OIP5 Interacts with NCK2 to Mediate Human Spermatogonial Stem Cell Self-Renewal and Apoptosis through Cell Cyclins and Cycle Progression and Its Abnormality Is Correlated with Male Infertility

Spermatogonial stem cells(SSCs)have important applications in both reproduction and regenerative medicine.Nevertheless,specific genes and signaling transduction pathways in mediating fate decisions of human SSCs remain elusive.Here,we have demonstrated for the first time that OIP5(Opa interacting protein 5)controlled the self-renewal and apoptosis of human SSCs.RNA sequencing identified that NCK2 was a target for OIP5 in human SSCs,and interestingly,OIP5 could interact with NCK2 as shown by Co-IP(co-immunoprecipitation),IP-MS(mass spectrometry),and GST pulldown assays.NCK2 silencing decreased human SSC proliferation and DNA synthesis but enhanced their apoptosis.Notably,NCK2 knockdown reversed the influence of OIP5 overexpression on human SSCs.Moreover,OIP5 inhibition decreased the numbers of human SSCs at S and G2/M phases,while the levels of numerous cell cycle proteins,including cyclins A2,B1,D1,E1 and H,especially cyclin D1,were remarkably reduced.Significantly,whole-exome sequencing of 777 patients with nonobstructive azoospermia(NOA)revealed 54 singlenucleotide polymorphism mutations of the OIP5 gene(6.95%),while the level of OIP5 protein was obviously lower in testes of NOA patients compared to fertile men.Collectively,these results implicate that OIP5 interacts with NCK2 to modulate human SSC self-renewal and apoptosis via cell cyclins and cell cycle progression and that its mutation and/or lower expression is correlated with azoospermia.As such,this study offers novel insights into molecular mechanisms underlying the fate determinations of human SSCs and the pathogenesis of NOA,and it provides new targets for treating male infertility.

Vagus nerve stimulation in cerebral stroke:biological mechanisms,therapeutic modalities,clinical applications,and future directions

Stroke is a major disorder of the central nervous system that poses a serious threat to human life and quality of life.Many stro ke victims are left with long-term neurological dysfunction,which adversely affects the well-being of the individual and the broader socioeconomic impact.Currently,poststroke brain dysfunction is a major and difficult area of treatment.Vagus nerve stimulation is a Food and Drug Administration-approved exploratory treatment option for autis m,refractory depression,epilepsy,and Alzheimer’s disease.It is expected to be a novel therapeutic technique for the treatment of stroke owing to its association with multiple mechanisms such as alte ring neurotransmitters and the plasticity of central neuro ns.In animal models of acute ischemic stroke,vagus nerve stimulation has been shown to reduce infarct size,reduce post-stroke neurological damage,and improve learning and memory capacity in rats with stroke by reducing the inflammatory response,regulating bloodbrain barrier permeability,and promoting angiogenesis and neurogenesis.At present,vagus nerve stimulation includes both invasive and non-invasive vagus nerve stimulation.Clinical studies have found that invasive vagus nerve stimulation combined with rehabilitation therapy is effective in im proving upper limb motor and cognitive abilities in stroke patients.Further clinical studies have shown that non-invasive vagus nerve stimulation,including ear/ce rvical vagus nerve stimulation,can stimulate vagal projections to the central nervous system similarly to invasive vagus nerve stimulation and can have the same effect.In this paper,we first describe the multiple effects of vagus nerve stimulation in stroke,and then discuss in depth its neuroprotective mechanisms in ischemic stroke.We go on to outline the res ults of the current major clinical applications of invasive and non-invasive vagus nerve stimulation.Finally,we provide a more comprehensive evaluation of the advantages and disadvantages of different types of vagus nerve stimulation in the

The"spleen"in traditional Chinese medicine is the"acquired foundation"and the"source of qi and blood biochemistry".Strengthening the"spleen"is one of the important treat‐ment methods in traditional Chinese medicine;The gut microbiota has been a hot topic in modern scientific research in recent years,and its nutritional and immune functions correspond to some of the functions of the spleen in traditional Chinese medicine.In terms of philosophy,traditional Chinese medicine emphasizes the balance between the spleen and the gut microbiota,emphasizing the importance of supporting the right and eliminating evil,preventing diseases before they occur,seeking the root cause of treat‐ment,and adapting measures to the three causes;Clinically,the treatment of obesity,con‐stipation,diarrhea and other diseases by invigorating the spleen and the treatment mechanism of acupuncture and moxibustion,chiropractic and invigorating the spleen are all related to microecology.This article combines relevant literature on traditional Chinese medicine's"spleen"and"spleen health"to explore the treatment concept,clinical application,and correlation with intestinal microbiota of traditional Chinese medicine's"spleen health".It also elaborates on the latest literature,aiming to exchange ideas with peers and promote research on the microbiota of traditional Chinese medicine's"spleen".

Early diagnosis and management of neonatal sepsis:a perspective

Neonatal sepsis is an illness caused by bloodstream bacteria or fungal infections that affect newborn infants under 28 days of age.It stands as a significant contributor to morbidity and mortality in neonatal intensive care units(NICUs).With the improved survival rates of very low-birth-weight or extremely premature infants,a growing number of these vulnerable infants are receiving intensive care and undergoing invasive procedures,making nosocomial infections a persistent challenge in NICUs.The mortality rate associated with neonatal sepsis varies depending on factors such as birth weight,gestational age,and other co-morbidities.It can also lead to a range of long-term health consequences,including neurodevelopmental disabilities like cerebral palsy,visual or hearing impairments,and cognitive problems.Furthermore,neonatal sepsis is linked to various adverse outcomes,including respiratory complications,bronchopulmonary dysplasia,nutritional and growth issues,and immunological dysfunction[1].due to its atypical clinical presentation and the low rate of specific diagnostic indicators,such as a positive blood culture,accurately diagnosing neonatal sepsis is challenging.Consequently,the appropriateness of antibiotic treatment is often a matter of controversy.

A new class of backward particle systems with sequential interaction is proposed to approximate the mean-field backward stochastic differential equations.It is proven that the weighted empirical measure of this particle system converges to the law of the McKean-Vlasov system as the number of particles grows.Based on the Wasserstein met-ric,quantitative propagation of chaos results are obtained for both linear and quadratic growth conditions.Finally,numerical experiments are conducted to validate our theoretical results.

miR‑423 sponged by lncRNA NORHA inhibits granulosa cell apoptosis

Background Atresia and degeneration,a follicular developmental fate that reduces female fertility and is triggered by granulosa cell(GC)apoptosis,have been induced by dozens of miRNAs.Here,we report a miRNA,miR-423,that inhibits the initiation of follicular atresia(FA),and early apoptosis of GCs.Results We showed that miR-423 was down-regulated during sow FA,and its levels in follicles were negatively correlated with the GC density and the P4/E2 ratio in the follicular fluid in vivo.The in vitro gain-of-function experiments revealed that miR-423 suppresses cell apoptosis,especially early apoptosis in GCs.Mechanically speaking,the miR-423 targets and interacts with the 3’-UTR of the porcine SMAD7 gene,which encodes an apoptosis-inducing factor in GCs,and represses its expression and pro-apoptotic function.Interestingly,FA and the GC apoptosis-related lncRNA NORHA was demonstrated as a ceRNA of miR-423.Additionally,we showed that a single base deletion/insertion in the miR-423 promoter is significantly associated with the number of stillbirths(NSB)trait of sows.Conclusion These results demonstrate that miR-423 is a small molecule for inhibiting FA initiation and GC early apoptosis,suggesting that treating with miR-423 may be a novel approach for inhibiting FA initiation and improving female fertility.

Nest complexity correlates with larger brain size but smaller body mass across bird species

Amniotes differ substantially in absolute and relative brain size after controlling for allometry,and numerous hypotheses have been proposed to explain brain size evolution.Brain size is thought to correlate with processing capacity and the brain’s ability to support complex manipulation such as nest-building skills.The increased complexity of nest structure is supposed to be a measure of an ability to manipulate nesting material into the required shape.The degree of nest-structure complexity is also supposed to be associated with body mass,partly because small species lose heat faster and delicate and insulated nests are more crucial for temperature control of eggs during incubation by small birds.Here,we conducted comparative analyses to test these hypotheses by investigating whether the complexity of species-typical nest structure can be explained by brain size and body mass(a covariate also to control for allometric effects on brain size)across 1353 bird species from 147 families.Consistent with these hypotheses,our results revealed that avian brain size increases as the complexity of the nest structure increases after controlling for a significant effect of body size,and also that a negative relationship exists between nest complexity and body mass.

Prediction of subsurface settlement induced by shield tunnelling in sandy cobble stratum

This study focuses on the analytical prediction of subsurface settlement induced by shield tunnelling in sandy cobble stratum considering the volumetric deformation modes of the soil above the tunnel crown.A series of numerical analyses is performed to examine the effects of cover depth ratio(C/D),tunnel volume loss rate(h t)and volumetric block proportion(VBP)on the characteristics of subsurface settle-ment trough and soil volume loss.Considering the ground loss variation with depth,three modes are deduced from the volumetric deformation responses of the soil above the tunnel crown.Then,analytical solutions to predict subsurface settlement for each mode are presented using stochastic medium theory.The influences of C/D,h t and VBP on the key parameters(i.e.B and N)in the analytical expressions are discussed to determine the fitting formulae of B and N.Finally,the proposed analytical solutions are validated by the comparisons with the results of model test and numerical simulation.Results show that the fitting formulae provide a convenient and reliable way to evaluate the key parameters.Besides,the analytical solutions are reasonable and available in predicting the subsurface settlement induced by shield tunnelling in sandy cobble stratum.

Effects of Number of Bleed Holes on Shock-Wave/Boundary-Layer Interactions in a Transonic Compressor Stator

An extensive numerical investigation is conducted to characterize the flow separation control in a transonic compressor cascade with a porous bleed.The bleed holes are arranged on the suction surface in a single row,two staggered rows and three staggered rows.For each bleed scheme,five bleed pressure ratios are examined at an inlet Mach number of 1.0.The results indicate that the aerodynamic performance of the cascade is significantly improved by the porous bleed.For the single-row scheme,the maximum reduction in total pressure losses is 57%.For the two-staggered-row and three-staggered-row schemes,there is an optimal bleed pressure ratio of 1.0,and the maximum reductions in total pressure loss are 68% and 75%,respectively.The low loss in the cascade is due to the well-controlled boundary layer.The new local supersonic region created by the bleed hole is the key reason for the improved boundary layer.The vortex induced by side bleeding provides another mechanism for delaying flow separation.Increasing the bleed holes could create multiple local supersonic regions,which reduce the range of the adverse pressure gradient that the boundary layer needs to withstand.This is the reason why cascades with more bleed holes perform better.

Gear walk instability caused by brake-disc friction characteristics

A multi-body dynamic rigid-flexible coupling model of landing gear is established to study the gear walk instability caused by the friction characteristics of the brake disc.After validating the model with the experimental results,the influence of the landing gear structure and braking system parameters on gear walk is further investigated.Among the above factors,the slope of the graph for the friction coefficient of the brake disc and the relative velocity of brake stators and rotors is the most influential factor on gear walk instability.Phase trajectory analysis verifies that gear walk occurs when the coupling of multiple factors causes the system to exhibit an equivalent negative damping trend.To consider a more realistic braking case,a back propagation neural network method is employed to describe the nonlinear behavior of the friction coefficient of the brake disc.With the realistic nonlinear model of the friction coefficient,the maximum error in predicting the braking torque is less than 10%and the effect of the brake disc temperature on gear walk is performed.The results reveal that a more negative friction slope may contribute to a more severe unstable gear walk,and reducing the braking pressure is an effective approach to avoid gear walk,which provides help for future braking system design.
