Simultaneous determination of ten nucleosides and bases in Ganoderma by micellar electrokinetic chromatography

Ganoderma(lingzhi)is a famous herbal medicine and edible supplement in oriental countries for a long history.In this study,a simple micellar electrokinetic chromatography(MEKC)method was established for the analysis of nucleosides and bases,the major bioactive components in Ganoderma for the first time.By optimizing the borate concentration,the sodium dodecyl sulfate(SDS)concentration and the pH value of running buffer,10 nucleosides and bases achieved an ideal separation.In real sample analysis,the developed method was successfully used to determine the 10 target analytes in 23 batches of Ganoderma samples from different regions.Results indicated that contents of 10 investigated analytes in each sample showed obvious variation.The principal components analysis(PCA)and hierarchical cluster analysis(HCA)analysis classified the samples into three groups,and the HCA tree visualized the relationships which was mainly contributed by geographical partition.The results indicated geographical origin to be an important factor that affect the accumulation of nucleosides and bases in Ganoderma.In summary,this study provides a simple and practical strategy for quality assessment and cultivation reference of Ganoderma.

Marine symbiotic fungi represent an intriguing source of discovery of novel secondary metabolites with various biological activities.Sea anemones are benthic marine invertebrates,however,the cultivable symbiotic fungi residing in the sea anemones are paid few attentions compared to those derived from their cnidarian counterparts.Here we show the identification of antimicrobial secondary metabolites from the sea anemone-derived symbiotic fungi.Out offive isolated fungal strains,only the strain SMA01 showed strong antimicrobial activities,which was assigned into the genus Emericella based on the morphological characteristics and the ITS sequencing.Media swift from liquid fermentation to solid rice medium presented little influence on its antibacterial activity.A chemical investigation of the ethyl acetate extract of the Emericella sp.SMA01 led to discovery of the primary antibiotic metabolite phenazine-1-carboxylic acid.The IC_(50) values of the phenazine-1-carboxylic acid against Phytophthora capsici,Gibberella zeae,and Verticillium dahliae were determined to be 23.26-53.89μg/mL.To the best of our knowledge,this was the first report of Emericella sp.in sea anemones.The current study may benefit understanding of the defensive chemical interactions between the symbiotic fungi and their host sea anemones.

The Paleozoic huoshenmiao iron skarn deposit in the Tongbai area of North Qinling Orogen,China:Insights from garnet U-Pb dating and geological constraints

The Qinling Orogen is a composite orogenic belt that can be subdivided into the North and South Qinling,broadly separated by the Shangdan suture zone.These two orogenic belts were generated by subduction-collisional processes in the Early Paleozoic and Late Triassic,respectively.During the Late Jurassic to Early Cretaceous,the eastern portion of the Qinling Orogen was tectonically reactivated due to westerly subduction of the Izanagi plate underneath the East China continental margin.The Qinling Orogen is well-endowed with numerous Au,Mo,Ag-Pb-Zn deposits that predominantly formed in the Late Triassic to Early Cretaceous,with rare Paleozoic varieties documented.In this study,we present garnet and zircon U-Pb dating results to show that the huoshenmiao iron skarn deposit in the Tongbai area of North Qinling orogenic belt formed in the Early Silurian and is genetically related to subduction-related magmatism.The huoshenmiao deposit consists of lenticular and stratiform orebodies that contain massive or densely disseminated magnetite variably associated with garnet,diopside,epidote,hornblende,and actinolite.Garnets from iron ores are andradite and grossular in composition(Ad_(83.1)Gr_(13.3)-Ad_(86.5)Gr_(10.2))and characterized by enrichment in light rare earth elements(ΣLREE=57.85-103.82 ppm)and depletion in heavy rare earth elements(ΣHREE=5.50-11.34 ppm),with significantly positive Eu and Ce anomalies(δEu of 1.09-1.89 andδCe of 1.39-1.69).These compositional signatures are distinctly different from those of garnets in the ore-hosting metamorphic rocks that are typically dominated by almandine,spessartine and grossular(Al_(47.4)Sp_(30.4)Gr_(13.8)-Al_(51.9)Sp_(24.8)Gr_(17.6)),depleted in LREE(0.14-0.69 ppm),enriched in HREE(95.68-841.60 ppm)and have pronounced negative Eu anomalies(δEu=0.24-0.51).In addition,garnets from iron ores of the huoshenmiao deposit contain abundant daughter mineral-bearing,multiphase fluid inclusions,further confirming their hydrothermal origin.Two samples of those hydrothermal garnets yield

Freeze-drying and hot-pressing strategy to embed two-dimensional Ti_(0.87)O_(2) monolayers in commercial polypropylene films with enhanced dielectric properties

The dielectric capacitor has been widely used in advanced electronic and electrical power systems due to its capability of ultrafast charging–discharging and ultrahigh power density.Nevertheless,its energy density is still limited by the low dielectric constant(≈2.2)of the commercial dielectric polypropylene(PP).The conventional enhancement strategy by embedding inorganic fillers in PP matrix is still difficult and challenging due to that PP hardly dissolves in any inorganic/organic solvent.In this work,we develop a new strategy including freeze-drying,surface functionalization,and hot-pressing to incorporate Ti_(0.87)O_(2) monolayers in PP film.A series of uniform composited Ti_(0.87)O_(2)@PP film has been successfully fabricated with Ti0.87O2 content range of 0–15 wt%.The maximum dielectric constant of the as-prepared Ti_(0.87)O_(2)@PP film is 3.27 when the Ti_(0.87)O_(2) content is 9 wt%,which is about 1.5 times higher than that of pure PP.Our study provides a feasible strategy to embed two-dimensional material into commercial PP thin-film with superior dielectric performance for practical application.

Fuzzy cluster analysis of water mass in the western Taiwan Strait in spring 2019

The classification of the springtime water mass has an important influence on the hydrography,regional climate change and fishery in the Taiwan Strait.Based on 58 stations of CTD profiling data collected in the western and southwestern Taiwan Strait during the spring cruise of 2019,we analyze the spatial distributions of temperature(T)and salinity(S)in the investigation area.Then by using the fuzzy cluster method combined with the T-S similarity number,we classify the investigation area into 5 water masses:the Minzhe Coastal Water(MZCW),the Taiwan Strait Mixed Water(TSMW),the South China Sea Surface Water(SCSSW),the South China Sea Subsurface Water(SCSUW)and the Kuroshio Branch Water(KBW).The MZCW appears in the near surface layer along the western coast of Taiwan Strait,showing low-salinity(<32.0)tongues near the Minjiang River Estuary and the Xiamen Bay mouth.The TSMW covers most upper layer of the investigation area.The SCSSW is mainly distributed in the upper layer of the southwestern Taiwan Strait,beneath which is the SCSUW.The KBW is a high temperature(core value of 26.36℃)and high salinity(core value of 34.62)water mass located southeast of the Taiwan Bank and partially in the central Taiwan Strait.

Synthesis and performance of carbon-modified LiFePO_4 using an in situ PVA pyrolysis procedure

LiFePOa/carbon composite cathode material was prepared by granulating and subsequent pyrolysis processing in N2 atmosphere with polyvinyl alcohol （PVA） as the carbon source. The influences of carbon content on the microstructure and battery performance were investigated. Single LiFePO4 phase and amorphous carbon can be found in the products. A special micro-morphology of the optimum sample was observed. The discharge capacity of the cell with the optimum cathode was 135 mAh.g^-1, close to the charge capacity of 153 mAh.g^-1 at 17 mA.g^-1. The influence of ambient temperature on the cell capacity was investigated. The temperature dependence of its electrochemical characteristic was evaluated by using AC impedance spectroscopy. A new equivalent circuit based on the charge and mass transfer control process in an electrode was proposed to fit the obtained AC impedance spectra. The tendency of every element in the equivalent circuit was used to interpret the temperature dependence of the capacity of the optimum cathode.

目的探讨茶多酚对心力衰竭大鼠肾素-血管紧张素-醛固酮系统和转化生长因子-β1(TGF-β1)/Smads信号通路的作用机制。方法SD大鼠随机分为假手术组(Sham组)、模型组(Model组)、卡托普利治疗组(PC组)和茶多酚治疗组(TP组)。Model组、PC组和TP组大鼠采用丝线结扎左冠状动脉前降支建立心肌梗死后心力衰竭大鼠模型,Sham组不进行结扎。超声心动图检测心功能,HE染色检测心肌病理变化,Masson染色检测心肌纤维化,免疫组织化学染色检测Ⅰ型和Ⅲ型胶原的表达,ELISA法检测血管紧张素Ⅱ(AngⅡ)、醛固酮(ALD)、肾素(PRA)、白细胞介素(IL)-1β、IL-6和肿瘤坏死因子-α(TNF-α)的水平,Western blot检测TGF-β1、磷酸化Smad(p-Smad)2、Smad2、p-Smad3和Smad3蛋白表达。结果与Sham组比较,Model组大鼠心肌细胞排列紊乱,有明显的炎性细胞浸润,心肌纤维化程度明显升高(t=9.748,P=0.001),Ⅰ型胶原(t=11.754,P=0.001)和Ⅲ型胶原(t=10.573,P=0.001)表达明显升高,而射血分数(t=13.174,P=0.002)和左心室短轴缩短率(t=11.853,P=0.001)则明显降低,AngⅡ(t=4.246,P=0.001)、ALD(t=5.385,P=0.004)、PRA(t=4.386,P=0.004)、IL-1β(t=4.393,P=0.001)、IL-6(t=6.375,P=0.002)和TNF-α(t=4.753,P=0.002)表达水平明显升高,TGF-β1(t=6.365,P=0.001)、p-Smad2/Smad2(t=13.755,P=0.001)和p-Smad3/Smad3(t=11.657,P=0.002)蛋白表达明显升高;与Model组比较,PC组和TP组大鼠心肌病理变化得到明显改善,左心室舒张末期内径(t=6.367,P=0.003;t=5.264,P=0.003)、左心室收缩末期内径(t=5.253,P=0.002;t=5.974,P=0.001)、心脏质量指数(t=5.012,P=0.007;t=4.953,P=0.005)、左心室质量指数(t=5.531,P=0.003;t=5.483,P=0.004)、心肌纤维化程度(t=6.734,P=0.001;t=5.362,P=0.001)均明显降低,Ⅰ型胶原(t=5.373,P=0.001;t=4.364,P=0.001)和Ⅲ型胶原(t=6.764,P=0.001;t=4.579,P=0.001)表达明显降低,而射血分数(t=11.264,P=0.002;t=10.356,P=0.001)和左心室短轴缩短率(t=8.246,P=0.002;t=7.824,P=0.001)表达水平则明显升高,AngⅡ(t=3.126,P=0.001;t=2.853,P=0.001)、ALD(t=3.854,P=0.004;t=3.164,P=0.004)、PRA(t=3.126,P=0.004;t=3.063,P=0.004)、IL-1β(t=2.964,P=0.001;t=2.765,P=0.001)、IL-6(t=4.865,P=0.002;t=4.275,P=0.002)和TNF-α(t=3.146,P=0.002;t=2.973,P=0.002)表达水平明显降低,TGF-β1(t=4.657,P=0.001;t=4.176,P=0.001)、p-Smad2/Smad2(t=9.687,P=0.001;t=6.753,P=0.001)和p-Smad3/Smad3(t=6.477,P=0.002;t=4.754,P=0.002)蛋白表达明显降低。结论茶多酚通过抑制肾素-血管紧张素-醛固酮系统和TGF-β1/Smads通路的激活,在心力衰竭中发挥保护作用。

A"pea-pod"-like composite of graphene-wrapped silicon nanoparticles(P–Si@GS)was successfully prepared via a new and facial technique utilizing freeze-drying and subsequent solvothermal process.The cross-linked"pods"formed by curled few-layer graphene sheets constituted a conductive network,in which silicon nanoparticles were tightly wrapped.The unique graphene-wrapped structure with mesopores on the surface can effectively enhance the conductivity of silicon,alleviate its volume effect and provide rapid transport channels for electrons and ions,thus resulting in excellent electrochemical performance.The strategy of combining freeze-drying and solvothermal methods is facile,low-cost and effective,which provides new ideas for the preparation of highperformance graphene-wrapped nanoparticles composites.

Effect of laser-discrete-quenching on bonding properties of electroplated grinding wheel with AISI 1045 steel substrate and nickel bond

To improve the bonding strength between the nickel bond and the hub of the electroplated diamond grinding wheel,a hybrid technique was proposed to combine laser prequenching steel substrate and post-electroplating nickel.To validate the effectiveness of the proposed technique,AISI 1045 substrate was nickel-coated.The bonding properties between the electroplated nickel coating and substrate with or without laser-discrete-quenching were discussed comparatively by scratch,indentation,and thermal shock tests.The results show that the prequenching treatment leads to phase transformation of AISI 1045 microstructure from the mixed pearlite and ferrite phases into the martensitic phase.Since the martensitic phase is characterized as a high corrosion resistance,the interface of substrate/coating is smooth and flat in the prequenched zone,and the coating is bonded well with the steel substrate.In contrast to the steel substrate without pre-quenching treatment,the proposed technique significantly enhanced the bonding strengths of the electroplated nickel-coating.On one hand,the average hardness of electroplated nickel-coating on the laser pre-quenched zone is increased by 18.7%,and the scratch depth with the same load become narrower and shallower.On the other hand,the coefficient of friction(CoF)and the vibration amplitude are reduced,and the coating is bonded effectively with the substrate to inhibit the crack initialization at the interface.This prevents effectively the coating from peeling off and improves significantly the thermal shock resistance property.

Intercalated 2D nanoclay for emerging drug delivery in cancer therapy

Natural two-dimensional （2D） kaolinite nanoclay has been incorporated into an emerging drug delivery system. The basal spacing of the kaolinite nanoclay was expanded from 0.72 to 4.16 nm through the intercalation of various organic guest species of different chain lengths, which can increase the efficiency in drug delivery and reduce the toxicity of doxorubicin （DOX）. Original kaolinite （Kaolin） and the Kaolin intercalation compounds exhibited a high level of biocompatibility and very low toxicity towards cells of pancreatic cancer, gastric cancer, prostate cancer, breast cancer, colorectal cancer, esophageal cancer, and differentiated thyroid cancer. However, lung cancer and hepatocellular cancer cells need more strict compositional, structural, and morphological modulations for drug delivery carriers. DOX-Kaolin and the DOX-Kaolin intercalation compounds showed dramatically faster drug release in moderately acidic solution than in neutral condition, and exhibited enhanced therapeutic effects against ten model cancer cell cultures in a dose-dependent manner. The use of 2D nanoclay materials for a novel drug delivery system could feasibly pave a way towards high-performance nanotherapeutics, with superior antitumor efficacy and significantly reduced side effects.
