Inhibition of hypoxia-inducible factor 1 with acriflavine sensitizes hypoxic tumor cells to photodynamic therapy with zinc phthalocyanine-encapsulating cationic liposomes

Photodynamic therapy （PDT） is a tumor treatment modality in which a tumor- localized photosensitizer is excited with light, which results in local production of reactive oxygen species, destruction of tumor vasculature, tumor hypoxia, tumor cell death, and induction of an anti-tumor immune response. However, pre-existing tumor hypoxia may desensitize tumors to PDT by activating the hypoxia-inducible factor 1 （HIF-1） survival pathway. Therefore, we hypothesized that inhibition of HIF-1 with acriflavine （ACF） would exacerbate cell death in human epidermoid carcinoma （A431） cells. PDT of A431 tumor cells was per- formed using newly developed and optimized PEGylated cationic liposomes containing the photosensitizer zinc phthalocyanine （ZnPC）. Molecular dock.ng revealed that ACF binds to the dimerization domain of HIF-la, and confocal microscopy confirmed translocation of ACF from the cytosol to the nucleus under hypoxia. HIF-1 was stabilized in hypoxic, but not normoxic, A431 cells following PDT. Inhibition of HIF-1 with ACF increased the extent of PDT-induced cell death under hypoxic conditions and reduced the expression of the HIF-1 target genes VEGF, PTGS2, and EDN1. Moreover, co-encapsulation of ACF in the aqueous core of ZnPC-containing liposomes yielded an adjuvant effect on PDT efficacy that was comparable to non-encapsulated ACF. In conclusion, HIF-1 contributes to A431 tumor cell survival following PDT with liposomal ZnPC. Inhibition of HIF-1 with free or liposomal ACF improves PDT efficacy.

Cytoplasmic expression of p27^(k.p1) is associated with a favourable prognosis in colorectal cancer patients

AIM: To evaluate the prognostic significance of p27k.p1 in colorectal cancer patients. METHODS: Cytoplasmic and nuclear p27k.p1 expression was evaluated in 418 colorectal cancers using tissue microarrays. Data were associated with k.own patient and tumor variables and long-term patient outcomes, providing further insight into the mechanisms by which p27k.p1 may influence tumor development. RESULTS: Nuclear and cytoplasmic p27k.p1 expressions were detected in 59% and 19% of tumors respectively. Cytoplasmic p27k.p1 was almost invariably associated with positive nuclear p27k.p1 expression. Neither case correlat- ed with k.own clinical or pathological variables, includ- ing tumor stage, grade or extramural vascular invasion. Furthermore, nuclear p27k.p1 expression had no impact on survival. However, we identified a significant correla- tion between expression of cytoplasmic p27k.p1 and longer disease-specific survival times. On multivariate analysis, TNM stage and extramural vascular invasion were highly significant independent prognostic factors, with positive cytoplasmic p27 expression showing a trend towards im- proved patient survival (P = 0.059).CONCLUSION: These findings support the recent evi- dence that cytoplasmic p27k.p1 has a distinct and impor- tant biological role that can influence tumor outcome.

The United States （U.S.） is facing a national opioid epidemic, and medical systems are in need of non- pharmacologic strategies that can be employed to decrease the public＇s opioid dependence. Acupuncture has emerged as a powerful, evidence-based, safe, cost-effective, and available treatment modality suitable to meeting this need. Acupuncture has been shown to be effective for the management of numerous types of pain conditions, and mechanisms of action for acupuncture have been described and are understandable from biomedical, physiologic perspectives. Further, acupuncture＇s cost-effectiveness can dramatically decrease health care expenditures, both from the standpoint of treating acute pain and through avoiding addiction to opioids that requires costly care, destroys quality of life, and can lead to fatal overdose. Numerous federal regulatory agencies have advised or mandated that healthcare systems and providers offer non-pharmacologic treatment options for pain. Acupuncture stands out as the most evidence-based, immediately available choice to fulfil these calls. Acupuncture can safely, easily, and cost-effectively be incorporated into hospital settings as diverse as the emergency department, labor and delivery suites, andneonatal intensive care units to treat a variety of commonly seen pain conditions. Acupuncture is already being successfully and meaningfully utilized by the Veterans Administration and various branches of the U.S. Military, in some studies demonstrably decreasing the volume of opioids prescribed when included in care.

Waxy apple cuticles predominantly accumulate ursane-type triterpenes,but the profile shifts with the induction of sk.n russeting towards lupane-type *** previously characterised several k.y enzymes in the ursane-type and lupane-type triterpene pathways,but this switch in triterpene metabolism associated with loss of cuticle integrity is not fully *** analyse the relationship between triterpene biosynthesis and russeting,we used microscopy,RNA-sequencing and metabolite profiling during apple fruit *** compared the sk.n of three genetically-close clones of‘Golden Delicious’(with waxy,partially russeted and fully russeted sk.n).We identified a unique molecular profile for the russet clone,including low transcript abundance of multiple cuticle-specific metabolic pathways in the early stages of fruit *** correlation analyses between gene transcription and metabolite concentration we found MYB transcription factors strongly associated with lupane-type triterpene *** showed how their transcription changed with the onset of cuticle crack.ng followed by russeting and that one factor,MYB66,was able to bind the promoter of the oxidosqualene cyclase OSC5,to drive the production of lupeol *** results provide insights into the break.own of cuticle integrity leading to russet and how this drives MYB-regulated changes to triterpene biosynthesis.
