The mitogen-activated protein kinase(MAPK)cascade pathway plays an important role in regulating stress responses.The function of the c-jun NH_(2)-terminal kinase(JNK),a component of the MAPK cascade pathway,in Apis cerana cerana(Acc)remains unclear.Here,JNK was isolated and identified from Acc.Bioinformatics analyses revealed there is a typical serine/threonine protein kinase catalytic domain in the AccJNK protein.An expression profile analysis showed that AccJNK was significantly induced by pesticide treatments.To further explore the functional mechanisms of AccJNK,a yeast 2-hybrid screen was performed,activator protein-1(AP-1)was screened as the interaction partner of AccJNK,and the interaction relationship was further verified by pull-down assay.Quantitative real-time polymerase chain reaction showed the expression pattern of AccAP-I was similar to that of AccJNK.After a knockdown of AccJNK or AccAP-I by RNA interference,the survival rate of Acc after pesticide treatments increased.Additionally,the expression levels of antioxidant-related genes and the activities of antioxidant enzymes increased,suggesting that the knockdown of AccJNK or AccAP-I increased the antioxidant capacity of bees.Our study revealed that the JNK-mediated MAPK pathway responds to pesticide stress by altering the antioxidant capacity of Acc.

AIM:To investigate whether anti-placental growth factor(PGF) can inhibit subretinal fibrosis and whether this effect is mediated by the inhibitory effect of PGF on epithelial-mesenchymal transition(EMT) of retinal pigment epithelial(RPE) cells.METHODS:Subretinal fibrosis model was established in laser induced choroidal neovascularization(CNV) mice on day 21 after laser photocoagulation.Immunofluorescence staining(IFS) of cryosections and enzyme-linked immunosorbent assay(EliSA) were used to detect the expression of PGF.IFS of whole choroidal flat-mounts was used to detect the degree of subretinal fibrosis.IFS of cryosections and EliSA were used to detect the expression of EMT related indicators in subretinal fibrosis lesions.RESULTS:The expression of PGF protein in subretinal fibrosis lesions was significantly up-regulated(Plizing antibody reduced the area of subretinal fibrosis and the ratio of fibrotic/angiogenic area significantly at the concentrations of 0.25,0.5,1.0,and 2.0 μg/μL(all Plication of anti-PGF neutralizing antibody could reverse these changes(Plizing antibody can significantly inhibit the degree of subretinal fibrosis in CNV mice,and this effect may be mediated by the inhibition of PGF on EMT of RPE cells.

A multiphase field model coupled with a lattice Boltzmann(PF-LBM)model is proposed to simulate the distribution mechanism of bubbles and solutes at the solid-liquid interface,the interaction between dendrites and bubbles,and the effects of different temperatures,anisotropic strengths and tilting angles on the solidified organization of the SCN-0.24wt.%butanedinitrile alloy during the solidification process.The model adopts a multiphase field model to simulate the growth of dendrites,calculates the growth motions of dendrites based on the interfacial solute equilibrium;and adopts a lattice Boltzmann model(LBM)based on the Shan-Chen multiphase flow to simulate the growth and motions of bubbles in the liquid phase,which includes the interaction between solid-liquid-gas phases.The simulation results show that during the directional growth of columnar dendrites,bubbles first precipitate out slowly at the very bottom of the dendrites,and then rise up due to the different solid-liquid densities and pressure differences.The bubbles will interact with the dendrite in the process of flow migration,such as extrusion,overflow,fusion and disappearance.In the case of wide gaps in the dendrite channels,bubbles will fuse to form larger irregular bubbles,and in the case of dense channels,bubbles will deform due to the extrusion of dendrites.In the simulated region,as the dendrites converge and diverge,the bubbles precipitate out of the dendrites by compression and diffusion,which also causes physical phenomena such as fusion and spillage of the bubbles.These results reveal the physical mechanisms of bubble nucleation,growth and kinematic evolution during solidification and interaction with dendrite growth.

Insurance against climate risk is essential for mitigating the adverse effects of climate change.However,theoretical consensus regarding climate risk insurance remains elusive,and the implementation of climate insurance policies varies markedly between countries owing to various challenges.This study conducted bibliometric analysis of 1082 relevant publications(1975–2022)to determine the theoretical basis,evolution of research hotspots,and methodologies associated with climate risk insurance.Climate insurance publications are growing at an average annual rate of 8.9%,with more than 2333 authors from 1103 organizations in 78 countries publishing on the subject.On the basis of milestones of global climate change assessment,i.e.,the publication of the Fourth and Fifth Assessment Reports of the Intergovernmental Panel on Climate Change,climate insurance research can be divided into three major phases.In the start-up phase(1975–2007),research schemes examined the feasibilities and potentials of the National Flood Insurance Program in the United States,and the socioeconomic implications of transferring climate risk through reinsurance.The methodologies used in these studies were relatively simple owing to lack of comprehensive data.Research on flood insurance increased rapidly during the development phase(2008–2014),with increasing emphasis on the possibility of developing a flood insurance market in the Netherlands.Studies utilized catastrophe modeling and probabilistic approaches to estimate natural disaster losses and financial impacts.The boom phase(2015–2022)involved more research on the affordability of climate risk insurance given income inequality.The topic of climate insurance and the scope of its impact have developed global and interdisciplinary characteristics in terms of journal,sector,and disciplinary base.In the future,a trend might develop whereby big data will be combined with artificial intelligence and machine learning to design and implement index insurance.

Objective To screen antigen targets for immunotherapy by analyzing over-expressed genes,and to identify significant pathways and molecular mechanisms in esophageal cancer by using bioinformatic methods such as enrichment analysis,protein-protein interaction(PPI)network,and survival analysis based on the Gene Expression Omnibus(GEO)database.Methods By screening with highly expressed genes,we mainly analyzed proteins MUC13 and EPCAM with transmembrane domain and antigen epitope from TMHMM and IEDB websites.Significant genes and pathways associated with the pathogenesis of esophageal cancer were identified using enrichment analysis,PPI network,and survival analysis.Several software and platforms including Prism 8,R language,Cytoscape,DAVID,STRING,and GEPIA platform were used in the search and/or figure creation.Results Genes MUC13 and EPCAM were over-expressed with several antigen epitopes in esophageal squamous cell carcinoma(ESCC)tissue.Enrichment analysis revealed that the process of keratinization was focused and a series of genes were related with the development of esophageal cancer.Four genes including ALDH3A1,C2,SLC6A1,and ZBTB7C were screened with significant P value of survival curve.Conclusions Genes MUC13 and EPCAM may be promising antigen targets or biomarkers for esophageal cancer.Keratinization may greatly impact the pathogenesis of esophageal cancer.Genes ALDH3A1,C2,SLC6A1,and ZBTB7C may play important roles in the development of esophageal cancer.

Gastrointestinal cancer(GIC)is the most common cancer with a poor prognosis.Currently,surgery is the main treatment for GIC.However,the high rate of postoperative recurrence leads to a low five-year survival rate.In recent years,immunotherapy has received much attention.As the only immunotherapy drugs approved by the Food and Drug Administration(FDA),immune checkpoint blockade(ICB)drugs have great potential in cancer therapy.Nevertheless,the efficacy of ICB treatment is greatly limited by the low immunogenicity and immunosuppressive microenvironment of GIC.Therefore,the targets of immunotherapy have expanded from ICB to increasing tumor immunogenicity,increasing the recruitment and maturation of immune cells and reducing the proportion of inhibitory immune cells,such as M2-like macrophages,regulatory T cells and myeloid-derived suppressor cells.Moreover,with the development of nanotechnology,a variety of nanoparticles have been approved by the FDA for clinical therapy,so novel nanodrug delivery systems have become a research focus for anticancer therapy.In this review,we summarize recent advances in the application of immunotherapy-based nanoparticles in GICs,such as gastric cancer,hepatocellular carcinoma,colorectal cancer and pancreatic cancer,and described the existing challenges and future trends.

Non-dispersive solvent extraction （NDSE） with p-xylene as extractant was employed as a novel separation methodto recover both ρ-toluic （PT） acid and water from purified terephthalic acid （PTA） wastewater. The mass transport behavior ofPT acid from aqueous solution to ρ-xylene was investigated by experiments and numerical simulation. Experiments showed thatNDSE is feasible and effective. Residual PT acid in the raffinate can be reduced to lower than the permitted limit of wastewaterre-use （100 g/m^3） with extraction time longer than 60 s in industrial conditions. A mathematical model of PT acid mass transportwas developed to optimize the membrane module performance. The model was validated with the experimental results withrelative errors of less than 6%. Numerical analysis for mass transfer through the lumen side, the porous membrane layer, and theshell side showed that PT acid transport in the aqueous solution is the rate determining step. The effects of the membrane andoperating parameters on membrane module performance were investigated by means of computational simulations. The keyparameters suggested for industrial NDSE design are： fiber inner radius r1=200-250 μm, extraction time tc=50-60 s, aqueous/organic volumetric ratio a/o=9.0, and temperature T-318 K.

Validity of Diagnostic Evidence for Deceased Cases in Hospitals

Objective To determine the validity of the diagnostic evidence for deceased cases in hospitals. Methods All information collected from medical records of the deceased cases in tertiary care health facilities was input into ottr database. Four diagnosis levels were determined based on level of diagnostic evidence： level Ⅰ was based on autopsy, pathology or operative exploration, level Ⅱ on physical and laboratory tests plus expert clinical judgment, level Ⅲ on expert clinical judgment, level Ⅳ on postmortem assumptions. After the diagnostic evidence of each deceased case was reviewed by a panel of three experts, the diagnostic level of each diagnosis was determined. Results Among the 2102 medical cases for verbal autopsy study, only 26 （1.24%） afforded diagnostic evidence for level Ⅲ Among the level Ⅲ evidence-based cases of death, the major causes of death were cardiovascular diseases, respiratory diseases, and gastroenterological diseases. According to some special symptoms and medical histories, these cases could be diagnosed by comprehensive clinical judgment. Only one case met the criteria for level Ⅳ. Conclusion Level Ⅰ diagnostic evidence is hard to attain in China because of the traditional concept and economic restriction. The causes for 2101 deaths can be validated by level Ⅱ or Ⅲ diagnostic evidence.
