Identification of key genes and metabolites involved in meat quality performance in Qinchuan cattle by WGCNA

Understanding the genetic and metabolic elements that impact meat quality is crucial to improving production and meeting consumer demands in the beef *** in meat quality among various muscle areas in beef cattle can impact pricing in the *** progress in genomics,the specific genes and metabolites that affect meat quality characteristics in Qinchuan cattle remain inadequately ***,this study aims to evaluate the meat quality characteristics of four specific muscle locations(tenderloin,striploin,high rib,and ribeye muscles)in Qinchuan bulls,including 10 traits(total protein content(TPC),intramuscular fat(IMF),non-esterified fatty acid(NEFA),meat color(L*,a*,and b*),shear force(SF),cooking loss(CL),pH0,and pH24).This experiment uses transcriptome,metabolome sequencing,and sophisticated analytical methodologies such as weighted gene co-expression network analysis(WGCNA)and protein–protein interaction networks(PPI)to identify the key genes and metabolites associated with specific *** findings highlight three notable genes(NDUFAB1,NDUFA12,and NDUFB7)linked to intramuscular fat(IMF),three key genes(CSRP3,ACAA3,and ACADVL)correlated with non-esterified fatty acids(NEFA),and one crucial gene(CREBBP)influencing meat *** conclusion,this investigation offers a new perspective on the differences in bovine muscle locations and contributes to the molecular understanding of bovine meat *** research endeavors could delve deeper into the identified genes and pathways to enhance beef cattle’s quality and yield.

Prognostic Role and Potential Mechanisms of N6-methyladenosine-related Long Noncoding RNAs in Hepatocellular Carcinoma

Background and Aims:Numerous studies have explored the important role of N6-methyladenosine(m^(6)A)in ***,the interaction between m^(6)A and long noncoding RNAs(lncRNAs)is poorly ***,we systematically analyzed the role and prognostic value of m^(6)A-related lncRNAs in hepatocellular carcinoma(HCC).Methods:The m^(6)A-related lncRNAs were identified based on the correlation coefficients with m^(6)A-related genes in HCC from The Cancer Genome ***,a novel risk score model was determined using the least absolute shrinkage and selection operator Cox regression *** and multivariate Cox analyses were used to identify independent prog-nostic factors for overall survival(OS)of HCC;thereafter,a prognostic nomogram was ***:A total of 259 lncRNAs showed significant correlations with m^(6)A in HCC,while 29 lncRNAs had prognostic ***,six critical m^(6)A-related lncRNAs(NRAV,SNHG3,KDM4A-AS1,AC074117.1,AC025176.1,and AL031985.3)were screened out to construct a novel risk score model which classified HCC patients into high-and low-risk *** analy-ses revealed that patients in the high-risk group exhibited worse OS,both in the training and validation *** risk score was also identified as an independent prognostic factor of OS,and a nomogram was established and verified with superior prediction ***,the risk score signifi-cantly correlated with the expression of immune checkpoint genes and immune ***:These findings indicated the significant role of m^(6)A-related lncRNAs in HCC and the potential application of the novel risk score model for prognostic prediction.

Raman Micro-Imaging of the Coexistence of sⅠ and sⅡ Hydrates Formed from a Mixed Methane-Propane Gas in a Confined Space

Natural gas hydrate contains a specific amount of heavy hydrocarbons, such as ethane, propane, etc., aside from the primary guest gas of methane. Although the coexistence of two or even three hydrate structures has been discovered at several hydrate sites, the requisite formation mechanism is still not well understood. In-situ observation of the formation process of mixed methane-propane hydrate in a confined space was conducted using confocal Raman imaging microscopy. The Raman imaging results reveal that sI methane hydrate and sII mixed methane-propane hydrate are formed and coexist in the reaction system. In the confined space, the sI hydrate originates from the dissolved gas in water, while the sII hydrate is formed from free gas. The results obtained can help explain the coexistence of sI and sII hydrates found in natural hydrate samples, as well as providing insights into a possible dynamic scenario of hydrate reservoirs in geological history.

Fatigue crack initiation for Al-Zn-Mg alloy welded joint

To investigate fatigue crack initiation characteristics of A1-Zn-Mg alloy welded joint, notched specimens were used in fatigue test for the base metal, welding bead and heat affected zone （HAZ）. The fatigue fracture surface near the fatigue crack initiation site was observed by scanning electron microscope （SEM）. The results show that the differences of fatigue crack initiation life among base metal, welding bead and HAZ are not obvious. Inhomogeneity in microstructure and mechanical performance of HAZ influences the fatigue crack initiation life. The ratio of fatigue crack initiation life （Ni） to fatigue failure life （Nf） for the base metal, welding bead and HAZ of A7N01 aluminium alloy welded joint are 26.32%, 40.21% and 60.67%, respectively. Fatigue crack initiation life can be predicted using a uniform model. Observation of fatigue fracture surfaces shows that for the welding bead a fatigue crack initiates from the smooth surface due to the welding process, the blowhole in HAZ causes fatigue crack and the crushed second phase particles play an important role in fatigue crack initiation for the base metal.

Adaptive Fuzzy Sliding Mode Controller with Nonlinear Observer for Redundant Manipulators Handling Varying External Force

An Adaptive Fuzzy Sliding Mode Controller with Nonlinear Observer （AFSMCO） for the redundant robotic manipulator is proposed in this paper. This approach could achieve a precise trajectory tracking for a robot manipulator in the presence of uncertainties and disturbances. Primarily, a suitable observer using the recursive algorithm is presented for an accurate estima- tion of external disturbances caused by the varying external force. Secondly, the adaptive fuzzy logic is designed to approximate the parameters of the sliding mode controller （AFSMC） to avoid chattering in real time. Moreover, it is shown using the Lyapunov theory that the tracking error asymptotically converges to zero. Finally, the effectiveness of the proposed control approach and theoretical discussion are demonstrated by simulation results on a 7-1ink robot and tested on a 7-DOF manipulator platform.
