Mast cell degranulation-triggered by SARS-CoV-2 induces tracheal-bronchial epithelial inflammation and injury

SARS-CoV-2 infection-induced hyper-inflammation is a key pathogenic factor of COVID-19.Our research,along with others',has demonstrated that mast cells(MCs)play a vital role in the initiation of hyper-inflammation caused by SARS-CoV-2.In previous study,we observed that SARS-CoV-2 infection induced the accumulation of MCs in the peri-bronchus and bronchioalveolar-duct junction in humanized mice.Additionally,we found that MC degranulation triggered by the spike protein resulted in inflammation in alveolar epithelial cells and capillary endothelial cells,leading to subsequent lung injury.The trachea and bronchus are the routes for SARS-CoV-2 transmission after virus inhalation,and inflammation in these regions could promote viral spread.MCs are widely distributed throughout the respiratory tract.Thus,in this study,we investigated the role of MCs and their degranulation in the development of inflammation in tracheal-bronchial epithelium.Histological analyses showed the accumulation and degranulation of MCs in the peri-trachea of humanized mice infected with SARS-CoV-2.MC degranulation caused lesions in trachea,and the formation of papillary hyperplasia was observed.Through transcriptome analysis in bronchial epithelial cells,we found that MC degranulation significantly altered multiple cellular signaling,particularly,leading to upregulated immune responses and inflammation.The administration of ebastine or loratadine effectively suppressed the induction of inflammatory factors in bronchial epithelial cells and alleviated tracheal injury in mice.Taken together,our findings confirm the essential role of MC degranulation in SARS-CoV-2-induced hyper-inflammation and the subsequent tissue lesions.Furthermore,our results support the use of ebastine or loratadine to inhibit SARS-CoV-2-triggered degranulation,thereby preventing tissue damage caused by hyper-inflammation.

To editor:This study reports a pregnant woman who was naturally conceived with monochorionic triamniotic(MCTA)combined with twin reversed arterial perfusion(TRAP).An ultrasound revealed that one of the fetuses had a cardiac malformation,and a connection between the umbilical arteries was suspected.To reduce the acardiac fetus,radiofrequency ablation was performed at 17+3 weeks of pregnancy.At 36+4 weeks,two healthy infants were delivered via elective cesarean section.Postoperative examination revealed a direct connection between the umbilical arteries,with the placental perfusion acting as a direct pump.It is essential to identify the intrauterine status of the blood-supplying infant and vascular connections in order to achieve successful outcomes in pregnancies with complications,such as edema and heart failure.The studies involving human participants were reviewed and approved by Human Research Ethics Committee of Tianjin Central Hospital of Obstetrics and Gynecology.The patient provided her written informed consent to participate in this study.

Massive lithospheric delamination in southeastern Tibet facilitating continental extrusion

Significant left-lateral movement along the Ailao Shan-Red River fault accommodated a substantial amount of the late Eocene to early Miocene India-Asia convergence.However,the activation of this critical strike-slip fault remains poorly understood.Here,we show key seismic evidence for the occurrence of massive lithospheric delamination in southeastern Tibet.Our novel observation of reflected body waves(e.g.P410P and P660P) retrieved from ambient noise interferometry sheds new light on the massive foundered lithosphere currently near the bottom of the mantle transition zone beneath southeastern Tibet.By integrating the novel seismic and pre-existing geochemical observations,we highlight a linkage between massive lithospheric delamination shortly after the onset of hard collision and activation of continental extrusion along the Ailao Shan-Re d River fault.This information provides critical insight into the early-stage evolution of the India-Asia collision in southeastern Tibet,which has significant implications for continental collision and its intracontinental response.

BACKGROUND Leukemia stem cells(LSCs)are found to be one of the main factors contributing to poor therapeutic effects in acute myeloid leukemia(AML),as they are protected by the bone marrow microenvironment(BMM)against conventional therapies.Gossypol acetic acid(GAA),which is extracted from the seeds of cotton plants,exerts anti-tumor roles in several types of cancer and has been reported to induce apoptosis of LSCs by inhibiting Bcl2.AIM To investigate the exact roles of GAA in regulating LSCs under different microenvironments and the exact mechanism.METHODS In this study,LSCs were magnetically sorted from AML cell lines and the CD34+CD38-population was obtained.The expression of leucine-rich pentatricopeptide repeat-containing protein(LRPPRC)and forkhead box M1(FOXM1)was evaluated in LSCs,and the effects of GAA on malignancies and mitochondrial RESULTS LRPPRC was found to be upregulated,and GAA inhibited cell proliferation by degrading LRPPRC.GAA induced LRPPRC degradation and inhibited the activation of interleukin 6(IL-6)/janus kinase(JAK)1/signal transducer and activator of transcription(STAT)3 signaling,enhancing chemosensitivity in LSCs against conventional chemotherapies,including L-Asparaginase,Dexamethasone,and cytarabine.GAA was also found to downregulate FOXM1 indirectly by regulating LRPPRC.Furthermore,GAA induced reactive oxygen species accumulation,disturbed mitochondrial homeostasis,and caused mitochondrial dysfunction.By inhibiting IL-6/JAK1/STAT3 signaling via degrading LRPPRC,GAA resulted in the elimination of LSCs.Meanwhile,GAA induced oxidative stress and subsequent cell damage by causing mitochondrial damage.CONCLUSION Taken together,the results indicate that GAA might overcome the BMM protective effect and be considered as a novel and effective combination therapy for AML.

Dry eye disease in systemic lupus erythematosus: a cross sectional study

AIM:To explore ocular surface manifestations of dry eye disease(DED)and its influencing factors in systemic lupus erythematosus(SLE)patients.METHODS:Ophthalmological examinations were conducted in SLE patients(n=43)and controls(n=41),including Ocular Surface Disease Index(OSDI),objective scatter index(OSI),tear meniscus height(TMH),lipid layer thickness(LLT),non-invasive Keratograph tear breakup time(NIKBUT),corneal fluorescein score(CFS),Schirmer I test.DED was diagnosed according to the Tear Film and Ocular Surface Society Dry Eye Workshop II Criteria.SLE patients were further divided into DED group and non-DED group,the disease activity,clinical manifestations and laboratory investigations were compared between the two groups.The disease activity was evaluated by Systemic Lupus Erythematosus Disease Activity Index 2000(SLEDAI-2K).Receiver operative characteristic(ROC)curve and multiplefactor binary logistic regression were performed.RESULTS:SLE patients showed higher OSDI[9.1(2.8-15.9)vs 6.3(2.2-7.5),P=0.035],higher OSI[1.67(1.09-2.60)vs 0.96(0.87-1.60),P=0.001],higher CFS[1(0-2)vs 0(0-1),P=0.001],lower LLT[65(42-100)vs 100(79.5-100),P=0.010],and lower NIKBUT[8.03(4.02-9.73)vs 9.67(5.26-12.71),P=0.030]than controls.The 32.6%of SLE patients had DED,which was higher than 12.2%of healthy controls.DED group showed higher SLEDAI-2K score[9.7±6.1 vs 5.4±3.4,P=0.025],higher anti-cardiolipin antibody(ACL)[8.7(3.5-13.2)vs 3.6(2.0-6.9),P=0.035],and higher proportion of patients with cutaneous eruption[42.9%vs 6.9%,P=0.015]than non-DED group.According to multiple-factor binary logistic regression analysis,the SLEDAI-2K score(OR=1.194,P=0.041)and cutaneous eruption(OR=7.094,P=0.045)could be consider as risk factors for DED in SLE patients.The ROC curve of the combined factors including age,disease duration,SLEDAI-2K score,ACL,and cutaneous eruption was analyzed,with a sensitivity of 0.786,a specificity of 0.793,and an area under curve of 0.820.CONCLUSION:Ocular surface affection is frequent in SLE patients,and patients wi

Enhanced cyclic stability of partially disordered spinel cathodes through direct fluorination with gaseous fluorine

Spinel-type cathodes are considered an optimal substitute for conventional layered oxide cathodes owing to their use of inexpensive and earth-abundant manganese as the redox-active element.Moreover,the introduction of cation disorder can effectively suppress the detrimental two-phase reaction to realize high capacities in a wide voltage range.However,the continuous capacity decay during cycles has hindered the widespread application of these cathode materials.Inorganic fluorides exhibit excellent electrochemical stability at high voltage;therefore,in this study,the direct F2 gas reaction with a partially disordered spinel cathode(Li1.6Mn1.6O3.7F0.3,LMOF1.6) was initially applied to investigate the impacts of fluorination on the surface structure and electrochemical performances.The inorganic fluorinated layer,mainly containing LiF,was distributed uniformly on the surface of LMOF1.6nanoparticles after fluorination for an appropriate time without the turbulence caused by the valency of manganese cation,which improved the capacity retention and rate capability by the suppression of structural damage,parasitic reaction,and cation dissolution.The LMOF1.6cathode fluorinated for 0.5 h exhibited a capacity of283.6 mAh·g-1at 50 mA·g-1and an enhanced capacity retention of 29.6% after 50 cycles in the voltage range of1.5-4.8 V,as compared to the pristine LMOF1.6 with only27.9% capacity retention.

Shear-wave velocity structures of the shallow crust beneath the Ordos and Sichuan Basins from multi-frequency direct P-wave amplitudes in receiver functions

As the two largest cratonic basins in China,the Ordos Basin and the Sichuan Basin are of key importance for understanding the evolutionary history of the Chinese continent.In this study,the shear-wave velocity(V_(S))structures of the shallow crust(depth up to 10 km)beneath the two basins are imaged based on the frequency-dependence of direct P-wave amplitudes in receiver functions.The teleseismic data used in the study came from 160 broadband seismic stations,including permanent and temporary stations.The results show that the V_(S) and the thickness of the sediments in the Ordos Basin and the Sichuan Basin are respectively lower and thicker in the west than in the east.In the Ordos Basin,the shallow crustal V_(S) increases gradually from 2.10 km s^(−1)in the northwest to 2.65 km s^(−1)in the southeast and the thickest sediments are 7–8 km in the northwest and 5 km in the east.In the Sichuan Basin,the shallow crustal V_(S) increases from 2.4 km s^(−1) in the west to 2.7 km s^(−1)in the east and the thickness of the sediments decreases from>7 km in the west to 6 km in the east.The east-west difference of the shallow crustal structures of the two basins may have been controlled by the Cenozoic India-Eurasia collision.The western parts of the basins near the collision have a higher deposition rate,while in the parts inside the basins far from the collision,the V_(S) slowly increases with depth,indicating that these areas have experienced a more uniform deposition process.In addition,both basins are characterized by velocity structures that are higher along the edges and lower inside of the basins.The edges of the basins suffered strong denudation due to the uplifting and deformation influenced by tectonic evolution.The downward gradient of the shear-wave velocity beneath the Ordos Basin is twice that of the Sichuan Basin,which may be caused by the different deposition and denudation rates of the two basins resulting from differences in structural evolution and thermal events.In addition,the northern Ordos Basin exhi

Time-dependent Global Attractors for the Nonclassical Diffusion Equations with Fading Memory

In this paper, we discuss the long-time behavior of solutions to the nonclassical diffusion equation with fading memory when the nonlinear term f satisfies critical exponential growth and the external force g(x)∈L2(Ω). In the framework of time-dependent spaces, we verify the existence of absorbing sets and the asymptotic compactness of the process, then we obtain the existence of the time-dependent global attractor A={At}t∈Rin Mt. Furthermore, we achieve the regularity of A, that is, Atis bounded in Mt1with a bound independent of t.

Prediction of suitable water content in granulation of sintering mixture based on Litster's model

Suitable water content plays a decisive role in the granulation of sintering mixtures.Herein,a method was proposed to predict the suitable water content for effective granulation on the basis of Litster's granulation model.The granulation effectiveness of a sintering mixture was predicted by the model,with the allowance error of±10%.The effects of the water absorption properties,particle size composition and content of adhesive particles on the suitable water content were studied.The results showed that the allowable error of prediction was within±0.5%compared to the experimentally determined suitable water content.With an increase in adhesive powder content of mixtures with higher water absorption,the suitable water content increased to achieve similar granulation effectiveness.Moreover,as the amount of concentrates increased,the suitable water content first increased and then remained steady.The influence of the water absorption characteristics of the adhesive particles on the suitable water content was less than that of their particle size composition in the mixture.

In this work,boron(B)was used to promote Fe^(3+)/peracetic acid(Fe^(3+)/PAA)for the degradation of sulfamethazine(SMT).An SMT degradation efficiency of 9.1%was observed in the Fe^(3+)/PAA system over 60 min,which was significantly increased to 99.3%in the B/Fe^(3+)/PAA system over 10 min.The B/Fe^(3+)/PAA process also exhibited superior resistance to natural substances,excellent adaptability to different harmful substances,and good removal of antibiotics in natural fresh water samples.The mechanism of action of boron for Fe^(3+)reduction was determined using scanning electron microscopy(SEM),X-ray photoelectron spectroscopy(XPS),Fourier transform infrared(FT-IR)spectroscopy,density functional theory(DFT)calculations,and electrochemical tests.The dominant role of^(·)OH was confirmed using quenching experiments,electron spin resonance(EPR)spectroscopy,and quantitative tests.Organic radicals(R-O^(·))and Fe(IV)also significantly contribute to the removal of SMT.DFT calculations on the reaction between Fe^(2+)and the PAA were conducted to further determine the contribution from ^(·)OH,R-O^(·),and Fe(IV)from the perspective of thermodynamics and the reaction pathways.Different boron dosages,Fe^(3+)dosages,and initial pH values were also investigated in the B/Fe^(3+)/PAA system to study their effect of SMT removal and the production of the reactive species.Fe(IV)production determined the k_(R-O·+Fe(IV))value suggesting that Fe(IV)may play a more important role than R-O^(·).A comparison of the results with other processes has also proved that the procedure described in this study(B/Fe^(3+)/PAA)is an effective method for the degradation of antibiotics.
