Development and validation of a prediction model for early screening of people at high risk for colorectal cancer

为了明确质体乙酰辅酶A羧化酶(ACCase)Trp-1999-Leu突变对小麦田菵草生长适合度的影响,以ACCase基因CT区域的1999位纯合突变型(Leu/Leu, RR)和纯合野生型(Trp/Trp, SS)菵草为对象,采用温室盆栽法探究两种基因型菵草在早期生长阶段及营养生长阶段的株高及地上生物量、成熟期的繁殖力,在室内与田间不同条件下测定两种基因型菵草与小麦竞争能力的差异,以期明确在无除草剂选择压力下该位点突变对菵草生长的影响。结果表明,在菵草生长早期的10~15 d,RR型菵草的生物量和株高分别比SS型低24.9%和11.5%;在营养生长阶段,RR型菵草的相对生长速率(RGR)和净同化率(NAR)分别比SS型菵草低18.2%和28.6%,叶面积比(LAR)高于SS型14.0%,但是二者的地上生物量无显著差异。在成熟期,RR分配给生殖器官的资源更少,无论是繁殖力还是平均植株总种子重量均显著低于SS,分别低13.1%、11.4%。竞争力方面,在室内条件下,随着小麦密度的升高,RR基因型菵草地上生物量及种子产量均显著低于SS型;在田间试验中也证实了室内研究的结果,RR型与SS型菵草相比,与小麦的竞争力更弱。因此,ACCase Trp-1999-Leu基因突变会使抗性菵草产生明显的繁殖力和竞争力适合度代价,研究结果可为该基因型菵草的进化和治理提供一定的理论基础。

Alleles contributing to acetyl coenzyme A carboxylase(ACCase)resistance in keng stiffgrass(Pseudosclerochloa kengiana)populations from China

Keng stiffgrass is a grass weed that affects wheat-rice cropping systems in China. The extensive reliance on acetyl coen- zyme A carboxylase （ACCase）-inhibiting herbicides has resulted in keng stiffgrass developing resistance to these herbicides. The objective of this research was to evaluate the resistance level of the putative resistant keng stiffgrass populations to ACCase-inhibiting herbicides and to identify their molecular resistance mechanism. Whole-plant dose-response experi- ments demonstrated that SD-4 （R）, SD-11 （R）, and JS-25 （R） populations were highly resistant to fenoxaprop, clodinafop, and fluazifop, moderately resistant to diclofop, had low resistance to sethoxydim and pinoxaden, but were sensitive to clethodim. Partial chloroplasticACCase sequences showed that there were two copies ofACCase gene in keng stiffgrass, and all homoeologous genes were expressed. The results of sequence analyses of the ACCase CT domain revealed an isoleucine-to-asparagine substitution at position 2041 in SD-4 （R） and SD-11 （R） populations, and a tryptophan-to-cysteine substitution at position 2027 in the JS-25 （R） population. To our knowledge, this is the first report of Ile-2041-Asn and Trp- 2027-Cys mutations in ACCase-resistant keng stiffgrass. In addition, three robust （derived） cleaved amplified polymorphic sequence （（d）CAPS） markers have been developed to rapidly identify these mutations in the ACCase gene of keng stiffgrass.

High-strength,stretchable,and NIR-induced rapid self-healing polyurethane nanocomposites with bio-inspired hybrid crosslinked network

Stretchable and self-healable materials with excellent mechanical performance hold great promise for applications in flexible functional *** rapid developments,achieving high mechanical strength,extreme stretchability,and rapid selfhealing capability in self-healing materials remains ***,inspired by the hierarchical structure and unique network of connective tissue,we fabricated a class of bionic nanocomposites with high stretchability,outstanding mechanical strength,and rapid self-healing ability by integrating the bottlebrush copolymer functionalized graphene oxide(BCP@GO)into a polyurethane(PU)matrix via in-situ *** bottlebrush copolymer(BCP)acted as a bond bridge for linking the GO nanosheets(noncovalent interaction)and PU chains(covalent and hydrogen-bond interaction).The covalent interactions were responsible for providing high mechanical strength,and the abundant hydrogen-bond-based cross-links realized extreme stretchability and rapid self-healing *** resultant BCP@GO/PU nanocomposite with only 0.5 wt.%GO loading exhibited excellent mechanical properties(tensile strength increased by 52.1%,up to 28.6 MPa;toughness increased by 70.8%,up to 256.9 MJ/m^(3);elongation at break increased by 12.8%,up to 1847.2%),exceptional rapid and efficient self-healing ability(~99%with 20 s NIR irradiation),as well as superior shape memory and recyclable *** study develops a new strategy for designing highperformance self-healing nanocomposites and unfolds broad application prospects in smart materials.

In this paper,we consider two kinds of extragradient methods to solve the pseudo-monotone stochastic variational inequality ***,we present the modified stochastic extragradient method with constant step-size(MSEGMC)and prove the convergence of *** the strong pseudo-monotone operator and the exponentially growing sample sequences,we establish the R-linear convergence rate in terms of the mean natural residual and the oracle complexity O(1/ǫ).Second,we propose a modified stochastic extragradient method with adaptive step-size(MSEGMA).In addition,the step-size of MSEGMA does not depend on the lipschitz constant and without any line-search ***,we use some numerical experiments to verify the effectiveness of the two algorithms.

Learning to Branch in Combinatorial Optimization With Graph Pointer Networks

Traditional expert-designed branching rules in branch-and-bound(B&B) are static, often failing to adapt to diverse and evolving problem instances. Crafting these rules is labor-intensive, and may not scale well with complex *** the frequent need to solve varied combinatorial optimization problems, leveraging statistical learning to auto-tune B&B algorithms for specific problem classes becomes attractive. This paper proposes a graph pointer network model to learn the branch rules. Graph features, global features and historical features are designated to represent the solver state. The graph neural network processes graph features, while the pointer mechanism assimilates the global and historical features to finally determine the variable on which to branch. The model is trained to imitate the expert strong branching rule by a tailored top-k Kullback-Leibler divergence loss function. Experiments on a series of benchmark problems demonstrate that the proposed approach significantly outperforms the widely used expert-designed branching rules. It also outperforms state-of-the-art machine-learning-based branch-and-bound methods in terms of solving speed and search tree size on all the test instances. In addition, the model can generalize to unseen instances and scale to larger instances.

Thioredoxin pathway regulated live-cell synthesis of CdSe quantum dots in Saccharomyces cerevisiae

Currently, the application of synthetic biology to artificially manipulate and utilize organisms for the synthesis of desired products such as nanomaterials with excellent fluorescence properties is attracting considerable attention. However, it is still difficult to obtain designed products efficiently due to insufficient knowledge of the biosynthetic mechanisms. The thioredoxin（TRX） and glutathione（GSH） pathways are generally conserved thiol-reductase systems that protect organisms from oxidative stress and are involved in selenium（Se） metabolism. In this study, we revealed the pivotal role of cytoplasmic TRX pathway in regulating the metabolism of Na2SeO3during the live-cell synthesis of cadmium-selenium quantum dots（Cd Se QDs） in Saccharomyces cerevisiae by regulating the expression level of genes related to TRX pathway and measuring the intracellular content of selenocysteine（Se Cys）. The determination of Se Cys metabolism in yeast with GSH pathway-related genes deleted demonstrated that the TRX pathway played a more significant role in Se Cys metabolism than GSH pathway. A 6.4-fold enhancement in the synthetic yield of Cd Se QDs was achieved through the overexpression of TRX pathway-related genes,improvement of synthetic procedure, and supplementation of GSH based on the understanding of biological *** the mechanism of CdSe QDs live-cell synthesis facilitates the precise manipulation of biological processes for the synthesis of inorganic nanomaterials.
