OBJECTIVE: To investigate the antidepressant-likeeffect of active fraction of Polyrhachis vicina Roger(AFPR) in a rat depression model, and to elucidate the underlying mechanism.METHODS: AFPR was extracted with ethanol followed by petroleum ether. Its antidepressant-like effect was investigated in mice by tail suspension test(TST), forced swimming test(FST) and open field test(OPT). A repeated dose of reserpine(0.5 mg/kg, daily for 14 d) was used to establish a rat depression model. Fluoxetine was used as positive control agent. The effect of AFPR on reserpine-induced ptosis, hypothermia and akinesia, the levels of monoamines and their metabolites, and the activity of monoamine oxidase(MAO) in hippocampus and prefrontal cortex were determined.RESULTS: Administration of AFPR by gavage at 160 and 320 mg/kg significantly reduced the duration of immobility in the FST and TST, and did not affect locomotor activity in the OPT. In the reserpine-induced depression model, AFPR attenuated anhedonia, demonstrated by reversing hypothermia, akinesia and sucrose consumption. AFPR significantly increased the concentration of monoamines, including dopamine, serotonin, noradrenaline and acetylcholine.CONCLUSION: AFPR normalized the metabolism rates of noradrenaline, serotonin and dopamine,and the activity of MAO, which were altered by chronic reserpine exposure. The findings suggest that modulation of the monoaminergic neurotransmitter system likely underlies the antidepressant-like effect of AFPR.

OBJECTIVE:To investigate efficacy of Lidan Tang(LDT)on gallstone induced by high fat diet in mice,and to study its underlying mechanism.METHODS:Mice were fed with high fat diet every day and treated with LDT(9.01 times of human clinic dosage).Mice were randomly divided into 6 groups as control group,gallstone model group(high-fat diet),positive control ursodeoxycholic acid(UDCA)group(80 mg·kg^-1·d^-1,i.g.),LDT low dose group(6 kg/d,i.g.),LDT middle dose group(12 kg/d,i.g.),and LDT high dose group(24 kg/d,i.g.).The whole experiment was lasted for 4 weeks.The levels of ALT,AST,LDH,CHO,HDL-C and LDL-C in serum were measured,the pathological sections were observed by hematoxylin-eosin staining,the activities of antioxidant enzymes were measured by kits,and the proteins related to oxidative stress and lipid transport were detected by Western blot analysis.RESULTS:LDT could significantly reduce the contents of ALT and AST in serum and improve the pathological tissue of liver.LDT could significantly reduce the content of MDA and LPO,and increase the level of GSH and GSH-PX in liver tissue.The data of Western blot showed that LDT had antioxidant effect promoting Keap1/Nrf2 pathway and regulated the process of lipid transport,which was statistically significant.In addition,LDT treatment inhibited the expression of ATP-binding cassette transports ABCG5/8 in liver,and reduced cholesterol transport from the hepatocytes to the gallbladder.CONCLUSION:LDT has protective effect on gallstones induced by high fat diet in mice,which might be based on the protective effect on liver,including enhancing the antioxidant capacity of liver and reducing the production of lipid peroxides.

人无德不立,育人的根本在于立德。在新形势下,“立德树人”是高等教育的根本任务。在专业课的教学中融入思政教育是实现协同育人的主要途径。药物分析是我国药学及相关专业本科人才培养方案中的重要专业必修课之一。为深刻挖掘药物分析课程中的思政元素,更好地通过该课程发挥协同育人作用,响应习近平总书记提出的“把思想政治工作贯穿教育教学全过程,实现全程育人、全方位育人”的号召,本文首先结合药物分析课程思政教育的现状,探讨在该课程中开展思政教育的必要性,然后按章节对课程中思政元素进行凝练,最后对该课程思政建设实施进行探索,以期培养更多有理想、有道德、有文化、有纪律的高素质药学人才,为社会的健康事业发展作出贡献。

The biopharmaceutical industry contributes substantially to volatile organic compounds(VOCs)emissions,causing growing concerns and social developmental conflicts.This study conducted an on-site investigation of the process-based emission of VOCs from three biopharmaceutical enterprises.In the workshops of the three enterprises,26 VOCs were detected,which could be sorted into 4 classes:hydrocarbons,aromatic hydrocarbons,oxygen-containing compounds,and nitrogen-containing compounds.Ketones were the main components of waste gases,accounting for 44.13%-77.85%of the overall VOCs.Process-based source profiles were compiled for each process unit,with the fermentation and extraction units of tiamulin fumarate being the main source of VOC emissions.Dimethyl heptanone,vinyl acetate,diethylamine,propylene glycol methyl ether(PGME),and benzene were screened as priority pollutants through a fuzzy comprehensive evaluation system.Ground level concentration simulation results of the Gauss plume diffusion model demonstrated that the diffusivity of VOCs in the atmosphere was relatively high,indicating potential non-carcinogenic and carcinogenic risks 1.5-2 km downwind.Furthermore,the process-based formation potentials of ozone and secondary organic aerosols(SOAs)were determined and indicated that N-methyl-2-pyrrolidone,dimethyl heptanone,and PGME should be preferentially controlled to reduce the ozone formation potential,whereas the control of benzene and chlorobenzene should be prioritized to reduce the generation of SOAs.Our results provide a basis for understanding the characteristics of VOC emission by biopharmaceutical industries and their diffusion,potentially allowing the development of measures to reduce health risks and secondary pollution.

Bioremediation became a promising technology to resolve arsenic(As)contamination in aquatic environment.Since monoculture such as microalgae or bacteria was sensitive to environmental disturbance and vulnerable to contamination,green microalgae Chlorella vulgaris and arsenite(As(Ⅲ))-oxidizing bacteria Pseudomonas sp.SMS11 were co-cultured to construct algal-bacterial consortia in the current study.The effects of algae-bacteria(A:B)ratio and exposure As(Ⅲ)concentration on algal growth,As speciation and metabolomic profile were investigated.Algal growth arrested when treated with 100 mg/L As(Ⅲ)without the co-cultured bacteria.By contrast,co-cultured with strain SMS11 significantly enhanced As tolerance in C.vulgaris especially with A:B ratio of 1:10.All the As(Ⅲ)in culture media of the consortia were oxidized into As(Ⅴ)on day 7.Methylation of As was observed on day 14.Over 1% and 0.5% of total As were converted into dimethylarsinic acid(DMA)after 21days cultivation when the initial concentrations of As(Ⅲ)were 1 and 10 mg/L,respectively.Metabolomic analysis was further performed to reveal the response of consortia metabolites to external As(Ⅲ).The enriched metabolomic pathways were associated with carbohydrate,amino acid and energy metabolisms.Tricarboxylic acid cycle and glyoxylate and dicarboxylate metabolism were upregulated under As stress due to their biological functions on alleviating oxidative stress and protecting cells.Both carbohydrate and amino acid metabolisms provided precursors and potential substrates for energy production and cell protection under abiotic stress.Alterations of the pathways relevant to carbohydrate or amino acid metabolism were triggered by energy requirement.

Owing to the complex lithology of unconventional reservoirs,field interpreters usually need to provide a basis for interpretation using logging simulation models.Among the various detection tools that use nuclear sources,the detector response can reflect various types of information of the medium.The Monte Carlo method is one of the primary methods used to obtain nuclear detection responses in complex environments.However,this requires a computational process with extensive random sampling,consumes considerable resources,and does not provide real-time response results.Therefore,a novel fast forward computational method(FFCM)for nuclear measurement that uses volumetric detection constraints to rapidly calculate the detector response in various complex environments is proposed.First,the data library required for the FFCM is built by collecting the detection volume,detector counts,and flux sensitivity functions through a Monte Carlo simulation.Then,based on perturbation theory and the Rytov approximation,a model for the detector response is derived using the flux sensitivity function method and a one-group diffusion model.The environmental perturbation is constrained to optimize the model according to the tool structure and the impact of the formation and borehole within the effective detection volume.Finally,the method is applied to a neutron porosity tool for verification.In various complex simulation environments,the maximum relative error between the calculated porosity results of Monte Carlo and FFCM was 6.80%,with a rootmean-square error of 0.62 p.u.In field well applications,the formation porosity model obtained using FFCM was in good agreement with the model obtained by interpreters,which demonstrates the validity and accuracy of the proposed method.

The unfolded protein response(UPR)is a cellular stress response pathway activated when the endoplasmic reticulum,a crucial organelle for protein folding and modification,encounters an accumulation of unfolded or misfolded proteins.The UPR aims to restore endoplasmic reticulum homeostasis by enhancing protein folding capacity,reducing protein biosynthesis,and promoting protein degradation.It also plays a pivotal role in coordinating signaling cascades to determine cell fate and function in response to endoplasmic reticulum stress.Recent research has highlighted the significance of the UPR not only in maintaining endoplasmic reticulum homeostasis but also in influencing various physiological processes in the nervous system.Here,we provide an overview of recent findings that underscore the UPR’s involvement in preserving the function and viability of neuronal and myelinating cells under physiological conditions,and highlight the critical role of the UPR in brain development,memory storage,retinal cone development,myelination,and maintenance of myelin thickness.
