CRISPR/Cas9 technology is a powerful genome manipulation tool in ***,little is k.own about whether mRNA and protein of a target gene are completely cleared in homozygous *** study generated homozygous mutants of the insulin receptor gene 2(NlinR2)in the brown planthopper(Nilaparvata lugens)using CRISPR/Cas9 genome *** frameshift mutants,E5_D17 and E6_I7,differentiated towards long wings,but there were differences in wing morphology,with E5_D17 showing wing *** investigations revealed the presence of residual expression of NlinR2 mRNA in both mutants,as well as the occurrence of spliceosomes featuring exon sk.pping splicing in E5_***,the E5_D17 exhibited the detection of N-terminally truncated NlinR2 *** interference experiments indicated that the k.ock.own of NlinR2 expression in the E5_D17 mutant line increased the proportion of wing deformities from 11.1 to 65.6%,suggesting that the residual NlinR2 mRNA of the E5_D17 mutant might have retained some genetic *** results imply that systematic characterization of residual protein expression or function in CRISPR/Cas9-generated mutant lines is necessary for phenotypic interpretation.

The aim of this study was to investigate the oxidation reactivity and behavior of exhaust particulate matter(PM)from diesel *** samples from two diesel engines(1k.CY4102)with different emission levels were collected by a thermophoretic system and a quartz *** oxidation reactivity,oxidation behaviors,and physicochemical properties of the PM samples were analyzed using thermogravimetric analysis(TGA),high-resolution transmission electron microscopy(HRTEM),Fourier-transform infrared spectrometry(FTIR),and Raman *** results showed that there was a great difference in the oxidation reactivity of soot particles emitted by the two different diesel engines.A qualitative analysis of the factors influencing oxidation reactivity showed that the nanostructure,degree of graphitization,and relative concentration of aliphatic C—H functional groups were the most important factors,whereas no significant correlation was found between the primary particle size and activation energy of the diesel *** on the oxidation behavior analysis,the diesel soot particles exhibited both internal and surface oxidation modes during the oxidation *** oxidation was dominant during the initial stage,and as oxidation progressed,the mode gradually changed to internal *** oxidation mode of soot particles from the 1k.engine was significantly higher than that of CY4102.

Groundwater plays a critical role in maintaining ecological balance and ensuring social operations,especially in arid and semiarid regions(Hu et al.,2023;Han et al.,2022;Ma et al.,2014).The over-exploitation of groundwater,coupled with the influx of pollutants from human activities into the soil,significantly degrades the environment in certain regions(Jia et al.,2019;Famiglietti,2014;Giordano,2009;k.nik.w and k.ndy,2005;Bohlk.,2002;Shah et al.,2000).

Purpose To report the probable association of motility coupling post placement and late porous polyethylene implant exposure. Design Retrospective, observatio nal case series. Methods This was a retrospective analysis of 27 patients who ha d primary porous polyethylene orbital implantation from February 1999 to Novembe r 2000. Data on demographics, previous surgery, ocular diagnosis, type of surger y, size of the implant, and motility coupling post placement were collected. Com plications of porous polyethylene implants and implant exposure were documented. Results Among the 27 patients, 18 eyes (66.7%) received motility coupling post insertion after primary porous polyethylene implantation. Implant exposure occu rred in six (33.3%) of the 18 eyes with motility coupling post insertion. None of the eyes without insertion had implant exposure. The mean interval between po rous polyethylene implantation and motility coupling post placement for the impl ant exposure group (6 of 18)was 6.5±0.4months,which was not statistically signi ficant compared with 7.2 ±0.6 months in the nonexposure group (12 of 18). For t hese 6 cases of implant exposure, the mean interval between implantation and imp lant exposure was 24.2±11.8months, and the mean interval between pegging and ex posure was 17.6 ±11.7 months. Among these 6 patients, 4 underwent removal of ex posed porous polyethylene implants and reimplantation of hydroxyapatite implant or dermis fat reconstruction. Conclusions We found a trend (P=.07) of increasing risk.of porous polyethylene implant exposure with motility coupling post placem ent. Although the pegging group did not show a statistically significant higher rate of exposure comparedwith the nonpegging group,we believe that more care was needed when performingmotility coupling post placement. In addition, longer pos toperative follow up is needed after insertion of a motility coupling post.

Sequential.l.tin hypercube designs(SlHDs) have recently received great attention for computer experiments, with much of the research restricted to invariant spaces. The related systematic construction methods are inflexible, and algorithmic methods are ineffective for large designs. For designs in contracting spaces, systematic construction methods have not been investigated yet. This paper proposes a new method for constructing SlHDs via good lattice point sets in various experimental spaces. These designs are called sequential good lattice point(SGlP) sets. Moreover, we provide efficient approaches for identifying the(nearly)optimal SGlP sets under a given criterion. Combining the linear level permutation technique, we obtain a class of asymptotically optimal SlHDs in invariant spaces, where the l1-distance in each stage is either optimal or asymptotically optimal. Numerical results demonstrate that the SGlP set has a better space-filling property than the existing SlHDs in invariant spaces. It is also shown that SGlP sets have less computational complexity and more adaptability.

Bioremediation became a promising technology to resolve arsenic(As)contamination in aquatic *** monoculture such as microalgae or bacteria was sensitive to environmental disturbance and vulnerable to contamination,green microalgae Chlorella vulgaris and arsenite(As(Ⅲ))-oxidizing bacteria Pseudomonas ***11 were co-cultured to construct algal-bacterial consortia in the current *** effects of algae-bacteria(A:B)ratio and exposure As(Ⅲ)concentration on algal growth,As speciation and metabolomic profile were *** growth arrested when treated with 100 mg/l As(Ⅲ)without the co-cultured *** contrast,co-cultured with strain SMS11 significantly enhanced As tolerance in *** especially with A:B ratio of 1:*** the As(Ⅲ)in culture media of the consortia were oxidized into As(Ⅴ)on day *** of As was observed on day *** 1% and 0.5% of total As were converted into dimethylarsinic acid(DMA)after 21days cultivation when the initial concentrations of As(Ⅲ)were 1 and 10 mg/l,*** analysis was further performed to reveal the response of consortia metabolites to external As(Ⅲ).The enriched metabolomic pathways were associated with carbohydrate,amino acid and energy *** acid cycle and glyoxylate and dicarboxylate metabolism were upregulated under As stress due to their biological functions on alleviating oxidative stress and protecting *** carbohydrate and amino acid metabolisms provided precursors and potential substrates for energy production and cell protection under abiotic *** of the pathways relevant to carbohydrate or amino acid metabolism were triggered by energy requirement.
