氯代乙烯的厌氧微生物还原脱氯特性

四氯乙烯(PCE)和三氯乙烯(TCE)是地下水中典型的卤代有机化合物,严重威胁生态环境与人体健康.为获得氯代乙烯高效厌氧降解菌剂并探究其在污染地下水中的应用潜能,利用某工业污染场地的地下水,通过投喂PCE或TCE进行长期富集培养,获得了可将PCE和TCE完全脱氯成无毒乙烯的厌氧菌剂W-1.菌剂W-1的PCE和TCE脱氯速率分别是(120.1±4.9)μmol·(L·d)^(-1)和(172.4±21.8)μmol·(L·d)^(-1).16S rRNA基因扩增子测序和qPCR结果表明,98.3μmol PCE还原脱氯至顺-1,2-二氯乙烯(cis-1,2-DCE)时,Dehalobacter丰度从1.9%增长至57.1%,基因拷贝数每释放1μmol Cl-增加1.7×10^(7)copies;cis-1,2-DCE完全还原脱氯至乙烯时,Dehalococcoides丰度从1.1%增长至53.8%;PCE完全还原脱氯至乙烯过程中Dehalococcoides基因拷贝数每释放1μmol Cl^(-)增加1.7×10^(8)copies.以上结果说明Dehalobacter与Dehalococcoides协同互作实现PCE完全降解解毒.当菌群W-1以TCE为电子受体时,222.8μmol TCE完全还原脱氯至乙烯时候,Dehalococcoides丰度从(29.1±2.4)%增长至(77.7±0.2)%,基因拷贝数每释放1μmol Cl-增加(1.9±0.4)×10^(8)copies.结合PCR和Sanger测序,获得了菌剂W-1中主要脱卤菌Dehalococcoides LWT1较完整的16S rRNA基因序列,其与*** strain 19516S rRNA基因序列相似度达100%.将菌群W-1添加至受TCE(418.7μmol·L^(-1))污染的地下水中,28 d内实现了(69.2±9.8)%的TCE被完全脱毒至乙烯,TCE脱氯速率为(10.3±1.5)μmol·(L·d)^(-1).研究成果可为PCE或TCE污染地下水开展厌氧微生物修复提供菌剂资源和理论指导.

The BL14W1 beamline at Shanghai Synchrotron Radiation Facility(SSRF) is an X-ray absorption finestructure(XAFS) beamline, for investigating atomic local structure, which is demanded extensively in the fields of physics, chemistry, materials science, environmental science and so on. The beamline is based on a 38-pole wiggler with the maximum magnetic field of 1.2 T. X-rays of 4.5–40 ke V can be extracted by the optical scheme consisting of white beam vertical collimating mirror, liquid-nitrogen-cooled double crystal monochromator of Si(111) and Si(311), toroidal focusing mirror and higher harmonics rejection mirror. The maximum photon flux about 5 × 1012photons/s at the sample at 10 ke V, with a beam size of 0.3 mm × 0.3 mm. The beamline is equipped with four types of detectors for experiments in either transmission or fluorescence mode. At present,quick-XAFS, grazing incidence XAFS, X-ray emission spectroscopy, high-pressure XAFS and time-resolved X-ray excited optical luminescence methods have been developed.

钛酸盐纳米管对水中氨氮的吸附特性

以P25和Na OH为原料,采用水热法制备钛酸盐纳米管(TNTs),利用X射线衍射(XRD)、透射电镜(TEM)对材料的组成和形貌进行表征,通过其对水中氨氮的静态吸附实验,考察TNTs对水中氨氮的吸附特性及规律.结果表明碱浓度为10mol·L^-1时,可以获得管长约120 nm,管径约为8 nm的钛酸盐纳米管,其对氨氮的平衡吸附量达到10.67 mg·g^-1.p H值介于3~8时,TNTs能有效地吸附水中的氨氮.吸附过程在1 h基本达到平衡,符合准二级动力学方程.颗粒内扩散方程拟合结果发现,TNTs对氨氮的吸附过程由表面吸附和颗粒内扩散共同控制.Temkin方程能较好地描述TNTs对氨氮的吸附行为.热力学实验表明钛酸盐纳米管对氨氮的吸附是自发进行的吸热过程.共存阴阳离子对氨氮的吸附具有抑制作用,分别表现为SO4^2->Cl^->H2PO4^-、K^+>Na^+>Ca^2+.再生的钛酸盐纳米管对氨氮循环吸附5次仍有88.64%的吸附效果.红外光谱(FT-IR)研究表明钛酸盐纳米管对氨氮的吸附机制是TNTs层间的Na+与溶液中的NH4+之间发生离子交换.钛酸盐纳米管的优良循环使用性能和大吸附容量使得其能有效地去除水中氨氮.

Marsdeniae tenacissimae extract(MTE), commonly known as xiao-Ai-ping in China, is a traditional Chinese herb medicine capable of inhibiting proliferation and metastasis and boosting apoptosis in various cancer cells. However, little is known about the contribution of MTE towards tumor angiogenesis and the underlying mechanism. The present study aimed to evaluate the effects of MTE on the proliferation and apoptosis of human umbilical vein endothelial cells(HUVECs) and the molecular ism. 3-(4,5-dimethylthiazol-2-yl)-5(3-carboxymethoxyphenyl)-2-(4-sulfopheny)-2H-tetrazolium, inner salt(MTS) and PI-stained flow cytometry assays revealed that MTE dose-dependently reduced the proliferation of HUVECs by arresting cell cycle at S phase(P line in Bcl-2 expression; caspase-3 was activated simultaneously in a dose-dependent manner(P lial growth factor(VEGF) receptor-2(VEGFR-2), P2Y6 receptor(P2Y6R), and chemokine(C-C motif) ligand 2(CCL-2), along with the activation of PKC δ and up-regulation of p53 in a dose-dependent manner in MTE-treated selected cells(P liferation by attenuating CCL-2-mediated VEGF/VEGFR2 interactions and promoted the apoptosis through PKCδ-induced p53-dependent mitochondrial pathway in HUVECs, supporting that MTE may be developed as a potent anti-cancer medicine.

Background： Atopic dermatitis （AD） is an inflammatory skin disease characterized by chronic recurrent dermatitis with profound itching. Most patients have personal and/or family history of atopic diseases. Several criteria have been proposed for the diagnosis of AD. Although the clinical features of childhood AD have been widely studied, there has been less large-scale study on adult/adolescent AD. The aim of this study was to investigate the clinical features of adult/adolescent patients with chronic symmetrical eczemaJAD and to propose Chinese diagnostic criteria for adult/adolescent AD. Methods： A hospital-based study was performed. Forty-two dermatological centers participated in this study. Adult and adolescent patients （12 years and over） with chronic symmetrical eczema or AD were included in this study. Questionnaires were completed by both patients and-investigators. The valid questionnaires were analyzed using EpiData 3.1 and SPSS 17.0 software. Results： A total of 2662 valid questionnaires were collected （1369 male and 1293 female）. Of all 2662 patients, 2062 （77.5%） patients had the disease after 12 years old, while only 600 （22.5%） patients had the disease before 12 years old, suggesting late-onset eczema/AD is common. Two thousand one hundred and thirty-nine （80.4%） patients had the disease for more than 6 months. One thousand one hundred and forty-four （43.0%） patients had a personal and/or family history of atopic diseases. One thousand five hundred and forty-eight （58.2%） patients had an elevated total serum IgE and/or eosinophilia and/or positive allergen-specific IgE. Based on these clinical and laboratory features, we proposed Chinese criteria for adult/adolescent AD. Of all 2662 patients, 60.3% were satisfied with our criteria, while only 48.2% satisfied with Hanifin Rajka criteria and 32.7% satisfied with Williams criteria, suggesting a good sensitivity of our criteria in adult/adolescent AD patients. Cholangiocarcinoma refers to malignant tumors that develop in epithelial lining of biliary system, and it is divided into two categories according to tumor location, intrahepatic cholangiocarcinoma （ICC） and extrahepatic cholangiocarcinoma （ECC）. ICC occurs from the epithelial cells of the intrahepatic bile duct, its branches and interlobular biliary tree; and ECC is divided into hilar cholangiocarcinoma and distal cholangiocarcinoma by the circumscription at the confluence of cystic duct and the common hepatic duct.
