Fracture sealing performance of granular lost circulation materials at elevated temperature:A theoretical and coupled CFD-DEM simulation study

Lost circulation is a common downhole problem of drilling in geothermal and high-temperature,high-pressure(HTHP)formations.Lost circulation material(LCM)is a regular preventive and remedial measure for lost circulation.However,conventional LCMs seem ineffective in high-temperature formations.This may be due to the changes in the mechanical properties of LCMs and their sealing performance under high-temperature conditions.To understand how high temperature affects the fracture sealing performance of LCMs,we developed a coupled computational fluid dynamics-discrete element method(CFD-DEM)model to simulate the behavior of granular LCMs in fractures.We summarized the literature on the effects of high temperature on the mechanical properties of LCMs and the rheological properties of drilling fluid.We conducted sensitivity analyses to investigate how changing LCM slurry properties affected the fracture sealing efficiency at increasing temperatures.The results show that high temperature reduces the size,strength,and friction coefficient of LCMs as well as the drilling fluid viscosity.Smaller,softer,and less frictional LCM particles have lower bridging probability and slower bridging initiation.Smaller particles tend to form dual-particle bridges rather than single-particle bridges.These result in a deeper,tighter,but unstable sealing zone.Reduced drilling fluid viscosity leads to faster and shallower sealing zones.

As a widely used plasticizer,di-(2-ethylhexyl)phthalate(DEHP)is known to induce significant testicular injury.However,the potential mechanism and effects of pubertal exposure to DEHP on testis development remain unclear.In vivo,postnatal day(PND)21 male rats were gavaged with 0,250,and 500 mg/kg DEHP for ten days.Damage to the seminiferous epithelium and disturbed spermatogenesis were observed after DEHP exposure.Meanwhile,oxidative stress-induced injury and pyroptosis were activated.Both endoplasmic reticulum(ER)stress and mitophagy were involved in this process.Monoethylhexyl phthalate(MEHP)was used as the biometabolite of DEHP in vitro.The GC-1 and GC-2 cell lines were exposed to 0,100μM,200μM,and 400μM MEHP for 24 h.Reactive oxygen species(ROS)generation,oxidative stress damage,ER stress,mitophagy,and pyroptosis were significantly increased after MEHP exposure.The ultrastructure of the ER and mitochondria was destroyed.X-box binding protein 1(XBP1)was observed to be activated and translocated into the nucleus.ROS generation was inhibited by acetylcysteine.The levels of antioxidative stress,ER stress,mitophagy,and pyroptosis were decreased as well.After the administration of the ER stress inhibitor 4-phenyl-butyric acid,both mitophagy and pyroptosis were inhibited.Toyocamycin-induced XBP1 down-regulation decreased the levels of mitophagy and pyroptosis.The equilibrium between pyroptosis and mitophagy was disturbed by XBP1 accumulation.In summary,our findings confirmed that DEHP induced a ROS-mediated imbalance in pyroptosis and mitophagy in immature rat testes via XBP1.Moreover,XBP1 might be the key target in DEHP-related testis dysfunction.

Protein homeostasis is the basis of normal life activities,and the proteasomefamily plays an extremely important function in this process.The proteasome 2os is a concentric circle structure with twoαrings and twoβrings overlapped.The proteasome 2os can perform both ATP-dependent and non-ATP-dependent ubiquitination proteasome degradation by binding to various subunits(such as 19S,11S,and 200 PA),which is performed by its active subunitβ1,β2,andβ5.The proteasome can degrade misfolded,excess proteins tomaintain homeostasis.At the same time,it can be utilized by tumors to degrade over-proliferate and unwanted proteins to support their growth.Proteasomes can affect the development of tumors from several aspects including tumor signaling pathways such as NF-kB and p53,cell cycle,immune regulation,and drug resistance.Proteasome-encoding genes have been found to be overexpressed in a variety of tumors,providing a potential novel target for cancer therapy.In addition,proteasome inhibitors such as bortezomib,carfilzomib,and ixazomib have been put into clinical application as the first-line treatment of multiple myeloma.More and more studies have shown that it also has different therapeutic effects in other tumors such as hepatocellular carcinoma,non-small cell lung cancer,glioblastoma,and neuroblastoma.However,proteasome inhibitors are not much effective due to their tolerance and singleness in other tumors.Therefore,further studies on their mechanisms of action and drug interactions are needed to investigate their therapeutic potential.

The appropriate regulation of band structure is an effective strategy in constructing efficient photocatalytic systems.Present photocatalytic system mainly employs powder photocatalysts,which makes their recovery reliant on expensive separation processes and severely limits their industrial application.Herein,we constructed a novel Cd S/Ni3S2heterostructure using free-standing and flexible nickel fiber paper as the matrix.The regulated energy band structure achieves effective electron–hole separation.The as-synthesized flexible photocatalyst exhibits considerable photocatalytic activity toward the H2evolution reaction under visible-light irradiation,with an H2production rate of5.63μmol·cm-2·h-1(14.1 mmol·g-1cat·h-1according to the catalyst loading content).Additionally,the otherwisewasted excited holes simultaneously drive organic transformations to yield value-added organic products,thus markedly improving the photocatalytic H2evolution rate.Such a photocatalytic system is scaled up further,where a self-supported 20 cm×25 cm sample achieves a champion H2production rate of 60–80μmol·h-1under practical sun irradiation.This newly developed self-supported photocatalyst produces opportunities for practical solar H2production with biomass upgrading.

Optimal Timing of Business Conversion for Solvency Improvement

In this paper,we study the optimal timing to convert the risk of business for an insurance company in order to improve its solvency.The cash flow of company evolves according to a jump-diffusion process.Business conversion option offers the company an opportunity to transfer the jump risk business out.In exchange for this option,the company needs to pay both fixed and proportional transaction costs.The proportional cost can also be seen as the profit loading of the jump risk business.We formulated this problem as an optimal stopping problem.By solving this stopping problem,we find that the optimal timing of business conversion mainly depends on the profit loading of the jump risk business.A larger profit loading would make the conversion option valueless.The fixed cost,however,only delays the optimal timing of business conversion.In the end,numerical results are provided to illustrate the impacts of transaction costs and environmental parameters to the optimal strategies.

Antibiotic resistance is an escalating global concern,leading to millions of annual fatalities.Antibiotic resistance genes(ARGs)present in bacteria equip them to withstand the effects of antibiotics.Intra-and interspecific ARGs transmission through horizontal gene transfer further exacerbates resistance dissemination.The presence of ARGs in the environment heightens the probability of human exposure via direct inhalation,ingestion,or contact with polluted air,food,or water,posing substantial biosafety and health hazards.Consequently,ARGs represent a critical focal point in public health and environmental safety and are classified as emerging contaminants.This perspective underscores the necessity to assess ARG exposure within the One Health framework and to accord greater attention to the mitigation strategies and tactics associated with ARGs.

Phase field modeling of ferroelastic variant switching in yttria-stabilized t'zirconia with strain gradient elasticity and interface tension

The 6–8 wt%yttria-stabilized zirconia with a tetragonal structure(t’-YSZ)is extensively employed in thermal barrier coatings.The exceptional fracture toughness of t’-YSZ can be attributed to its distinctive ferroelastic toughening mechanism.Microstructure and interface tension play a critical role in ferroelastic variant switching at the micro-and nano-scale.This paper presents an original thermodynamically consistent phase field(PF)theory for analyzing ferroelastic variant switching at the micro-and nano-scale of t’-YSZ.The theory incorporates strain gradient elasticity using higher-order elastic energy and interface tension tensor via geometric nonlinearity to represent biaxial tension resulting from interface energy.Subsequently,a mixed-type formulation is employed to implement the higher-order theory through the finite element method.For an interface in equilibrium,the effects of strain gradient elasticity result in a more uniform distribution of stresses,whereas the presence of interface tension tensor significantly amplifies the stress magnitude at the interface.The introduction of an interface tension tensor increases the maximum value of stress at the interface by a factor of 4 to 10.The nucleation and evolution of variants at a pre-existing crack tip in a mono-phase t’-YSZ have also been studied.The strain gradient elasticity is capable of capturing the size effect of ferroelastic variant switching associated with microstructures in experiments.Specifically,when the grain size approaches that of the specimen,the critical load required for variant switching at the crack tip increases,resulting in greater dissipation of elastic energy during ferroelastic variant switching.Moreover,the interface tension accelerates the evolution of variants.The presented framework exhibits significant potential in modeling ferroelastic variant switching at the micro-and nano-scale.

Fabrication of patterned transparent conductive glass via laser metal transfer for efficient electrical heating and antibacteria

Vapor deposition and three-dimensional(3D)printing technology are considered to be conventional methods to achieve patterned metal film preparation through the assistance of masks and high temperature.Therefore,there are still some challenges in fabricating metal films in template-free and normal temperature environment.In this work,we report a flexible and rapid laser metal transfer(LMT)technique for fabricating the various metal films(Cu,Ni,Sn,Al,Fe,and Ag)with different patterns without templates on arbitrary substrates(glass,polyimide(PI)films,and aluminum nitride(AlN)ceramic).Especially,the obtained transparent conductive glass displays high transmittance(more than 90%)and adjustable resistances(≈5Ω).According to the Joule effect,the interface resistance between Cu particles and copper oxide coating produces the high temperature approximately 280℃ at 2 V in a short time(≈60 s)and remains stable at 120℃ over 12 h.At last,the multifunctional glass with Cu patterns also shows excellent bactericidal activity(≈95%).This work demonstrates that laser metal transfer is an exceeding effective means of fabricating the micro/nano structures with potential applications in functional devices.

BACKGROUND The development and progression of gastric cancer(GC)are closely linked to the nutritional status of patients.Although immunotherapy has been demonstrated to be clinically effective,the relationships of sarcopenia and myosteatosis with the use of immune checkpoint inhibitors(ICIs)in patients with gastric cancer remain to be characterized.METHODS We performed a retrospective study of patients who were undergoing immuno-therapy for GC.For the evaluation of sarcopenia,the optimal cut-off value for the skeletal muscle index was established using receiver operating characteristic analysis of data obtained from pre-treatment computed tomography images at the L3 vertebral level.Myosteatosis was defined using the mean skeletal muscle density(SMD),with a threshold value ofliable predictor of the clinical outcomes of patients with GC who are undergoing treatment with an ICI.

The growth-promoting hormones brassinosteroids(BRs)and their key signaling component BZR1 play a vital role in balancing normal growth and defense reactions.Here,we discovered that BRs and OsBZR1 up-regulated sakuranetin accumulation and conferred basal defense against Magnaporthe oryzae infection under normal conditions.Resource shortages,including phosphate(Pi)deficiency,potentially disrupt this growth-defense balance.OsSPX1 and OsSPX2 have been reported to sense Pi concentration and interact with the Pi signal mediator OsPHR2,thus regulating Pi starvation responses.In this study,we discovered that OsSPX1/2 interacts with OsBZR1 in both Pi-sufficient and Pi-deficient conditions,inhibit-ing BR-responsive genes.When Pi is sufficient,OsSPX1/2 is captured by OsPHR2,enabling most of OsBzR1 to promote plant growth and maintain basal resistance.In response to Pi starvation,more OsSPX1/2 is released from OsPHR2 to inhibit OsBZR1 activity,resulting in slower growth.Collectively,our study reveals that the OsBZR1-SPX1/2 module balances the plant growth-immunity trade-off in responsetoPiavailability.He Y.,Zhao Y.,Hu J.,Wang L.,li L.,Zhang X.,zhou Z.,Chen L.,Wang H.,Wang J.,and hong G.(2024).The OsBZR1-OsSPX1/2 module fine-tunes the growth-immunity trade-off in adaptation to phosphate availability in esponse to Pi availability.
