Developing a mucosal vaccine against SARS-CoV-2 is critical for combatting the ***,we investigated long-term immune responses and protection against SARS-CoV-2 for the intranasal vaccination of a triple receptor-binding domain(RBD)scaffold protein(3R-NC)adjuvanted with a flagellin protein(KFD)(3R-NC+KFDi.n).In mice,the vaccination elicited RBD-specific broad-neutralizing antibody responses in both serum and mucosal sites sustained at high level over a *** long-lasting humoral immunity was correlated with the presence of long-lived RBD-specific IgG-and IgA-producing plasma cells,alongside the Th17 and Tfh17-biased T-cell responses driven by the KFD *** upon these preclinical findings,an open labeled clinical trial was conducted in individuals who had been primed with the inactivated SARS-CoV-2(IAV)*** a favorable safety profile,the 3R-NC+KFDi.n boost elicited enduring broad-neutralizing IgG in plasma and IgA in salivary *** meet the challenge of frequently emerged variants,we further designed an updated triple-RBD scaffold protein with mutated RBD combinations,which can induce adaptable antibody responses to neutralize the newly emerging variants,including *** findings highlight the potential of the KFD-adjuvanted triple-RBD scaffold protein is a promising prototype for the development of a mucosal vaccine against SARS-CoV-2 infection.

Capsaicin alleviates the hepatic clock gene disruption and gut microbiota dysbiosis in circadian rhythm disorder mouse model

As the body’s internal clock,the circadian rhythm regulates the energy expenditure,appetite,and *** exists a close relationship between the host circadian rhythm and gut *** this work,a circadian disorder mouse model induced by constant darkness(CD)was constructed to investigate the regulating effects of capsaicin(CAP)on disturbances of metabolism homeostasis and gut microbiota in the respect of circadian rhythm-related *** results indicated that CAP reduced weight gain induced by circadian rhythm disorder in mice by inhibiting fat accumulation in liver and adipose *** rhythmic expressions of circadian clock genes and lipid-metabolism related genes in liver were also recovered by *** study using 16S rRNA sequencing revealed that CAP modulated the gut microbiota richness,diversity and composition,and restored diurnal oscillations of gut microbes at the phylum and family *** results indicated that CAP could alleviate CD-induced hepatic clock gene disruption and gut microbiota dysbiosis in mice,providing theoretical basis for CAP to be used as a muti-functional ingredient with great healthpromoting effects.

The rich phase behavior of block copolymers(BCPs)has drawn great attention in recent ***,the double diamond(DD)phase is rarely obtained because of the competition between the minimization of interfacial energy and packing ***,a rod-coil BCP containing mesogen-jacketed liquid crystalline polymer is designed to acquire ordered bicontinuous network *** reduction of internal energy originating from the orientational interaction among the rod blocks can compensate for the free energy penalty of packing frustration to stabilize the DD *** resulting BCP can also experience lamellae-to-DD and double gyroid-to-lamellae transitions by changing the annealing *** results make the rod-coil BCP an excellent candidate for the self-assembly of ordered network structures,demonstrating great potential in nanopatterning and metamaterials.

细胞内存在大量性质各异的大分子复合物,它们是控制生命活动的核心信号枢纽。荧光显微成像因其分子特异性、细胞原位可视化和多色标记解析等优势,已成为当今生物学研究不可或缺的技术手段。而突破光学衍射极限的超分辨光学成像技术为进一步理解多种重要生物百纳米信号体的原位结构和功能调节提供了亚细胞尺度甚至单分子精度的研究思路和方案。首先阐述了超分辨成像和单分子定位显微镜的基本原理,介绍了近年来在多项生命科学研究中的代表性应用案例,结合实际研究经验总结了超分辨光学成像技术在使用中面临的诸多挑战和未来发展方向,提出了基于原位纳米成像解析信号枢纽组织结构将有力促进新型疾病治疗靶点和策略的开发。

The coronavirus disease 2019(COVID-19)pandemic,triggered by the novel coronavirus severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),has ravaged the globe,resulting in a staggering loss of life and wreaking havoc on the worldwide economy.[1,2]Sepsis is a cascade of abnormal responses provoked by infection,leading to a critical deterioration in organ function that poses a life-threatening risk.[3]However,it is unclear from published reports whether COVID-19 and sepsis are commonly aff ected by molecular ***,we performed a bioinformatics analysis to uncover shared diagnostic genes and potential mechanisms between COVID-19 and sepsis.

Higher palaeoelevation in the Baoshan Basin:Implications for landscape evolution at the southeastern margin of the Tibetan Plateau

Surface uplift at the southeastern margin of the Tibetan Plateau has been widely studied,but more palaeoaltimetry data are required to better understand the elevation history of this geologically complex *** this study,fossil leaves of Abies(Pinaceae),a cool-temperate element,recovered from the latest Miocene-Pliocene Yangyi Formation of the southern Baoshan Basin,were used as a proxy to estimate the local *** on the regional modern altitude range(2100-4280 m)of the genus as well as regional temperature discrepancy(1.5℃)between the past and present,the palaeoelevation of the study area was calculated to be>2360 m above sea level as compared to 1670 m at *** result suggests that the southern Baoshan Basin experienced pronounced uplift prior to the time of fossil deposition,probably as a result of crustal shortening and thickening of the northern Baoshan Terrane during the *** infer that surface growth in areas south of the Dali Basin may have been greater than previously interpreted,and that a widespread plateau or plateau patches higher than 2000 m probably extended southwards into at least the Baoshan Basin by the latest *** also infer that the elevation of the southern Baoshan Basin has decreased by at least 690 m since then,in contrast to most other scenarios in which the elevation of the southeastern margin of the Tibetan Plateau has increased or remained close to modern levels since the late *** major cause of the inferred altitude decline is likely tectonic *** a transtensional graben basin,the Baoshan Basin has experienced pull-apart and base-fall movement since the late Miocene,which would reduce the altitude of its southern part located on the hanging *** erosion associated with the increased summer rainfall might also have played a role especially in reducing the local relief,although its contribution can be *** study provides one of the few palaeoelevation

The organ-specific toxicity resulting from microplastic(MP)exposure has been extensively explored,particularly concerning the gut,liver,testis,and ***,under natural conditions,these effects are not restricted to specific organs or *** whether MP exposure presents a systemic threat to an entire organism,impacting factors such as lifespan,sleep,and fecundity,is *** this study,we investigated the effects of dietary exposure to two different doses of MPs(1–5μm)using the terrestrial model organism Drosophila *** indicated that the particles caused gut damage and remained within the digestive *** MP exposure significantly shortened the lifespan of adult *** short-term exposure disrupted sleep patterns,increasing the length of daytime sleep ***,one week of MP exposure reduced ovary size,with a trend towards decreased egg-laying in mated *** MPs did not penetrate the brain or ovaries,transcriptome analysis revealed altered gene expression in these *** the ovary,Gene Ontology(GO)analysis indicated genotoxic effects impacting inflammation,circadian regulation,and metabolic processes,with significant impacts on extracellular structure-related *** the brain,GO analysis identified changes in pathways associated with proteolysis and carbohydrate ***,this study provides compelling evidence of the systemic negative effects of MP exposure,highlighting the urgent need to address and mitigate environmental MP pollution.

An improved simple model for α decay half-lives

In this paper,using theαparticle preformation probabilities Pαfrom Xu et al.[Xu and Ren,***.A 760,303(2005)],which were extracted by fitting experimental half-lives ofαdecay,based on a phenomenological harmonic oscillator potential model(HOPM)[Bayrak,j Phys G 47,025102(2020)],refitting 178αdecay half-lives of even-even nuclei obtained from the latest nuclear property table NUBASE2020,we obtain the only one adjustable parameter V0-162.6 MeV in the HOPM,i.e.,the depth of nuclear *** corresponding root-mean-square(rms)deviation isσ-***,to consider the contribution of centrifugal potential to unfavoredαdecay half-lives,adding a new term■(d and l are the adjustable parameter and orbital angular momentum carried away by emittedαparticle)to the logarithmic form of favoredαdecay half-lives under the HOPM framework,we propose an improved simple model(ISM)for calculating favored and unfavoredαdecay *** the experimental half-lives of 205 unfavoredαdecay,we obtain *** ISM is used to calculate the unfavoredαdecay half-lives of 128 odd-A and 77 odd-odd *** results improve by 54.2%and 53.6%,respectively,compared with *** addition,we extend the ISM to predict theαdecay half-lives of 144 nuclei with Z=117,118,119,and *** comparison,the improved model with eight parameters(DUR)proposed by Deng et al.[Deng,***.C 101,034307(2020)]and the modified universal decay law(MUDL)proposed by Soylu et al.[Soylu,***.A 1013,122221(2021)]are also *** predictions of these models and/or formulas are generally consistent with each other.
