quantitatively describe fluid movement inside the hydrodynamic torque converter. The particle movement inside the hydrodynamic torque converter is decomposed into meridional component movement and torus component movement, and a universal meridional streamline equation is derived. According to the relationship between the converter wheel velocity polygon and its blade angle, a torus streamline differential equation is established. The universal meridional streamline equation is approximated with square polynomials. The approximation error curve is given and the percentage error is not greater than 0.86%. Considered as a function of polar angle, the blade angle cotangent of each converter wheel varies linearly with polar angle. By using integral calculus, torus streamline equations are obtained. As a result, the problem of difficult flow description of the hydrodynamic torque converter is solved and a new analytic research system is established.

IMPROVE THE KINETIC PERFORMANCE OF THE PUMP CONTROLLED CLAMPING UNIT IN PLASTIC INJECTION MOLDING MACHINE WITH ADAPTIVE CONTROL STRATEGY

The kinetic characteristics of the clamping unit of plastic injection molding machine that is controlled by close loop with newly developed double speed variable pump unit are investigated. Considering the wide variation of the cylinder equivalent mass caused by the transmission ratio of clamping unit and the severe instantaneous impact force acted on the cylinder during the mold closing and opening process, an adaptive control principle of parameter and structure is proposed to improve its kinetic performance. The adaptive correlation between the acceleration feedback gain and the variable mass is derived. The pressure differential feedback is introduced to improve the dynamic performance in the case of small inertia and heavy impact load. The adaptation of sum pressure to load is used to reduce the energy loss of the system. The research results are verified by the simulation and experiment, The investigation method and the conclusions are also suitable for the differential cylinder system controlled by the traditional servo pump unit.

Prognosis of gestational choriocarcinoma diagnosed incidentally during laparoscopy for a presumed cornual pregnancy:a report of five cases

The clinical presentations of gestational choriocarcinoma vary markedly, and a misdiagnosis could be made in atypical patients if simply relying on clinical features. Laparoscopic resection of uterine mass lesion is rarely used in gestational choriocarcinoma diagnosis because of the fear of heavy blood loss and distant metastasis. Five patients who were preoperatively diagnosed as having cornual pregnancy underwent laparoscopic resection of mass lesion and then proved to have gestational choriocarcinoma based on pathological examinations. Chemotherapy was started within two days after surgery, and the rate of complete remission was 100%. The mean follow-up time was 29.8±19.1 months, and no patient showed signs of relapse. Laparoscopic resection of uterine mass followed by timely postoperative chemotherapy may be an effective and safe way to obtain pathologic results in patients with suspected gestational choriocarcinoma.

The development of spatial transcriptomics(ST)technologies has transformed genetic research from a single-cell data level to a two-dimensional spatial coordinate system and facilitated the study of the composition and function of various cell subsets in different environments and *** large-scale data generated by these ST technologies,which contain spatial gene expression information,have elicited the need for spatially resolved approaches to meet the requirements of computational and biological data *** requirements include dealing with the explosive growth of data to determine the cell-level and gene-level expression,correcting the inner batch effect and loss of expression to improve the data quality,conducting efficient interpretation and in-depth knowledge mining both at the single-cell and tissue-wide levels,and conducting multi-omics integration analysis to provide an extensible framework toward the in-depth understanding of biological ***,algorithms designed specifically for ST technologies to meet these requirements are still in their ***,we review computational approaches to these problems in light of corresponding issues and challenges,and present forward-looking insights into algorithm development.

Construction of Phosphorus-Functionalized Multichannel Carbon Interlayers for Dendrite-Free Metallic Zn Anodes

Zn metal anodes are usually subject to grave dendrite growth during platting/stripping,which dramatically curtails the lifespan of aqueous Zn-ion batteries and *** address above problems,in our work,a novel phosphorus-functionalized multichannel carbon interlayer was designed and covered on Zn *** results demonstrated that the multichannel structure combined with the three-dimensional meshy skeleton can provide more sufficient space for Zn deposition,thereby effectively inhibiting the growth of zinc ***,theoretical calculations also confirmed that the P-C and P=O functional groups from phosphorus-functionalized multichannel carbon interlayer have the decisive influence in reducing the zinc nucleation potential and depositing uniformly ***,the symmetrical battery assembled with phosphorus-functionalized multichannel carbon interlayer-covered Zn anodes possessed a long lifetime of 3300 h at 2 mA cm^(-2)with 1 mAh cm^(-2).Furthermore,the full cell with activated carbon cathodes exhibited a high specific capacity of 80.5 mAh g^(-1)and outstanding cycling stability without capacity decay after 15000 cycles at a high current density of 5 A g^(-1).The superior electrochemical performance exceeded that of most reported ***,our synthesized zincophilic interlayer with the unique structure has superior prospects for application in stabilizing zinc anodes and prolonging the lifespan of batteries.

Resource Sharing by the Different Clonal White Clover Plant(Trifolium repens L.)

Resource sharing in the different parts of the clonal white clover plant (Trifolium repens L.) was investigated in this study. The results showed that the integration of water and nitrogen metabolism occured among parental, young and old branches of a plant when water passed through its stolons. Under the sufficient supply of light, water and nutrients from soil the newly developed branches showed similar capacity in taking in sufficient water and nitrogen to that of old branches. The development of the new branches did not affect the growth of the old and parental branches attached. The results also indicated that energy values in roots was significantly related with plant age, however, the energy values were various in the different parts of a plant.

Production of β-1,3-glucanase and chitinase of two biocon-trol agents and their possible modes of action

Pichia membranefaciens Hansen and Candida guilliermondii (Cast) Langeronet Guerra are two antagonists of R. stolonifer on harvested nectarine and peach fruits. In this study, β-1,3-glucanase and chitinase activities of the antagonists were induced in vitro and in vivo. The highest β-1, 3-glucanase activity was detected in Lilly-Barnett minimal salt medium supplemented with glucose in combination with CWP of R. stolonifer as a carbon source. The β-1,3-glucanase activity of P. membranefaciens reached the maximum level, being 114.0 SU (specific activity unit), and that of C. guilliermondii reached 103.2 SU. The lowest β-1,3-glucanase activity was observed in the medium containing glucose as sole car-bon source. P. membranefaciens was able to produce signifi-cantly higher levels of chitinase (exochitinase and endochiti-nase) in vitro than C. guilliermondii grown in Czapeck mini-mal medium.
