The Sulfate Transporter Family in Wheat： Tissue-Specific Gene Expression in Relation to Nutrition

Sulfate uptake and distribution in plants are managed by the differential expression of a family of transporters, developmentally, spatially, and in response to sulfur nutrition. Elucidation of the signaling pathways involved requires a knowledge of the component parts and their interactions. Here, the expression patterns of the full complement of sulfate transporters in wheat, as influenced by development and sulfur nutrition, are described. The 10 wheat sulfate transporters characterized here are compared to the gene families for both rice and Brachypodium, for whom full genome information is available. Expression is reported in young seedlings with a focus on roles in uptake from nutrient solution and differential expression in relation to sulfate deprivation. In addition, patterns of expression in all organs at the grain filling stage are reported and indicate differential responses to nutritional signals of the individual transporters in specific tissues and an overall coordination of uptake, storage, and remobilization to deliver sulfur to the developing grain.

AIM:To support probe-based confocal laser endomi-croscopy (pCLE) diagnosis by designing software for the automated classification of colonic polyps. METHODS:Intravenous fluorescein pCLE imaging of colorectal lesions was performed on patients under-going screening and surveillance colonoscopies, followed by polypectomies. All resected specimens were reviewed by a reference gastrointestinal pathologist blinded to pCLE information. Histopathology was used as the criterion standard for the differentiation between neoplastic and non-neoplastic lesions. The pCLE video sequences, recorded for each polyp, were analyzed off-line by 2 expert endoscopists who were blinded to the endoscopic characteristics and histopathology. These pCLE videos, along with their histopathology diagnosis, were used to train the automated classification software which is a content-based image retrieval technique followed by k-nearest neighbor classification. The performance of the off-line diagnosis of pCLE videos established by the 2 expert endoscopists was compared with that of automated pCLE software classification. All evaluations were performed using leave-one-patient- out cross-validation to avoid bias. RESULTS:Colorectal lesions (135) were imaged in 71 patients. Based on histopathology, 93 of these 135 lesions were neoplastic and 42 were non-neoplastic. The study found no statistical significance for the difference between the performance of automated pCLE software classification (accuracy 89.6%, sensitivity 92.5%, specificity 83.3%, using leave-one-patient-out cross-validation) and the performance of the off-line diagnosis of pCLE videos established by the 2 expert endoscopists (accuracy 89.6%, sensitivity 91.4%, specificity 85.7%). There was very low power (< 6%) to detect the observed differences. Boosting the superconducting spin valve effect in a metallic superconductor/ferromagnet heterostructure

Superconducting spin valves based on the superconductor/ferromagnet （S/F） proximity effect are considered to be a key element in the emerging field of superconducting spintronics. Here, we demonstrate the crucial role of the morphology of the superconducting layer in the operation of a multilayer S/F1/F2 spin valve. We study two types of superconducting spin valve heterostructures, with rough and with smooth superconducting layers, using transmission electron microscopy in combination with transport and magnetic characterization. We find that the quality of the S/F interface is not critical for the S/F proximity effect, as regards the suppression of the critical temperature of the S layer. However, it appears to be of paramount importance in the performance of the S/F1/F2 spin valve. As the morphology of the S layer changes from the form of overlapping islands to a smooth case, the magnitude of the conventional superconducting spin valve effect significantly increases. We attribute this dramatic effect to a homogenization of the Green function of the superconducting condensate over the S/F interface in the S/F1/F2 valve with a smooth surface of the S laver.

Densification of Lithium Disilicate under High Pressure Investigated by XPS

The aim of this work was to investigate by X-ray photoelectron spectroscopy the effect of high pressure on the chemical environments of Si 2p, O 1s and Li 1s in lithium disilicate glass ceramic with stoichiometric composition Li2O·2SiO2 (LS2). A group of samples was processed at 2.5 GPa, 4 GPa and 7.7 GPa at room temperature and a second group was crystallized under high pressure and high temperature. Large shifts of the binding energy toward higher energies were observed in the X-ray photoelectron spectroscopy spectra for samples of the first group after densification at 2.5 and 4 GPa. For samples processed at 7.7 Gpa, the major component of the binding energy for the Si 2p environment remained practically unchanged compared to the pristine sample but new components, with smaller intensities, appeared in the spectra, indicating the existence of distinct Q-species induced by high pressure. This behavior may be related to changes in the number of bridged and non-bridged oxygen atoms in the glass structure. The results for the second group of samples, crystallized under high pressure, showed evidences of three binding energies for the O atoms, one of them related to non-bridged and two of them to bridged O atoms.

Standardized high-throughput biomonitoring using DNA metabarcoding: Strategies for the adoption of automated liquid handlers

Reliable and comprehensive monitoring data are required to trace and counteract biodiversity loss. Highthroughput metabarcoding using DNA extracted from community samples (bulk) or from water orsediment (environmental DNA) has revolutionized biomonitoring, given the capability to assess biodiversity across the tree of life rapidly with feasible effort and at a modest price. DNA metabarcoding canbe upscaled to process hundreds of samples in parallel. However, while automated high-throughputanalysis workflows are well-established in the medical sector, manual sample processing still predominates in biomonitoring laboratory workflows limiting the upscaling and standardization for routinemonitoring applications. Here we present an automated, scalable, and reproducible metabarcodingworkflow to extract DNA from bulk samples, perform PCR and library preparation on a liquid *** features are the independent sample replication throughout the workflow and the use of manynegative controls for quality assurance and quality control. We generated two datasets: i) a validationdataset consisting of 42 individual arthropod specimens of different species, and ii) a routine monitoringdataset consisting of 60 stream macroinvertebrate bulk samples. As a marker, we used the mitochondrialCOI gene. Our results show that the developed single-deck workflow is free of laboratory-derivedcontamination and produces highly consistent results. Minor deviations between replicates are mostlydue to stochastic differences for low abundant OTUs. Thus, we successfully demonstrated that roboticliquid handling can be used reliably from DNA extraction to final library preparation on a single deck,thereby substantially increasing throughput, reducing costs, and increasing data robustness for biodiversity assessments and monitoring.

目的：对比研究泌尿系肿瘤患者根治术前后骨髓中散在的微转移肿瘤细胞。方法：术前及术后采集肾癌、前列腺癌和尿道膀胱癌共102例患者的骨髓标本，应用细胞角蛋白的免疫细胞化学方法及自动显微图像对比检测分析。结果：根治术前后骨髓中微转移肿瘤细胞检出率密切相关（P＜0．005），术前肿瘤细胞总检出率为31．4％，术后为34．3％，无显著性差别（P＝0．65）；三种泌尿肿瘤患者根治术前后检出率比较亦均无明显差别（P＝0．70P＝0．10P＝0．59） 但术后检出的肿瘤细胞数多于术前者总数分别为64和44，3个以上阳性细胞仅在术后出现，两次阳性的细胞数均值分别为1．31±0．48和1．95±1．02（P＝0．009 9）12例患者手术8天后检出细胞数（22）明显多于术前者（7），P＝0．01。结论：免疫细胞化学方法可作为检测骨髓中隐蔽的微转移肿瘤细胞的敏感标准方法，根治术前后检出率虽无明显改变 但术后及手术8天以后检出细胞数明显多于术前者，提示手术可能会促使肿瘤细胞在骨髓中的播散。
