Success of susceptibility-guided eradication of Helicobacter pylori in a region with high secondary clarithromycin and levofloxacin resistance rates

Goat casein hydrolysates alleviated insomnia in mice via modulating the level of neurotransmitters and the composition of gut microbiota

insomnia is associated with neurotransmitters and intestinal dysbiosis. Though studies have demonstrated the ameliorative effects of milk hydrolysates on insomnia， the underlying mechanisms require further exploration. in this study， we investigated how papain hydrolysates of goat casein (CPH) and whey protein (WPH) affected mice’s sleeplessness. Here， we show that CPH effectively improved the total sleep time in 12 h and restoring neurotransmitters (5-hydroxytryptamine (5-HT)， γ-aminobutyric acid (GABA)， dopamine (DA)， and norepinephrine (NE)) in mice. Further gut microbiota analysis revealed a significant increase in the relative abundance of Helicobacter and Escherichia-Shigella and a decrease in the relative abundance of Lactobacillus in the insomnia model mice (model). Compared to the model group， both CPH and WPH significantly increased the relative abundance of Akkermansia and Lactobacillus while lowering the relative abundance of Helicobacter and Escherichia-Shigella. Notably， while diazepam (DZP) increased mouse sleep duration， it also increased the relative abundance of Colidextribacter， Parasutterella， Muribaculaceae， and Prevotella. Additionally， the gene expression and protein expression of GABAAreceptor， cAMP-response element binding protein (CREB)， and brain-derived neurotrophic factor (BDNF) were upregulated in the hypothalamus. We also discovered a link between intestinal gut microbiota and neurotransmitters. Overall， our results suggest that goat milk hydrolysates， especially CPH， can effectively improve insomnia， providing a theoretical basis for further experimentation and individualized designs.

Glaucoma drainage device implantation and cyclophotocoagulation in the management of refractory glaucoma after Descemet-stripping automated endothelial keratoplasty

AiM:To compare the surgical outcomes of glaucoma drainage device implantation(GDi)and trans-scleral neodymium:YAG cyclophotocoagulation(CPC)in the management of refractory glaucoma after Descemetstripping automated endothelial keratoplasty(DSAEK).METHODS:This retrospective study on observational case series enrolled 29 patients who underwent DSAEK and posterior anti-glaucoma surgery(15 with GDi and 14 with CPC).The main outcome measures were intraocular pressure(iOP),glaucoma surgery success rate(defined as iOP of 6–21 mm Hg without additional anti-glaucoma operation),number of glaucoma medications,endothelial graft status,and best-corrected visual acuity(BCVA).RESULTS:The mean follow-up time was 34.1 and 21.0mo for DSAEK or glaucoma surgeries,both for the GDi and CPC *** groups showed significant iOP reduction after glaucoma *** GDi group presented a significantly higher success rate in iOP control than the CPC group(60%vs 21.4%,P=0.03).Both procedures significantly decreased the number of glaucoma medications(P=0.03).Forty percent and 57%of cases in the GDi and the CPC group,respectively,experienced endothelial graft failure during follow-up(P=0.36).Significantly worse BCVA after surgery was observed in the CPC group but not in the GDi ***:Both GDi and CPC significantly decrease iOP in eyes with glaucoma after *** is preferable to CPC in refractory glaucoma cases after DSAEK,as it manifests a significantly higher success rate for iOP control,similar endothelial graft failure rate,and relatively preserves BCVA than CPC.

生长激素(growth hormone,GH)不仅能调节动物的代谢、生长、发育和生殖,也能调节动物的免疫系统。本研究旨在通过转录组测序及生物信息学技术分析GH影响J亚群禽白血病病毒(subgroup J avian leukosis viruses,ALV-J)感染的鸡胚成纤维细胞系(chicken fibroblast cells,DF-1)中的关键基因和信号通路。将空载和GH的过表达质粒分别转染至DF-1,接着均接种ALV-J毒株SCAU-HN06,采用转录组测序技术检测相关的基因表达谱变化。结果显示,与对照组相比,实验组中有上调差异表达基因(differentially expressed genes,DEGs)122个(49%),下调DEGs 127个(51%)。通过GO功能富集分析发现,DEGs主要富集于新陈代谢过程、生物调节、细胞过程、细胞部分、催化活性和结合等过程。经KEGG富集分析发现DEGs主要在免疫系统、信号转导、信号分子和相互作用、细胞生长和死亡等通路上富集。进一步构建蛋白质-蛋白质相互作用(protein-protein interaction,PPi)网络,筛选出7个免疫相关基因进行实时定量PCR(RT-qPCR)验证,其中TRAF1、DSTYK、HESX1基因的表达趋势与转录组测序技术(RNA sequencing,RNA-seq)的测序结果相一致。本研究通过生物信息学分析挖掘出可能参与调控ALV-J的关键基因和信号通路,以期为禽白血病病毒-宿主相互作用的关系提供新的见解,为禽白血病的净化提供新的思路。

Quantifying peripapillary vessel density and retinal nerve fibre layer in type 1 diabetic children without clinically detectable retinopathy using OCTA

AiM:To quantify changes in radial peripapillary capillary vessel density(ppVD)and the peripapillary retinal nerve fiber layer(pRNFL)in children with type 1 diabetes without clinical diabetic retinopathy by optical coherence tomography angiography(OCTA),providing a basis for early retinopathy in children with type 1 ***:This was a retrospective study.A total of 30 patients(3–14y)with type 1 diabetes without clinical diabetic retinopathy(NDR group)were included.A total of 30 age-matched healthy subjects were included as the normal control group(CON group).The HbA1c level in the last 3mo was measured once in the NDR *** pRNFL thickness and ppVD were automatically measured,and the mean pRNFL and ppVD were calculated in the nasal,inferior,temporal,and superior *** changes in ppVD and pRNFL in the two groups were ***:Compared with CON group,the nasal and superior ppVDs decreased in the NDR group(all Pickness of the nasal pRNFL decreased significantly(Pile the inferior,temporal and superior pRNFLs slightly decreased but not significant in the NDR group(all P>0.05).Person and Spearman correlation analysis of ppVD and pRNFL thickness in each quadrant of the NDR group showed a positive correlation between nasal and superior(all Pile inferior and temporal had no significant correlation(all P>0.05).There was no significant correlation between the HbA1c level and ppVD and pRNFL in any quadrant(all P>0.05).There was no significant correlation between the course of diabetes mellitus and ppVD and pRNFL in any quadrant(all P>0.05).CONCLUSiON:ppVD and pRNFL decrease in eyes of children with type 1 diabetes before clinically detectable retinopathy and OCTA is helpful for early monitoring.
