Protective mechanism of quercetin compounds against acrylamide-induced hepatotoxicity

Quercetin compounds have antioxidant,anti-inflammatory and anticancer pharmacological *** exposure to acrylamide(AA)can cause liver injury and endanger human ***,whether quercetin compounds can attenuate AA-induced liver injury and the specific mechanism are not ***,we studied the mechanism and structure-activity relationship of quercetin compounds in reducing AA-induced hepatotoxicity in vivo and in *** vivo studies found that quercetin-like compounds protect against AAinduced liver injury by reducing oxidative stress levels,activating the Akt/m TOR signaling pathway to attenuate autophagy,and improving mitochondrial apoptosis and endoplasmic reticulum stress-mediated *** vitro studies found that quercetin compounds protected Hep G2 cells from AA by attenuating the activation of AA-induced autophagy,lowering reactive oxygen species(ROS)levels by exerting antioxidant effects and thus attenuating oxidative stress,increasing mitochondrial membrane potential(MMP),and improving apoptosis-related proteins,thus attenuating AA-induced ***,the conformational differences between quercetin compounds correlated with their protective capacity against AA-induced hepatotoxicity,with quercetin showing the best protective capacity due to its strongest antioxidant *** conclusion,quercetin compounds can protect against AA-induced liver injury through multiple pathways of oxidative stress,autophagy and apoptosis,and their protective capacity correlates with antioxidant activity.

冠状动脉造影是评估冠状动脉解剖学特征和指导经皮冠状动脉介入治疗(PCI)的传统影像技术。然而,其不能评估动脉壁、血管尺寸、斑块特征和支架置入结果。腔内成像(IVI)技术可提供血管壁结构、斑块病变和置入物等重要信息,其辅助冠心病诊治的临床价值已得到广泛认可。通过准确评估斑块形态可识别斑块易损特征。在体确定急性冠脉综合征患者"罪犯"斑块并提供影像学特征,精准指导个体化治疗。IVI指导PCI获益的证据日益增多,IVI可规划PCI手术策略、优化支架选择,并提升手术成功率。对支架失败患者进行IVI检查可了解其潜在机制、制定后续治疗方案。不同IVI技术具有各自的优势和局限性,未来各种迭代新技术、影像融合、人工智能融合技术会进入临床应用。

ASD (autism spectrum disorder) is a neurodevelopmental disorder influenced by genes and the environment. This study investigated the protective effects of bovine MFGM (BMFGM) and goat MFGM (GMFGM) supplementation on ASD model mice. Analysis of phospholipid composition showed higher levels of phosphatidylcholine in BMFGM and phosphatidylethanolamine in GMFGM. Behavioral results indicated MFGMs ameliorated social deficits， with GMFGM being more effective. MFGMs mitigated neuroinflammation by suppressing microglial overactivation and proinflammatory cytokines expression， meanwhile GMFGM increased the anti-inflammatory factor IL-10. MFGMs also altered gut microbiota composition and maintained gut barrier integrity. Uniquely， GMFGM increased butyrate production. Correlation analysis revealed positive associations between social behavior， levels of phosphatidylcholine， sphingomyelin， and the abundance of Allobaculum， Clostridium＿sensu＿stricto， and Turicibacter. Overall， these findings revealed the protective effects of MFGMs on neurodevelopment in ASD animal models and the underlying mechanism could be partly explained by their regulation of gut microbiota by the phospholipid components in MFGMs.

为克服姜黄素和表没食子儿茶素没食子酸酯(EGCG)两种生物活性成分在稳定性和代谢动力学方面的局限性,进一步发挥其协同抗氧化和抗炎作用,采用薄膜超声法构建共递送姜黄素和EGCG的脂质体。以粒径、PDI和电位为评价指标,分析磷脂与胆固醇质量比、磷脂与吐温80质量比、水合时间及超声时间对脂质体的影响,并通过静电吸附作用对脂质体进行乳铁蛋白和透明质酸修饰。使用脂多糖构建BV2小胶质细胞炎症模型,探讨乳铁蛋白和透明质酸修饰的共递送姜黄素和EGCG的脂质体对神经炎症的改善作用。在磷脂与胆固醇质量比为10∶1,磷脂和吐温80质量比为10∶3,水合时间40 min,以及超声时间15 min时,成功制备了呈均匀球形、平均粒径为(131.53±0.258)nm的共递送姜黄素和EGCG的脂质体,在乳铁蛋白体积为0.3 mL,透明质酸体积为0.5 mL时对脂质体进行修饰。实验结果显示,乳铁蛋白和透明质酸修饰的共递送姜黄素和EGCG的脂质体显著提高了姜黄素和EGCG的稳定性和抗氧化特性,抑制了脂多糖诱导的细胞形态的改变,恢复线粒体功能障碍,降低细胞内活性氧的产生,改善了脂多糖诱导的BV2小胶质细胞的炎症作用。研究结果旨在为进一步开发具有神经保护作用的功能食品提供新的思路。

目的探讨芪贞归脾汤辅助治疗对食管癌伴锁骨上淋巴结转移患者细胞钙黏素(E-cadherin)表达的影响。方法选取2016年1月—2021年1月食管癌伴锁骨上淋巴结转移病例96例,随机编号方法分两组,各48例,对照组采用常规放疗;在此基础上,观察组采用芪贞归脾汤辅助治疗,持续治疗8周。治疗前后评价中医证候积分、匹兹堡睡眠质量指数(Pittsburgh sleep quality index,PSQI)、免疫功能、细胞角蛋白19片段(cytokeratin 19 fragment,CYFRA21-1)、癌胚抗原(carcinoembryonic antigen,CEA)、癌抗原(cancer antigen 125,CA125)、E-cadherin和不良反应情况。结果观察组治疗后咽下困难、胸闷不舒、形体消瘦、气短乏力等积分低于对照组,差异有统计学意义(均P0.05)。结论芪贞归脾汤辅助治疗可以提高食管癌伴锁骨上淋巴结转移患者的免疫功能和睡眠质量,降低肿瘤标志物及E-cadherin的表达,安全性高。

Chicoric acid(CA),a natural phenolic acid,has been used as a nutraceutical food ingredient due to its powerful antioxidant,anti-HIV and anti-diabetic *** could be partly metabolized into caffeic acid(CFA)and caftaric acid(CTA)on cytochrome P450s in rat liver *** compare the protective effects of CA and its metabolites on biomolecules and inflammatory responses,oxidative damage induced by free radicals in vitro and microglial inflammation triggered by lipopolysaccharide in BV2 cells were *** showed that CA,CTA and CFA all significantly inhibited protein degradation and carbonylation induced by hydroxyl radicals and alcoxyl radicals,and suppressed hemin/nitrite/H2O2 ***,CA,CTA and CFA all exerted remarkable inhibition capacities on linoleic acid and soybean lecithin liposomes peroxidation in a dose-dependent manner,and restrained the oxidation of herring sperm DNA,as well as the breakage of pBR322 plasmid ***,CA and its metabolites suppressed lipopolysaccharide-induced decline of BV2 cell viability and the production of NO and ***,bioactivities of CA were significantly stronger than those of its metabolites within a certain concentration *** study provides scientific basis for the application of CA and its metabolites as nutrition and natural antioxidants.

Bone loss caused by ageing has become one of the leading health risk factors worldwide. Wheat germ (WG) is consists of high amounts of bioactive peptides， polyunsaturated fatty acids， and dietary fibre. Currently， WG has been proven to possess strong antioxidant and anti-inflammatory properties. We recently explored the beneficial effects and relevant mechanisms of a WG-rich diet (2.5% and 5% WG) (wt:wt) on bone homeostasis in aged rats. Our results showed that 5% WG supplementation for 12 months effectively attenuated ageing-induced microstructural damage and differentiation activity changes in the femur. The 5% WG supplementation also significantly increased the levels of T-AOC， GSH-Px (P < 0.01)， and SOD (P < 0.05)， and decreased inflammatory cytokine levels (TNF-α and IL-6) (P < 0.01). Furthermore， the WG-rich diet reshaped the composition of the gut microbiota， enhancing SCFAs-producing microbes and reducing inflammation-related microbes. In addition， metabolomics analysis showed that 5% WG supplementation improved plasma metabolites related to bone metabolism. Conclusively， our study purports long-term WG-rich diet may preserve bone homeostasis by regulating gut microbiota and plasma metabolites in aged rats.
