Arg-X Activity in Trypsin-like Complexes of the Nuclear Rroteins in the Suprastructures of Interphase Chromatin During Induction of Growth Morphogenesis Mature Germs of Wheat

The purpose of this study was to analyze spatio-temporal dynamics of localization of protease-sensitive sites Arg-X in non-histone and histone blocks of heteropolymer suprastructures （nucleoplasm, chromatin, nuclear matrix） as possible zones affecting the conformational rearrangements of the total interphase chromatin at the induction of increasing morphogenesis of mature embryos-germs of spring and transformed from its winter wheat. Germinated embryos-germs were detached from endosperm after 24 hours from the start of soaking. Cell nuclei have been allocated from embryos-germs and cleared, and then from their heteropolymer suprastructures （nucleoplasm, chromatin loosely bound with nuclear matrix and chromatin tightly bound with nuclear matrix, and nuclear matrix） were extracted by increasing ionic strength of solution. From isolated nuclear suprastructures, non-histone proteins were separated from histones using ion exchange chromatography. Trypsin-like complexes from non-histone proteins and histone blocks were isolated using the affinity chromatography. The Arg-X （tryptase） activity was assessed by cleavage of Arg-X bonds in the arginine-enriched protein protamine. Hypersensitivity to the Arg-X proteolysis in trypsin-like complexes detected at the level suprastructures of chromatin tightly bound with the nuclear matrix was shown. The most active changes of the nuclear proteome have occurred at the level of the non-histone proteins and the core histones （H2A ＋ H2B）（H3 ＋ H4） of induced to growth embryos-seedlings of winter wheat （compared to the initial spring form of wheat）. Perhaps hypersensitivity to the Arg-X activity of the trypsin-like complexes in the non-histone proteins and the core blocks of chromatin tightly bound with nuclear matrix have been entrenched during the transforming of the winter wheat from the initial spring wheat.

Colorectal peritoneal metastases:Optimal management review

The peritoneum is a common site of dissemination for colorrectal cancer, with a poorer prognosis than other sites of metastases. In the last two decades, it has been considered as a locoregional disease progression and treated as such with curative intention treatments. Cytoreductive surgery(CRS) and hyperthermic intraperitoneal chemotherapy(HIPEC) is the actual reference treatment for these patients as better survival results have been reached as compared to systemic chemotherapy alone, but its therapeutic efficacy is still under debate. Actual guidelines recommend that the management of colorectal cancer with peritoneal metastases should be led by a multidisciplinary team carried out in experienced centers and consider CRS + HIPEC for selected patients. Accumulative evidence in the last three years suggests that this is a curative treatment that may improve patients disease-free survival, decrease the risk of recurrence, and does not increase the risk of treatment-related mortality. In this review we aim to gather the latest results from referral centers and opinions from experts about the effectiveness and feasibility of CRS + HIPEC for treating peritoneal disease from colorectal malignancies.

Rye(Secale cereale)is an important crop with multiple uses and a valuable genetic resource for wheat ***,due to its complex genome and outcrossing nature,the origin of cultivated rye remains *** geneticist *** proposed that cultivated rye had been domesticated from weedy rye,rather than directly from wild species like other *** the domestication history of rye will extend our understanding of crop evolution and upend our inherent understanding of agricultural *** this end,in this study we generated the 8.5 Tb of whole-genome resequencing data from 116 worldwide accessions of wild,weedy,and cultivated rye,and demonstrated that cultivated rye was domesticated directly from weedy relatives with a similar but enhanced genomic selection by *** found that a repertoire of genes that experienced artificial selection is associated with important agronomic traits,including shattering,grain yield,and disease ***,we identified a composite introgression in cultivated rye from the wild perennial Secale strictum and detected a 2-Mb introgressed fragment containing a candidate ammonium transporter gene with potential effect on the grain yield and plant growth of *** together,our findings unravel the domestication history of cultivated rye,suggest that interspecific introgression serves as one of the likely causes of obscure species taxonomy of the genus Secale,and provide an important resource for future rye and wheat breeding.

Effect of Some Bioproducts on Winter Mortality of Grafted Buds and the Number of Maiden Fruit Trees Produced in an Organic Nursery

In an organic nursery, various bioproducts were used to stimulate plant growth： Fertigo （granulated bovine manure）, Micosat, Humus UP （humic acids microbiologically enriched）, Humus Active ＋ Aktywit PM （beneficial microorganisms）, BioFeed Amin, BioFeed Quality, Tytanit, and Vinassa. One of the aims of the study was to determine the effect of these products on the ability of grafted buds of apple and sour cherry cultivars to survive the winter. Losses resulting from the freezing of buds during winter dormancy were calculated as the difference between the number of buds deemed to have taken in autumn and the number of live buds that began to develop properly in the spring of the following year. In autumn, the number of maiden trees obtained in each fertilization combination was also determined in the same nursery. The greatest tendency to die in winter was shown by apple and sour cherry buds in those combinations in which the rootstocks were supplied with a mineral fertilizer with NPK components, and the smallest where humic preparations （so-called vermiculites） were used. The type of fertilizer had an indirect influence not only on the number of grafted buds that died in winter as a result of freezing, but also on the final number of maiden trees obtained from a given area of the nursery.

The Quality of Sour Cherry Maidens Fertilized with Various Biopreparations in an Organic Nursery

The study was conducted in an organic nursery of sour cherry cultivars ＂Debreceni Botermo＂ and ＂Sabina＂ in the years 2010 and 2011. Cherry trees grafted on Mahaleb （Prunus mahaleb L.） seedlings were grown at a spacing of 25 crn × 1.0 m. In the nursery, an assessment was carried out of the effect of various biopreparations on such growth characteristics as the thickness and height of the maidens, the formation of lateral shoots and the total increase in their length. The control trees were not treated at all, but fertilized with an NPK fertilizer only （second control）. To stimulate growth, use was made of granular manure, mycorrhizal fungi in the form of the preparation Micosat, humic preparations or so-called vermicomposts （Humus UP, Humus Active）, extracts of marine algae （BF Quality） and of terrestrial plants （BF Arnin）, a product derived from yeast （Vinassa） and Tytanit. The specific biological effects of the preparations used in the sour cherry nursery for each morphological feature are presented in this paper in graphical form. A beneficial effect on the increase in the number of lateral shoots and the increase in their length was produced by the use of such products as： Micosat, Humus UP, Humus Active ＋ Aktywit PM and Vinassa. The most effective in this respect was BF Amin.

Influence of Biopreparations on Maiden Growth of Sour Cherry （Prunus cerasus L.） in Organic Nursery- Preliminary Results

A study was conducted in an organic nursery in 2010 and 2011 on the maiden growth of sour cherry cultivars ＂Debreceni Botermo＂ and ＂Sabina＂ grafted on Mahaleb cherry seedlings （Prunus mahaleb L.）. The young trees were grown at a spacing of 25 cm × 1.0 m on a podzolic soil. The study focused on the differences in diameter and height of the maiden stock, the branch number, lateral shoots in the crown and their length, among treatments with various bioproducts applied to the maidens once or twice, such as granulated manure, Micosat, Humus UP （humus cultivated）, Humus Active ＋ Aktywit PM （positive microorganisms）, BioFeed Amin, BioFeed Quality, Tytanit and Vinassa. The control maidens were not fertilized at all, or fertilized with NPK （nitrogen, potassium, phosphorus）. The first time when these products were applied in the nursery was mid-May, and the second time was one month later. Repeating the application of humic preparations （vermiculites） and a formulation containing mycorrhizal fungi （Micosat） gave better results than applying them only once, but in the case of BF Quality and the preparation Tytanit treating the plants for the second time did not increase their growth vigour compared to the plants treated only once. And the effectiveness of the biopreparations was different for the treated cultivars changed.

Dear Editor,Microcalcifications(MCs)are common findings in mammography and can be indicative of different degrees of malignancy,thus requiring multiple stereotaxis vacuumassisted biopsies under mammography to identify and characterize breast cancer[1,2].In many cases,however,the presence of MCs is not associated with the presence of a ***,a better understanding of the physical-chemical properties of MCs is needed to assess the relationship between their presence and breast cancer.

Analgesic and anti-inflammatory effects of UP1304,a botanical composite containing standardized extracts of Curcuma longa and Morus alba

OBJECTIVE： Though the initial etiologies of arthritis are multifactorial, clinically, patients share the prime complaints of the disease, pain. Here the authors assessed the analgesic and anti-inflammatory effects of UP1304, a composite that contains a standardized blend of extracts from the rhizome of Curcuma /onga and the root bark of Morus a/ba, on rats with carrageenan-induced paw edema. METHODS： A plant library was screened for bradykinin receptor antagonists./n vivo, the anti- inflammatory and analgesic effects of the standardized composite, UP1304, were evaluated in rats with carrageenan-induced paw edema using oral dose ranges of 100-400 mg/kg. Ibuprofen, at a dose of 200 mg/kg, was used as a reference compound. In vitro, cycleoxygenase （COX） and lipoxygenase （LOX） inhibition assays were performed to evaluate the degree of inflammation. RESULTS： Statistically significant improvements in pain resistance and paw edema suppression were observed in animals treated with UP1304, when compared to vehicle-treated rats. Results from the highest dose of UP1304 （400 mg/kg） were similar to those achieved by ibuprofen treatment at 200 mg/kg. /n vitro, UP1304 showed dose-dependent inhibition of the enzymatic activities of COX and LOX. A half-maximal inhibitory concentration of 9.6 tJg/mL for bradykinin B1 inhibition was calculated for the organic extract of C./onga. Curcumin showed Ki values of 2.73 and 58 IJg/mL for bradykinin receptors B1 and B2, respectively. CONCLUSION： Data presented here suggest that UP1304, analgesic and anti-inflammatory agent of botanical origin, acted as a bradykinin receptor B1 and B2 antagonist, and inhibited COX and LOX enzyme activities. This compound should be considered for the management of symptoms associated with arthritis.
