LAPTM4B,which is a proto-oncogene,plays important roles in cancer progression,including clinical stages and pathologic grades.[1]The LAPTM4B gene encodes the lysosomal-associated protein transmembrane-4 beta(LAPTM4B)protein and LAPTM4B is cloned from hepatocellular carcinoma(HCC).[2]There is increasing evidence that LAPTM4B is a novel oncogene,which is over-expressed in many human solid tumors.[3]Furthermore,the up-regulation of LAPTM4B is associated with chemotherapy resistance,as well as poor clinical outcomes and prognoses.[4,5]LAPTM4B also promotes cell survival,growth,proliferation,metastasis,tolerance to metabolic stress,and autophagy.[6]Thus,the LAPTM4B protein may represent an excellent candidate for individualized treatment and tumor monitoring.

Industrial-use VOx-based catalysts usually have a higher active temperature window (>250-300℃),which becomes a“bottleneck”for the practical application of PCDD/Fs catalytic degradation technology.In this work,VO_(x)-FeO_(x)/TiO_(2) catalyst prepared via mechanochemically method was investigated for the catalytic removal of PCDD/Fs.The removal efficiency of 1,2-DCBz,pure PCDD/Fs gas generated in the lab,PCDD/Fs from actual fue gas,long-term were studied,and the degradation mechanism was explored using FTIR and TOFMS.The degradation efficiency of 1,2-DCBz and PCDD/Fs on VO_(x)-FeO_(x)/TiO_(2) were higher than that of VO_(x)/TiO_(2)catalyst,and the optimal FeOx addition ratio was 3 wt.%.The characterization results show that the addition of FeOx can effectively improve the pore structure,surface acidity,and VOx dispersion of the catalyst,thus contributing to increasing the V^(5+)content and surface-active oxygen,which is conducive to the improvement of adsorption and redox performance of the catalyst.Under the actual MSWI (municipal solid waste incineration)fue gas,the PCDD/Fs removal efficiency over VTi-3Fe-MC maintained long-term stability,higher than 85%for 240 min.This result was not significantly reduced compared with the data obtained in the laboratory.According to the analysis results of intermediate products by FTIR and GC-TOFMS,it can be inferred that the epoxidation fracture of benzene ring is the rate-limiting step of dioxin catalytic degradation reaction.This work gives an in-depth view into the PCDD/Fs removal over VO_(x)-FeO_(x)/TiO_(2) catalysts and could provide guidelines for the rational design of reliable catalysts for industrial applications.

The Co-Movements Between Crude Oil Price and Internet Concerns:Causality Analysis in the Frequency Domain

With the boom of web technology,Internet concerns(IC)have become emerging drivers of crude oil price.This paper makes the first attempt to measure the frequency-varying co-movements between crude oil price and IC in five domains(i.e.,fundamentals,supply-demand,crisis,war and weather)by using the frequency causality test method.Based on the monthly Brent spot price and search volumes(SVs)captured by Google Trends from January 2004 to September 2019,new and complementary insights regarding the co-movements between crude oil price and IC are obtained.1)The co-movements between crude oil price and the IC of supply-demand,war,and weather support a neutral hypothesis at all frequencies due to the characteristics(low value or volatility)of these IC data.2)There is a unidirectional causal relationship between crude oil price and the IC of fundamentals,running from the latter to the former at low frequencies(long-term).3)There is a feedback relationship between crude oil price and the IC of crisis,with the IC of crisis driving crude oil price at medium and low frequencies(mid-and long-term)and crude oil price causing the IC of crisis to change permanently.The conclusions of this paper provide important implications for both oil market economists and investors.

While hallmarks of rodent spermatogonia stem cell biomarkers' heterogeneity have recently been identified, their stage and subset distributions remain unclear. Furthermore, it is currently difficult to accurately identify subset-specific SSC marker distributions due to the poor nuclear morphological characteristics associated with fixation in 4% paraformaldehyde. In the present study, testicular cross-sections and whole-mount samples were Bouin fixed to optimize nuclear resolution and visualized by immunohistochemistry (IHC) and immunofluorescence (IF). The results identified an expression pattern of PLZFhighc-KITpos in A1 spermatogonia, while A2–A4-differentiating spermatogonia were PLZFlowc-KITpos. Additionally, this procedure was used to examine asymmetrically expressing GFRA1 and PLZF clones, asymmetric Apr and false clones were distinguished based on the presence or absence of TEX14, a molecular maker of intercellular bridges, despite having identical nuclear morphology and intercellular distances that were litates the accurate identification of spermatogonium subsets based on their molecular profiles and is capable of distinguishing asymmetric and false clones. Therefore, the findings presented herein will facilitate further morphological and functional analysis studies and provide further insight into spermatogonium subtypes.

Calcium-containing versus calcium-free replacement solution in regional citrate anticoagulation for continuous renal replacement therapy:a randomized controlled trial

Background: A simplified protocol for regional citrate anticoagulation (RCA) using a commercial calcium-containing replacement solution, without continuous calcium infusion, is more efficient for use in continuous renal replacement therapy (CRRT). We aim to design a randomized clinical trial to compare the safety and efficacy between calcium-free and calcium-containing replacement solutions in CRRT with RCA.Methods: Of the 64 patients receiving RCA-based postdilution continuous venovenous hemodiafiltration (CVVHDF) enrolled from 2017 to 2019 in West China Hospital of Sichuan University, 35 patients were randomized to the calcium-containing group and 29 to the calcium-free replacement solution group. The primary endpoint was circuit lifespan and Kaplan-Meier survival analysis was performed. Secondary endpoints included hospital mortality, kidney function recovery rate, and complications. The amount of 4% trisodium citrate solution infusion was recorded. Serum and effluent total (tCa) and ionized (iCa) calcium concentrations were measured during CVVHDF.Results: A total of 149 circuits (82 in the calcium-containing group and 67 in the calcium-free group) and 7609 circuit hours (4335 hvs. 3274 h) were included. The mean circuit lifespan was 58.1 h (95% CI 53.8-62.4 h) in the calcium-containing groupvs. 55.3 h (95% CI 49.7-60.9 h, log rankP = 0.89) in the calcium-free group. The serum tCa and iCa concentrations were slightly lower in the calcium-containing group during CRRT, whereas the postfilter iCa concentration was lower in the calcium-free group. Moreover, the mean amounts of 4% trisodium citrate solution infusion were not significantly different between the groups (171.1 ± 15.9 mL/hvs. 169.0 ± 15.1 mL/h,P = 0.49). The mortality (14/35 [40%]vs. 13/29 [45%],P = 0.70) and kidney function recovery rates of AKI patients (19/26, 73%vs. 14/24, 58%,P = 0.27) were comparable between the calcium-containing and calcium-free group during hospitalization, respectively. Regulating antioxidative stress pathways to augment oxidative stress and enhance antitumor therapy is highly desirable but very challenging.Herein,we initiated a multifunctional nanoparticle to regulate the Keap1-Nrf2 antioxidative stress pathway to promote cancer cell apoptosis.The OPFV-SnMP@GE11 nanoparticles were assembled by enzyme-activated OPFV-TLQ,tin mesoporphyrin(SnMP),and DSPEPEG-GE11.OPFV-SnMP@GE11 accumulated at tumor sites through specific targeting with GE11.OPFV-TLQ was specifically reduced to a photosensitizer OPFVNH2 by endocellular NAD(P)H:quinone oxidoreductase 1(NQO1).Under irradiation,OPFV-NH2 greatly produced reactive oxygen species(ROS)through a type I mechanism,which activated the Keap1-Nrf2 signal pathway and enhanced the transcription of NQO1,resulting in a continuous and explosive generation of ROS.Additionally,SnMP inhibited the activity of heme oxygenase-1(HO-1),further depressing antioxidative stress.This strategy provides insight into the regulation of the signal pathway to amplify oxidative stress,paving the way to studying the molecular mechanisms of cellular activities to enhance cancer therapy.

Polychlorinated dibenzo-p-dioxins and polychlorinated dibenzofurans(PCDD/Fs)have attracted widespread concern due to their high toxicity,and their difficult manipulation in laboratories has made the research process tough.Thus,in our work,furan is selected as the model compound owing to the same structure of a central oxygenate ring.Although catalytic oxidation is regarded as an effective and applicable method for the abatement of PCDD/Fs,the synthesis of low-temperature catalysts is still a challenging problem in practical applications.Considering this situation,we prepared a novel V_(2)O_(5)/TiO_(2)catalyst modified with N-doped hierarchical porous carbon(NHPC)via a wet impregnation method.The V/T-1%NHPC catalyst could achieve expectant low-temperature performances with 50%furan conversion at 150℃and a complete conversion at 200℃,which decreased 23℃and 40℃compared to the V/T catalyst respectively.Moreover,the addition of NHPC presented lifting chemical stability during long-time test.The addition of NHPC in V/T catalysts decreased the formation of crystalline V_(2)O_(5) and increased the percentages of V^(5+)and O_(lat),which improved the utilization of vanadium ions and the catalytic activity.Simultaneously,the higher binding energy shift of O_(lat) implied more reaction possibility with other oxidise reactants.Importantly,this work proved the lifting catalytic activity by the interaction between catalysts and NHPC,and proposed the promoting effects of the N element.The results showed that the content of the pyridinic N and graphitic N in NHPC changed after combining with V/T catalyst,which played crucial roles in the excellent catalytic performance.Overall,this work provides comprehensive research of the V/T-1%NHPC catalyst toward furan oxidation at low temperature and explain the effects of N-doped biomass carbon in catalytic activity clearly,which gave a new thought to design low-temperature catalysts in PCDD/Fs degradation.Besides,the internal functional mechanisms of N species are worth further exploratio

Long-Term Reproducibility of SIMS Zircon U-Pb Geochronology

Secondary ion mass spectrometry(SIMS) zircon U-Pb dating has been widely used to confine the absolute ages of the magmatic or metamorphic events and to distinguish multiple events.Here,a data set consisting of the zircon standards dating data(~15 000 Ple?ovice and ~8 000 Qinghu items) accumulated for more than 8 years using the CAMECA IMS 1280 HR of the Institute of Geology and Geophysics,Chinese Academy of Sciences(IGGCAS) is compiled to evaluate the long-term external reproducibility.With Ple?ovice zircon as the calibration standard and Qinghu zircon as an unknown sample,the analytical uncertainties for single-spot(n = 7 723) and session analyses(n = 691,with more than 7 single spot analyses in one session) are 2.6% and 0.9%(2 RSD,relative standard deviation),respectively.It means that single-spot U-Pb dating for a standard-level zircon sample could vary 5.2% at 95% confidence level,while the average value(n > 7) for each session may vary 1.8%.Thus,these values should be considered as the minimum uncertainty when comparing single spot and individual session analysis results for multiple dating works on a certain geological event.

Targeted delivery of therapeutics for spinal cord injury(SCI)has been a long-term challenge due to the complexity of the pathological procession.Macrophage,as an immune cell,can selectively accumulate at the trauma site after SCI.This intrinsic targeting,coupled with good immune-escaping capacity makes macrophages an ideal source of biomimetic delivery carrier for SCI.Worth mentioning,macrophages have multiple polarization states,which may not be ignored when designing macrophage-based delivery systems.Herein,we fabricated macrophage membrane-camouflaged liposomes(RM-liPs)and evaluated their abilities to extend drug circulation time and target the injured spinal cord.Specially,we detected the expression levels of the two main targeted receptors Mac-1 and integrinα4 in three macrophage subtypes,including unactivated(M0)macrophages,classically activated(M1)macrophages and alternatively activated(M2)macrophages,and compared targeting of these macrophage membrane-coated nanoparticles for SCI.The macrophage membrane camouflage decreased cellular uptake of liposomes in RAW264.7 immune cells and strengthened binding of the nanoparticle to the damaged endothelial cells in vitro.RM-liPs can prolong drug circulation time and actively accumulate at the trauma site of the spinal cord in vivo.Besides,RM-liPs loaded with minocycline(RM-liP/MC)showed a comprehensive therapeutic effect on SCI mice,and the anti-pyroptosis was found to be a novel mechanism of RM-liP/MC treatment of SCI.Moreover,the levels of Mac-1 and integrinα4 in macrophages and the targeting of RM-liP for SCI were found to be independent of macrophage polarization states.Our study provided a biomimetic strategy via the biological properties of macrophages for SCI targeting and treatment.

Spermatogenesis is a complex process in which spermatogonial stem cells differentiate and develop into mature spermatozoa.The transcriptional regulatory network involved in fish spermatogenesis remains poorly understood.Here,we demonstrate in Nile tilapia that the Sox transcription factor family member Sox30 is specifically expressed in the testes and mainly localizes to spermatocytes and spermatids.CRISPR/Cas9-mediated sox30 mutation results in abnormal spermiogenesis,reduction of sperm motility,and male subfertility.Comparative transcriptome analysis shows that sox30 mutation alters the expression of genes involved in spermatogenesis.Further chromatin immunoprecipitation followed by high-throughput sequencing(Ch IP-seq),Ch IP-PCR,and luciferase reporter assays revealed that Sox30 positively regulates the transcription of ift140 and ptprb,two genes involved in spermiogenesis,by directly binding to their promoters.Our data,taken together,indicate that Sox30 plays an essential role in Nile tilapia spermatogenesis by directly regulating the transcription of the spermiogenesis-related genes ift140 and ptprb.
