This study aims to investigate characteristics of continental shelf wave(CSW)on the northwestern continental shelf of the South China Sea(SCS)induced by winter storms in *** and cruise observations,tidal gauge data at stations Hong Kong,Zhapo and Qinglan and sea surface wind data from January 1 to February 28,2021 are used to examine the relationship between along-shelf wind and sea level *** events of CSWs driven by the along-shelf sea surface wind are detected from wavelet spectra of tidal gauge *** signals are triply peaked at periods of 56 h,94 h and 180 h,propagating along the coast with phase speed ranging from 6.9 m/s to18.9 m/*** dispersion relation shows their property of the Kelvin mode of *** develop a simple method to estimate amplitude of sea surface fluctuation by along-shelf *** results are comparable with the observation data,suggesting it is *** mode 2 CSWs fits very well with the m.oring current velocity *** results from rare current help to understand wave-current interaction in the northwestern SCS.

Aerosol liquid water content(ALWC)plays an im.ortant role in secondary aerosol *** this study,a whole year field campaign was conducted at Shanxi in north Zhejiang Province during *** estimated by ISoRRoPIA-Ⅱ was then investigated to explore its characteristics and relationship with secondary *** exhibited a highest value in spring(66.38μg/m^(3)),followed by winter(45.08μg/m^(3)),summer(41.64μg/m^(3)),and autumn(35.01μg/m^(3)),*** was supposed that the secondary inorganic aerosols(SIA)were facilitated under higher ALWC conditions(RH>80%),while the secondary organic species tended to form under lower ALWC *** RH(>80%)promoted the No_(3)^(-)formation via gas-particle partitioning,while So_(4)^(2-)was generated at a relative lower RH(>50%).The ALWC was more sensitive to No_(3)^(-)(R=0.94)than So_(4)^(2-)(R=0.90).Thus,the self-amplifying processes between the ALWC and SIA enhanced the particle mass *** sensitivity of ALWC and oX(No_(2)+o_(3))to secondary organic carbon(SoC)varied in different seasons at Shanxi,more sensitive to aqueous-phase reactions(daytime R=0.84;nighttime R=0.54)than photochemical oxidation(daytime R=0.23;nighttime R=0.41)in wintertime with a high level of oX(daytime:130-140μg/m^(3);nighttime:100-140μg/m^(3)).The self-amplifying process of ALWC and SIA and the aqueous-phase formation of SoC will enhance aerosol formation,contributing to air pollution and reduction of visibility.

Background:Bitter taste receptors(Tas2rs)are generally considered to sense various bitter com.ounds to escape the intake of toxic *** taste receptors have been found to widely express in extraoral tissues and have im.ortant physiological functions outside the gustatory system in ***:To investigate the physiological functions of the bitter taste receptor cluster Tas2r106/Tas2r104/Tas2r105/Tas2r114 in lingual and extraoral tissues,multiple Tas2rs mutant mice and Gnat3 were produced using CRISPR/Cas9 gene-editing technique.A mixture containing Cas9 and sgRNA mRNAs for Tas2rs and Gnat3 gene was microinjected into the cytoplasm.of the ***,T7EN1 assays and sequencing were used to screen genetic mutation at the target sites in founder *** real-time polymerase chain reaction(qRT-PCR)and immunostaining were used to study the expression level of taste signaling cascade and bitter taste receptor in taste *** to taste substance was also studied using twobottle preference ***:We successfully produced several Tas2rs and Gnat3 mutant mice using the CRISPR/Cas9 *** results showed that the expression of GNAT3 and PLCB2 was not altered in Tas2rs mutant *** qRT-PCR results revealed the changed expression profile of m Tas2rs gene in taste buds of these mutant *** two-bottle preference tests,these mutant mice eliminate responses to cycloheximide due to genetic mutation of *** addition,these mutant mice showed a loss of taste perception to quinine dihydrochloride,denatonium benzoate,and cucurbitacin B(CuB).Gnat3-mediated taste receptor and its signal pathway contribute to CuB ***:These findings implied that these mutant mice would be a valuable means to understand the biological functions of TAS2Rs in extraoral tissues and investigate bitter com.ound-induced responses mediated by these TAS2Rs in many extraoral tissues.

The gingiva is a key oral barrier that protects oral tissues from various stimuli.A loss of gingival tissue hom.ostasis causes periodontitis,one of the most prevalent inflammatory diseases in *** human gingiva exists as a complex cell network comprising specialized *** understand the tissue-specific pathophysiology of the gingiva,we applied a recently developed spatial enhanced resolution omics-sequencing(Stereo-seq)technique to obtain a spatial transcriptome(ST)atlas of the gingiva in healthy individuals and periodontitis *** utilizing Stereo-seq,we identified the major cell types present in the gingiva,which included epithelial cells,fibroblasts,endothelial cells,and immune cells,as well as subgroups of epithelial cells and immune *** further observed that inflammation-related signalling pathways,such as the JAK-STAT and NF-κB signalling pathways,were significantly upregulated in the endothelial cells of the gingiva of periodontitis patients compared with those of healthy ***,we characterized the spatial distribution of periodontitis risk genes in the gingiva and found that the expression of IFI16 was significantly increased in endothelial cells of inflamed *** conclusion,our Stereo-seq findings may facilitate the development of innovative therapeutic strategies for periodontitis by mapping periodontitis-relevant genes and pathways and effector cells.
