Dear Editor,Electric vehicle(EV)sales have significantly grown over the years to fulfill growing demands for economic travel and greenhouse gas mitigation.1 However,the surge in the number of EVs has led to charging anxiety as users struggle to find an available charging station before running out of electricity,resulting in longer reserve and waiting times.2 Moreover,severe mobility restrictions caused by infectious diseases,such as coronavirus disease 2019(COVID-19),have greatly affected people’s travel behavior3,4 and hindered their willingness to use EVs,given that charging in public spaces consumes time and increases the risk of contracting the virus.5 This implies that in the postpandemic era,in which individuals coexist with the virus,the interplay between the two important trends,namely vehicle electrification and mobility restrictions,can extensively affect people’s daily commuting by using EVs.

Background:Liver cancer is largely resistant to chemotherapy.This study aimed to identify the effective chemotherapeutics forβ-catenin-activated liver cancer which is caused by gain-of-function mutation of catenin beta 1(CTNNB1),the most frequently altered proto-oncogene in hepatic neoplasms.Methods:Constitutiveβ-catenin-activated mouse embryonic fibroblasts(MEFs)were established by deleting exon 3(β-catenin^(Δ(ex3)/+)),the most common mutation site in CTNNB1 gene.A screening of 12 widely used chemotherapy drugs was conducted for the ones that selectively inhibitedβ-catenin^(Δ(ex3)/+)but not for wild-type MEFs.Untargeted metabolomics was carried out to examine the alterations of metabolites in nucleotide synthesis.The efficacy and selectivity of methotrexate(MTX)onβ-catenin-activated human liver cancer cells were determined in vitro.Immuno-deficient nude mice subcutaneously inoculated withβ-catenin wild-type or mutant liver cancer cells and hepatitis B virus(HBV);β-catenin^(lox(ex3)/+)mice were used,respectively,to evaluate the efficacy of MTX in the treatment ofβ-catenin mutant liver cancer.Results:MTX was identified and validated as a preferential agent against the proliferation and tumor formation ofβ-catenin-activated cells.Boosted nucleotide synthesis was the major metabolic aberration inβ-catenin-active cells,and this alteration was also the target of MTX.Moreover,MTX abrogated hepatocarcinogenesis of HBV;β-catenin^(lox(ex3)/+)mice,which stimulated concurrent Ctnnb1-activated mutation and HBV infection in liver cancer.Conclusion:MTX is a promising chemotherapeutic agent forβ-catenin hyperactive liver cancer.Since repurposing MTX has the advantages of lower risk,shorter timelines,and less investment in drug discovery and development,a clinical trial is warranted to test its efficacy in the treatment ofβ-catenin mutant liver cancer.

An Extreme Gale Event in East China under the Arctic Potential Vorticity Anomaly through the Northeast China Cold Vortex

Arctic changes influence not only temperature and precipitation in the midlatitudes but also contribute to severe convection.This study investigates an extreme gale event that occurred on 30 April 2021 in East China and was forced by an Arctic potential vorticity(PV)anomaly intrusion.Temperature advection steered by storms contributed to the equatorward propagation of Arctic high PV,forming the Northeast China cold vortex(NCCV).At the upper levels,a PV southward intrusion guided the combination of the polar jet and the subtropical jet,providing strong vertical wind shear and downward momentum transportation to the event.The PV anomaly cooled the upper troposphere and the northern part of East China,whereas the lower levels over southern East China were dominated by local warm air,thus establishing strong instability and baroclinicity.In addition,the entrainment of Arctic dry air strengthened the surface pressure gradient by evaporation cooling.Capturing the above mechanism has the potential to improve convective weather forecasts under climate change.This study suggests that the more frequent NCCV-induced gale events in recent years are partly due to high-latitude waviness and storm activities,and this hypothesis needs to be investigated using more cases.

Micelle-directed self-assembly of single-crystal-like mesoporous stoichiometric oxides for high-performance lithium storage

Due to their uncontrollable assembly and crystallization process,the synthesis of mesoporous metal oxide single crystals remains a formidable challenge.Herein,we report the synthesis of single-crystal-like mesoporous Li2TiSiO5by using soft micelles as templates.The key lies in the atomic-scale self-assembly and step-crystallization processes,which ensure the formation of single-crystal-like mesoporous Li2TiSiO5microparticles via an oriented attachment growth mechanism under the confinement of an in-situ formed carbon matrix.The mesoporous Li2TiSiO5anode achieves a superior rate capability(148 mAh g-1at5.0 A g-1) and outstanding long-term cycling stability(138 mAh g-1after 3000 cycles at 2.0 A g-1) for lithium storage as a result of the ultrafast Li+diffusion caused by penetrating mesochannels and nanosized crystal frameworks（5-10 nm）.In comparison,bulk Li2TiSiO5exhibits poor rate capability and cycle performance due to micron-scale diffusion lengths.This method is very simple and reproducible,heralding a new way of designing and synthesizing mesoporous single crystals with controllable frameworks and chemical functionalities.

To the Editor:Cardiac amyloidosis(CA)is an infiltrative cardiomyopathy characterized by extracellular deposition of misfolded insoluble amyloid fibrils.^([1])Electrocardiography(ECG)is an essential diagnostic test characterized by low QRS voltages in CA,observed in 20–74%of patients.^([2])Cipriani et al^([3])reported that low QRS voltages could independently predict cardiovascular death.But the mechanisms underlying low QRS voltages in CA remain unclear.

Confinement fluorescence effect of an aggregation-induced emission luminogen in crystalline polymer

Despite the impressive progress of stimuli-responsive fluorescent materials,little is known about the influence of confinement created by crystalline polymer over the fluorescence properties of fluorescent molecules.The effects of confinement on the fluorescence of an aggregation-induced emission luminogen(AIEgen)are investigated using computational simulations,which reveal that the confined space induces the AIEgens to take a more planar conformation,resulting in a red-shifted emission spectrum.With this property,the study is extended to explore the confinement generated by various polymer crystalline forms,and it is shown that different fluorescence colors are activated.This confinement fluorescence effect is attributed to the different spatial dimensions of the polymer amorphous layer between lamellar crystals where the AIEgens are located.These results indicate the immediate association between crystalline structure and fluorescence signals,activating unprecedented photophysical properties of luminescent materials,and also providing the possibility for crystalline structure visualization,it is important for the many polymer crystallization processes occurring in the materials processing.

Carbon-free hydrogen as a promising clean energy source can be produced with electrocatalysts via water electrolysis.Oxygen evolution reaction(OER)as anodic reaction determines the overall efficiency of water electrolysis due to sluggish OER kinetics.Thus,it’s much desirable to explore the efficient and earth-abundant transition-metal-based OER electrocatalysts with high current density and superior stability for industrial alkaline electrolyzers.Herein,we demonstrate a significant enhancement of OER kinetics with the hybrid electrocatalyst arrays in alkaline via judiciously combining earth-abundant and ultrathin NiCo-based layered double hydroxide(NiCo LDH)nanosheets with nickel cobalt sulfides(NiCoS)with a facile metal-organic framework(MOF)-template-involved surface sulfidation process.The obtained NiCo LDH/NiCoS hybrid arrays exhibits an extremely low OER overpotential of 308 mV at 100 mA·cm^(−2),378 mV at 200 mA·cm^(−2)and 472 mV at 400 mA·cm^(−2)in 1 M KOH solution,respectively.A much low Tafel slope of 48 mV·dec^(−1)can be achieved.Meanwhile,with the current density from 50 to 250 mA·cm^(−2),the NiCo-LDH/NiCoS hybrid arrays can run for 25 h without any degradation.Our results demonstrate that the construction of hybrid arrays with abundant interfaces of NiCo LDH/NiCoS can facilitate OER kinetics via possible modulation of binding energy of O-containing intermediates in alkaline media.The present work would pave the way for the development of lowcost and efficient OER catalysts and industrial application of water alkaline electrolyzers.

目的:探讨补阳还五汤是否通过调控核转录因子E2相关因子2(Nrf2)/血红素加氧酶-1(HO-1)通路抑制糖尿病肾病(DKD)小鼠氧化应激,从而发挥减轻肾组织损伤,延缓肾脏纤维化的作用。方法:将73只C57BL/6小鼠随机分为造模组63只和正常对照组10只,造模组小鼠高糖高脂饲料喂养6 w后,连续5 d以50 mg/kg腹腔注射链脲佐菌素(STZ)制备糖尿病模型后继续高糖高脂饲料喂养8 w,当小鼠出现尿蛋白阳性则DKD模型制备成功。随机将DKD小鼠分为模型对照组、罗格列酮7.05×10^(-4)g/kg组、补阳还五汤3.21、6.41、12.82 g/kg组,各组灌胃相应药物或生理盐水,1次/d,连续8 w。药物干预期间监测小鼠体质量、血糖、尿量、24 h尿蛋白定量(24 h-UTP),给药结束后采用马松(Masson)染色及高碘酸希夫(PAS)染色观察小鼠肾组织病理学改变;测定超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活力;荧光探针标记法检测小鼠肾组织活性氧簇(ROS)水平;蛋白免疫印迹法(Western blot)检测小鼠肾组织转化生长因子β1(TGF-β1)、α-平滑肌肌动蛋白(α-SMA)、纤连蛋白(FN)、Ⅰ型胶原(CollagenⅠ)、Nrf2、HO-1蛋白表达。结果:与正常对照组比较,模型对照组小鼠体质量下降,血糖升高,尿量、24 h-UTP增加,肾小球囊壁增厚,有明显的胶原纤维增生,肾小球内系膜增生,肾小球周围可见糖原颗粒沉积,SOD、CAT活力降低,ROS水平增加,Nrf2、HO-1蛋白表达下调,TGF-β1、α-SMA、FN、CollagenⅠ蛋白表达上调(P<0.05或P<0.01);与模型对照组比较,补阳还五汤各组及罗格列酮7.05×10^(-4)g/kg组小鼠体质量增加,尿量、24 h-UTP降低,肾组织病理学损伤可见不同程度改善,SOD、CAT活力明显增加,ROS水平明显降低,Nrf2、HO-1表达明显上调,TGF-β1、α-SMA、FN、CollagenⅠ表达明显下调(P<0.05或P<0.01)。结论:补阳还五汤可通过激活Nrf2/HO-1通路抑制氧化应激,缓解DKD小鼠肾纤维化,减轻小鼠病理损伤。

目的建立超高效液相色谱-串联质谱法同时测定活血化瘀类中成药中非法添加抗心绞痛、抗凝、抗血小板聚集以及抗炎镇痛等30个化学药物的方法。方法样品采用60%甲醇超声提取,以0.1%甲酸溶液-乙腈为流动相进行梯度洗脱,经色谱柱Phenomenex Kinetex C_(18)(2.1 mm×100 mm,2.6μm)分离,流速为0.3 mL·min^(-1),柱温为40℃。质谱采用电喷雾电离,正负离子同时扫描,多反应监测的模式采集数据。结果30个化合物在考察的浓度范围内线性关系良好,相关系数均>0.999,各化合物的检出限为1.3~238.4 ng·g^(-1),平均回收率为63.7%~108.2%,RSD为1.0%~6.7%。采用该方法对90批舒筋活血片进行检测,共有3批检出对乙酰氨基酚,其余化合物均未检出。对乙酰氨基酚检出量为每片0.16~0.93μg,远低于对应药品日服用的最小剂量,排除人为故意添加。可能的原因是企业共线生产清洁验证不彻底引起的残留。结论该方法快速准确,灵敏度高,高选择性,可用于活血化瘀类中成药非法添加30个化合物的定性筛查和定量检测。

采用质性研究的访谈法、参与式观察法和文献查阅法获取巢湖流域洪灾的相关资料,分析洪灾中所表现出的灾害社会资本现状。研究表明,社会资本内部和外部良好互动能够有效减小甚至避免灾害带来的伤害,表现为互惠和信任的社会信任支持度、公众参与的非正式支持网络、资本转化的良好循环、共同命运的利他社会规范。最后,通过促进资本之间的良性循环、强化社会信任与支持、规范利他主义3方面来挖掘和提升区域内的资本存量,帮助负责规划和政策框架的人员有效管理和应对未来的灾害,提高最脆弱人群成功吸收冲击和压力的能力。
