目的:研究γ-突触核蛋白(γ-synuclein)对内质网应激诱导的结肠癌细胞自噬和凋亡的影响及其细胞保护作用。方法:针对稳定表达γ-synuclein siRNA的人结肠癌HCT116细胞和空载体HCT116细胞,通过基因表达谱芯片分析γ-synuclein下游差异基因,找到可能的自噬及凋亡相关分子。用毒胡萝卜素(thapsigargin,TG)模拟内质网应激,采用Western blot检测γ-synuclein蛋白、自噬相关蛋白[微管相关蛋白1轻链3(microtubule-associated protein 1 light chain 3,LC3)、beclin-1、自噬相关蛋白5(autophagy-related protein 5,ATG5)和ATG7]及凋亡相关蛋白{多腺苷二磷酸核糖聚合酶[poly(ADP-ribose)polymerase,PARP]、pro-caspase-3和pro-caspase-9}的表达,CCK-8法检测细胞活力,荧光显微镜观察吖啶橙染色的酸性囊泡细胞器和绿色荧光蛋白标记的LC3,检测人结肠癌HCT116和SW480细胞自噬、凋亡及活力的变化。分别采用γ-synuclein siRNA、细胞外信号调节激酶(extracellular signal-regulated kinase,ERK)抑制剂PD98059、c-Jun氨基末端激酶(c-Jun N-terminal kinase,JNK)抑制剂SP600125和JNK激活剂anisomycin预处理,Western blot检测γ-synuclein及ERK和JNK信号通路相关蛋白表达,检测伴随的HCT116细胞自噬和凋亡的变化,研究γ-synuclein通过调控相关信号通路对细胞自噬的影响及其细胞保护作用。结果:TG模拟内质网应激诱导的结肠癌细胞自噬主要发生在早期(0~24 h),细胞凋亡主要发生在晚期(36~48 h)。内质网应激上调γ-synuclein的表达,并伴有自噬的增强。γ-synuclein早期(0~24 h)通过激活ERK和JNK通路促进自噬,晚期(36~48 h)通过抑制JNK通路抑制凋亡,从而起到保护细胞的作用。γ-synuclein可能在内质网应激诱导的细胞自噬和凋亡之间的过渡发挥一定的作用。结论:在内质网应激环境下,γ-synuclein通过调控ERK和JNK信号通路来促进自噬、抑制凋亡,发挥针对结肠癌细胞的保护作用,这为抗肿瘤治疗提供了一个潜在的思路。

Brain encoding and decoding via functional magnetic resonance imaging(fMRI)are two important aspects of visual perception *** previous researchers have made significant advances in brain encoding and decoding models,existing methods still require improvement using advanced machine learning *** example,traditional methods usually build the encoding and decoding models separately,and are prone to overfitting on a small *** fact,effectively unifying the encoding and decoding procedures may allow for more accurate *** this paper,we first review the existing encoding and decoding methods and discuss the potential advantages of a“bidirectional”modeling ***,we show that there are correspondences between deep neural networks and human visual streams in terms of the architecture and computational ***,deep generative models(e.g.,variational autoencoders(VAEs)and generative adversarial networks(GANs))have produced promising results in studies on brain encoding and ***,we propose that the dual learning method,which was originally designed for machine translation tasks,could help to improve the performance of encoding and decoding models by leveraging large-scale unpaired data.

Inspired by the multi-layer architecture of mammal skins,interfacial robust,stretchable,and entirely healable gel-elastomer hybrids hold great potential in diverse fields including biomedical devices,wearable electrical devices,and soft ***,existing gel-elastomer hybrids have numerous limitations including low interfacial bonding toughness,complex and time-consuming preparation process,unhealable,and ***,we propose a simple and general chemical strategy through the interfacial dynamic bonding between gel and elastomer to simultaneously address the abovementioned *** covalent bonds readily and repeatably covalent bonding ionogel and elastomer(interfacial toughness:390 J m^(-2)),endowed the hybrids with entire self-healing features like skin and enabled discretionary assembly and ***,this strategy resolved the troublesome contradiction between interfacial stability and *** advantage of the aforementioned features,we readily constructed a multi-module,self-healing,self-powered,and realtime monitoring of personal status integrated elastic electronics,which could simply reconfigure the output signal of elastic electronics into an input signal of the devices-braille keyboard.

In^(3+)-doped Sr_(2)Fe_(1.5)Mo_(0.5)O_(6−δ)cathode with improved performance for an intermediate-temperature solid oxide fuel cell

Promoting the oxygen reduction reaction(ORR)is critical for commercialization of intermediate-temperature solid oxide fuel cells(IT-SOFCs),where Sr_(2)Fe_(1.5)Mo_(0.5)O_(6)−δ(SFM)is a promising cathode by working as a mixed ionic and electronic *** this work,doping of In^(3+)greatly increases the oxygen vacancy concentration and the content of adsorbed oxygen species in Sr_(2)Fe_(1.5)Mo_(0.5−x)InxO_(6−δ)(SFMInx),and thus effectively promotes the ORR *** a typical example,SFMIn_(0.1)reduces the polarization resistance(R_(p))from 0.089 to 0.046Ω∙cm^(2)at 800°C,which is superior to those doped with other metal *** addition,SFMIn0.1 increases the peak power density from 0.92 to 1.47 W∙cm^(−2)at 800°C with humidified H_(2)as the fuel,indicating that In3+doping at the Mo site can effectively improve the performance of SOFC cathode material.

外源添加物对桑黄液体发酵菌丝体和多糖含量的影响

为了探究外源添加物对桑黄液体发酵菌丝体产量及胞内、胞外多糖含量的影响,在桑黄液体发酵培养基中分别加入不同浓度的油酸、α-萘乙酸、维生素B2和桑白皮水提物4种添加物,经液体发酵后,测定了菌丝体产量及胞内、胞外多糖含量。试验结果显示,4种外源添加物都能显著增加桑黄菌丝体产量和多糖含量(P<0.05),其中最优外源添加物为油酸,当油酸浓度2.4 mL/L时,菌丝体产量最高,达2.86 mg/mL,是空白对照的7.15倍,在此油酸浓度下,胞外多糖含量最大,达56.04 mg/mL,是空白对照的7.71倍;当油酸浓度1.2 mL/L时,桑黄胞内多糖含量最高,达60.07 mg/g,是空白对照的3.96倍。其他3种添加物也可以不同程度地提高桑黄菌丝体及胞内、胞外多糖含量(P<0.05)。4种外源添加物是有效提高桑黄液体发酵菌丝体产量和多糖含量的诱导因子。
