Genetic and lipidomic analyses reveal the key role of lipid metabolism for cold tolerance in maize

Lipid remodeling is crucial for cold tolerance in plants.However,the precise alternations of lipidomics during cold responses remain elusive,especially in maize(Zea mays L.).In addition,the key genes responsible for cold tolerance in maize lipid metabolism have not been identified.Here,we integrate lipidomic,transcriptomic,and genetic analysis to determine the profile of lipid remodeling caused by cold stress.We find that the homeostasis of cellular lipid metabolism is essential for maintaining cold tolerance of maize.Also,we detect 210 lipid species belonging to 13 major classes,covering phospholipids,glycerides,glycolipids,and free fatty acids.Various lipid metabolites undergo specific and selective alterations in response to cold stress,especially mono-/di-unsaturated lysophosphatidic acid,lysophosphatidylcholine,phosphatidylcholine,and phosphatidylinositol,as well as polyunsaturated phosphatidic acid,monogalactosyldiacylglycerol,diacylglycerol,and triacylglycerol.In addition,we identify a subset of key enzymes,including ketoacyl-acyl-carrier protein synthase II(KAS II),acyl-carrier protein 2(ACP2),male sterility33(Ms33),and stearoyl-acyl-carrier protein desaturase 2(SAD2)involved in glycerolipid biosynthetic pathways are positive regulators of maize cold tolerance.These results reveal a comprehensive lipidomic profile during the cold response of maize and provide genetic resources for enhancing cold tolerance in crops.

Design and test of the key components for a combing-type tobacco harvester

In response to the problems of low efficiency,high labor intensity,and low mechanization in manual tobacco harvesting,a comb-off tobacco picking device for southern hilly tobacco areas was designed following the agronomic requirements and the principle of manual picking of tobacco harvesting in southern China.The device was composed of the power chassis,picking mechanism,and storage mechanism.This study involved the theoretical analysis,structural design,and modeling of the key components,such as chassis structure,combing-type picking mechanism,and power synchronization mechanism.The results of the motion analysis and calculation of the picking process demonstrated that the adjustment range of the comb chain elevation angles was from 12.4°to 20.9°,the comb rod installation distance was 76.2 mm,and the synchronizing mechanism transmission ratio was 3:10.One-factor test and three-factor three-level orthogonal test was performed with the forward speed of the chassis,the distance between the picking device baffles and the elevation angle of the chain with the combing bar as test factors,and the rate of broken and missed tobacco picking as evaluation indicators.It was revealed that the optimal combination of the forward speed of the chassis,the distance between the baffles,and the chain elevation angle were 1.5 km/h,75 mm,and 12.4°,respectively.Moreover,verification tests suggested that the breakage rate of tobacco leaves was 9.99%,and the probability of missed tobacco picking was 7.31%,both of which satisfy the agriculture requirements and the operational requirements in the agricultural machinery certification syllabus.

Environmental,economic and exergy analysis of separation of ternary azeotrope by variable pressure extractive distillation based on multi-objective optimization

In this work,the ternary azeotrope of tert-butyl alcohol/ethyl acetate/water is separated by extractive distillation(ED)to recover the available constituents and protect the environment.Based on the conductor like shielding model and relative volatility method,ethylene glycol was selected as the extractant in the separation process.In addition,in view of the characteristic that the relative volatility between components changes with pressure,the multi-objective optimization method based on nondominated sorting genetic algorithm II optimizes the pressure and the amount of solvent cooperatively to avoid falling into the optimal local solution.Based on the optimal process parameters,the proposed heat-integrated process can reduce the gas emissions by 29.30%.The heat-integrated ED,further coupled with the pervaporation process,can reduce gas emission by 42.36%and has the highest exergy efficiency of 47.56%.In addition,based on the heat-integrated process,the proposed two heat pump assisted heat-integrated ED processes show good economic and environmental performance.The double heat pump assisted heat-integrated ED can reduce the total annual cost by 28.78%and the gas emissions by 55.83%compared with the basis process,which has a good application prospect.This work provides a feasible approach for the separation of ternary azeotropes.

Epigenetic pathways play a critical role in the initiation, progression, and metastasis of cancer. Over the past few decades, significant progress has been made in the development of targeted epigenetic modulators(e.g., inhibitors). However, epigenetic inhibitors have faced multiple challenges,including limited clinical efficacy, toxicities, lack of subtype selectivity, and drug resistance. As a result,the design of new epigenetic modulators(e.g., degraders) such as PROTACs, molecular glue, and hydrophobic tagging(Hy T) degraders has garnered significant attention from both academia and pharmaceutical industry, and numerous epigenetic degraders have been discovered in the past decade. In this review,we aim to provide an in-depth illustration of new degrading strategies(2017-2023) targeting epigenetic proteins for cancer therapy, focusing on the rational design, pharmacodynamics, pharmacokinetics, clinical status, and crystal structure information of these degraders. Importantly, we also provide deep insights into the potential challenges and corresponding remedies of this approach to drug design and development. Overall, we hope this review will offer a better mechanistic understanding and serve as a useful guide for the development of emerging epigenetic-targeting degraders.

Detecting bulk and edge exceptional points in non-Hermitian systems through generalized Petermann factors

Non-orthogonality in non-Hermitian quantum systems gives rise to tremendous exotic quantum phenomena,which can be fundamentally traced back to non-unitarity.In this paper,we introduce an interesting quantity(denoted asη)as a new variant of the Petermann factor to directly and efficiently measure non-unitarity and the associated non-Hermitian physics.By tuning the model parameters of underlying non-Hermitian systems,we find that the discontinuity of bothηand its first-order derivative(denoted as■η)pronouncedly captures rich physics that is fundamentally caused by non-unitarity.More concretely,in the 1D non-Hermitian topological systems,two mutually orthogonal edge states that are respectively localized on two boundaries become non-orthogonal in the vicinity of discontinuity ofηas a function of the model parameter,which is dubbed"edge state transition".Through theoretical analysis,we identify that the appearance of edge state transition indicates the existence of exceptional points(EPs)in topological edge states.Regarding the discontinuity of■η,we investigate a two-level non-Hermitian model and establish a connection between the points of discontinuity of■ηand EPs of bulk states.By studying this connection in more general lattice models,we find that some models have discontinuity of■η,implying the existence of EPs in bulk states.

Calcium-dependent protein kinase CPK32 mediates calcium signaling in regulating Arabidopsis flowering time

Appropriate flowering time is critical for the reproductive success of plant species.Emerging evidence indicates that calcium may play an important role in the regulation of flowering time.However,the underlying molecular mechanisms remain unclear.In this study,we demonstrate that calcium-dependent protein kinase 32(CPK32) regulates flowering time by affecting the alternative polyadenylation of FLOWERING CONTROL LOCUS A(FCA) and altering the transcription of FLO WERING LOCUS C(FLC),a central repressor of flowering time.The knockdown of CPK32 results in an obvious late flowering phenotype and dramatically enhanced FL C transcription.CPK32 interacts with FCA,and phosphorylates the serine592of FCA in a Ca2+-dependent manner.Moreover,the ratio of abundance of the FCA transcripts(FCA-D and FCA-P) changes significantly in the cpk32 mutant,which subsequently affects FLC expression and consequently regulates floral transition.The present evidence demonstrates that CPK32 modulates flowering time by regulating FCA alternative polyadenylation and consequent FL C expression.

Nasopharyngeal carcinoma(NPC)is a malignancy originating from the nasopharyngeal epithelial cells.Although some studies have identified potential biomarkers for early diagnosis of NPC based on metabolomics data(chen et al.,2023;Zhou et al.,2023)and Epstein-Barr virus subtypes(Xu et al.,2019),their effectiveness and applicability need further investigation,particularly when consideringthe roles of oral microbiota.

Cytokinin signaling promotes salt tolerance by modulating shoot chloride exclusion in maize

Excessive accumulation of chloride(Cl^(-))in the aboveground tissues under saline conditions is harmful to crops.Increasing the exclusion of Cl^(-) from shoots promotes salt tolerance in various crops.However,the underlying molecular mechanisms remain largely unknown.In this study,we demonstrated that a type A response regulator(ZmRR1)modulates Cl^(-) exclusion from shoots and underlies natural variation of salt tolerance in maize.ZmRR1 negatively regulates cytokinin signaling and salt tolerance,likely by interacting with and inhibiting His phosphotransfer(HP)proteins that are key mediators of cytokinin signaling.A naturally occurring non-synonymous SNP variant enhances the interaction between ZmRR1 and ZmHP2,conferring maize plants with a salt-hypersensitive phenotype.We found that ZmRR1 undergoes degradation under saline conditions,leading to the release of ZmHP2 from ZmRR1 inhibition,and subsequently ZmHP2-mediated signaling improves salt tolerance primarily by promoting Cl^(-) exclusion from shoots.Furthermore,we showed that ZmMATE29 is transcriptionally upregulated by ZmHP2-mediated signaling under highly saline conditions and encodes a tonoplast-located Cl^(-) transporter that promotes Cl^(-) exclusion from shoots by compartmentalizing Cl^(-) into the vacuoles of root cortex cells.Collectively,our study provides an important mechanistic understanding of the cytokinin signaling-mediated promotion of Cl^(-) exclusion from shoots and salt tolerance and suggests that genetic modification to promote Cl^(-) exclusion from shoots is a promising route for developing salt-tolerant maize.

Lithium–oxygen battery with ultrahigh theoretical energy density is considered a highly competitive next-generation energy storage device,but its practical application is severely hindered by issues such as difficult decomposition of discharge products at present.Here,we have developed N-doped carbon anchored atomically dispersed Ru sites cathode catalyst with open hollow structure(h-RuNC)for Lithium–oxygen battery.On one hand,the abundance of atomically dispersed Ru sites can effectively catalyze the formation and decomposition of discharge products,thereby greatly enhancing the redox kinetics.On the other hand,the open hollow structure not only enhances the mass activity of atomically dispersed Ru sites but also improves the diffusion efficiency of catalytic molecules.Therefore,the excellent activity from atomically dispersed Ru sites and the enhanced diffusion from open hollow structure respectively improve the redox kinetics and cycling stability,ultimately achieving a high-performance lithium–oxygen battery.

Tendon injuries often lead to joint dysfunction due to the limited self-regeneration capacity of tendons.Repairing tendons is a major challenge for surgeons and imposes a signif-icant financial burden on society.Therefore,there is an urgent need to develop effective stra-tegies for repairing injured tendons.Tendon tissue engineering using hydrogels has emerged as a promising approach that has attracted considerable interest.Hydrogels possess excellent biocompatibility and biodegradability,enabling them to create an extracellular matrix-like growth environment for cells.They can also serve as a carrier for cells or other substances to accelerate tendon repair.In the past decade,numerous studies have made significant prog-ress in the preparation of hydrogel scaffolds for tendon healing.This review aims to provide an overview of recent research on the materials of hydrogel-based scaffolds used for tendon tis-sue engineering and discusses the delivery systems based on them.
