Wide‑field swept‑source OCT angiography of the periarterial capillary‑free zone before and after anti‑VEGF therapy for branch retinal vein occlusion

Background:The aim of the study was to investigate the changes in the periarterial capillary-free zone(paCFZ)after anti-vascular endothelial growth factor(VEGF)therapy in patients with branch retinal vein occlusion(BRVO)by widefield swept-source optical coherence tomography angiography(SS-OCTA)and assess their associations with clinical outcomes.Methods:In this retrospective observational study of 54 treatment-naive BRVO patients with macular edema,we reviewed the findings of 12×12 mm^(2)SS-OCTA at baseline,3,6,and 12 months after intravitreal ranibizumab injections.The paCFZ and major retinal artery areas were measured on SS-OCTA images.The paCFZ area to artery area(P/A)ratio was calculated.Results:The paCFZ areas and P/A ratios of first-and second-order arteries were significantly greater in BRVO eyes than in contralateral eyes(all P<0.01),but there were no differences in the first-and second-order artery areas(P=0.20 and 0.25,respectively).The paCFZ areas and P/A ratios decreased significantly at 3,6,and 12 months after anti-VEGF therapy(all P<0.01).The baseline P/A ratio was significantly correlated with the baseline best-corrected visual acuity(BCVA),central retinal thickness,and their improvements at 3,6,and 12 months(all P<0.05).Baseline BCVA and P/A ratios of first-and second-order arteries were independently associated with the final BCVA in multivariate linear regression.Conclusions:Wide-field SS-OCTA shows that anti-VEGF therapy can lead to a significant improvement in the paCFZ parameters in BRVO.Smaller baseline P/A ratios on SS-OCTA tend to predict better visual outcomes at 12 months after anti-VEGF therapy.

Huoxue Jiedu Huayu recipe(活血解毒化瘀方) inhibits macrophage-secreted vascular endothelial growth factor-a on angiogenesis and alleviates renal fibrosis in the contralateral kidneys of unilateral ureteral obstruction rats

OBJECTIVE: To elucidate the mechanism by which Huoxue Jiedu Huayu recipe( 活血解毒化瘀方, HJHR) regulates angiogenesis in the contralateral kidney of unilateral ureteral obstruction(UUO) rats and the mechanism by which it reduces of renal fibrosis. METHODS: Male Wistar rats were randomly divided into 4 groups: the sham group, UUO group(180 d of left ureter ligation), UUO plus eplerenone(EPL) group, and UUO plus HJHR group. After 180 d of oral drug administration, blood and contralateral kidneys were collected for analysis. Angiogenesis-and fibrosis-related indexes were detected. RESULTS: HJHR and EPL improved structural damage and renal interstitial fibrosis in the contralateral kidney and reduced the protein expression levels of α-smooth muscle actin(α-SMA), vimentin and collagen I. Moreover, these treatments could reduce the expression of vascular endothelial growth factor-A(VEGFA) by inhibiting the infiltration of macrophages. Furthermore, HJHR and EPL significantly reduced the expression of CD34 and CD105 by downregulating VEGFA production, which inhibited angiogenesis. Finally, the coexpressions of CD34, CD105 and α-SMA were decreased in the HJHR and EPL groups, indicating that endothelial-to-mesenchymal transition was inhibited. CONCLUSIONS: These findings confirm that HJHR alleviates contralateral renal fibrosis by inhibiting VEGFAinduced angiogenesis, encourage the use of HJHR against renal interstitial fibrosis and provide a theoretical basis for the clinical management of patients with CKD.

The spatial and temporal effects of Fog-Haze pollution on the influenza transmission

The energy-intensive industrial economy results in frequent Fog-Haze weather which increases the risk of influenza spread and brings new challenges for the prevention and control of influenza.Therefore,the study of spread mechanics and controlling strategies of influenza based on Fog-Haze will be scientifically meaningful.Considering that when the concentration of Fog-Haze is low,Fog-Haze contributes to the number of infectious individuals and Fog-Haze suppresses the transmission of the influenza virus when the concentration of Fog-Haze is high,we establish the Fog-Haze dynamics model.Then we prove the global existence and boundedness of the solution,and the global asymptotic stability of the solution is given by constructing a suitable Lyapunov functional.Under the Fog-Haze weather,we study the inffuenza virus transmission model that incorporates the incidence rate to reflect the Fog-Haze-dependent saturation effect and investigate the effect of Fog-Haze pollution on the transmission of influenza.We show that the basic reproduction number R_(0) determines the global dynamics of the system:if R_(0)1.Simulations are carried out to validate the theoretical results.Our study provides further understanding of the dynamics of Fog-Haze and the effect of Fog-Haze pollution on the transmission of influenza.

p53 is mutated in half of cancer cases.However,no p53-targeting drugs have been approved.Here,we reposition decitabine for triple-negative breast cancer(TNBC),a subtype with frequent p53 mutations and extremely poor prognosis.In a retrospective study on tissue microarrays with 132 TNBC cases,DNMT1 overexpression was associated with p53 mutations(P=0.037)and poor overall survival(OS)(P=0.010).In a prospective DEciTabinE and Carboplatin in TNBC(DETECT)trial(NCT03295552),decitabine with carboplatin produced an objective response rate(ORR)of 42%in 12 patients with stage IV TNBC.Among the 9 trialed patients with available TP53 sequencing results,the 6 patients with p53 mutations had higher ORR(3/6 vs.0/3)and better OS(16.0 vs.4.0 months)than the patients with wild-type p53.In a mechanistic study,isogenic TNBC cell lines harboring DETECT-derived p53 mutations exhibited higher DNMT1 expression and decitabine sensitivity than the cell line with wild-type p53.In the DETECT trial,decitabine induced strong immune responses featuring the striking upregulation of the innate immune player IRF7 in the p53-mutated TNBC cell line(upregulation by 16-fold)and the most responsive patient with TNBC.Our integrative studies reveal the potential of repurposing decitabine for the treatment of p53-mutated TNBC and suggest IRF7 as a potential biomarker for decitabine-based treatments.

Research progress on short-term mechanical properties of FRP bars and FRP-reinforced concrete beams

Fiber-reinforced polymer(FRP)bars have been increasingly recognized in the field of civil engineering due to their advantages of light weight,high strength and excellent durability.FRP bars can replace steel bars in concrete beams and effectively improve the durability of beams.In this paper,the literature relevant to the short-term mechanical properties of FRP bars and FRP-reinforced concrete beams was reviewed based on previous studies and practical engineering application.Firstly,the mechanical properties of FRP bars were reviewed.Different types of fibers or steel and fibers can be combined to obtain hybrid fiber-reinforced polymer(HFRP)or steel-fiber composite bars(SFCB)with excellent mechanical performance,respectively.The bond performance and bond-slip model between FRP bars and concrete were discussed.Several common bond-slip models were usually used to study the bond performance between carbon fiber-reinforced polymer(CFRP)bars or glass fiber-reinforced polymer(GFRP)bars and concrete,but changing the type of FRP bars will lead to larger dispersion.Then,the experimental studies,theoretical calculation methods and finite element simulation methods of flexural/shear behavior of FRP-reinforced concrete beams were presented.Finally,their applications in practical engineering were discussed and the prospects of further research were proposed.It is pointed out that FRP-reinforced ultra-high performance concrete(UHPC)beams,FRP-reinforced geopolymer concrete(GPC)beams,engineered cementitious composites(ECC)-FRP-reinforced concrete beams,prestressed FRP-reinforced concrete beams and steel/FRP hybrid-reinforced concrete beams can effectively improve the deformation resistance and poor ductility of pure FRP-reinforced concrete beams.

Presently,integrating multi-omics information into a prediction model has become a ameliorate strategy for genomic selection to improve genomic prediction accuracy.Here,we set the genomic and transcriptomic data as the training population data,using BSLMM,TWAS,and eQTL mapping to prescreen features according to |β_(b)|>0,top 1%of phenotypic variation explained(PVE),expression-associated single nucleotide polymorphisms(eSNPs),and egenes(false discovery rate(FDR)<0.01),where these loci were set as extra fixed effects(named GBLUP-Fix)and random effects(GFBLUP)to improve the prediction accuracy in the validation population,respectively.The results suggested that both GBLUP-Fix and GFBLUP models could improve the accuracy of longissimus dorsi muscle(LDM),water holding capacity(WHC),shear force(SF),and pH in Huaxi cattle on average from 2.14 to 8.69%,especially the improvement of GFBLUP-TWAS over GBLUP was 13.66%for SF.These methods also captured more genetic variance than GBLUP.Our study confirmed that multi-omics-assisted large-effects loci prescreening could improve the accuracyofgenomic prediction.

Inverse-time Backup Protection Based on Unified Characteristic Equation for Distribution Networks with High Proportion of Distributed Generations

The setting work of backup protection using steady-state current is tedious,and mismatches occasionally occur due to the increased proportion of distributed generations(DGs)connected to the power grid.Thus,there is a practical need to study a backup protection technology that does not require step-by-step setting and can be adaptively coordinated.This paper proposes an action sequence adaptive to fault positions that uses only positive sequence fault component(PSFC)voltage.Considering the influence of DGs,the unified time dial setting can be obtained by selecting specific points.The protection performance is improved by using the adjacent upstream and downstream protections to meet the coordination time interval in the case of metallic faults at the near-and far-ends of the line.Finally,the expression and implementation scheme for inverse-time backup protection(ITBP)based on the unified characteristic equation is given.Simulation results show that this scheme can adapt to DG penetration scenarios and can realize the adaptive coordination of multi-level relays.

Cell replacement with stem cell-derived retinal ganglion cells from different protocols

Glaucoma,characterized by a degenerative loss of retinal ganglion cells,is the second leading cause of blindness worldwide.There is currently no cure for vision loss in glaucoma because retinal ganglion cells do not regenerate and are not replaced after injury.Human stem cell-derived retinal ganglion cell transplant is a potential therapeutic strategy for retinal ganglion cell degenerative diseases.In this review,we first discuss a 2D protocol for retinal ganglion cell differentiation from human stem cell culture,including a rapid protocol that can generate retinal ganglion cells in less than two weeks and focus on their transplantation outcomes.Next,we discuss using 3D retinal organoids for retinal ganglion cell transplantation,comparing cell suspensions and clusters.This review provides insight into current knowledge on human stem cell-derived retinal ganglion cell differentiation and transplantation,with an impact on the field of regenerative medicine and especially retinal ganglion cell degenerative diseases such as glaucoma and other optic neuropathies.

The chirality structure of a single-walled carbon nanotube(SWNT)strongly depends on the composition of catalyst used in the chemical vapor deposition process.In this study,we develop a porous magnesia supported manganese-rhenium(MnRe/MgO)catalyst for chirality-selective synthesis of SWNTs.Detailed characterizations reveal that(6,5)tubes with a selectivity higher than 70%are grown from the Re-rich MnRe/MgO catalyst.By comparing the SWNT growth results with those of monometallic Mn or Re,the formation of sigma phase,an intermetallic compound occurring in transition-metal alloy systems,is revealed to be crucial for the dominant synthesis of(6,5)SWNTs.This work not only extends the application of sigma phase alloy for catalytic synthesis of SWNTs,but also sheds lights on the growth of SWNTs with a high chirality selectivity.

Event-triggered H_(∞) consensus control for input-constrained multi-agent systems via reinforcement learning

This article presents an event-triggered H_(∞) consensus control scheme using reinforcement learning (RL) for nonlinear second-order multi-agent systems (MASs) with control constraints. First, considering control constraints, the constrained H_(∞) consensus problem is transformed into a multi-player zero-sum game with non-quadratic performance functions. Then, an event-triggered control method is presented to conserve communication resources and a new triggering condition is developed for each agent to make the triggering threshold independent of the disturbance attenuation level. To derive the optimal controller that can minimize the cost function in the case of worst disturbance, a constrained Hamilton–Jacobi–Bellman (HJB) equation is defined. Since it is difficult to solve analytically due to its strongly non-linearity, reinforcement learning (RL) is implemented to obtain the optimal controller. In specific, the optimal performance function and the worst-case disturbance are approximated by a time-triggered critic network;meanwhile, the optimal controller is approximated by event-triggered actor network. After that, Lyapunov analysis is utilized to prove the uniformly ultimately bounded (UUB) stability of the system and that the network weight errors are UUB. Finally, a simulation example is utilized to demonstrate the effectiveness of the control strategy provided.
