Microfluidic reactors for advancing the MS analysis of fast biological responses

The response of cells to physical or chemical stimuli is complex,unfolding on time-scales from seconds to days,with or without de novo protein synthesis,and involving signaling processes that are transient or *** combining the technology of microfluidics that supports fast and precise execution of a variety of cell handling operations,with that of mass spectrometry detection that facilitates an accurate and complex characterization of the protein complement of cells,in this work,we developed a platform that supports(near)real-time sampling and proteomelevel capturing of cellular responses to a perturbation such as treatment with *** geometric design of the chip supports three critical features:(a)capture of a sufficient number of cells to meet the detection limit requirements of mass spectrometry instrumentation,(b)fluid delivery for uniform stimulation of the resident cells,and(c)fast cell recovery,lysis and processing for accurate sampling of time-sensitive cellular responses to a *** simulations and microscopy were used to predict and evaluate the flow behavior inside the microfluidic *** analysis of the cellular extracts generated by the chip experiments revealed that the identified proteins were representative of all cellular locations,exosomes,and major biological processes related to proliferation and signaling,demonstrating that the device holds promising potential for integration into complex lab-on-chip workflows that address systems biology *** applicability of the chips to study time-sensitive cellular responses is discussed in terms of technological challenges and biological relevance.

Space systems have become a key enabler for a wide variety of applications that are vital to the functioning of advanced societies. The trend is one of quantitative and qualitative increase of this dependence, so much so that space systems have been described as a new example of critical infrastructure. This article argues that the existence of critical space infrastructures implies the emergence of a new category of disasters related to disruption risks. We inventory those risks and make policy recommendations for what is, ultimately, a resilience governance issue.

Photonic crystals(PCs)exhibit promising structural coloration properties and possess extensive application prospects in diverse optical ***,state-of-the-art inorganic or polymeric PCs show limited adaptivity as their configurations are fixed once ***,bio-organic adaptive PCs are fabricated via drop-casting of amphiphilic guanine-based peptide nucleic acid selfassembled *** high formation activation energy of up to 81.8 kJ·mol−1 suggests that the self-assembly step dominates the entire ***,the configurations along with the structural coloration of the supramolecular PCs are sensitive to self-assembly influencing parameters,showing temperature-encoded structural color evolution and solvent polaritydependent *** findings demonstrate that the supramolecular PCs are adaptive,thus showing promising potential for detection of organic solvents of different polarities in a visual and real-time manner for environmental protection or optical applications.

Development of the sexually dimorphic nucleus of the preoptic area and the influence of estrogen-like compounds

One of the well-defined sexually dimorphic structures in the brain is the sexually dimorphic nucleus, a cluster of cells located in the preoptic area of the hypothalamus. The rodent sexually dimorphic nucleus of the preoptic area can be delineated histologically using conventional Nissl staining or immunohistochemically using calbindin D28K immunoreactivity. There is increasing use of the cal- bindin D28K-delineated neural cluster to define the sexually dimorphic nucleus of the preoptic area in rodents. Several mechanisms are proposed to underlie the processes that contribute to the sexual dimorphism （size difference） of the sexually dimorphic nucleus of the preoptic area. Recent evidence indicates that stem cell activity, including proliferation and migration presumably from the 3^rd ventricle stem cell niche, may play a critical role in the postnatal development of the sexually dimorphic nucleus of the preoptic area and its distinguishing sexually dimorphic feature： a signifi-cantly larger volume in males. Sex hormones and estrogen-like compounds can affect the size of the sexually dimorphic nucleus of the preoptic area. Despite considerable research, it remains un-clear whether estrogen-like compounds and/or sex hormones increase size of the sexually dimor-phic nucleus of the preoptic area via an increase in stem cell activity originating from the 3^rd ventricle stern cell niche.

Despite a decrease in incidence over past decades,gastric cancer remains a major global health problem. In the more recent period, survival has shown only minor improvement, despite significant advances in diagnostic techniques, surgical and chemotherapeutic approaches, the development of novel therapeutic agents and treatment by multidisciplinary teams. Because multiple genetic mutations, epigenetic alterations, and aberrant molecular signalling pathways are involved in the development of gastric cancers, recent research has attempted to determine the molecular heterogeneity responsible for the processes of carcinogenesis, spread and metastasis. Currently, some novel agents targeting a part of these dysfunctional molecular signalling pathways have already been integrated into the standard treatment of gastric cancer, whereas others remain in phases of investigation within clinical trials. It is essential to identify the unique molecular patterns of tumours and specific biomarkers to develop treatments targeted to the individual tumour behaviour. This review analyses the global impact of gastric cancer, as well as the role of Helicobacter pylori infection and the efficacy of bacterial eradication in preventing gastric cancer development. Furthermore, the paper discusses the currently available targeted treatments and future directions of research using promising novel classes of molecular agents for advanced tumours.

Hydrogen bonds are non-covalent interactions and essential for assembling supermolecules into ordered structures in biological systems,endowing crystals with fascinating physical properties,and inspiring the construction of eco-friendly electromechanical ***,the interplay between hydrogen bonding and the physical properties is not fully understood at the molecular ***,we demonstrate that the physical property of biological crystals with double-layer structures could be enhanced by rationally controlling hydrogen bonding interactions between amino and carboxyl *** hydrogen bonding interactions result in various thermal,mechanical,electronic,and piezoelectric *** particular,the weak interaction between O and H atoms contributes to low mechanical strength that permits important ion displacement under stress,giving rise to a strong piezoelectric *** study not only reveals the correlation between the hydrogen bonding and physical properties in double-layer structures of biological crystals but also demonstrates the potential of these crystals as functional biomaterials for high-performance energy-harvesting *** calculations and experimental verifications in this work provide new insights into the rational design of biomaterials with desirable physical properties for bioelectrical devices by modulating intermolecular interactions.

Stem cell-derived exosomes-an emerging tool for myocardial regeneration

Cardiovascular diseases(CVDs) continue to represent the number one cause of death and disability in industrialized countries. The most severe form of CVD is acute myocardial infarction(AMI), a devastating disease associated with high mortality and disability. In a substantial proportion of patients who survive AMI, loss of functional cardiomyocytes as a result of ischaemic injury leads to ventricular failure, resulting in significant alteration to quality of life and increased mortality. Therefore, many attempts have been made in recent years to identify new tools for the regeneration of functional cardiomyocytes. Regenerative therapy currently represents the ultimate goal for restoring the function of damaged myocardium by stimulating the regeneration of the infarcted tissue or by providing cellsthat can generate new myocardial tissue to replace the damaged tissue. Stem cells(SCs) have been proposed as a viable therapy option in these cases. However, despite the great enthusiasm at the beginning of the SC era, justified by promising initial results, this therapy has failed to demonstrate a significant benefit in large clinical trials. One interesting finding of SC studies is that exosomes released by mesenchymal SCs(MSCs) are able to enhance the viability of cardiomyocytes after ischaemia/reperfusion injury, suggesting that the beneficial effects of MSCs in the recovery of functional myocardium could be related to their capacity to secrete exosomes. Ten years ago, it was discovered that exosomes have the unique property of transferring miRNA between cells, acting as miRNA nanocarriers. Therefore, exosomebased therapy has recently been proposed as an emerging tool for cardiac regeneration as an alternative to SC therapy in the post-infarction period. This review aims to discuss the emerging role of exosomes in developing innovative therapies for cardiac regeneration as well as their potential role as candidate biomarkers or for developing new diagnostic tools.

This article analyses the literature regarding the value of computer-assisted systems in esogastroduodenoscopy-quality monitoring and the assessment of gastric *** data show promising results in upper-endoscopy quality control and a satisfactory detection accuracy of gastric premalignant and malignant lesions,similar or even exceeding that of experienced ***,artificial systems enable the decision for the best treatment strategies in gastriccancer patient care,namely endoscopic vs surgical resection according to tumor *** so doing,unnecessary surgical interventions would be avoided whilst providing a better quality of life and prognosis for these *** these performance data have been revealed by numerous studies using different artificial intelligence(AI)algorithms in addition to white-light endoscopy or novel endoscopic techniques that are available in expert endoscopy *** is expected that ongoing clinical trials involving AI and the embedding of computer-assisted diagnosis systems into endoscopic devices will enable real-life implementation of AI endoscopic systems in the near future and at the same time will help to overcome the current limits of the computer-assisted systems leading to an improvement in *** benefits should lead to better diagnostic and treatment strategies for gastric-cancer ***,the incorporation of AI algorithms in endoscopic tools along with the development of large electronic databases containing endoscopic images might help in upper-endoscopy assistance and could be used for telemedicine purposes and second opinion for difficult cases.
