A COVID-19 T-Cell Response Detection Method Based on a Newly Identified Human CD8+T Cell Epitope from SARS-CoV-2—Hubei Province,China,2021

Introduction:Similar to antibody detection,severe acute respiratory syndrome coronavirus type 2(SARSCoV-2)-specific T-cell response evaluation is also pivotal among the coronavirus disease 2019(COVID-19)convalescents and the vaccinated ***(N)protein is one of the main structural proteins of SARS-CoV-2 and can trigger T-cell responses in ***:An overlapping peptide pool covering the full length of the N protein was designed,peptides with positive T-cell activating potency in COVID-19 convalescents were screened,and CD8^(+)T cell epitopes were further *** epitope was used to detect the SARS-CoV-2-specific CD8^(+)T cell responses in COVID-19 convalescents based in intracellular cytokine staining and tetramer staining in flow ***:A human leukocyte antigen A(HLAA)*1101-restricted CD8^(+)T cell epitope,which could stimulate the production of IfN-γvia peripheral blood mononuclear cells(PBMCs)of the convalescents was defined,and the tetramer generated with this epitope could detect SARS-CoV-2-specific T cells in the PBMCs of the *** structural investigation eliminated that the epitope was a typical HLA-A^(*)1101-restricted T-cell epitope which was conserved among all the ***:The newly identified SARS-CoV-2-derived T-cell epitope was helpful to detect the cellular immunity against different sarbecoviruses including SARS-CoV and *** study provided an evaluation method and also a peptide candidate for the research and development of T-cell based vaccine for the virus.

The grain boundary character distribution(GBCD) optimization and its effect on the intergranular stress corrosion cracking(IGSCC) resistance in a cold-rolled and subsequently annealed fe-18 Cr-17 Mn-2 Mo-0.85 N high-nitrogen nickel-free austenitic stainless steel were systematically *** results show that stacking faults and planar slip bands appearing at the right amount of deformation(lower than 10%) are beneficial cold-rolled microstructures to the GBCD *** proportion of special boundaries gradually increases in the subsequent stages of recrystallization and grain growth,accompanying with the growth of twin-related domain in the experimental *** this way,the fraction of low ∑ coincidence site lattice(CSL) boundaries can reach as high as 82.85% for the specimen cold-rolled by 5% and then annealed at 1423 K for 72 *** GBCD optimization,low ∑ CSL boundaries and the special triple junctions(j2,j3) of high proportion can greatly hinder the nitride precipitation along grain boundaries and enhance the capability for intergranular crack arrest,thus improving the IGSCC resistance of the experimental steel.

Laser interference induced crystallization of amorphous silicon （a-Si） on the glass substrate was performed using a Q-switched Nd：YAG （yttrium aluminum garnet） laser. White light interferometer （WLI） and atomic force microscope （AfM） were used to characterize the morphology of the structured films, while X-ray diffraction （XRD）, combined with the AfM, was used to analyse the crystalline structure of the film. The experimental results show that the laser energy density above a certain threshold, in the range of 400-500 mj/cm2,triggers the patterned crystallizations which take the form similar to the laser intensity distribution. for the patterned crystallization under multipulse exposure, a definite polycrystalline structure with individual phases was observed by XRD. The difference in feature form, e.g., deepened craters or heightened lines, is related to the laser energy density relative to the threshold of evaporation of the material.

As a classical technique for chaos suppression,the time-delayed feedback controlling strategy has been widely developed by stabilizing unstable periodic orbits(UPOs)embedded in chaotic systems.A critical issue for achieving high controlling precision is to search for an appropriate time *** paper proposes a simple yet effective approach,based on incremental harmonic balance method,to determine the optimal time delay in the delayed feedback *** time delay is adjusted within the iterative scheme provided by the proposed method,and finally converges to the period of the target *** long as the optimal time delay is fixed,moreover,the attained solution makes it quite convenient to analyze its stability according to the floquet theory,which further provides the effective interval of the feedback gain.

lNovel lithium doped tris 8 hydroxylquinoline aluminium(Alq3:Li) layer is deposited between emission layer and electron injection aluminium electrode as an electron injection assistant layer in different organic light emitting diodes(OLED) to lower the electron injection barrier. In these devices, Alq3 is used as emission layer, and a bilayer film of N,N bis (1 naphhyl) N,N diphenyl 1,1 biphenyle 4,4' diamine(NPB) and 4,4,4' tris(3 methyl phenylphenylamino) triphenylamine( m MTDATA) used as hole transport layer(HTL). The electroluminescent performance of devices with different thicknesses of Alq3∶Li shows that the insertion of the lithium doped Alq3 layer can reduce the turn on voltage by at least 2 volts, and the stability of devices with this lithium doped Alq3 layer is improved too. It can also change the efficiency of devices. Compared with an ultra thin lithium fluoride(Lif) layer, Alq3∶Li sheet gives similar effects but higher efficiency and can be much thicker and hence it is easier to control the deposition.

Predicting the variation of stacking fault energy for binary Cu alloys by first-principles calculations

The variation of stacking fault energy(SfE)in a number of binary Cu alloys is predicted through considering the Suzuki segregation by the full potential linearly augmented plane wave(fPLAPW)*** calculated results show that some solute atoms(Mg,Al,Si,Zn,Ga,Ge,Cd,Sn,and Pb),which prefer to form the Suzuki segregation,may decrease the value of SfE;while the others(Ti,Mn,fe,Ni,Zr,Ag,and Au),which do not cause the Suzuki segregation may not decrease the ***,it is interesting to find that the former alloying elements are located on the right of Cu group while the latter on the left of Cu group in the periodic table of *** intrinsic reasons for the new findings can be traced down to the valences electronic structure of solute and Cu atoms,i.e.,the similarity of valence electronic structure between solute and Cu atoms increases the value of SfE,while the difference decreases the value of SfE.
