Pancreatic cancer is one of the most aggressive cancers with a median survival time of less than 5 months,and conventional chemotherapeutics are the main treatment ***(ADP-ribose)polymerase(PARP)inhibitors have been recently approved for BRCA1/2-mutant pancreatic cancer,opening a new era for targeted therapy for this ***,most pancreatic cancer patients carry wild-type BRCA1/2 with resistance to PARP ***,we reported that mammalian target of rapamycin complex 2(mTORC2)kinase is overexpressed in pancreatic cancer tissues and promotes pancreatic cancer cell growth and ***,we found that knockdown of the mTORC2 obligate subunit Rictor sensitized pancreatic cancer cells to the PARP inhibitor ***,we showed that mTORC2 positively regulates homologous recombination(HR)repair by modulating BRCA1 recruitment to DNA double-strand breaks(DSBs).In addition,we confirmed that combination treatment with the mTORC2 inhibitor PP242 and the PARP inhibitor olaparib synergistically inhibited pancreatic cancer growth in ***,this study provides a novel target and strategy for optimizing PARP inhibitor efficiency in pancreatic cancers.

目的:探讨机器人辅助阴道骶骨固定术治疗重度盆腔器官脱垂的效果及安全性。方法:纳入2021年1月—2023年12月在青岛大学附属医院妇科接受机器人辅助阴道骶骨固定术(RASC)治疗的54例重度盆腔器官脱垂患者为研究对象。32例(59.26%)患者因合并阴道后壁重度脱垂接受改良的机器人辅助下肛提肌筋膜-阴道骶骨固定术,22例(40.74%)患者行常规阴道骶骨固定术,其中有5例患者因合并SUI同时行TVT-O术。记录所有患者的围术期相关资料,收集患者术后1个月、6个月及12个月的随访数据。对比了患者术前、术后盆腔脏器脱垂的治疗效果、安全性及可行性。采用盆底功能障碍问卷(PFDI-20)和盆底障碍影响简易问卷(PFIQ-7)评估患者术后主观满意度,并采用PISQ-12问卷评价其性生活满意度。结果:所有手术均顺利完成,无中转开腹,未见大血管及邻近器官损伤。平均手术时间(186.11±68.91)min,术中平均出血量(40.37±19.62)ml,平均术后住院时间(3.63±1.70)d。所有患者均完成术后12个月随访,最长随访时间22个月。患者术后6个月、12个月POP-Q评分均较术前有明显的改善,无复发,术后客观及主观治愈率均为100%。PFDI-20及PFIQ-7调查结果显示,患者术后6个月及12个月生活质量均得到显著改善(P<0.001);术后12个月有40例患者恢复性生活,所有患者均未述性生活疼痛及不适,PISQ-12评分在手术前后无显著差异。术后阴道残端感染2例(3.7%),持续阴道残端网片暴露1例(1.85%),于术后1年再次缝合后愈合。结论:达芬奇机器人手术系统治疗重度盆腔器官脱垂,尤其是合并重度阴道后壁脱垂及肠疝疗效确切、出血量少、手术时间短,可作为该类疾病的有效治疗方案之一。

Developing platinum-group-metal(PGM)catalysts possessing strong metal-support interaction and controllable PGM size is urgent for the sluggish oxygen reduction reaction(ORR)in proton-exchange membrane fuel ***,we propose an in-situ self-assembled reduction strategy to successfully induce highly-dispersed sub-3nm platinum nanoparticles(Pt NPs)to attach on resin-derived atomic Co coordinated by N-doped carbon substrate(Pt/Co_(SA)-N-C)for *** be specific,the interfacial electron interaction effect,along with a highly robust Co_(SA)-N-C support endow the as-fabricated Pt/Co_(SA)-N-C catalyst with significantly enhanced catalytic properties,i.e.,a mass activity(MA)of 0.719 A/mgPt at 0.9 ViR-free and a reduction of 24.2%in MA after a 20,000-cycles *** functional theory(DFT)calculations demonstrate that the enhanced electron interaction between Pt and Co_(SA)-N-C support decreases the dband center of Pt,which is in favor of lowering the desorption energy of ^(*)OH on Pt/Co_(SA)-N-C surface and accelerating the formation of H_(2)O,thus enhance the instinct activity of ***,the higher binding energy between Pt and Co_(SA)-N-C compared to Pt and C indicates that the migration of Pt has been suppressed,which theoretically explains the improved durability of Pt/Co_(SA)-*** work offers an enlightenment on constructing composite Pt-based catalysts with multiple active sites.

Grassland biomass is an important parameter of grassland *** complexity of the grassland canopy vegetation spectrum makes the long-term assessment of grassland growth a *** studies have explored the original spectral information of typical grasslands in Inner Mongolia and examined the influence of spectral information on aboveground biomass(AGB)*** order to improve the accuracy of vegetation index inversion of grassland AGB,this study combined ground and Unmanned Aerial Vehicle(UAV)remote sensing technology and screened sensitive bands through ground hyperspectral data transformation and correlation *** narrow band vegetation indices were calculated,and ground and airborne hyperspectral inversion models were ***,the accuracy of the model was *** results showed that:(1)The vegetation indices constructed based on the ASD FieldSpec 4 and the UAV were significantly correlated with the dry and fresh weight of AGB.(2)The comparison between measured R^(2) with the prediction R^(2) indicated that the accuracy of the model was the best when using the Soil-Adjusted Vegetation Index(SAVI)as the independent variable in the analysis of AGB(fresh weight/dry weight)and four narrow-band vegetation *** SAVI vegetation index showed better applicability for biomass monitoring in typical grassland areas of Inner Mongolia.(3)The obtained ground and airborne hyperspectral data with the optimal vegetation index suggested that the dry weight of AGB has the best fitting effect with airborne hyperspectral data,where y=17.962e^(4.672x),the fitting R^(2) was 0.542,the prediction R^(2)was 0.424,and RMSE and REE were 57.03 and 0.65,***,established vegetation indices by screening sensitive bands through hyperspectral feature analysis can significantly improve the inversion accuracy of typical grassland biomass in Inner *** with ground monitoring,airborne hyperspectral monitoring better reflec

The Betula genus contains pentacyclic triterpenoid betulin known for its environmental adaptation and medicinal ***,the mechanisms underlying betulin biosynthesis responding to climate change remain *** this study,the role of epigenetic modification(DNA methylation) in betulin biosynthesis was examined and how climatic factors influence ***-genome bisulfite sequencing was performed for greenhouse-grown Chinese white birch(Betula platyphylla Sukaczev) treated with DNA methylation inhibitor zebularine(ZEB) and a natural birch population in Northeast *** treatment significantly affected the CHH methylation level of transposable elements and betulin content in a hormesis dose-dependent *** methylation and expression of bHLH9,a key transcriptional factor controlling betulin biosynthesis,were also consistently affected by ZEB treatment as a hormetic *** the natural population,there was a positive correlation between promoter methylation of bHLH9 and summer precipitation,while winter temperature was negatively *** climate-dependent methylation of bHLH9 regulates the expression of downstream genes involved in betulin *** study highlights the role of environmental signals to induce epigenetic changes that result in betulin production,possibly helping to develop resilient plants to combat ongoing climate change and enhance secondary metabolite production.

Dear Editor,Immune checkpoint therapies manipulating the immune system to eliminate tumor cells have shown remarkable clinical efficacy in treating various ***47,an emerging efficient immune checkpoint,is crucial for cancer cells to evade macrophagemediated phagocytosis by interaction with signal-regulatory proteinα(SIRPα).Antibodies blocking the CD47/SIRPαinteraction have been effective to promote macrophage-mediated phagocytosis in various types of cancer in mice and ***47 is not only highly expressed in tumor cells,but also normal cells,such as red blood cells(RBCs).Thus,during clinical trials involving cancer patients,anti-CD47 antibodies may promote the macrophagesmediated phagocytosis of RBCs,ultimately inducing undesirable anemia side *** contrast,small molecule inhibitors interrupting CD47/SIRPαaxis have shown potential to overcome the anemia,possibly due to their lower immunogenicity and shorter half-life compared to antibodies.1 Hence,developing the novel strategies,especially those without the anemia side effect,to intervene in CD47/SIRPαinteraction will benefit cancer immunotherapy.

The primary objective of this investigation was to scrutinize the prepregnancy conditions and lifestyles of 2046 women residing in Liuzhou City, with the aim of delineating the determinants of delivery methods. Evidently, the study unearthed substantial correlations between prepregnancy body mass index, educational attainment, exposure to passive smoking, medical history, and other variables with the mode of delivery. Furthermore, a predictive nomogram model was formulated to accurately forecast the likelihood of cesarean section. These discernments equip pertinent authorities with the means to institute targeted screening and supportive measures for women contemplating pregnancy based on these identified factors. Moreover, provision of services such as prepregnancy counseling and clinical risk assessments could be instrumental in curbing the incidence of cesarean section.
