Coal is one of the world’s main energy resources,accounting for approximately 68%of China’s current total power generation.However,several studies have demonstrated that dust,exhaust fumes,and other harmful factors in coal mines increase the risk of cardiovascular disease(CVD)among miners^([1]).Arterial stiffness(AS)is an independent risk factor of CVD,and epidemiological studies have shown that AS plays a vital role in assessing the risk of CVD^([2]).

Background:Findings on the association of genetic factors and colorectal cancer(CRC)survival are limited and inconsistent,and revealing the mechanism underlying their prognostic roles is of great importance.This study aimed to explore the relationship between functional genetic variations and the prognosis of CRC and further reveal the possible mechanism.Methods:We first systematically performed expression quantitative trait locus(eQTL)analysis using The Cancer Genome Atlas(TCGA)dataset.Then,the Kaplan-Meier analysis was used to filter out the survival-related eQTL target genes of CRC patients in two public datasets(TCGA and GSE39582 dataset from the Gene Expression Omnibus database).The seven most potentially functional eQTL single nucleotide polymorphisms(SNPs)associated with six survival-related eQTL target genes were genotyped in 907 Chinese CRC patients with clinical prognosis data.The regulatory mechanism of the survival-related SNP was further confirmed by functional experiments.Results:The rs71630754 regulating the expression of endoplasmic reticulum aminopeptidase 1(ERAP1)was significantly associated with the prognosis of CRC(additive model,hazard ratio[HR]:1.43,95%confidence interval[CI]:1.08-1.88,P=0.012).The results of dual-luciferase reporter assay and electrophoretic mobility shift assay showed that the A allele of the rs71630754 could increase the binding of transcription factor 3(TCF3)and subsequently reduce the expression of ERAP1.The results of bioinformatic analysis showed that lower expression of ERAP1 could affect the tumor immune microenvironment and was significantly associated with severe survival outcomes.Conclusion:The rs71630754 could influence the prognosis of CRC patients by regulating the expression of the immune-related gene ERAP1.Trial Registration:No.NCT00454519(https://clinicaltrials.gov/)

许多新型反应堆由于能谱较硬,核数据引起的不确定度已成为反应堆物理设计主要不确定度的来源。之前针对快能谱反应堆的不确定度分析主要集中在显式敏感性系数计算及不确定度分析,对隐式效应的分析较少,很少对隐式效应的影响给出分析。本文在深入研究反应堆截面处理方法的基础上,提出了一种新的基于本底截面迭代方法的截面隐式敏感性效应分析方法,由于该截面隐式敏感性系数计算主要与LN函数相关,因此也称为LN函数隐式敏感性计算方法。并利用基准题对新方法计算的截面隐式效应进行了分析,证明了方法的正确性。针对提出的五核素快谱基准题,采用多群截面计算,在共振自屏效应强烈的能群,部分核截面敏感性系数隐式效应修正最大可达50%,针对少群截面,对于大多数反应道,考虑隐式效应后计算得到的k eff相对敏感性系数和不确定度计算精度均有明显提高,但有部分反应道可能是由于Bondarenko方法和窄共振近似本身问题,导致敏感性系数和不确定度精度改善不明显。

Integrated interactome and transcriptome analysis reveals key host factors critical for SARS-CoV-2 infection

The coronavirus disease 2019(COVID-19)pandemic,caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),has seriously threatened global public health and caused huge economic losses.Omics studies of SARS-CoV-2 can help understand the interaction between the virus and host,thereby providing a new perspective in guiding the intervention and treatment of the SARS-CoV-2 infection.Since large amount of SARS-CoV-2 omics data have been accumulated in public databases,this study aimed to identify key host factors involved in SARSCoV-2 infection through systematic integration of transcriptome and interactome data.By manually curating published studies,we obtained a comprehensive SARS-CoV-2-human protein-protein interactions(PPIs)network,comprising 3591 human proteins interacting with 31 SARS-CoV-2 viral proteins.Using the RobustRankAggregation method,we identified 123 multiple cell line common genes(CLCGs),of which 115 up-regulated CLCGs showed host enhanced innate immunity and chemotactic response signatures.Combined with network analysis,co-expression and functional enrichment analysis,we discovered four key host factors involved in SARS-CoV-2 infection:IFITM1,SERPINE1,DDX60,and TNFAIP2.Furthermore,SERPINE1 was found to facilitate SARSCoV-2 replication,and can alleviate the endoplasmic reticulum(ER)stress induced by ORF8 protein through interaction with ORF8.Our findings highlight the importance of systematic integration analysis in understanding SARS-CoV-2-human interactions and provide valuable insights for future research on potential therapeutic targets against SARS-CoV-2 infection.

甲醛是一种对人体健康造成巨大威胁的污染物。气态甲醛浓度的准确检测对大气空气治理评估和室内环境安全均有重要意义,而高效传感器的研发则是甲醛分子检测技术优化的关键。本研究通过基于第一性原理的DFT计算软件VASP,对甲醛分子在Mn掺杂Zigzag(8,0)型单臂碳纳米管上的吸附特性进行研究。结果表明:Mn掺杂Zigzag(8,0)型单臂碳纳米管是一种稳定的分子构型。区别于在原始CNT上的物理吸附过程,甲醛分子在Mn掺杂CNT上的吸附键长更短,吸附能更大,属于化学吸附过程。同时,甲醛分子在Mn掺杂CNT上的吸附过程还伴随着明显的电荷转移。吸附过程发生后,吸附样品的光吸收曲线在580~705 nm以及365~447 nm的范围内出现明显的蓝移,在307~360 nm的范围内出现了明显的红移。

Splicing factor proline and glutamine-rich is a prognostic biomarker and correlated with clinical pathologic features and immune infiltrates in hepatocellular carcinoma

Background:Hepatocellular carcinoma(HCC)is the fourth leading cause of cancer-related deaths globally.Splicing factor proline and glutamine-rich(SFPQ)is a multifunctional protein that controls various biological functions.As a potential therapeutic target and a promising prognostic indicator,the potential effects and processes of SFPQ in HCC require further investigation.Methods:The RNA sequencing data were obtained from the Gene Expression Omnibus,International Cancer Genome Consortium,and The Cancer Genome Atlas databases to analyze SFPQ expression and differentially expressed genes(DEGs).We utilized the LinkedOmics database to identify co-expressed genes.A Venn diagram was constructed to determine the overlapping genes between the DEGs and the co-expressed genes.Functional enrichment analysis was performed on the overlapping genes and DEGs.Furthermore,our study involved functional enrichment analysis,a protein-protein interaction network analysis,and an analysis of immune cell infiltration.The cBioPortal and Tumor Immune Single-cell Hub were utilized to investigate the genetic alterations of SFPQ and the single-cell transcriptome visualization of the tumor microenvironment.A ceRNA network was established with the assistance of the ENCORI website.Finally,we elucidated the clinical significance of SFPQ in HCC by employing Kaplan-Meier survival analysis,univariate and multivariate Cox regression,and prognostic nomogram models.Results:The expression of SFPQ in HCC tissues was significantly elevated compared to normal tissues.GSEA results indicated that increased expression of SFPQ was associated with pathways related to HCC.The ceRNA network,including SFPQ,hsa-miR-101-3p,AC023043.4,AC124798.1,AC145207.5,and GSEC,was constructed with the assistance of ENCORI.High SFPQ expression was related to a poor prognosis in HCC and its subtypes.Univariate and multivariate Cox regression analysis showed that elevated SFPQ expression is an independent predictive factor.Conclusions:The overexpression of SFPQ may serve as a poten

The adenosine subfamily G protein-coupled receptors A_(2A)R and A_(2B)R have been identified as promising cancer immunotherapy candidates.One of the A_(2A)R/A_(2B)R dual antagonists,AB928,has progressed to a phaseⅡclinical trial to treat rectal cancer.However,the precise mechanism underlying its dual-antagonistic properties remains elusive.Herein,we report crystal structures of the A_(2A)R complexed with AB928 and a selective A_(2A)R antagonist 2-118.The structures revealed a common binding mode on A_(2A)R,wherein the ligands established extensive interactions with residues from the orthosteric and secondary pockets.In contrast,the cAMP assay and A_(2A)R and A_(2B)R molecular dynamics simulations indicated that the ligands adopted distinct binding modes on A_(2B)R.Detailed analysis of their chemical structures suggested that AB928 readily adapted to the A_(2B)R pocket,while 2-118 did not due to intrinsic differences.This disparity potentially accounted for the difference in inhibitory efficacy between A_(2B)R and A_(2A)R.This study serves as a valuable structural template for the future development of selective or dual inhibitors targeting A_(2A)R/A_(2B)R for cancer therapy.

Organic solar cells(OSCs)have gained conspicuous progress during the past few decades due to the development of materials and upgrading of the device structure.The power conversion efficiency(PCE)of the single-junction device had surpassed 19%.The cathode interface layer(CIL),by optimizing the connection between the active layer and the cathode electrode,has become a momentous part to strengthen the performances of the OSCs.Simultaneously,CIL is also indispensable to illustrating the working mechanism of OSCs and enhancing the stability of the OSCs.In this essay,hybrid CILs in OSCs have been summarized.Firstly,the advancement and operating mechanism of OSCs,and the effects and relevant design rules of CIL are briefly concluded;secondly,the significant influence of CIL on enhancing the stability and PCE of OSCs is presented;thirdly,the characteristics of organic hybrid CIL and organic-inorganic hybrid CIL are introduced.Finally,the conclusion and outlook of CIL are summarized.

Influence of acceleration stage electrode voltage on the performance of double-stage Hall effect thruster with adjustable zero magnetic point

The configuration of electrode voltage and zero magnetic point position has a significant impact on the performance of the double-stage Hall effect thruster. A 2D-3V model is established based on the two-magnetic peak type double-stage Hall thruster configuration, and a particle-in-cell simulation is carried out to investigate the influences of both acceleration electrode voltage value and zero magnetic point position on the thruster discharge characteristics and performances.The results indicate that increasing the acceleration voltage leads to a larger potential drop in the acceleration stage, allowing ions to gain higher energy, while electrons are easily absorbed by the intermediate electrode, resulting in a decrease in the anode current and ionization rate. When the acceleration voltage reaches 500 V, the thrust and efficiency are maximized, resulting in a 15%increase in efficiency. After the acceleration voltage exceeds 500 V, a potential barrier forms within the channel, leading to a decrease in thruster efficiency. Further study shows that as the second zero magnetic point moves towards the outlet of the channel, more electrons easily traverse the zero magnetic field region, participating in the ionization. The increase in the ionization rate leads to a gradual enhancement in both thrust and efficiency.
