Intelligent lesion blood–brain barrier targeting nano-missiles for Alzheimer's disease treatment by anti-neuroinflammation and neuroprotection

The treatment of Alzheimer's disease(AD)is one of the most difficult challenges in neurodegenerative diseases due to the insufficient blood‒brain barrier(BBB)permeability and unsatisfactory intra-brain distribution of drugs.Therefore,we established an ibuprofen and FK506 encapsulated drug co-delivery system(Ibu&FK@RNPs),which can target the receptor of advanced glycation endproducts(RAGE)and response to the high level of reactive oxygen species(ROS)in AD.RAGE is highly and specifically expressed on the lesion neurovascular unit of AD,this property helps to improve targeting specificity of the system and reduce unselective distribution in normal brain.Meanwhile,these two drugs can be specifically released in astrocytes of AD lesion in response to high levels of ROS.As a result,the cognition of AD mice was significantly improved and the quantity of Aβplaques was decreased.Neurotoxicity was also alleviated with structural regeneration and functional recovery of neurons.Besides,the neuroinflammation dominated by NF-κB pathway was significantly inhibited with decreased NF-κB and IL-1βin the brain.Overall,Ibu&FK@RNPs can efficiently and successively target diseased BBB and astrocytes in AD lesion.Thus it significantly enhances intracephalic accumulation of drugs and efficiently treats AD by anti-neuroinflammation and neuroprotection.

A gap-free reference genome reveals structural variations associated with flowering time in rapeseed(Brassica napus)

Allopolyploid oilseed rape(Brassica napus)is an important oil crop and vegetable.However,the latest version of its reference genome,with collapsed duplications,gaps,and other issues,prevents comprehensive genomic analysis.Herein,we report a gap-free assembly of the rapeseed cv.Xiang5A genome using a combination of ONT(Oxford Nanopore Technologies)ultra-long reads,PacBio high-fidelity reads,and Hi-C datasets.It includes gap-free assemblies of all 19 chromosomes and telomere-to-telomere assemblies of eight chromosomes.Compared with previously published genomes of B.napus,our gap-free genome,with a contig N50 length of 50.70 Mb,has complete assemblies of 9 of 19 chromosomes without manual intervention,and greatly improves contiguity and completeness,thereby representing the highest quality genome assembly to date.Our results revealed that B.napus Xiang5A underwent nearly complete triplication and allotetraploidy relative to Arabidopsis thaliana.Using the gap-free assembly,we found that 917 flowering-related genes were affected by structural variation,including BnaA03.VERNALIZATION INSENSITIVE 3 and BnaC04.HIGH EXPRESSION OF OSMOTICALLY RESPONSIVE GENES 1.These genes may play crucial roles in regulating flowering time and facilitating the adaptation of Xiang5A in the yangtze River Basin of China.This reference genome provides a valuable genetic resource for rapeseed functional genomic studies and breeding.

Rechargeable Na-CO_(2) Batteries Starting from Cathode of Na2CO_(3) and Carbon Nanotubes

Na-CO_(2) batteries have attracted signifcant attentions due to their high energy density and efective utilization of greenhouse gas CO_(2).However,all reported Na-CO_(2) batteries employ excessive preloaded metal Na,which will lead to safety issues such as dendrite formation and short circuit.In addition,the charging mechanism of reported Na-CO_(2) batteries is not very clear.Here we report the Na-CO_(2) batteries,starting from the cathode of cheap Na2CO_(3) and multiwalled carbon nanotubes(CNTs).Due to the efective electron transfer and high reactivity,the decomposition of Na2CO_(3) and CNTs could take place under 3.8 V.Te charging mechanism of 2Na2CO_(3)+C�→4Na+3CO_(2) without any side reactions is revealed by in/ex situ techniques such as Raman,gas chromatograph,and optical microscope.Dendrite-free Na can quantitatively deposit on the Super P/Al anode because of large specifc surface area and low nucleation barrier of the anode for Na plating.Te batteries could deliver an energy density of 183 Wh kg−1(based on the whole mass of the pouch-type batteries,4 g)with stable cycling performance.Tis work reveals that safe rechargeable Na-CO_(2) batteries could be constructed by cheap Na2CO_(3) and multiwalled carbon nanotubes.

高压氧对初发型溃疡性结肠炎患者的早期疗效观察

目的观察高压氧(hyperbaric oxygen,HBO)对初发型疡性结肠炎(ulcerative colitis,UC)患者的早期临床疗效。方法收集2019年5月至2021年5月于锦州医科大学附属第一医院入院行肠镜检查并确诊为初发型、轻中度、病变局限于左半结肠的UC患者60例,随机分为对照组(A组,n=30人)和实验组(B组,n=30人),对照组在常规治疗的基础上加用美沙拉嗪肠溶片,实验组在对照组的基础上加用HBO治疗,治疗36 d后,复查肠镜,比较两组患者的治疗总有效率和血清中白细胞介素(interleukin,IL)-2,IL-4,IL-6,IL-10、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)及Thl7细胞(CD4^(+)IL-17^(+)T细胞)的表达水平。进行统计学分析。结果实验组治疗的总有效率较对照组更高(P<0.05);实验组患者较对照组患者血清中IL-2、IL-4、IL-6细胞及Thl7细胞水平下降(P<0.05),TNF-α及IL-10水平上升(P<0.05),差异具有统计学意义。结论HBO可抑制血清中炎症因素的表达、提高免疫应答、促进肠道黏膜的修复,对初发型轻中度UC患者产生较好的治疗效果,值得进一步在临床进行推广和应用。

Durable superamphiphobic silica aerogel surfaces for the culture of 3D cellular spheroids

The 3D multicellular spheroids with intact cell–cell junctions have major roles in biological research by virtue of their unique advantage of mimicking the cellular physiological environments. In this work, a durable superamphiphobic silica aerogel surface(SSAS) has been fabricated for the upward culture of 3D multicellular spheroids. Poly(3,4-ethylenedioxythiophene)(PEDOT) was first electrodeposited on a conductive steel mesh as a first template for porous silica coating. Soot particles were then applied as a second template to construct a cauliflower-like silica aerogel nanostructure. After fluorination, a hierarchical structure with re-entrant curvature was finally fabricated as a durable superamphiphobic surface. This superamphiphobic surface also presented excellent antifouling towards biomacromolecules and cells, which has been demonstrated by the successful upward culture of cell spheroids. The upward culture makes the observation of cellular behavior in situ possible, holding great potential for 3D cellular evaluation in vitro.

降水变化既影响地上植被动态,也影响地下碳循环过程,尤其以干旱半干旱生态系统对降水的响应更为敏感。然而极端降水如何影响土壤碳固存潜力仍未得出明确结果。本研究在黄土高原西部荒漠草原样地实施了为期3年的降水控制实验,该实验包含5个降水梯度(即自然降水(对照),以及在自然降水基础上的减水40%、减水20%、增水20%、增水40%)。通过对不同降水处理下植物生长指标、地上净初级生产力(ANPP)、土壤水分和土壤呼吸(Rs)进行监测,采用双侧不对称模型揭示ANPP和Rs对降水变化的响应规律;采用结构方程模型,分析降水变化下影响ANPP和Rs的直接和间接因素。研究结果表明,ANPP对极端干旱的响应比极端湿润更敏感,在干旱和湿润年份均符合负向不对称模型。ANPP的变化主要受到降水的直接影响,同时,干旱年份植物密度的变化也对ANPP产生了影响。在湿润年份,Rs对降水变化的响应也呈负向不对称性。然而,干旱年份,Rs对降水变化表现出正向不对称响应,即对降水增加响应的敏感性高于降水减少,这可能与植物生长和ANPP对增水处理的正响应增加使自氧呼吸增强,及降水事件对异氧呼吸具有较强的‘Birch效应’有关。在干旱年份Rs对极端干旱(减水40%处理)表现出饱和响应。ANPP和Rs对降水格局改变的响应模式差异表明荒漠草原区极端湿润或干旱可能降低研究区土壤碳固存的潜力。

The dearomative[5+1]annulation of 2-methylindoles with new five-membered synthons O-alkyl orthooxybenzaldehydes was developed unprecedentedly through cascade[1,5]-hydride transfer/dearomativecyclization in HFIP for the synthesis of spirochromanes bearing the 2-methylindolenine skeleton.Inaddition,the dual alkylation of the methyl group of 2-methylindolenines was achieved by sequential operation through the redox neutral[5+1]annulation with the second five-membered synthon N-alkylortho-aminobenzaldehyde,providing the chromane and tetrahydroquinoline fused spiroindolenines ingood yields.Furthermore,the auxiliary group that facilitates the hydride transfer process could be simplyremoved.

A spurious positive Indian Ocean Dipole in 2017

The Indian Ocean Dipole (IOD) is an intrinsic ocean-atmosphere coupled mode in the tropical Indian Ocean (IO)[1,2]. It features a sea surface temperature (SST) anomaly gradient, defined by the dipole mode index (DMI), the difference of area-averaged SST anomaly between the western tropical IO (50°–70°E, 10°N–10°S)and the southeastern tropical IO (90°–110°E, Eq.-10°S). The IOD event tends to develop and mature in the boreal summer and fall seasons. When a positive IOD (p IOD) occurs, SST warms up in the west and cools down in the east, which sets up an east-west pressure gradient. The atmosphere hence responds to the

Revisiting ENSO impacts on the Indian Ocean SST based on a combined linear regression method

The El Nino-Southern Oscillation(ENSO)has great impacts on the Indian Ocean sea surface temperature(SST).In fact,two major modes of the Indian Ocean SST namely the Indian Ocean Basin(IOB)and the Indian Ocean Dipole(IOD)modes,exerting strong influences on the Indian Ocean rim countries,are both influenced by the ENSO.Based on a combined linear regression method,this study quantifies the ENSO impacts on the IOB and the IOD during ENSO concurrent,developing,and decaying stages.After removing the ENSO impacts,the spring peak of the IOB disappears along with significant decrease in number of events,while the number of events is only slightly reduced and the autumn peak remains for the IOD.By isolating the ENSO impacts during each stage,this study reveals that the leading impacts of ENSO contribute to the IOD development,while the delayed impacts facilitate the IOD phase switch and prompt the IOB development.Besides,the decadal variations of ENSO impacts are various during each stage and over different regions.These imply that merely removing the concurrent ENSO impacts would not be sufficient to investigate intrinsic climate variability of the Indian Ocean,and the present method may be useful to study climate variabilities independent of ENSO.
