Cloning, expression and chitin-binding activity of two peritrophin-like protein genes in the common cutworm, Spodoptera fitura

Insect midgut secretes a semi-permeable peritrophic membrane （PM）, which plays important roles in protecting the midgut and helping with food digestion. The lep- idopteran larvae produce type 1 PM, which is degraded when insects develop into the metamorphic stages. However, the underlying mechanism is unclear. In the present study, two peritrophin-like proteins （peritrophin-57 and 37） were identified from the midgut expression sequence tag library and transcriptome of the common cutworm, Spodoptera litura. The temporal and spatial expression patterns and responses to the induction of 20- hydroxyecdysone （20E） and starvation were examined by real-time quantitative polymerase chain reaction according to their common sequence region. The chitin-binding activity was also studied using a competitor, calcofluor. The open reading frames are 1 554 and 1 020 bp, respectively. They shared four highly conserved peritrophin-A domains and were ex- pressed only in the midgut rather than in the other tissues, including fat body, epidermis, Malpighian tube and hemolymph. Their transcriptional expression could only be detected at the larval stages rather than in eggs, prepupae, pupae and adults. The purified protein of peritrophin-37 bound to chitin in a dose-dependent manner. These results indicate that the two proteins are peritrophins, the structural components of PM. In addition, the messenger RNA levels of the two peritrophins were significantly down-regulated by 20E injection, whereas feeding/starvation had no effect on the expression. These findings suggest that the increase of20E titer may be an important factor which controls the degradation of PM during metamorphosis.

We comment on an article by Koizumi et ***(EFN)is a dual pero-xisome proliferator-activated receptorα/δ*** experimental results from Koizumi et al demonstrated that EFN significantly increases intestinal barrier function and ameliorates liver *** positive outcomes suggest that EFN could be a promising therapeutic option for alcohol-associated liver disease(ALD).However,this study has limitations that necessitate further research to evaluate the efficacy of *** studies should consider the use of more appropriate animal models and cell types,optimize the administration routes and dosages of the drug,and conduct an in-depth investigation into the underlying mechanisms of action to determine the therapeutic effects of EFN in *** sustained and in-depth research,EFN has the potential to emerge as a novel therapeutic agent for the treatment of ALD.

Cretaceous-Neogene Exhumation of the Daqing Shan,North China Constrained by Apatite Fission Track Thermochronology

The Daqing Shan(DQS)located in the Yinshan-Yanshan Orogenic Belt plays an important role in the Mesozoic to Cenozoic evolution of the North China ***,the cooling and exhumation history since the Cretaceous is still *** the apatite fission track(AFT)data in both this study and previous works,a three-stage exhumation history from Cretaceous to Neogene of the DQS is proposed.(1)The first stage is composed of the early exhumation during Early Cretaceous driven by the collision between the North China and Siberia cratons(ca.148-132 Ma)and the far-field effect of the subduction of the Pacific Plate(ca.132-114 Ma).(2)Due to the subsidence of the Hetao Basin and the subsequent compensation between the DQS and the Hetao Basin,the DQS experienced the second rapid exhumation from Early Eocene to Early Oligocene(ca.54-29 Ma).(3)Since the Late Miocene(ca.13.5 Ma),the third rapid cooling and exhumation of the DQS occurred due to the far-field effect of the uplift of the Tibetan Plateau and the subduction of the Pacific Plate.

A non-structural protein encoded by Rice Dwarf Virus targets to the nucleus and chloroplast and inhibits local RNA silencing

RNA silencing is a potent antiviral mechanism in plants and animals. As a counter-defense, many viruses studied to date encode one or more viral suppressors of RNA silencing(VSR). In the latter case, how different VSRs encoded by a virus function in silencing remains to be fully understood. We previously showed that the nonstructural protein Pns10 of a Phytoreovirus, Rice dwarf virus(RDV), functions as a VSR. Here we present evidence that another nonstructural protein, Pns11, also functions as a VSR. While Pns10 was localized in the cytoplasm, Pns11 was localized both in the nucleus and chloroplasts. Pns11 has two bipartite nuclear localization signals(NLSs), which were required for nuclear as well as chloroplastic localization. The NLSs were also required for the silencing activities of Pns11. This is the first report that multiple VSRs encoded by a virus are localized in different subcellular compartments, and that a viral protein can be targeted to both the nucleus and chloroplast. These findings may have broad significance in studying the subcellular targeting of VSRs and other viral proteins in viral-host interactions.

Polylactide(PLA),methyl methacrylate-butadiene-styrene copolymer(MBS),and poly(propylene carbonate)polyurethane(PPCU)were blended and subjected to blown film *** rheological,mechanical,morphological,thermal,and crystalline properties of the PLA/MBS/PPCU ternary blends and the mechanical properties of the resulting films were *** of mechanical test showed that PPCU and MBS could synergistically toughen *** impact strength of 50/10/40 PLA/MBS/PPCU blend(74.7 k J/m^2)was about 7.5 times higher than that of the neat PLA(10.8 k J/m^2),and the elongation at break of 50/10/40 PLA/MBS/PPCU blend(276.5%)was higher by about 45 times that of PLA(6.2%).The tear strength of PLA/MBS/PPCU films was 20 k N/m higher than that of PLA,and the elongation at break(MD/TD)of 50/10/40 PLA/MBS/PPCU films was 271.1%/222.3%,whereas that of PLA was only 2.7%/3.0%.POM observations displayed that the density of spherulite nucleation increased and the size of crystalline particles decreased with the addition of *** increasing PPCU content from 5%to 20%,the density of spherulite nucleation increased and the size of crystalline particles decreased continuously,but the nucleation density of spherulites was slightly lowered with increasing PPCU content from 30%to 40%.The PLA/MBS/PPCU films exhibited excellent mechanical properties,which expanded the application range of these biodegradable films.

CELL DIVISION CONTROL PROTEIN48(CDC48)is essential for membrane fusion,protein degradation,and other cellular ***,we revealed the crucial role of CDC48B in regulating periclinal cell division in roots by analyzing the recessive gen1 *** identified the GEN1 gene through map-based cloning and verified that GEN1 encodes ***1 showed severely inhibited root growth,increased periclinal cell division in the endodermis,defective middle cortex(MC)formation,and altered ground tissue patterning in *** with these phenotypes,CYCliND 6;1(CYCD6;1),a periclinal cell division marker,was upregulated in gen1 compared to *** ratio of SHR_(pro):SHR-GFP fluorescence in pre-dividing nuclei versus the adjacent stele decreased by 33%in gen1,indicating that the trafficking of SHORT-ROOT(SHR)decreased in gen1 when endodermal cells started to *** findings suggest that the loss of function of CDC48B inhibits the intercellular trafficking of SHR from the stele to the endodermis,thereby decreasing SHR accumulation in the *** findings shed light on the crucial role of CDC48B in regulating periclinal cell division in roots.

Hepatic segmentectomy combined with major hepatic vein resection for preserving remnant liver lobe

BACKGROUND: Impairment of liver function is the most serious complication that occurs after liver resection or in cirrhotic liver. Postoperative hepatic failure, which is mainly preceded by insufficient remnant liver function and/or postoperative septic complications, is the major cause of hospital mortality. This study was undertaken to evaluate hepatic segmentectomy combined with major hepatic vein (MHV) resection for preserving the remnant liver lobe in the treatment of resectable primary liver cancer. METHODS: From 1997 to 2007, six patients with primary liver cancer underwent hepatic segmentectomy with MHV resection, and three patients with hepatic vein injury had ligation of the MHV. The remnant liver lobe was preserved after hepatic segmentectomy combined with MHV resection or ligation. RESULTS: The preserved liver lobe with normal structure could maintain hepatic function and showed no evidence of atrophy or swelling after hepatic segmentectomy combined with MHV resection or ligation. CONCLUSIONS: After the right inferior hepatic vein is confirmed, and the MHV is occluded experimentally before hepatic segmentectomy combined with MHV resection, progressively deteriorating congestion does not occur in the preserved segment. ligation or resection of the two MHVs must be avoided in patients with hepatic cirrhosis who have to undergo hepatic segmentectomy combined with MHV resection. ligation of the MHV in patients with juxtahepatic vein injury is a simple and effective therapeutic modality.
