Rice stripe virus （RSV） causes severe diseases in Oryza sativa （rice） in many Eastern Asian countries. Diseasespecific protein （SP） of RSV is a non-structural protein and its accumulation level in rice plant was shown to determine the severity of RSV symptoms. Here, we present evidence that expression of RSV SP alone in rice or Nicotiana benthamiana did not produce visible symptoms. Expression of SP in these two plants, however, enhanced RSV- or Potato virus X （PVX）- induced symptoms. Through yeast two-hybrid screening, GST pull-down, and bimolecular fluorescence complementation assays, we demonstrated that RSV SP interacted with PsbP, a 23-kDa oxygen-evolving complex protein, in both rice and N. benthamiana. Furthermore, our investigation showed that silencing of PsbP expression in both plants increased disease symptom severity and virus accumulation. Confocal microscopy using N, benthamiana protoplast showed that PsbP accu- mulated predominantly in chloroplast in wild-type N. benthamiana cells. In the presence of RSV SP, most PsbP was recruited into cytoplasm of the assayed cells. In addition, accumulation of SP during RSV infection resulted in alterations of chloroplast structure and function. Our findings shed light on the molecular mechanism underlying RSV disease symptom development.

Phycocyanin attenuates X-ray-induced pulmonary inflammation via the TLR2-MyD88-NF-κB signaling pathway

Phycocyanin (PC), a natural algal protein, is reported for having anti-oxidant and antiinfl ammatory properties. We investigated its ability to attenuate lung infl ammation in mice subjected to X-ray radiation. Male C57BL/6 mice were assigned to the control, total body irradiation, PC pretreatment, and PC treatment groups. Mice in the PC pretreatment group were gavaged with 200 mg/kg PC for 7 consecutive days before irradiation, and those in the PC treatment group were gavaged with 200 mg/kg PC for 7 consecutive days after irradiation. lungs were collected on Day 7 after irradiation exposure. Hematoxylin and eosin staining of mouse lung sections showed considerable infl ammation damage 7 days after irradiation compared with the control lung but a reduction in pathological injury in the PC treatment group. Pretreatment or treatment with PC signifi cantly decreased levels of interleukin-6 and tumor necrosis factor-α in the lung, and also increased the relative mRNA expression of superoxide dismutase and glutathione. In vivo, PC signifi cantly reduced the expression of Toll-like receptor TLR2, myeloid diff erentiation primary response Myd88, and nuclear factor NF-κB, at both the transcriptional and translation level. Taken together, these data indicated that PC attenuated lung infl ammatory damage induced by radiation by blocking the TLR2- MyD88-NF-κB signaling pathway. Therefore, PC could be a protective agent against radiation-induced infl ammatory damage in normal tissues.

The liver, which is a metabolic organ, plays a pivotal role in tolerance induction. Hepatic stellate cells(Hp SCs), which are unique non-parenchymal cells, exert potent immunoregulatory activity during cotransplantation with allogeneic islets effectively protecting the islet allografts from rejection. Multiple mechanisms participate in the immune tolerance induced by Hp SCs, including the marked expansion of myeloid-derived suppressor cells(MDSCs), attenuation of effector T cell functions and augmentation of regulatory T cells. Hp SC conditioned MDSC-based immunotherapy has been conducted in mice with autoimmune disease and the results show that this technique may be promising. This article demonstrates how Hp SCs orchestrate both innate immunity and adaptive immunity to build a negative network that leads to immune tolerance.

Monoterpenoids are typically present in the secretory tissues of higher plants,and their biosynthesis is catalyzed by the action of monoterpene synthases(MTSs).However,the knowledge about these enzymes is restricted in a few plant *** are responsible for the complex cyclization of monoterpene precursors,resulting in the production of diverse monoterpene *** enzymatic reactions are considered exceptionally complex in ***,it is crucial to understand the catalytic mechanism of MTSs to elucidate their ability to produce diverse or specific monoterpenoid *** our study,we analyzed thirteen genomes of Dipterocarpaceae and identified 38 MTSs that generate a variety of monoterpene *** focusing on four MTSs with different product spectra and analyzing the formation mechanism of acyclic,monocyclic and bicyclic products in MTSs,we observed that even a single amino acid mutation can change the specificity and diversity of MTS products,which is due to the synergistic effect between the shape of the active cavity and the stabilization of carbon-positive intermediates that the mutation ***,residues N340,I448,and phosphoric acid groups were found to be significant contributors to the stabilization of intermediate terpinyl and pinene *** in these residues,either directly or indirectly,can impact the synthesis of single monoterpenes or their *** revealing the role of key residues in the catalytic process and establishing the interaction model between specific residues and complex monoterpenes in MTSs,it will be possible to reasonably design and engineer different catalytic activities into existing MTSs,laying a foundation for the artificial design and industrial application of MTSs.

Over the past years, it has been found that the epigenetic silence of tumor suppressor genes induced by overexpression of histone deacetylases （HDACs） plays an important role in carcinogenesis. Thus, HDAC inhibitors have emerged as the accessory therapeutic agents for multiple human cancers, since they can block the activity of specific HDACs, restore the expression of some tumor suppressor genes and induce cell differentiation, growth arrest and apoptosis. To date, the precise mechanisms by which HDAC inhibitors induce cell death have not yet been fully elucidated and the roles of individual HDAC inhibitors have not been identified. Moreover, the practical uses of HDAC inhibitors in cancer therapy, as well as their synergistic effects with other therapeutic strategies are yet to be evaluated. In this review article, we discuss briefly the recent advances in studies of the developments of anti-cancer HDAC inhibitors and their potential clinical value.

Geochronology and Geochemical Characteristics of Intrusion in the Jinchangliang Gold Deposit,Inner Mongolia and Their Tectonic Significance

Discovered and mined in recent years, the Jinchangliang gold deposit has not yet been studied in its genetic type. In this paper, the geological features of ore deposit, S isotopic composition, metallogenic age and elements geochemical of the granite closely related to mineralization were discussed. The results of the geological features of ore deposit and S isotopic composition show that ore-bearing hydrothermal solution was closely related with the intrusion of magmatic. The granite is characteristic of high silica SiO2=72.38%–72.98%, high aluminum and Al2O3=14.22%–14.35%, low calcium CaO=0.16% –0.26%, and low value of FeOT/MgO（6.86–7.73）, and rich in alkalis Na2O+K2O=9.11%–9.24%, suggesting that it is high-K calc-alkaline, highly fractionated, weak aluminum A-type granite. The REE patterns are inclined to right and show intense fractionation between LREE and HREE, without obvious negative Eu anomaly（δEu=0.80–0.84）. The primitive mantle-normalized spidergrams are characterized by depletion of Ba, U, Ta, Nb, Zr, Ti and P, which implies that the granite has the characteristics of the crust-mantle mixing. S isotopes also indicate that the material source of gold deposit is closely related to the granite rocks. The LA-ICP-MS Zircon UPb age of the Damiao rock mass medium-fine grained monzogranite（belonging to the early IndoChina） is（245±1） Ma. It shows that Jinchangliang gold deposit was not formed in Yanshanian, but the early Indo-China. Specifically speaking, the deposit was formed in the collision stage of the North China plate and the Siberian plate.

Hepatitis B virus (HBV)-induced hepatocellular carcinoma (HCC) is a major cause of cancer-related deaths in Asia and Africa. Developing effective and non-invasive biomarkers of HCC for individual patients remains an urgent task for early diagnosis and convenient monitoring. Analyzing the transcriptomic profiles of peripheral blood mononuclear cells from both healthy donors and patients with chronic HBV infection in different states (i.e. HBV carrier, chronic hepatitis B, cirrhosis, and HCC), we identified a set of 19 candidate genes according to our algorithm of dynamic network biomarkers. These genes can both characterize different stages during HCC progression and identify cirrhosis as the critical transition stage before carcinogenesis. The interaction effects (i.e. coexpressions) of candidate genes were used to build an accurate prediction model: the so-called edge-based biomarker. Considering the convenience and robustness of biomarkers in clinical applications, we performed functional analysis, validated candidate genes in other independent samples of our collected cohort, and finally selected COL5A1, HLA-DQB1, MMP2, and CDK4 to build edge panel as prediction models. We demonstrated that the edge panel had great performance in both diagnosis and prognosis in terms of precision and specificity for HCC, especially for patients with alpha-fetoprotein-negative HCC. Our study not only provides a novel edge-based biomarker for non-invasive and effective diagnosis of HBV-associated HCC to each individual patient but also introduces a new way to integrate the interaction terms of individual molecules for clinical diagnosis and prognosis from the network and dynamics perspectives.

Background:Through long-term research on Huangjiu fermentation,it has been found that the quality of Huangjiu is closely associated with Chinese 24-solar ***,this study was conducted to explore the characteristic and property index differences of Huangjiu,such as aroma and flavor profile,and physicochemical properties—brewed in different solar terms by choosing five representative fermentation times of Shaoxing ***:Huangjiu samples in the current study all met the national standards of traditional semi-dry *** are significant differences in physicochemical properties such as acidity and amino nitrogen among different solar-term *** three detected volatiles were applied by principal component analysis and partial least squares discriminant analysis analyses to differentiate the main *** mainly loaded to four principal components,which accounted for 86.5%.Nineteen volatiles were discriminated to significantly differentiate solar-term *** activity values(OAV)analysis found 14 compounds with OAV>1,while correlation analysis between volatiles and the outcomes of sensory evaluation displayed the various properties of Huangjiu on aroma and flavor due to the various combinations of volatiles,reducing sugar,acidity and amino *** a national wine inspector evaluated and scored Huangjiu fermented in different solar terms,Huangjiu brewed in Winter Solstice exhibited the highest performance,with a score of 91.0,and praised as a kind of gold medal ***:The methodology of this study can help to produce more types of appealing flavor and aroma of alcoholic beverages to consumers,build varietal Huangjiu or other alcoholic beverages through fermenting guidance by solar term,and even expand the applications of the traditional Chinese 24-solar term.
