Efficiently tracking of stem cells in vivo using different kinds of superparamagnetic iron oxide in swine with myocardial infarction

Background Superparamagnetic iron oxide （SPIO） particles have shown much promise as a means to visualize labeled cells using molecular magnetic resonance imaging （MRI）. Micrometer-sized superparamagnetic iron oxide （MPIO）particles and nanometer-sized ultrasmall superparamagnetic iron oxide （USPIO） are two kinds of SPIO widely used for monitoring stem cells migration. Here we compare the efficiency of two kinds of SPIO during the use of stem cells to treat acute myocardial infarction （AMI）.Methods An AMI model in swine was created by 60 minutes of balloon occlusion of the left anterior descending coronary artery. Two kinds of SPIO particles were used to track after intracoronary delivered 107 magnetically labeled mesenchymal stem cells （MR-MSCs）. The distribution and migration of the MR-MSCs were assessed with the use of 3.0T MR scanner and then the results were confirmed by histological *** MR-MSCs appeared as a local hypointense signal on T2 -weighted MRI and there was a gradual loss of the signal intensity after intracoronary transplantation. All of the hypointense signals in the USPIO-labeled group were found on T2 -weighted MRI, contrast to noise ratio （CNR） decreased in the MPIO-labeled group （16.07±5.85 vs. 10.96±1.34）and USPIO-labeled group （11.72±1.27 vs. 10.03±0.96） from 4 to 8 weeks after transplantation. However, the hypointense signals were not detected in MPIO-labeled group in two animals. MRI and the results were verified by histological *** We demonstrated that two kinds of SPIO particles in vitro have similar labeling efficiency and *** is more suitable for labeling stem cells when they are transplanted via a coronary route.

Aphis gossypii has become increasingly difficult to manage due to its strong insecticide *** the laboratory,we established sulfoxaflor-resistant and acetamiprid-resistant strains in two *** populations with different basal insecticide resistance levels,and evaluated the effects of basal insecticide resistance on the resistance development and cross-resistance,as well as differences in *** the same selection pressure,Yarkant ***(with low basal insecticide resistance)evolved resistance to sulfoxaflor and acetamiprid more quickly than Jinghe ***(with high basal insecticide resistance),and the evolution of *** resistance to sulfoxaflor developed faster than acetamiprid in both Yarkant and Jinghe,Xingjiang,*** sulfoxaflor-resistant strains selected from Yarkant and Jinghe developed significant cross-resistance to acetamiprid,imidacloprid,thiamethoxam and pymetrozine;while the acetamiprid-resistant strains developed significant cross-resistance to sulfoxaflor,imidacloprid,thiamethoxam,pymetrozine,and *** relative fitness of *** decreased as the resistance to sulfoxaflor and acetamiprid *** relative fitness levels of the sulfoxaflor-resistant strains(Yarkant-SulR and Jinghe-SulR)were lower than those of the acetamipridresistant strains(Yarkant-AceR and Jinghe-AceR).In addition,the relative fitness levels of sulfoxaflor-and acetamiprid-resistant strains were lower in Jinghe than in *** summary,basal insecticide resistance of *** and insecticide type affected the evolution of resistance to insecticides in ***,as well as cross-resistance to other *** sulfoxaflor-and acetamiprid-resistant *** strains had obvious fitness *** results of this work will contribute to the insecticide resistance management and integrated management of ***.

Transplantation of magnetically labeled mesenchymal stem cells improves cardiac function in a swine myocardial infarction model

Background Mesenchymal stem cells （MSCs） transplantation provides a new approach for myocardial repair. However, many important fundamental questions about MSCs transplantation remain unanswered. There is an urgent need to identify MSCs from the beating heart and analyze the efficacy of this new approach. This study aimed to localize the magnetically labeled MSCs （MR-MSCs） and monitor the restorative effects of MR-MSCs with magnetic resonance （MR） imaging. Methods Acute myocardial infarction （AMI） was created in swine by a balloon occlusion of the left anterior descending coronary artery. Cells were delivered via intracoronary infusion after myocardial infarction. Infarct size change and cardiac function were assessed with 3.0T MR scanner. The results were then confirmed by histological and western blot analysis. All statistical procedures were performed with Systat （SPSS version 12.01）. Results A total of 26 swine were divided into four groups （sham-operated group, n=6; AMI group with PBS transplantation, n=6; labeled MSCs group, n=7; unlabeled MSCs group, n=7）. MSCs, MR-MSCs （10~cells） or PBS were delivered by intracoronary injection after MI and serial cardiac MR imaging studies were performed at 0, 4 and 8 weeks after transplantation. MR imaging demonstrated MI size decreased after MSCs transplantation in labeled and unlabeled groups, however, increases were seen in the AMI group at 8 weeks after MI. The left ventricular ejection fraction （LVEF） was slightly increased in the AMI group （（41.87~2.45）% vs （39.04~2.80）%, P 〉0.05）, but significantly improved in the MR-MSCs group （（56.85~1.29）% vs （40.67~2.00）%, P 〈0.05） and unlabeled group （（55.38~1.07）% vs （41.78~2.08）%, P 〈0.05） at 8 weeks after treatment. MR-MSCs were further confirmed by Prussian blue and immunofluorescent staining. 目的比较不同生物制剂维持治疗中重度溃疡性结肠炎(UC)患者的疗效和安全性的网状meta分析。方法检索中国知网、万方数据知识服务平台、维普资讯、中国生物医学文献数据库、PubMed、the Cochrane Library、Embase数据库中的RCTs,检索时间自数据库建库至2023年7月,收集不同生物制剂治疗中重度UC患者的相关研究。按照纳入和排除标准筛选文献、质量评价、提取数据资料后,采用RevMan 5.3和Stata16.0软件进行网状meta分析。结果共检索到1374篇文献,最终纳入17篇,涉及18项RCTs,5939例中重度UC患者,包括安慰剂、英夫利西单抗静脉注射、阿达木单抗皮下注射、维多珠单抗静脉注射、维多珠单抗皮下注射、戈利木单抗皮下注射、乌司奴单抗皮下注射每8周1次、乌司奴单抗皮下注射每12周1次、依妥珠单抗皮下注射、米吉珠单抗皮下注射10种干预措施,7种药品。网状meta分析结果显示,每项有效性指标中,不同用药方案有不同的表现,综合内镜下黏膜愈合和总不良事件发生情况,发现维多珠单抗皮下注射既能保证较好的黏膜愈合疗效,又是不良事件发生率最低的干预措施,而维多珠单抗静脉注射和米吉珠单抗皮下注射次之。结论治疗UC的生物制剂主要有3种作用通路,其中整合素拮抗剂维多珠单抗的疗效和安全性表现较好,且有皮下注射和静脉注射两种剂型供临床选择,白细胞介素抑制剂米吉珠单抗可以作为次选。

文中针对大型主动主反射面天线在满足工程精度要求时存在形面精度冗余和成本高昂的问题,提出一种主动面板和非主动面板混合安装的反射面天线设计思想。首先,基于反射面变形区间安装主动面板和非主动面板,分析混合面板反射面天线的形面调控策略;然后,分别以天线质量为约束、以天线形面精度为目标以及以天线形面精度为约束、以天线质量为目标进行天线结构优化设计。结果,某35m反射面天线在满足精度要求的情况下背架结构质量下降了20.4%,在满足背架结构质量要求的情况下形面精度提升了18.7%。这充分证明了该设计思想在大口径、高精度反射面天线设计中的可行性和有效性。

In the pursuit of high-performance single-molecule magnets（SMMs）,incorporating intramolecular magnetic coupling emerges as a pivotal *** these,d-f SMMs have garnered significant attention due to their remarkable versatility,which lies in their ability to tune coordination environments and facilely substitute metal ***,achieving performance-centric d-f SMMs through the synergistic interplay between highly anisotropic f ions and d-f magnetic interactions remains a formidable *** mononuclear hexagonal bipyramidal(D6h) DyⅢSMMs have been successfully isolated,the exploration of d-f SMMs featuring D6h-lanthanide metal centers remains uncharted *** this study,we employed planar bipodal ligands in conjunction with “staple-like” axial phenoxide ligands to synthesize the first hexagonal bipyramidal d-f ***,this compound exhibits alternating-current magnetic susceptibilities peaking up to 68 K with an energy barrier surpassing 1,200 K,thus establishing a new benchmark within the heterometallic d-f SMM landscape inclusive of complexes with diamagnetic d metals and paramagnetic f ***,the ferromagnetic interaction at the d-f sites engenders oscillating relaxation times contingent on the magnetic field—a characteristic distinct from mononuclear *** findings shed light on a deliberate design approach for d-f SMMs,emphasizing the cooperative utilization of high-barrier lanthanide modules alongside d ions through magnetic *** synergy significantly enhances and diversifies the magnetic dynamics of these intriguing molecular systems.

An integrated strategy for comprehensive characterization of metabolites and metabolic profiles of bufadienolides from Venenum Bufonis in rats

Comprehensive characterization of metabolites and metabolic profiles in plasma has considerable significance in determining the efficacy and safety of traditional Chinese medicine(TCM)in ***,this process is usually hindered by the insufficient characteristic fragments of metabolites,ubiquitous matrix interference,and complicated screening and identification procedures for *** this study,an effective strategy was established to systematically characterize the metabolites,deduce the metabolic pathways,and describe the metabolic profiles of bufadienolides isolated from Venenum Bufonis in *** strategy was divided into five ***,the blank and test plasma samples were injected into an ultra-high performance liquid chromatography/linear trap quadrupole-orbitrap-mass spectrometry(MS)system in the full scan mode continuously five times to screen for valid matrix compounds and ***,an extension-mass defect filter model was established to obtain the targeted precursor ions of the list of bufadienolide metabolites,which reduced approximately 39%of the interfering ***,an acquisition model was developed and used to trigger more tandem MS(MS/MS)fragments of precursor ions based on the targeted ion *** acquisition mode enhanced the acquisition capability by approximately four times than that of the regular data-dependent acquisition ***,the acquired data were imported into Compound Discoverer software for identification of metabolites with metabolic network *** main in vivo metabolic pathways of bufadienolides were elucidated.A total of 147 metabolites were characterized,and the main biotransformation reactions of bufadienolides were hydroxylation,dihydroxylation,and ***,the main prototype bufadienolides in plasma at different time points were determined using LC-MS/MS,and the metabolic profiles were clearly *** strategy could be widely used to elucidate the metabolic profiles

针对污水处理厂洪灾恢复具有超越常规工程建设的复杂性特点,提出了基于复杂系统管理思维的分析方法。以河北省污水处理厂灾后恢复为例,从工程复杂性和系统复杂性两方面进行分析,利用关联切割方法和目标适用性思维对问题进行难度降解,将灾后恢复工程分为功能性恢复和现常态化运营两个阶段,对各要素关联相对薄弱处进行切割来降低和分解该工程的固有复杂性,并按预期实现了该污水处理厂通水和达标目标。文章基于复杂系统管理思维形成的复杂工程分析方法可为类似工程提供借鉴和参考。

Quality influencing factors of dispersion curves from short period dense arrays based on a convolutional neural network across the north section of the Xiaojiang fault area

The number of dispersion curves increases significantly when the scale of a short-period dense array *** to a substantial increase in data volume,it is important to quickly evaluate dispersion curve quality as well as select the available dispersion ***,this study quantitatively evaluated dispersion curve quality by training a convolutional neural network model for ambient noise tomography using a short-period dense *** model can select high-quality dispersion curves that exhibit a≤10%difference between the results of manual screening and the proposed *** addition,this study established a dispersion curve loss function by analyzing the quality of the dispersion curve and the corresponding influencing factors,thereby estimating the number of available dispersion curves for the existing observation ***,a Monte Carlo simulation experiment is used to illustrates the station-pair interval distance probability density function,which is independent of station number in the observational system with randomly deployed *** results suggested that the straight-line length should exceed 15 km to ensure that loss rate of dispersion curves remains<0.5,while maintaining the threshold ambient noise tomography accuracy within the study area.
