Association of DNA methylation/demethylation with the functional outcome of stroke in a hyperinflammatory state

Inflammation is closely related to stroke prognosis, and high inflammation status leads to poor functional outcome in stroke. DNA methylation is involved in the pathogenesis and prognosis of stroke. However, the effect of DNA methylation on stroke at high levels of inflammation is unclear. In this study, we constructed a hyperinflammatory cerebral ischemia mouse model and investigated the effect of hypomethylation and hypermethylation on the functional outcome. We constructed a mouse model of transient middle cerebral artery occlusion and treated the mice with lipopolysaccharide to induce a hyperinflammatory state. To investigate the effect of DNA methylation on stroke, we used small molecule inhibitors to restrain the function of key DNA methylation and demethylation enzymes. 2,3,5-Triphenyltetrazolium chloride staining, neurological function scores, neurobehavioral tests, enzyme-linked immunosorbent assay, quantitative reverse transcription PCR and western blot assay were used to evaluate the effects after stroke in mice. We assessed changes in the global methylation status by measuring DNA 5-mc and DNA 5-hmc levels in peripheral blood after the use of the inhibitor. In the group treated with the DNA methylation inhibitor, brain tissue 2,3,5-triphenyltetrazolium chloride staining showed an increase in infarct volume, which was accompanied by a decrease in neurological scores and worsening of neurobehavioral performance. The levels of inflammatory factors interleukin 6 and interleukin-1 beta in ischemic brain tissue and plasma were elevated, indicating increased inflammation. Related inflammatory pathway exploration showed significant overactivation of nuclear factor kappa B. These results suggested that inhibiting DNA methylation led to poor functional outcome in mice with high inflammation following stroke. Worldwide,the incidence of major depressive disorder(MDD)is increasing annually,resulting in greater economic and social burdens.Moreover,the pathological mechanisms of MDD and the mechanisms underlying the effects of pharmacological treatments for MDD are complex and unclear,and additional diagnostic and therapeutic strategies for MDD still are needed.The currently widely accepted theories of MDD pathogenesis include the neurotransmitter and receptor hypothesis,hypothalamicpituitary-adrenal(HPA)axis hypothesis,cytokine hypothesis,neuroplasticity hypothesis and systemic influence hypothesis,but these hypothesis cannot completely explain the pathological mechanism of MDD.Even it is still hard to adopt only one hypothesis to completely reveal the pathogenesis of MDD,thus in recent years,great progress has been made in elucidating the roles of multiple organ interactions in the pathogenesis MDD and identifying novel therapeutic approaches and multitarget modulatory strategies,further revealing the disease features of MDD.Furthermore,some newly discovered potential pharmacological targets and newly studied antidepressants have attracted widespread attention,some reagents have even been approved for clinical treatment and some novel therapeutic methods such as phototherapy and acupuncture have been discovered to have effective improvement for the depressive symptoms.In this work,we comprehensively summarize the latest research on the pathogenesis and diagnosis of MDD,preventive approaches and therapeutic medicines,as well as the related clinical trials.

The response and anti-stress mechanisms of nitrifying sludge under long-term exposure to Cd Se quantum dots

The wide application of CdSe quantum dots(QDs)increases its stress risk to sewage treatment systems.This study evaluated the response of nitrification performance,floc characteristics and microbial community of nitrifying sludge under long-term exposure to CdSe QDs.Results showed CdSe QDs(≥1 mg/L)would decrease the activity of ammonia monooxygenase(AMO).Under the stress of 30 mg/L CdSe QDs,the activity of AMO was reduced by 66%,while the activities of hydroxylamine oxidase and nitrite oxidoreductase were enhanced by 19.1%and 26%,respectively.Thus,the final nitrification effects were not adversely affected,and the production rates of NO_(2)^(-)-N and NO_(3)^(-)-N were accelerated.Additionally,CdSe QDs improved biomass concentration in sludge and maintained the stability of sludge settleability.High throughput sequencing analysis showed that CdSe QDs evidently reduced the abundance and diversity of microbial community in nitrifying sludge.The abundances of amino acid metabolism and lipid metabolism were enriched.Moreover,CdSe QDs decreased the fluorescence intensity of tryptophan-like protein from 2,326 to 1,179 a.***.in loosely bound extracellular polymeric substances(EPSs)and from 3,792 to 3,117 a.***.in tightly bound EPSs.To relieve CdSe QD stress,the polysaccharide content increased from0.31 to 0.61 mg/g MLSS and intracellular antioxidant defense was activated.With CdSe QD level increasing to 30 mg/L,the total antioxygenic capacity and the activities of catalase were enhanced up to 411%and 143.2%,respectively.Thereby,CdSe QDs had little adverse effects on cell membrane integrity,microbial metabolism and the abundance of Nitrospirae.

Number of Solitons Emerged in the Initial Profile of Shallow Water Using Convolutional Neural Networks

The soliton resolution conjecture proposes that the initial value problem can evolve into a dispersion part and a soliton part.However,the problem of determining the number of solitons that form in a given initial profile remains unsolved,except for a few specific cases.In this paper,the authors use the deep learning method to predict the number of solitons in a given initial value of the Korteweg-de Vries(KdV)equation.By leveraging the analytical relationship between Asech^(2)(x)initial values and the number of solitons,the authors train a Convolutional Neural Network(CNN)that can accurately identify the soliton count from spatio-temporal data.The trained neural network is capable of predicting the number of solitons with other given initial values without any additional assistance.Through extensive calculations,the authors demonstrate the effectiveness and high performance of the proposed method.

Background:Hepatic ischemia-reperfusion injury(HIRI)remains a common complication during liver transplantation(LT)in patients.As a key downstream effector of the Hippo pathway,Yes-associated protein(YAP)has been reported to be involved in various physiological and pathological processes.However,it remains elusive whether and how YAP may control autophagy activation during ischemia-reperfusion.Methods:Human liver tissues from patients who had undergone LT were obtained to evaluate the correlation between YAP and autophagy activation.Both an in vitro hepatocyte cell line and in vivo liver-specific YAP knockdown mice were used to establish the hepatic ischemia-reperfusion models to determine the role of YAP in the activation of autophagy and the mechanism of regulation.Results:Autophagy was activated in the post-perfusion liver grafts during LT in patients,and the expression of YAP positively correlated with the autophagic level of hepatocytes.liver-specific knockdown of YAP inhibited hepatocytes autophagy upon hypoxia-reoxygenation and HIRI(Pling through binding to the transcriptional enhanced associate domain(TEAD).Conclusions:YAP protects against HIRI by inducing autophagy via JNK signaling that suppresses the apoptosis of hepatocytes.Targeting Hippo(YAP)-JNK-autophagy axis may provide a novel strategy for the prevention and treatment of HIRI.

Modeling and Performance Analysis of UAV-Aided Millimeter Wave Cellular Networks with Stochastic Geometry

UAV-aided cellular networks,millimeter wave(mm-wave) communications and multi-antenna techniques are viewed as promising components of the solution for beyond-5G(B5G) and even 6G communications.By leveraging the power of stochastic geometry,this paper aims at providing an effective framework for modeling and analyzing a UAV-aided heterogeneous cellular network,where the terrestrial base stations(TBSs) and the UAV base stations(UBSs) coexist,and the UBSs are provided with mm-wave and multi-antenna techniques.By modeling the TBSs as a PPP and the UBSs as a Matern hard-core point process of type Ⅱ(MPH-Ⅱ),approximated but accurate analytical results for the average rate of the typical user of both tiers are derived through an approximation method based on the mean interference-to-signal ratio(MISR) gain.The influence of some relevant parameters is discussed in detail,and some insights into the network deployment and optimization are revealed.Numerical results show that some trade-offs are worthy of being considered,such as the antenna array size,the altitude of the UAVs and the power control factor of the UBSs.

Performance Design of High-Temperature Chloride Salts as Thermal Energy Storage Material

The chloride salts have great potential used as high-temperature thermal energy storage(TES) medium for the concentrated solar power system.In this study,liCl,KCl and CaCl_(2) were selected as energy storage materials in order to further broaden the working temperature of ternary chloride salt and improve its energy storage density.The new high-temperature energy storage ternary chloride composed of liCl,KCl,and CaCl_(2) was developed based on the phase diagram generated by FactSage.Three components of liCl-KCl-CaCl_(2) with the mass ratio of 37.85%-53.38%-8.77%,30.90%-13.82%-55.28% and 1.78%-18.61%-79.61% were developed,of which the corresponding melting temperature(T_(m)) 340.93,433.57 and 626.85℃,respectively,was obtained either.Considering that T_(m) of the third group of salts was too high,only the first two groups of salts were tested.DSC test showed that the actual melting point was only 0.46% and 1.64% different from the melting point predicted by Factsage.The thermal properties of the two ternary chloride salts were also compared.The solid and liquid-specific heat of ternary salts was determined by DSC using sapphire as the standard reference.The vapor pressure and decomposition temperature of ternary chloride salts were investigated.The results showed that the vapor pressure of salt 1 was almost constant below 650℃ by FactSage.Meanwhile,the TG results showed that the upper working temperature of salt 1 was 650℃ under the air atmosphere.In addition,the ternary chloride salts after short-term cycling still exhibited excellent thermal properties,which revealed that these good thermal properties make them have broad application prospects in high-temperature thermal energy storage systems.

Vanadium oxide nanospheres encapsulated in N-doped carbon nanofibers with morphology and defect dual-engineering toward advanced aqueous zinc-ion batteries

Vanadium-based electrodes are regarded as attractive cathode materials in aqueous zinc ion batteries(ZIBs)caused by their high capacity and unique layered structure.However,it is extremely challenging to acquire high electrochemical performance owing to the limited electronic conductivity,sluggish ion kinetics,and severe volume expansion during the insertion/extraction process of Zn^(2+).Herein,a series of V_(2)O_(3)nanospheres embedded N-doped carbon nanofiber structures with various V_(2)O_(3)spherical morphologies(solid,core-shell,hollow)have been designed for the first time by an electrospinning technique followed thermal treatments.The N-doped carbon nanofibers not only improve the electrical conductivity and the structural stability,but also provides encapsulating shells to prevent the vanadium dissolution and aggregation of V_(2)O_(3)particles.Furthermore,the varied morphological structures of V_(2)O_(3)with abundant oxygen vacancies can alleviate the volume change and increase the Zn^(2+)pathway.Besides,the phase transition between V_(2)O_(3)and Zn_XV_(2)O_(5-m)·n H_(2)O in the cycling was also certified.As a result,the as-obtained composite delivers excellent long-term cycle stability and enhanced rate performance for coin cells,which is also confirmed through density functional theory(DFT)calculations.Even assembled into flexible ZIBs,the sample still exhibits superior electrochemical performance,which may afford new design concept for flexible cathode materials of ZIBs.

The development of highly active,stable and inexpensive electrocatalysts for hydrogen production by defects and morphology engineering remains a great challenge.Herein,S vacancies-rich Ni_(3)S_(2)@Cu_(2)S nan-otube heterojunction arrays were in-situ grown on copper foam(V_(s)-Ni_(3)S_(2)@V_(s)-Cu_(2)S NHAs/CF)for efficient electrocatalytic overall water splitting.With the merits of nanotube arrays and efficient electronic mod-ulation drived by the OD vacancy defect and 2D heterojunction defect,the resultant V_(s)-Ni_(3)S_(2)@V_(s)-Cu_(2)S NHAs/CF electrocatalyst exhibits excellent electrocatalytic activity with a low overpotential of 47 mV for the hydrogen evolution reaction(HER)at 10 mA cm^(-2) current density,and 263 mV for the oxygen evolution reaction(OER)at 50 mA cm^(-2) current density,as well as a cell voltage of 1.48 V at 10 mA cm^(-2).Moreover,the nanotube heterojunction arrays endows V_(s)-Ni_(3)S_(2)@V_(s)-Cu_(2)S NHAs/CF with outstanding stability in long-term catalytic processes,as confirmed by the continuous chronopotentiom-etry tests at current densities of 10 mA cm^(-2) for 100 h.

Femtosecond laser fabrication of 3D vertically aligned micro-pore network on thick-film li4Ti5O12 electrode for high-performance lithium storage

The development of energy storage devices with high energy density relies heavily on thick film electrodes, but it is challenging due to the limited ion transport kinetics inherent in thick electrodes. Here,we report on the preparation of a directional vertical array of micro-porous transport networks on LTO electrodes using a femtosecond laser processing strategy, enabling directional ion rapid transport and achieving good electrochemical performance in thick film electrodes. Various three-dimensional(3D)vertically aligned micro-pore networks are innovatively designed, and the structure, kinetics characteristics, and electrochemical performance of the prepared ion transport channels are analyzed and discussed by multiple characterization and testing methods. Furthermore, the rational mechanisms of electrode performance improvement are studied experimentally and simulated from two aspects of structural mechanics and transmission kinetics. The ion diffusion coefficient, rate performance at 60 C,and electrode interface area of the laser-optimized 60-15% micro-porous transport network electrodes increase by 25.2 times, 2.2 times, and 2.15 times, respectively than those of untreated electrodes.Therefore, the preparation of 3D micro-porous transport networks by femtosecond laser on ultra-thick electrodes is a feasible way to develop high-energy batteries. In addition, the unique micro-porous transport network structure can be widely extended to design and explore other high-performance energy materials.
