Downregulation of Na^＋/Ca^2＋ Exchanger Isoform 1 Protects Isolated Hearts by Sevoflurane Postconditioning but Not by Delayed Remote Ischemic Preconditioning in Rats

Background：Calcium regulatory proteins-L-type Ca^2＋ channels （LTCCs）,ryanodine receptor 2 （RyR2）,and Na^＋/Ca^2＋ exchanger isoform 1 （NCX1） have been recognized as important protective mechanisms during myocardial ischemia-reperfusion injury （I/RI）.Both sevofturane postconditioning （SevoPoC） and delayed remote ischemic preconditioning （DRIPC） have been shown to protect the heart against I/RI.In this study,we aimed to compare the effects of SevoPoC and DRIPC on the expression of the three calcium regulatory proteins in an isolated rat heart model.Methods：After 30-min balanced perfusion,isolated hearts from rats were subjected to 30-min ischemia followed by 60-min reperfusion.Totally 40 isolated hearts were randomly assigned to four groups （n =10/group）：time control group,I/RI group,SevoPoC group,and DRIPC group.The effect of SevoPoC （3% v/v） and DRIPC were observed.Myocardial infarct size （IS）,cardiac troponin I level,and heart function were measured.The protein and messenger RNA levels of LTCCs,RyR2,and NCX1 were determined.Results：Both SevoPoC and DRIPC improved the recovery of myocardial function,and reduced cardiac troponin Ⅰ release after I/RI.The decrease in IS was more significant in the SevoPoC group than that in the DRIPC group （16.50% ± 4.54% in the SevoPoC group [P =0.0006],and 22.34% ± 4.02% in the DRIPC group [P =0.0007] vs.35.00% ± 5.24% in the I/RI group,respectively）.SevoPoC,but not DRIPC significantly inhibited the activity of NCX 1 （0.59 ＋ 0.09 in the I/RI group vs.0.32 ± 0.16 in the SevoPoC group,P =0.006;vs.0.57 ± 0.14 in the DRIPC group,P =0.072）.No statistical significant differences were observed in the expression of LTCCs and RyR2 between SevoPoC and DRIPC.In addition,subsequent correlation analysis showed a significantly positive relationship between the cardiac troponin Ⅰ level and the protein expression ofNCX1 （r =0.505,P =0.023）.Conclusion：SevoPoC may be more effective in the cardioprotection than DRIPC partly due to the deactivation of NCX1.

基于CRISPR/Cas9的棉花GhbHLH71基因编辑突变体的分析

碱性/螺旋-环-螺旋(basic/helix-loop-helix,bHLH)转录因子在植物的生长发育、次生代谢、信号转导、逆境胁迫等方面起重要的调控作用。棉花GhbHLH71基因c DNA全长996 bp,编码331个氨基酸残基,蛋白序列中含有保守的bHLH结构域,属于bHLH转录因子家族成员。实时荧光定量分析结果表明,GhbHLH71基因在棉花纤维快速伸长期3~12 DPA (days post anthesis,DPA)相对高表达,暗示其主要在棉花纤维发育过程中发挥作用。构建该基因的CRISPR/Cas9基因编辑载体进行了棉花遗传转化,T_0代再生株经过Cas9基因的PCR检测以及靶位点突变情况检测分析,获得6个T_0代基因编辑突变体。T_1代不同突变株系在棉花生长发育期的表型性状与对照相比无明显差异,但成熟纤维的长度与对照相比皆明显变短。T_2代突变株系可以稳定遗传T_1代株系纤维变短的表型。其中4#和8#株系连续2代的纤维长度与对照相比缩短比率皆达20%以上,表明GhbHLH71基因的突变主要影响了棉花纤维细胞的伸长。本研究为深入了解棉花中bHLH转录因子的生物学功能以及棉花纤维发育的分子机制提供了一定的参考。

为探究新乡市大气PM_(2.5)中水溶性无机离子(WSIIs)的污染演变、来源特征及其气象影响,利用URG-9000在线监测系统于2022年1月(冬季)、4月(春季)、7月(夏季)和10月(秋季)对PM_(2.5)组分进行在线观测.结果表明,TWSIIs(总水溶性无机离子)与PM_(2.5)的季节变化特征一致,季度ρ(TWSIIs)均值变化范围为19.62~72.15μg·m^(-3),在PM_(2.5)中的占比超过66%,WSIIs是大气PM_(2.5)的重要组分.年均NO_(3)^(-)/SO_(4)^(2-)(质量浓度比)为2.11,且呈现逐年增加的趋势,移动源对二次无机气溶胶(SNA)的影响不容忽视,年均[NH_(4)^(+)]/[NO_(3)^(-)](量比)为1.95,说明农业源是大气中氮的主要贡献者.后向轨迹分析表明,在盛行东北风且风速较大时,PM_(2.5)中Ca^(2+)和Mg^(2+)的浓度较高.低温高湿的气象条件下(T60%),SOR和NOR值均较高,更多的气态前体物SO_(2)和NO_(2)转化为颗粒态的SO_(4)^(2-)和NO_(3)^(-).与SOR不同,在高温条件下(T>24℃),NOR并没有表现出高值特征,与高温条件下NH_(4)NO_(3)的分解有关.结合PMF和后向轨迹分析,来自西北方向的气团所对应的扬尘源对WSIIs的贡献较大,观测站点周边区域的低空低速气团所对应的二次硫酸盐以及二次硝酸盐和生物质源对WSIIs的贡献较大.

Beneficial effects of silicon on photosynthesis of tomato seedlings under water stress

Silicon can improve drought tolerance of plants,but the mechanism still remains unclear.Previous studies have mainly concentrated on silicon-accumulating plants,whereas less work has been conducted in silicon-excluding plants,such as tomato（Solanum lycopersicum L.）.In this study,we investigated the effects of exogenous silicon（2.5 mmol L^（-1））on the chlorophyll fluorescence and expression of photosynthesis-related genes in tomato seedlings（Zhongza 9）under water stress induced by 10%（w/v）polyethylene glycol（PEG-6000）.The results showed that under water stress,the growth of shoot and root was inhibited,and the chlorophyll and carotenoid concentrations were decreased,while silicon addition improved the plant growth and increased the concentrations of chlorophyll and carotenoid.Under water sterss,chlorophyll fluorescence parameters such as PSII maximum photochemical efficiency（F_v/F_m）,effective quantum efficiency,actual photochemical quantum efficiency（Ф_（PSII））,photosynthetic electron transport rate（ETR）,and photochemical quenching coefficient（q_P）were decreased;while these changes were reversed in the presence of added silicon.The expressions of some photosynthesis-related genes including PetE,PetF,PsbP,PsbQ,PsbW,and Psb28 were down-regulated under water stress,and exogenous Si could partially up-regulate their expressions.These results suggest that silicon plays a role in the alleviation of water stress by modulating some photosynthesis-related genes and regulating the photochemical process,and thus promoting photosynthesis.

Non-Convex Optimization of Resource Allocation in Fog Computing Using Successive Approximation

Fog computing can deliver low delay and advanced IT services to end users with substantially reduced energy consumption.Nevertheless,with soaring demands for resource service and the limited capability of fog nodes,how to allocate and manage fog computing resources properly and stably has become the bottleneck.Therefore,the paper investigates the utility optimization-based resource allocation problem between fog nodes and end users in fog computing.The authors first introduce four types of utility functions due to the diverse tasks executed by end users and build the resource allocation model aiming at utility maximization.Then,for only the elastic tasks,the convex optimization method is applied to obtain the optimal results;for the elastic and inelastic tasks,with the assistance of Jensen’s inequality,the primal non-convex model is approximated to a sequence of equivalent convex optimization problems using successive approximation method.Moreover,a two-layer algorithm is proposed that globally converges to an optimal solution of the original problem.Finally,numerical simulation results demonstrate its superior performance and effectiveness.Comparing with other works,the authors emphasize the analysis for non-convex optimization problems and the diversity of tasks in fog computing resource allocation.

Enhanced properties of stone coal-based composite phase change materials for thermal energy storage

Phase change materials (PCMs) can be incorporated with low-cost minerals to synthesize composites for thermal energy storage in building applications.Stone coal (SC) after vanadium extraction treatment shows potential for secondary utilization in composite preparation.We prepared SC-based composite PCMs with SC as a matrix,stearic acid (SA) as a PCM,and expanded graphite (EG) as an additive.The combined roasting and acid leaching treatment of raw SC was conducted to understand the effect of vanadium extraction on promoting loading capacity.Results showed that the combined treatment of roasting at 900℃ and leaching increased the SC loading of the composite by 6.2%by improving the specific surface area.The loading capacity and thermal conductivity of the composite obviously increased by 127%and 48.19%,respectively,due to the contribution of 3wt% EG.These data were supported by the high load of 66.69%and thermal conductivity of 0.59 W·m^(-1)·K-1of the designed composite.The obtained composite exhibited a phase change temperature of 52.17℃,melting latent heat of 121.5 J·g^(-1),and good chemical compatibility.The SC-based composite has prospects in building applications exploiting the secondary utilization of minerals.

A molecular insight into coke formation process of vacuum residue in thermal cracking reaction

Understanding the coking behaviors has been considered to be really essential for developing better vacuum residue processing technologies. A battery of thermal cracking tests of typical vacuum residue at410°C with various reaction time were performed to evaluate the coke formation process. The total yields of ideal components including naphtha, atmospheric gas oil(AGO) and vacuum gas oil(VGO) of thermal cracking reactions increased from 10.89% to 40.81%, and the conversion ratios increased from8.05% to 43.33% with increasing the reaction time from 10 to 70 min. The asphaltene content increased from 12.14% to a maximum of 22.39% and then decreased, and this maximum of asphaltene content occurred at the end of the coking induction period. The asphaltenes during the coking induction period,at the end and after coking induction period of those tested thermal cracking reactions were characterized to disclose the structure changing rules for coke formation process, and the coke formation pathways were discussed to reveal the coke formation process at molecular level.

First record of abnormal body coloration in a rockfish Sebastes koreanus(Scorpaenoidei:Sebastidae)from coastal water of China based on morphological characteristics and DNA barcoding

The first record of abnormal body coloration in Sebastes koreanus Kim and Lee,1994,from the Yellow Sea of China,was documented based on morphological characteristics and DNA barcoding.The two rockfish specimens were collected from the coastal waters of Qingdao,China,and the whole body and all fins of them were red.Of the two red-colored rockfish,there were tiny deep red spots on each fin,2 red radial stripes behind and below the eyes and 1 large deep red blotch on the opercula,while the similar stripe and spot patterns are also present in the S.koreanus specimens with normal body coloration.The countable characteristics of the two specimens are in the range of the morphometry of S.koreanus.To further clarify the species identity and taxonomic status of the two specimens,DNA barcode analysis was carried out.The genetic distance between the red-colored rockfish and S.koreanus was 0,and the minimum net genetic distances between the red-colored rockfish and other Sebastes species except for S.koreanus were 3.0%,which exceeds the threshold of species delimitation.The phylogenetic analysis showed that the DNA barcoding sequences of the two red-colored rockfish clustered with the S.koreanus sequences.The above results of DNA barcode analysis also support that the two red-colored rockfish could be identified as the species of S.koreanus.The mechanism of color variation in S.koreanus is desirable for further research and the species could be an ideal model to study the color-driven speciation of the rockfishes.

LFM Radar Source Passive Localization Algorithm Based on Range Migration

Traditional single-satellite passive localization algorithms are influenced by frequency and angle measurement accuracies,resulting in error estimation of emitter position on the order of kilometers.Subsequently,a single-satellite localization algorithm based on passive synthetic aper-ture(PSA)was introduced,enabling high-precision positioning.However,its estimation of azimuth and range distance is considerably affected by the residual frequency offset(RFO)of uncoopera-tive system transceivers.Furthermore,it requires data containing a satellite flying over the radia-tion source for RFO search.After estimating the RFO,an accurate estimation of azimuth and range distance can be carried out,which is difficult to achieve in practical situations.An LFM radar source passive localization algorithm based on range migration is proposed to address the dif-ficulty in estimating frequency offset.The algorithm first provides a rough estimate of the pulse repetition time(PRT).It processes intercepted signals through range compression,range interpola-tion,and polynomial fitting to obtain range migration observations.Subsequently,it uses the changing information of range migration and an accurate PRT to formulate a system of nonlinear equations,obtaining the emitter position and a more accurate PRT through a two-step localization algorithm.Frequency offset only induces a fixed offset in range migration,which does not affect the changing information.This algorithm can also achieve high-precision localization in squint scenar-ios.Finally,the effectiveness of this algorithm is verified through simulations.

We present the case of a patient with iridoschisis complicated with cataract,peripheral anterior synechiae(PAS),secondary glaucoma,and corneal endothelial damage.The patient was initially misdiagnosed with acute angle-closure glaucoma.Iridoschisis is a rare condition characterized by the splitting of the iris into two layers:the anterior layer breaks down into fibers,floating freely in the anterior chamber with a“shredded wheat”appearance.
