Biogenesis of flavor-related linalool is diverged and genetically conserved in tree peony (Paeonia × suffruticosa)

Floral scent is an important and genetically complex trait in horticultural *** peony(Paeonia×suffruticosa)originates in the Pan-Himalaya and has nine wild species divided into two subsections,Delavayanae and *** f lowers are beloved worldwide for their sweet f loral fragrance,yet the f lavor-related volatiles and underlying biosynthetic pathways remain ***,we characterized the volatile blends of all wild tree peony species and found that the flavor-related volatiles were highly divergent,but linalool was a unique monoterpene in *** detection of volatiles in 97 cultivars with various genetic backgrounds showed that linalool was also the characteristic aroma component in Paeonia delavayi hybrid progenies,suggesting that linalool was conserved and dominant within *** and its hybrids,instead of species and cultivars from *** transcriptome analysis of all wild tree peony species and 60 cultivars revealed five candidate genes that may be involved in key steps of linalool biosynthesis;especially the expressions of three TPS genes,PdTPS1,PdTPS2,and PdTPS4,were significantly positively correlated with linalool emissions across tree peony *** biochemical evidence demonstrated that PdTPS1 and PdTPS4were the pivotal genes determining the species-specific and cultivar-specific emission of *** study revealed a new insight into floral scent divergence in tree peony and would greatly facilitate our understanding of the phylogeny and evolution of Paeonia.

American lotus(Nelumbo lutea)is one of the two species in Nelumbo and has only yellow *** of total flavonoids showed wild American lotus contained almost only flavonols with quercetin 3-O-glucuronide to be the dominant *** variation tendency of the total flavonol content was coincident with yellow color variation of petals during flower *** understand the mechanism of accumulation and constituent of pigments in petals,three pivotal genes,NlFLS1,NlFLS2 and NlFLS3,which were predicted to encode flavonol synthases were isolated and characterized by analyses of basic bioinformatics,temporal and spatial expression patterns and enzymatic *** temporal expression levels showed the same variation tendency,which was also consistent with the development-dependent variation of total flavonol *** expression patterns indicated the three genes should function in *** the three proteins were demonstrated to be bifunctional dioxygenase,possessing both flavonol synthase activity and flavanone 3-hydroxylase ***,other flavonol biosynthesis related genes were also investigated on their expression levels to give more clues on the *** preferences of the three FLSs,substrate competitions between the FLSs and other flavonol biosynthesis related enzymes,and the greatly differential expression levels between F3’H(flavonoid 3-hydroxylase)and F3’5’H(flavonoid 3,5-hydroxylase)contributed to the flavonol constituent in the petals of America lotus,namely abundant quercetin-derivatives while very few kaempferol-derivatives and myricetin-derivatives.

TaMADS29 interacts with TaNF-YB1 to synergistically regulate early grain development in bread wheat

Grain development is a crucial determinant of yield and quality in bread wheat(Triticum aestivum L.).However,the regulatory mechanisms underlying wheat grain development remain *** we report how Ta MADS29 interacts with Ta NF-YB1 to synergistically regulate early grain development in bread *** tamads29 mutants generated by CRISPR/Cas9 exhibited severe grain filling deficiency,coupled with excessive accumulation of reactive oxygen species(ROS)and abnormal programmed cell death that occurred in early developing grains,while overexpression of Ta MADS29 increased grain width and1,000-kernel *** analysis revealed that Ta MADS29 interacted directly with Ta NF-YB1;null mutation in Ta NF-YB1caused grain developmental deficiency similar to tamads29 *** regulatory complex composed of Ta MADS29 and Ta NF-YB1 exercises its possible function that inhibits the excessive accumulation of ROS by regulating the genes involved in chloroplast development and photosynthesis in early developing wheat grains and prevents nucellar projection degradation and endosperm cell death,facilitating transportation of nutrients into the endosperm and wholly filling of developing ***,our work not only discloses the molecular mechanism of MADS-box and NF-Y TFs in facilitating bread wheat grain development,but also indicates that caryopsis chloroplast might be a central regulator of grain development rather than merely a photosynthesis *** importantly,our work offers an innovative way to breed high-yield wheat cultivars by controlling the ROS level in developing grains.

OspTAC2 encodes a pentatricopeptide repeat protein and regulates rice chloroplast development

Functional chloroplast generation depends on the precise coordination of gene expression between the plastid and the nucleus and is essential for plant growth and development. In this study, a rice（Oryza sativa） mutant that exhibited albino and seedling-lethal phenotypes was isolated from a60Co-irradiated rice population. The mutant gene was identified as an ortholog of the Arabidopsis plastid transcriptionally active chromosome protein 2（p TAC2） gene, and the mutant strain was designated osptac2. Sequence and transcription analyses showed that Osp TAC2 encodes a putative chloroplast protein consisting of 10 pentratricopeptide repeat（PPR） domains and a C-terminal small Mut S-related（SMR） domain. Cytological observations via microscopy showed that the Osp TAC2-green fluorescent fusion protein is localized in the chloroplasts. Transmission electron microscopy revealed that the chloroplast of the osptac2 mutant lacks an organized thylakoid membrane. The transcript levels of all investigated PEP（plastid-encoded RNA polymerase）-dependent genes were dramatically reduced in the osptac2 mutant, whereas the transcript levels of NEP（nuclear-encoded polymerase）-dependent genes were increased. These results suggest that Osp TAC2 plays a critical role in chloroplast development and indicate that the molecular function of the Osp TAC2 gene is conserved in rice and Arabidopsis.

Comparative genetic analysis of Arabidopsis purple acid phosphatases AtPAP10, AtPAP12, and AtPAP26 provides new insights into their roles in plant adaptation to phosphate deprivation

Induction and secretion of acid phosphatases （APases） is thought to be an adaptive mechanism that helps plants survive and grow under phosphate （Pi） deprivation, in Arabidopsis, there are 29 purple acid phosphatase （AtPAP） genes. To systematically investigate the roles of different AtPAPs, we first identified knockout or knock-down T-DNA lines for all 29 AtPAP genes. Using these atpap mutants combined with in-gel and quantitative APase enzyme assays, we demonstrated that AtPAP12 and AtPAP26 are two major intracellular and secreted APases in Arabidopsis while AtPAPlo is mainly a secreted APase. On Pi-deficient （P-） medium or P- medium supplemented with the organophosphates ADP and fructose-6-phosphate （Fru-6-P）, growth of atpaplo was significantly reduced whereas growth of atpap12 was only moderately reduced, and growth of atpap26 was nearly equal to that of the wild type （WT）. Overexpression of the AtPAP12 or AtPAP26 gene, however, caused plants to grow better on P- or P- medium supplemented with ADP or Fru-6-P. Interest-ingly, Pi levels are essentially the same for the WT and overexpressing lines, although these two types of plants have significantly different growth phenotypes. These results suggest that the APases may have other roles besides enhancing internal Pi recycling or releasing Pi from external organophosphates for plant uptake.

Invertebrate species are a natural reservoir of viral genetic diversity,and invertebrate pests are widely distributed in crop ***,information on viruses infecting invertebrate pests of crops is *** this report,we describe the deep metatranscriptomic sequencing of 88 invertebrate samples covering all major invertebrate pests in rice *** identified 296 new RNA viruses and 13 known RNA *** viruses clustered within 31 families,with many highly divergent viruses constituting potentially new families and *** the identified viruses,13 RNA viruses clustered within the Fiersviridae family of bacteriophages,and 48 RNA viruses clustered within families and genera of *** detected known rice viruses in novel invertebrate hosts at high ***,some novel RNA viruses have genome structures closely matching to known plant viruses and clustered within genera of several plant virus *** potential insect pathogenic RNA viruses were detected in invertebrate *** analysis revealed that host taxonomy plays a major role and geographical location plays an important role in structuring viral ***-species transmission of RNA viruses was detected between invertebrate *** identified viral genomes showed extensive variation for invertebrate viral families or ***,the large-scale metatranscriptomic analysis greatly expands our understanding of RNA viruses in rice invertebrate species,the results provide valuable information for developing efficient strategies to manage insect pests and virus-mediated crop diseases.

Chemical Mechanism of Flower Color Microvariation in Paeonia with Yellow Flowers

Paeonia species are important ornamental plants,including three types:tree peony,herbaceous peony,and Itoh hybrid *** with yellow color is desired and becoming ***,the chemical mechanism of the formation of yellow flowers in Paeonia is still *** this study,14 representative samples were selected:four samples of three species of Paeonia,four herbaceous peony cultivars,three lutea hybrid tree peony cultivars,and three Itoh hybrid peony *** petal extracts of the samples were determined by HPLC-DAD and HPLCQ-TOF-MS/*** total,29 flavonoids were isolated and identified,including 28 flavonol glycosides and one chalcone *** were significant differences in the composition and content of flavonoids in petals of different cultivars(species).The total content of flavonoids was between 19.430 and 143.043 mg·g^−1 *** glycosides were detected in all *** 2′-O-glucoside was detected from one herbaceous peony cultivar‘Golden Wheel’lutea hybrid tree peony cultivars,and Itoh peony *** content of chalconaringenin 2′-O-glucoside in the samples was more than 43% of total *** pigment type and content of lutea hybrid tree peonies and Itoh hybrid peonies are *** flavonoids,especially quercetin 3-O-galloylglucoside and chalconaringenin 2′-O-glucoside contribute greatly to the formation of yellow flowers of *** results of this study provide a comprehensive understanding of the chemical mechanism for yellow flower coloration of Paeonia,and lay the foundation for yellow flower breeding of Paeonia.
