Although considerable conservation resources have been committed to develop and use law enforcement monitoring and management tools such as SMART,measures of success are ill-defined and,to date,few reports detail results ***,we present 4 case studies from protected areas with Amur tigers(Panthera tigris altaica)in Russia,in which indicators of success were defined and evaluated at *** ultimate goal was an increase in tiger numbers to 1 individual/100 km^(2)at each *** predicted that improvements in law enforcement effectiveness would be followed by increases in prey numbers and,subsequently,tiger *** used short-term and long-term indicators of success,including:(i)patrol team effort and effectiveness;(ii)catch per unit effort indicators(to measure reductions in threats);and(iii)changes in target species *** addition to implementing a monitoring system,we focused on improving law enforcement management using an adaptive management *** 4 years,we noted clear increases in patrol effort and a partial reduction in *** we did not detect clear trends in ungulate numbers,tiger populations remained stable or increased,suggesting that poaching of tigers may be more limiting than prey *** effectiveness is needed before a clear reduction in threats can be noted,and more time is needed before detecting responses in target ***,delineation of concrete goals and indicators of success provide a means of evaluating progress and *** monitoring should be a central component of law enforcement strategies for protected areas.

Modulation of liver tolerance by conventional and nonconventional antigen-presenting cells and regulatory immune cells

The liver is a tolerogenic organ with exquisite mechanisms of immune regulation that ensure upkeep of local and systemic immune tolerance to self and foreign antigens, but that is also able to mount effective immune responses against pathogens. The immune privilege of liver allografts was recognized first in pigs in spite of major histo-compatibility complex mismatch, and termed the ＂liver tolerance effect＂. Furthermore, liver transplants are spontaneously accepted with only low-dose immunosuppression, and induce tolerance for non-hepatic co-transplanted allografts of the same donor. Although this immunotolerogenic environment is favorable in the setting of organ transplantation, it is detrimental in chronic infectious liver diseases like hepatitis B or c, malaria, schistosomiasis or tumorigenesis, leading to pathogen persistence and weak anti-tumor effects. The liver is a primary site of T-cell activation, but it elicits poor or incomplete activation of T cells, leading to their abortive activation, exhaustion, suppression of their effector function and early death. This is exploited by pathogens and can impair pathogen control and clearance or allow tumor growth. Hepatic priming of T cells is mediated by a number of local conventional and nonconventional antigen-presenting cells （APcs）, which promote tolerance by immune deviation, induction of T-cell anergy or apoptosis, and generating and expanding regulatory T cells. This review will focus on the communication between classical and nonclassical APcs and lymphocytes in the liver in tolerance induction and will discuss recent insights into the role of innate lymphocytes in this process.

Effects and Potential Mechanisms of Danzhi xiaoyao Pill(丹栀逍遥丸) on Proliferation of McF-7 Human Breast cancer cells in vitro

Objective： To investigate the effects of 50% ethyl alcohol （EtOH） extracts from Danzhi xiaoyao Pill （丹栀逍遥丸, DxP） on the proliferation of McF-7 human breast cancer cells and potential mechanisms. Methods： ATP-Lite assay was performed to test the proliferation of the McF-7 breast cancer cell line; and antioxidant activity was measured by the oxygen radical absorbance capacity （ORAc）. The effects of DxP on nitric oxide （NO） production were tested by lipopolysaccharide （LPS）- stimulated RAW 264.7 murine macrophages using the Griess reaction. Results： The 50% EtOH DxP extracts displayed a cytotoxic response on McF-7 cells at 0.10, 0.25 and 0.50 mg/mL dosedependently with the proliferation inhibited by more than 85%. The ORAc value of the DxP was 820μ moL Trolox equivalent/g, about 40% of the vitamin c value. DxP extracts had significant inhibitory effect on NO production at the concentration from 0.0625 mg/mL to 0.5 mg/mL （P〈0.05, P〈0.01）. conclusion： The extracts of DxP could significantly inhibit the proliferation of McF-7 cells, with the effect possibly related to its antioxidant activity and the inhibition of NO production.

Aim:The cluster of differentiation 44(cD44)transmembrane protein is cleaved byγ-secretase,the inhibition of which blocks cD44 *** study aimed to determine the biological consequence of cD44 cleavage and its potential interaction with Runt-related transcription factor(RUNx2)in a sequence-specific manner in Pc3 prostate cancer ***:Using full-length and c-terminal deletion constructs of cD44-IcD(D1-D5)expressed as stable green fluorescent protein-fusion proteins in Pc3 cells,we located possible RUNx2-binding ***:chromatin immunoprecipitation assays demonstrated that the c-terminal amino acid residues between amino acids 671 and 706 in D1 to D3 constructs were indispensable for sequence-specific binding of *** binding was minimal for sequences in the D4 and D5 ***,an increase in matrix metalloprotease-9(MMP-9)expression was observed at the mRNA and protein levels in Pc3 cells stably expressing D1–D3 ***:These results provide biochemical evidence for the possible sequence-specific cD44-IcD/RUNx2 interaction and its functional relationship to MMP-9 transcription in the promoter region.

Myeloid cells,such as neutrophils,are produced in the bone marrow in high quantities and are important in the pathogenesis of vascular diseases such as pulmonary hypertension(PH).Although neutrophil recruitment into sites of inflammation has been well studied,the mechanisms of neutrophil egress from the bone marrow are not well *** computational flow cytometry,we observed increased neutrophils in the lungs of patients and mice with ***,we found elevated levels of IL-6 in the blood and lungs of patients and mice with *** observed that transgenic mice overexpressing Il-6 in the lungs displayed elevated neutrophil egress from the bone marrow and exaggerated neutrophil recruitment to the lungs,resulting in exacerbated pulmonary vascular remodeling,and dysfunctional ***,we found that IL-6-induced neutrophil egress from the bone marrow was dependent on interferon regulatory factor 4(IRF-4)-mediated cx3cR1 expression in ***,cx3cr1 genetic deficiency in hematopoietic cells in Il-6-transgenic mice significantly reduced neutrophil egress from bone marrow and decreased neutrophil counts in the lungs,thus ameliorating pulmonary remodeling and *** summary,these findings define a novel mechanism of IL-6-induced neutrophil egress from the bone marrow and reveal a new therapeutic target to curtail neutrophil-mediated inflammation in pulmonary vascular disease.

马氏珠母贝（Pinctada fucata martensii）是生产海水珍珠主要物种。通过植核手术生产珍珠在世界范围内备受关注,马氏珠母贝生产的珍珠占珍珠产量的90%以上。我们通过群体选育培育了红、黄、黑与白壳色群体。前期研究表明,红色、黄色、黑色、白色和棕色（最常见）壳色群体表现出不同的生长和繁殖能力,例如植入核后黑色壳色群体具有较高的成活率、留核率等。本研究中,以对照养殖群体为参考,比较了植核后黑壳色群体免疫相关基因的表达量、免疫酶活力与产珠性能差异。1.黑壳色群体和对照群体植核后的存活率、留核率和免疫酶活性比较本研究比较了BG和cG的总类胡萝卜素含量（Tcc）。对BG进行了植核实验。比较移植后不同时间点BG与cG的存活率、留珠率、免疫力和抗氧化能力。结果表明,BG组的Tcc显著大于对照组（P＜0.05）。植核后第7、30、360天,BG组存活率和留珠率均显著高于cG组（P＜0.05）。360天时,BS组珍珠厚度显著大于cG组（P＜0.05）。BG在植核后0h、12h、1d、3d、5d、7d和30d的AcP、AKP、SOD、cAT、TAOc和LZ活性均高于cG组。与cG相比,Fascin、SOD、cDK-7、cDAP-1、IRAK-1、GST2M、α-1、TRAF-3和caspase-2的m RNA在BG组高表达。2.不同年龄黑壳色选群体的产珠性能、酶活性及免疫相关基因表达的比较本研究评估了年龄对黑壳色选群体产珠性能和先天免疫反应的影响。对两组（A组,1.5龄;B组,2.5龄）进行植核手术。于植核后3d检测两组的免疫酶活性、抗氧化活性和免疫相关基因的表达水平。在珍珠收获时比较两组的存活率、保留率和生物矿化能力。结果表明,植核后3d,A组碱性磷酸酶（AKP）、酸性磷酸酶（AcP）、超氧化物歧化酶（SOD）、过氧化氢酶（cAT）、谷胱甘肽过氧化物酶（GPx）活性及SOD、GPx、HSP70、HSP90基因表达均明显高于B组（P＜0.05）。A组在第3、7、30天和采珠时的存活率均显著高于B组。收获珍珠时,A组的留珠率和珍珠层厚度显著高于B组。除Piff-177外,A组生物矿化基因Nacrein和Pearlin的表达水平均显著高于B组（P＜0.05）。3.硫氧还蛋白样蛋白-5的特性及植核后两个群体的差异分析硫氧还蛋白样蛋白5（Trxlp-5）是一种硫氧还蛋白亚型,通过硫醇二硫还原酶的活性参与细胞氧化还原动态平衡。在本研究中,克隆Trxlp-5马氏珠母贝基因全长,检测Poly（I:c）和LPS刺激后Pm Trxlp-5的表达,并比较黑壳色群体（BS）与对照群体（cG）植核后Pm Trxlp-5的表达差异。Pm Trxlp-5开放阅读框（ORF）长1167bp,编码388个氨基酸,5‘-UTR为41bp,3’-UTR为846bp。Pm Trxlp-5显示了一个保守的Wcxxc功能基序,与其他物种的硫氧还蛋白相似。组织分析显示Pm Trxlp-5m RNA在血细胞中的相对表达量最高。有趣的是,移植后,血液中Pm Trxlp-5的m RNA表达在移植后6h达到峰值,表明该基因在移植后的早期高度参与了免疫反应。在移植后24h、6d和30d,BS组的表达显著高于cG组。Pm Trxlp-5经LPS和Poly I:c处理后,m RNA表达也呈波浪型。这些结果表明,Pm Trxlp-5在细胞氧化还原、动态平衡和免疫应答中起重要作用。4.细胞周期蛋白依赖性激酶-7的特性及及植核后两个群体的差异分析细胞周期蛋白依赖性激酶-7（cDK-7）是一种参与细胞周期、细胞分化和增殖、凋亡和炎症反应的蛋白激酶。本研究克隆了马氏珠母贝Pm cdk-7的全长c DNA序列。Pm cdk-7全长1473bp,开放阅读框（ORF）915bp,编码304个氨基酸,5’-UTR为58bp,3’-UTR为500bp。Pm cdk-7的推导氨基酸序列包括丝氨酸/苏氨酸蛋白激酶、具有蛋白激酶ATP结合区特征的催化（S＿TKc）结构域（14-38aa）和结构域内的丝氨酸/苏氨酸蛋白激酶活性位点特征（129-141aa）。组织分布分析显示Pm cdk-7m RNA在血细胞中有较高的相对表达量。有趣的是,移植手术后,Pm cdk-7m RNA在BS组和cG组之间差异表达。移植后各时间点BS均较cG高表达,在3d达到高峰,提示其DNA复制和炎症反应增强。在LPS和poly I:c刺激后,Poly I:c组Pm cdk-7相对表达在24h高表达,12h下调,LPS组18h高表达,24h表达最低。我们的研究结果表明,Pm cdk-7通过帮助DNA复制在细胞周期中发挥重要作用,也有助于对由于移植手术切口和长期暴露于免疫刺激剂（病原体）而产生的炎症反应。

conversion of Australian Food composition Data from AUSNUT1999 to 2007 in the clinical Trial context

An Australian food composition database, AUSNUT1999, does not include long chain omega-3 polyunsaturated fatty acid （Lc omega-3 PUFA） data. Measurement of the fatty acid content of diets initially analysed using AUSNUT1999 requires conversion to AUSNUT2007, an updated database inclusive of Lc omega-3 PUFA. The aim of this study was to convert clinical trial dietary data from AUSNUT1999 to AUSNUT2007 and measure Lc omega-3 PUFA intake. clinical trial diet history （DH） data was converted from AUSNUTI999 to 2007 using a staged approach. Macronutrient intake from AUSNUTI999 and 2007 were calculated and compared via paired t-tests and Wilcoxon Signed Ranks tests. Mean dietary Lc omega-3 PUFA intake and the percentage contribution of food groups to total Lc omega-3 PUFA were then calculated. DHs were collected at baseline （n = 118）, three months （n = 86）, and 12 months （n = 64）. The accuracy of the conversion process improved with time, with no significant difference between most macronutrients at 12 months. Mean Lc n-3 PUFA intake was 441.87 mg at baseline, 521.07 mg at 3 months, and 442.40 mg at 12 months, and was predominantly provided by fish and seafood, followed by meat products. This study allowed for the measurement of Lc omega-3 intake, which was previously impossible using the AUSNUT 1999 database.

Data on global population distribution are a strategic resource currently in high demand in an age of new Development Agendas that call for universal inclusiveness of ***,quality,detail,and age of census data varies significantly by country and suffers from shortcomings that propagate to derived population grids and their *** this work,the improved capabilities of recent remote sensing-derived global settlement data to detect and mitigate major discrepancies with census data is *** layers mapping builtup presence were used to revise census units deemed as‘unpopulated’and to harmonize population distribution along *** procedures to detect and mitigate these anomalies,while minimizing changes to census geometry,preserving the regional distribution of population,and the overall counts were developed,tested,and *** two procedures employed for the detection of deficiencies in global census data obtained high rates of true positives,after verification and *** also show that the targeted anomalies were significantly mitigated and are encouraging for further uses of free and open geospatial data derived from remote sensing in complementing and improving conventional sources of fundamental population statistics.

Association between inherited monogenic liver disorders and chronic hepatitis c

AIM:To determine the frequencies of mutations that cause inherited monogenic liver disorders in patients with chronic hepatitis c. METHODS:This study included 86 patients with chronic hepatitis c(55 men, 31 women; mean age at diagnosis, 38.36 ± 14.52 years) who had undergone antiviral therapy comprising pegylated interferon and ribavirin. Viral load, biochemical parameter changes, and liver biopsy morphological data were evaluated in all patients. The control group comprised 271 unrelated individuals representing the general population of Latvia for mutation frequency calculations. The most frequent mutations that cause inherited liver disorders [gene(mutation): ATP7B(H1069Q), HFE(c282Y, H63D),UGT1A1(TA)7, and SERPINA1(PiZ)] were detected by polymerase chain reaction(PcR), bidirectional PcR allele-specific amplification, restriction fragment length polymorphism analysis, and sequencing. RESULTS: The viral genotype was detected in 80 of the 86 patients. Viral genotypes 1, 2, and 3 were present in 61(76%), 7(9%), and 12(15%) patients, respectively. Among all 86 patients, 50(58%) reached an early viral response and 70(81%) reached a sustained viral response. All 16 patients who did not reach a sustained viral response had viral genotype 1. casecontrol analysis revealed a statistically significant difference in only the H1069Q mutation between patients and controls(patients, 0.057; controls, 0.012; odds ratio, 5.514; 95%cI: 1.119-29.827, P = 0.022). However, the H1069Q mutation was not associated with antiviral treatment outcomes or biochemical indices. The(TA) 7 mutation of the UGT1A1 gene was associated with decreased ferritin levels(beta regression coefficient =-295.7, P = 0.0087). cONcLUSION: Genetic mutations that cause inherited liver diseases in patients with hepatitis c should be studied in detail.
