NKX2-3 and IRGM variants are associated with diseasesu sceptibility to IBD in Eastern European patients

AIM: To investigate variants of immunity-related GT-Pase family M (IRGM) and NKX2-3 genes and genotype-phenotype in Eastern European patients with inflammatory bowel disease (IBD).METHODS: We analyzed 1707 Hungarian and Czech subjects with Crohn’s disease (CD) (n = 810, age: 37.1 ± 12.6 years, duration: 10.7 ± 8.4 years) and ulcerative colitis (UC) (n = 428, age: 43.7 ± 15.0 years, duration: 12.6 ± 9.9 years), as well as 469 healthy controls. IRGM rs13361189, NKX2-3 rs10883365 and ECM1 rs13294 polymorphisms were tested by LightCy-cler allele discrimination. Detailed clinical phenotypes were determined by reviewing the medical charts. RESULTS: NKX2-3 rs10883365 variant allele was as-sociated with increased risk for CD (P = 0.009, OR = 1.24, 95% CI = 1.06-1.48) and UC (P = 0.001, OR = 1.36, 95% CI = 1.13-1.63), whereas variant IRGM allele increased risk for CD (P = 0.029, OR = 1.36, 95% CI = 1.03-1.79). In contrast, ECM1 rs13294 was not associat-ed with either CD or UC. In CD, the variant IRGM allele was associated with a colon-only location (P = 0.02, OR = 1.62, 95% CI = 1.07-2.44), whereas in UC, the ECM1 variant was associated with cutaneous manifestations (P = 0.002, OR = 3.36, 95% CI = 1.48-7.63). variant alleles did not predict resistance to steroids or azathio-prine, efficacy of infliximab, or need for surgery. CONCLUSION: NKX2-3 and IRGM are susceptibility locifor IBD in Eastern European patients. Further studies are needed to confirm the reported phenotype-genotype associations.

Regeneration of neurotransmission transcriptome in a model of epileptic encephalopathy after antiinflammatory treatment

Inflammation is an established etiopathogenesis factor of infantile spasms（IS）, a therapy-resistant epileptic syndrome of infancy. We investigated the IS-associated transcriptomic alterations of neurotransmission in rat hypothalamic arcuate nucleus, how they are corrected by antiinflamatory treatments and whether there are sex differences. IS was triggered by repeated intraperitoneal administration of N-methyl-D-aspartic acid following anti-inflammatory treatment（adreno-cortico-tropic-hormone（ACTH） or PMX53）or normal saline vehicle to prenatally exposed to betamethasone young rats. We found that treatments with both ACTH and PMX53 resulted in substantial recovery of the genomic fabrics of all types of synaptic transmission altered by IS. While ACTH represents the first line of treatment for IS, the even higher efficiency of PMX53（an antagonist of the complement C5 a receptor） in restoring the normal transcriptome was not expected. In addition to the childhood epilepsy, the recovery of the neurotransmission genomic fabrics by PMX53 also gives hope for the autism spectrum disorders that share a high comorbidity with IS. Our results revealed significant sex dichotomy in both IS-associated transcriptomic alterations（males more affected） and in the efficiency of PMX53 anti-inflammatory treatment（better for males）. Our data further suggest that anti-inflammatory treatments correcting alterations in the inflammatory transcriptome may become successful therapies for refractory epilepsies.

A dynamical approach to generate chaos in a micromechanical resonator

Chaotic systems,presenting complex and nonreproducible dynamics,may be found in nature,from the interaction between planets to the evolution of weather,but can also be tailored using current technologies for advanced signal ***,the realization of chaotic signal generators remains challenging due to the involved dynamics of the underlying *** this paper,we experimentally and numerically present a disruptive approach to generate a chaotic signal from a micromechanical *** technique overcomes the long-established complexity of controlling the buckling in micro/nanomechanical structures by modulating either the amplitude or the frequency of the driving force applied to the resonator in the nonlinear *** experimental characteristic parameters of the chaotic regime,namely,the Poincarésections and Lyapunov exponents,are directly comparable to simulations for different *** results confirm that this dynamical approach is transposable to any kind of micro/nanomechanical resonator,from accelerometers to *** demonstrate a direct application exploiting the mixing properties of the chaotic regime by transforming an off-the-shelf microdiaphragm into a true random number generator conforming to the National Institute of Standards and Technology *** versatility of this original method opens new paths to combine the unique properties of chaos with the exceptional sensitivity of microstructures,leading to emergent microsystems.

We report on an experiment performed at the FLASH2 free-electron laser(FEL)aimed at producing warm dense matter via soft x-ray isochoric *** the experiment,we focus on study of the ions emitted during the soft x-ray ablation process using time-of-flight electron multipliers and a shifted Maxwell–Boltzmann velocity distribution *** find that most emitted ions are thermal,but that some impurities chemisorbed on the target surface,such as protons,are accelerated by the electrostatic field created in the plasma by escaped *** morphology of the complex crater structure indicates the presence of several ion groups with varying *** find that the ion sound velocity is controlled by the ion temperature and show how the ion yield depends on the FEL radiation attenuation length in different materials.

The Western Carpathians are located out of world main natural hazardous zones. Human casualties are related more to snow avalanches in connection with mountain hiking, some individuals yearly by flooding and rarely by forest fires. Economic lost about 0.1 to 0.2 %, exceptionally up to 0.8 % of the gross domestic product (GDP) proportionally to the Carpathian regions of particular countries. Natural disasters are linked, except of the above mentioned events, to infrequent small and medium scale earthquakes, landslides, and erosion. Records of the most harmful natural events are found in archives since the 16th century. Their systematic study and factor analysis started from the end of the 19th century, and protective measures and organization of impact mitigation developed during the 20th century to minimize the risk.

Acute appendicitis in a patient after a uterus transplant: A case report

BACKGROUND Acute appendicitis in a solid organ transplant recipient is a rare occurrence, and experience remains limited. Appendicitis in uterine transplant recipients has never been reported. Immunocompromised patients with acute abdomen often present late and with attenuated symptoms. The differential diagnosis in a transplanted patient is broad and challenging due to possible existing complications associated with the graft, effects of immunosuppression, and altered anatomical *** SUMMARY A 26-year-old woman suffering from absolute uterine factor infertility received a uterus transplant. In the post-transplant period, she suffered from leukopenia and recurrent acute cellular rejection. Her compliance was suboptimal. She travelled to an exotic destination despite the physician's recommendation not to do so. Following her vacation, she presented with abdominal discomfort, nausea and diarrhoea. There was no sign of acute abdomen;the abdominal ultrasound was negative on day 0. Clostridium difficile colitis was verified and treated with perorally administered vancomycin. On day 4, the discomfort changed to pain;the ultrasound scan revealed a finding suggestive of appendicitis. Surgical exploration revealed perforated appendicitis, and appendectomy was *** a surgical point of view, the patient's follow-up was uneventful. The patient underwent a successful embryo transfer 6 months after the appendectomy. The patient gave birth to a healthy boy at the 35 th week of *** A high index of suspicion of an atypical course and symptomatology of acute abdomen should be maintained in immunosuppressed patients.

The production of octahydrocoumarin,which can serve as a replacement for toxic coumarin,was investigated using 5% Ru on active carbon(Ru/C) as the catalyst for the hydrogenation of coumarin. The hydrogenation was studied by optimizing the reaction conditions(pressure,solvent and coumarin concentration). The activity and selectivity of the Ru/C catalyst were compared for different solvents. The mechanism of coumarin hydrogenation was deduced. The formation of side products was explained. The optimal hydrogenation reaction conditions were: 130 °C,10 MPa,60 wt% coumarin in methanol,and 0.5 wt%(based on coumarin) of Ru/C catalyst. At the complete conversion of coumarin,the selectivity to the desired product was 90%.

Conversion of ethanol to 1,3-butadiene over Ag-ZrO_(2)/SiO_(2) catalysts:The role of surface interfaces

A series of Ag-ZrO_(2)/SiO_(2) catalysts with different metal-support interfaces were synthesized in an effort to elucidate the roles of specific interfaces in controlling the ethanol to 1,3-butadiene conversion and *** to the results of detailed characterizations(***/pyridine-DRIFTS,XPS,TEM,NH3-TPD,and ^(1)H MAS NMR),it was found that the Ag-O-Si interfaces significantly enhanced the dehydrogenation of ethanol while the presence of ZrO_(2) improved the interaction between Ag and ZrO_(2)/SiO_(2),creating more Ag^(δ+)active *** high dispersion of ZrO_(2) on SiO_(2) generated abundant Zr-O-Si interfaces with medium and weak Lewis acidity,promoting the condensation of acetaldehyde to *** Zr-O-Si interfaces in close interaction with Ag^(δ+)species played a critical role in the enhanced H transfer during the MPv reduction of crotonaldehyde to crotyl *** synergies among the interfaces resulted in retarded ethanol dehydration reactivity,balanced ethanol dehydrogenation and condensation reactions,and a subsequent high 1,3-butadiene yield.

The article deals with potential use of waste materials in construction industry, specifically use of high density polyethylene （HDPE）. The article is focused in particular on recycled polyethylene application in products designed for construction industry, especially for passive houses. Currently certain building details of passive houses are not perfect or their solution results in higher economic demands related to house purchase and its further use. For the purpose of this thesis details of windows installation in external walls and elimination of thermal bridges in wall footing have been chosen. Products were subject to mathematic modelling of thermal technique and statics. The executed mathematic models documented that products are fully functional and that the suggested product successfully eliminate insufficiencies of some currently applied solutions.

Osteopontin: A non-invasive parameter of portal hypertension and prognostic marker of cirrhosis

AIM: To investigate the relationship between osteopontin plasma concentrations and the severity of portal hypertension and to assess osteopontin prognostic ***: A cohort of 154 patients with confirmed liver cirrhosis (112 ethylic, 108 men, age 34-72 years) were enrolled in the study. Hepatic venous pressure gradient (HvPG) measurement and laboratory and ultrasound examinations were carried out for all patients. HvPG was measured using a standard catheterization method with the balloon wedge technique. Osteopontin was measured using the enzyme-linked immunosorbent assay (ELISA) method in plasma. Patients were followed up with a specific focus on mortality. The control group consisted of 137 healthy age- and sex- matched ***: The mean value of HvPG was 16.18 ± 5.6 mmHg. Compared to controls, the plasma levels of osteopontin in cirrhotic patients were significantly higher (P vels of osteopontin were positively related to HvPG (P = 0.0022, r = 0.25) and differed among the individual Child-Pugh groups of patients. The cut-off value of 80 ng/mL osteopontin distinguished patients with significant portal hypertension (HvPG above 10 mmHg) at 75% sensitivity and 63% specificity. The mean follow-up of patients was 3.7 ± 2.6 years. The probability of cumulative survival was 39% for patients with HvPG > 10 mmHg and 65% for those with HvPG ≤ 10 mmHg (P = 0.0086, odds ratio (OR), 2.92, 95% confidence interval (CI): 1.09-7.76). Osteopontin showed a similar prognostic value to HvPG. Patients with osteopontin values above 80 ng/mL had significantly lower cumulative survival compared to those with osteopontin ≤ 80 ng/mL (37% vs 56%, P = 0.00035; OR = 2.23, 95%CI: 1.06-4.68).CONCLUSION: Osteopontin is a non-invasive parameter of portal hypertension that distinguishes patients with clinically significant portal hypertension. It is a strong prognostic factor for survival.
