Aberrant epithelial cell interaction promotes esophageal squamous-cell carcinoma development and progression

Epithelial-mesenchymal transition(EMT)and proliferation play important roles in epithelial cancer formation and progression,but what molecules and how they trigger EMT is largely unknown.Here we performed spatial transcriptomic and functional analyses on samples of multistage esophageal squamous-cell carcinoma(ESCC)from mice and humans to decipher these critical issues.By investigating spatiotemporal gene expression patterns and cell–cell interactions,we demonstrated that the aberrant epithelial cell interaction via EFNB1-EPHB4 triggers EMT and cell cycle mediated by downstream SRC/ERK/AKT signaling.The aberrant epithelial cell interaction occurs within the basal layer at early precancerous lesions,which expands to the whole epithelial layer and strengthens along the cancer development and progression.Functional analysis revealed that the aberrant EFNB1-EPHB4 interaction is caused by overexpressedΔNP63 due to TP53 mutation,the culprit in human ESCC tumorigenesis.Our results shed new light on the role of TP53-TP63/ΔNP63-EFNB1-EPHB4 axis in EMT and cell proliferation in epithelial cancer formation.

Engineering transition metal compounds(TMCs)catalysts with excellent adsorption-catalytic ability has been one of the most effec-tive strategies to accelerate the redox kinetics of sulfur cathodes.Herein,this review focuses on engineering TMCs catalysts by cation doping/anion doping/dual doping,bimetallic/bi-anionic TMCs,and TMCs-based heterostructure composites.It is obvious that introducing cations/anions to TMCs or constructing heterostructure can boost adsorption-catalytic capacity by regulating the electronic structure including energy band,d/p-band center,electron filling,and valence state.Moreover,the elec-tronic structure of doped/dual-ionic TMCs are adjusted by inducing ions with different electronegativity,electron filling,and ion radius,resulting in electron redistribution,bonds reconstruction,induced vacancies due to the electronic interaction and changed crystal structure such as lat-tice spacing and lattice distortion.Different from the aforementioned two strategies,heterostructures are constructed by two types of TMCs with different Fermi energy levels,which causes built-in electric field and electrons transfer through the interface,and induces electron redistribution and arranged local atoms to regulate the electronic structure.Additionally,the lacking studies of the three strategies to comprehensively regulate electronic structure for improving catalytic performance are pointed out.It is believed that this review can guide the design of advanced TMCs catalysts for boosting redox of lithium sulfur batteries.

Gut microbiota-mediated modulation of lipid metabolism by punicic acid in diet-induced hepatic steatosis

Pomegranate seed oil (PSO) mainly consisted of punicic acid (PunA)， has gained attention due to rich conjugated linolenic acids and potential application in the improvement of various diseases. The aim of this study was to explore the ability of PunA to modulate lipid metabolism associated with non-alcoholic fatty liver disease (NAFLD) and explore the emerging role of the gut microbiota in modulating hepatic lipid metabolism. In this regard， PunA’s capacity was evaluated through C57BL/6J fed with a choline and L-amino acid defined， high fat diet (CDAHFD). Mice were randomly divided into four groups: NC group， MC group， PSO group and Cur group. Collected results indicated that supplement of PunA improved hepatic lipid profile. PunA administration also ameliorated hepatic steatosis and lipid accumulation. mRNA analysis showed that PunA exerted ameliorative e?ect on hepatic lipid accumulation by downregulating the mRNA expression of Dgat1， Scd-1， Srebp-1c， but increasing expression level of Cpt-1α. Metagenomic analysis discovered that PunA increased species diversity and richness of intestinal microbiota， promoted proliferation of beneficial commensals and restraining the endotoxin-producing microorganisms in cecum of CDAHFD-administrated mice. Furthermore， PunA suppressed lipid accumulation and lipid droplet formation in a cell NAFLD model in vitro. Overall， PunA exhibited anti-lipogenic and lipid-lowering effects， improvement of intestinal flora. In summary， PunA could be a candidate for the exploitation of functional foods or nutraceuticals aiming NAFLD.

Natural variation of an autophagy-family gene among rice subspecies affects grain size and weight

Elucidating the genetic basis of natural variation in grain size and weight among rice varieties can help breeders develop high-yielding varieties.We identified a novel gene,GW3a(Grain Weight 3a)(LOC_Os03g27350),that affects rice grain size and weight.gw3a mutants showed higher total starch content and dry matter accumulation than the wild type(WT),Nipponbare,suggesting that GW3a negatively regulates grain size and weight.Moreover,our study found that GW3a interacted with OsATG8 by cleaving it,suggesting that GW3a may be involved in the assembly of autophagosomes and starch degradation in plants.The haplotype analysis of GW3a showed functional differences between indica and japonica rice.Taken together,we conclude that GW3a is expressed in the autophagosome pathway regulating starch metabolism in rice,affecting yield-related traits,such as grain size,grain weight and thousand grain weight(TGW).Our findings also shed new light on autophagy-mediated yield trait regulation,proposing a possible strategy for the genetic improvement of high-yield germplasm in rice.

Background:Colorectal cancer(CRC)still ranks the top in morbidity and mortality of cancers worldwide,posing a huge threat and burden to the society.We aimed to determine the age-standardized incidence,mortality,and disability-adjusted life years(DALYs)of CRC and explore potential changes in the temporal trends of the CRC burden in Shanghai during 2002 to 2016.Methods:The cancer statistics and demographics were obtained from the Cancer Registry and the Statistics Bureau of Pudong New Area,respectively.Data from 2002 to 2016 were included and analyzed retrospectively.DALYs were calculated using DisMod and the age-standardized rates(ASRs)were obtained according to Segi world standard population.Joinpoint regression was used to measure the trends in CRC incidence and to estimate the annual percent change.Results:The increasing trend of CRC ASR incidence halted after 2014,coinciding with the introduction of the Shanghai CRC screening program.The ASRs of mortality and DALYs increased,at 0.42%(P74 years.The benefits of the screening program have been partially proven by the ASRs of CRC incidence,providing important insights into better and wider application of screening programs.

A Triple Human-Digital Twin Architecture for Cyber-Physical Systems

With the development of information and communication technology and the advent of the Internet of Things(IoT)era,cyber-physical system(CPS)is becoming the trend of products or systems.The deep integration and real-time interaction between the physical world and the virtual world expand system functions.Although there are some CPS implementation guidelines,the virtual world is still relatively abstract compared to the concrete physical world that can be touched through the IoT.Besides that,human is a non-negligible CPS endogenous interactive intelligent component.In this paper,we propose a triple human-digital twin architecture,where the physical objects and the digital twins that are the projections of the physical entities establish the cornerstone of human functioning together.And the hierarchically distributed digital twins grow dynamically with the physical entities along the lifecycle.Furthermore,the interaction and collaboration among the physical objects,the digital twins,and the humans in their respective worlds(the expected world,the interpreted world,and the physical world)integrate the full value chain of the products in anticipation of seamless synergy.Finally,we present a power management digital companion platform for the lunar probe to demonstrate the efficacy of the architecture.

UV-induced senescence of human dermal fibroblasts restrained by low-stiffness matrix by inhibiting NF-κB activation

As a hallmark of skin aging,senescent human dermal fibroblasts(HDFs)are known to lose the ability to divide.However,they can still interact with their cellular environment and the surrounding matrix.As the skin ages,the progressive slowing down of HDFs function decreases the skin’s structural integrity,which is more serious than if there is the dermal collagen matrix eroded.This leads to matters of the unbalanced barrier under the skin,skin fragility,inadequate wound healing,as well as other cosmetic issues.It is also well documented that skin aging comes with significant stiffness increases.Therefore,understanding the interactions between HDFs and the surrounding microenvironments during senescence may provide insights into skin aging.Here we aim to inves-tigate matrix stiffness’effect on HDF senescence and elucidate possible mechanisms that make HDFs senescent.In our experiments,HDFs were cultivated on Polydimethylsiloxane(PDMS)with various stiffnesses and exposed to UV light to trigger senescence.Results show that HDFs are significantly affected by senescence when cultured on a matrix with stiffness.However,the cells are not significantly affected when cultured on a low stiffness matrix.The following characterization revealed cells cultured on stiffsubstrates under UV exposure had stimu-lated the nucleus factor kappa-B(NF-κB)activation.In contrast,cells on a matrix of softness only displayed low activation of NF-κB.NF-κB activity suppression with ammonium pyrrolidine dithiocarbamate(PDTC)decreases UV-induced HDFs senescence on stiffsubstrates.Taken together,we demonstrated that soft matrix defends HDFs against ultraviolet-induced senescence by inhibiting the activation of NF-κB.
