Juvenile hormone induces reproduction via miR-1175-3p in the red imported fire ant, Solenopsis invicta

Juvenile hormone(JH)acts in the regulation of caste differentiation between queens and workers(i.e.,with or without reproductive capacity)during vitellin synthesis and oogenesis in social insects.However,the regulatory mechanisms have not yet been elucidated.Here,we identified a highly expressed microRNA(miRNA),miR-1175-3p,in the red imported fire ant,Solenopsis invicta.We found that miR-1175-3p is prominently present in the fat bodies and ovaries of workers.Furthermore,miR-1175-3p interacts with its target gene,broad-complex core(Br-C),in the fat bodies.By utilizing miR-1175-3p agomir,we successfully suppressed the expression of the Br-C protein in queens,resulting in reduced vitellogenin expression,fewer eggs,and poorly developed ovaries.Conversely,decreasing miR-1175-3p levels led to the increased expression of Br-C and vitellogenin in workers,triggering the“re-development”of the ovaries.Moreover,when queens were fed with JH,the expression of miR-1175-3p decreased,whereas the expression of vitellogenin-2 and vitellogenin-3 increased.Notably,the suppression of fertility in queens caused by treatment with agomir miR-1175-3p was completely rescued by the increased vitellogenin expression induced by being fed with JH.These results suggest the critical role of miR-1175-3p in JH-regulated reproduction,shedding light on the molecular mechanism underlying miRNA-mediated fecundity in social insects and providing a novel strategy for managing S.invicta.

The temporal-spatial expression and functional analysis of three gustatory receptor genes in Solenopsis invicta using sweet and bitter compounds

The insect gustatory system participates in identifying potential food sources and avoiding toxic compounds.During this process,gustatory receptors(GRs)recognize feeding stimulant and deterrent compounds.However,the GRs involved in recognizing stimulant and deterrent compounds in the red imported fire ant,Solenopsis invicta,remain unknown.Therefore,we conducted a study on the genes SinvGR1,SinvGR32b,and SinvGR28a to investigate the roles of GRs in detecting feeding stimulant and deterrent compounds.In this current study,we found that sucrose and fructose are feeding stimulants and the bitter compound quinine is a feeding deterrent.The fire ant workers showed significant behavior changes to avoid the bitter taste in feeding stimulant compounds.Reverse transcription quantitative real-time polymerase chain reaction results from developmental stages showed that the SinvGR1,SinvGR32b,and SinvGR28a genes were highly expressed in fire ant workers.Tissue-specific expression profiles indicated that SinvGR1,SinvGR32b,and SinvGR28a were specifically expressed in the antennae and foreleg tarsi of workers,whereas SinvGR32b gene transcripts were also highly accumulated in the male antennae.Furthermore,the silencing of SinvGRI or SinvGR32b alone and the co-silencing of both genes disrupted worker stimulation and feeding on sucrose and fructose.The results also showed that SinvGR28a is required for avoiding quinine,as workers with knockdown of the SinvGR28a gene failed to avoid and fed on quinine.This study first identified stimulant and deterrent compounds of fire ant workers and then the GRs involved in the taste recognition of these compounds.This study could provide potential target gustatory genes for the control of the fire ant.

We investigate the asymptotic stability of the solutions to Boussinesq equations without heat conduction with the initial data near a specific stationary solution in the three-dimensional domain Ω=R^(2)×(0,1).It is shown that the solution starting from a small perturbation to the stationary solution converges to it with explicit algebraic rates as time tends to infinity.

Clinical observation on 90 cases of primary dysmenorrhea treated by buccal acupuncture therapy:a randomized controlled study

OBJECTIVE:To explore the short-term and long-term efficacy of buccal acupuncture therapy in the treatment of patients with primary dysmenorrhea(PD).METHODS:A total of 90 patients with PD who were admitted to the Second Affiliated Hospital of Zhejiang University of Traditional Chinese Medicine from January 2020 to December 2021 were included in this study and randomly divided into three groups,including the observation group,the control group,and the placebo group,with 30 patients in each group.Patients in the observation group were treated according to the syndrome types of Traditional Chinese Medicine,those in the control group were treated with Saridon,and those in the placebo group received a placebo.The visual analog scale(VAS)scores of these patients were evaluated at different time points(before treatment,5 min after needle retention,30 min after needle retention,1 h after treatment,2 h after treatment,1 d after treatment,and 2 d after treatment).The Cox Menstrual Symptom Scale(CMSS)scores were assessed before the treatment and after three courses of treatment.Besides,these two scales were also evaluated during follow-up.Moreover,the pain score and adverse reactions of these patients were assessed during treatment.RESULTS:After treatment,the VAS and CMSS scores in the observation group decreased significantly(P<0.05)5 and 30 min after needle retention and after treatment.CONCLUSION:Buccal acupuncture therapy is verified to have short-term efficacy in the treatment of PD.In addition,this therapy has an immediate analgesic effect and long-term efficacy in reducing the severity and frequency of PD.

Improved PSO-Extreme Learning Machine Algorithm for Indoor Localization

Wi Fi and fingerprinting localization method have been a hot topic in indoor positioning because of their universality and location-related features.The basic assumption of fingerprinting localization is that the received signal strength indication(RSSI)distance is accord with the location distance.Therefore,how to efficiently match the current RSSI of the user with the RSSI in the fingerprint database is the key to achieve high-accuracy localization.In this paper,a particle swarm optimization-extreme learning machine(PSO-ELM)algorithm is proposed on the basis of the original fingerprinting localization.Firstly,we collect the RSSI of the experimental area to construct the fingerprint database,and the ELM algorithm is applied to the online stages to determine the corresponding relation between the location of the terminal and the RSSI it receives.Secondly,PSO algorithm is used to improve the bias and weight of ELM neural network,and the global optimal results are obtained.Finally,extensive simulation results are presented.It is shown that the proposed algorithm can effectively reduce mean error of localization and improve positioning accuracy when compared with K-Nearest Neighbor(KNN),Kmeans and Back-propagation(BP)algorithms.

Investigations on High-Speed Flash Boiling Atomization of Fuel Based on Numerical Simulations

Flash boiling atomization(FBA)is a promising approach for enhancing spray atomization,which can generate a fine and more evenly distributed spray by increasing the fuel injection temperature or reducing the ambient pressure.However,when the outlet speed of the nozzle exceeds 400 m/s,investigating high-speed flash boiling atomization(HFBA)becomes quite challenging.This difficulty arises fromthe involvement ofmany complex physical processes and the requirement for a very fine mesh in numerical simulations.In this study,an HFBA model for gasoline direct injection(GDI)is established.This model incorporates primary and secondary atomization,as well as vaporization and boilingmodels,to describe the development process of the flash boiling spray.Compared to lowspeed FBA,these physical processes significantly impact HFBA.In this model,the Eulerian description is utilized for modeling the gas,and the Lagrangian description is applied to model the droplets,which effectively captures the movement of the droplets and avoids excessive mesh in the Eulerian coordinates.Under various conditions,numerical solutions of the Sauter mean diameter(SMD)for GDI show good agreement with experimental data,validating the proposed model’s performance.Simulations based on this HFBA model investigate the influences of fuel injection temperature and ambient pressure on the atomization process.Numerical analyses of the velocity field,temperature field,vapor mass fraction distribution,particle size distribution,and spray penetration length under different superheat degrees reveal that high injection temperature or low ambient pressure significantly affects the formation of small and dispersed droplet distribution.This effect is conducive to the refinement of spray particles and enhances atomization.

CFD-DEM study on the synergistic effect of coke consumption and particle dynamics in the blast furnace raceway

In this study,the gas-solid flow process in the blast furnace raceway is numerically simulated using coupled computational fluid dynamics and the discrete element method(CFD-DEM).The coke reaction kinetics data are imported into the DEM model to reproduce the consumption process of each coke particle.The effects of inlet gas velocity and angle on the morphology of the raceway,coke consumption rate,coke bed temperature,and particle size distribution in the blast process are systematically investigated and analyzed.The results show that the consumption of coke particles promotes the formation of raceways during the blast process.At the same time,a coke mixture layer is produced at the edge of the raceway.The higher the inlet gas velocity,the thicker the coke mixture layer in the middle and upper parts of the raceway region,and the larger the proportion of small particles in the coke mixture layer.The effect of the inlet gas angle on the raceway region is less than the inlet gas velocity.However,with the increase in the inlet gas angle,the high-temperature region of the coke bed extends downward gradually,which is conducive to activating the hearth.

The MW5.5 earthquake on August 6,2023,in Pingyuan,Shandong,China:A rupture on a buried fault

On August 6,2023,a magnitude MW5.5 earthquake struck Pingyuan County,Dezhou City,Shandong Province,China.This event was significant as no large earthquakes had been recorded in the region for over a century,and no active fault had been previously identified.This study collects 1309 P-wave arrival times and 866 S-wave arrival times from 74 seismic stations less than 200 km to the epicenter to constrain the spatial distribution of the mainshock and its 125 early aftershocks by the double difference earthquake relocation method,and selects 864 P-waveforms from 288 stations located within 800 km of the epicenter to constrain the focal mechanism solution of the mainshock through centroid moment tensor inversion.The relocation and the inversion indicate,the Pingyuan MW5.5 earthquake was caused by a rupture on a buried fault,likely an extensive segment of the Gaotang fault.This buried fault exhibited a dip of approximately 75°to the northwest,with a strike of 222°,similar to the Gaotang fault.The rupture initiated at the depth of 18.6 km and propagated upward and northeastward.However,the ground surface was not broken.The total duration of the rupture was~6.0 s,releasing the scalar moment of 2.5895×1017 N·m,equivalent to MW5.54.The moment rate reached the maximum only 1.4 seconds after the rupture initiation,and the 90%scalar moment was released in the first 4.6 s.In the first 1.4 seconds of the rupture process,the rupture velocity was estimated to be 2.6 km/s,slower than the local S-wave velocity.As the rupture neared its end,the rupture velocity decreased significantly.This study provides valuable insights into the seismic characteristics of the Pingyuan MW5.5 earthquake,shedding light on the previously unidentified buried fault responsible for the seismic activity in the region.Understanding the behavior of such faults is crucial for assessing seismic hazards and enhancing earthquake preparedness in the future.

Reduced-Order Observer-Based LQR Controller Design for Rotary Inverted Pendulum

The Rotary Inverted Pendulum(RIP)is a widely used underactuated mechanical system in various applications such as bipedal robots and skyscraper stabilization where attitude control presents a significant challenge.Despite the implementation of various control strategies to maintain equilibrium,optimally tuning control gains to effectively mitigate uncertain nonlinearities in system dynamics remains elusive.Existing methods frequently rely on extensive experimental data or the designer’s expertise,presenting a notable drawback.This paper proposes a novel tracking control approach for RIP,utilizing a Linear Quadratic Regulator(LQR)in combination with a reduced-order observer.Initially,the RIP system is mathematically modeled using the Newton-Euler-Lagrange method.Subsequently,a composite controller is devised that integrates an LQR for generating nominal control signals and a reduced-order observer for reconstructing unmeasured states.This approach enhances the controller’s robustness by eliminating differential terms from the observer,thereby attenuating unknown disturbances.Thorough numerical simulations and experimental evaluations demonstrate the system’s capability to maintain balance below50Hz and achieve precise tracking below1.4 rad,validating the effectiveness of the proposed control scheme.

Predicting microbial extracellular electron transfer activity in paddy soils with soil physicochemical properties using machine learning

Soil extracellular electron transfer(EET)is a pivotal biological process within the realm of soil.Unfortunately,EET suffers from a lack of predictive models.Herein,an intricately crafted machine learning model has been developed for the purpose of predicting soil EET by using the physicochemical properties of soil as independent input variables and the EET capabilities in terms of current density(j_(max))and Coulombic charge(C_(out))as dependent output variables.An autoencoder ensemble stacking(AES)model was developed to address the aforementioned issue by integrating support vector machine,multilayer perceptron,extreme gradient boosting,and light gradient boosting machine algorithms as the stacking algorithms.With 10-fold crossvalidation,the AES model exhibited notable improvements in predicting j_(max)and C_(out),with average test R^(2)values of 0.83 and 0.84,respectively,surpassing those of single machine learning(ML)models and the basic ensemble model.By utilizing partial correlation plots(PDPs),Shapley Additive explanations(SHAP)values,and SHAP decision plots,we quantitatively explained the impact and contribution of the input molecules on the AES model’s predictions of j_(max)and C_(out).In the context of the SHAP method for the AES model,total carbon(TC)was identified as the most correlated descriptor for j_(max),while total organic carbon(TOC)stood out as the most relevant descriptor for C_(out).In the prediction tasks of j_(max)and C_(out)within the AES model,employing a multitask ML approach allowed the model to benefit from the shared information of input variables,thereby enhancing its overall generalizability.This study provides a feasible tool for the prediction of soil EET from soil physiochemical properties and an advanced understanding of the relationship between soil physiochemical properties and EET capability.
