Metabolic responses of indigenous bacteria in chicken faeces and maggots to multiple antibiotics via heavy water labeled single-cell Raman spectroscopy

The use of maggots derived from chicken faeces as fish diets might serve as a vehicle for the widespread of multiple antibiotic resistant bacteria(ARB) in the environment. Heavy water labeled single-cell Raman spectroscopy(D_(2)O-Raman) was applied to detect the metabolic responses of indigenous bacteria in chicken faeces and maggots to different concentrations of combined colistin, kanamycin, and vancomycin. By incubating the samples with D_(2)O and antibiotics, metabolically active bacterial cells to antibiotics were distinguished from those inactive by the exhibition of C-D Raman band. Using the C-D band as a universal metabolic biomarker, 96% and 100% of cells in chicken faeces and maggots were revealed to be metabolically active to 1 × minimum inhibition concentration(MIC) of the aforementioned antibiotics. A noticeable decrease in the percentage of active cells from 96% to 76% in faeces and 100% to 93% in maggots was observed at 5 × MIC of antibiotics. However, these ratios were still far above that obtained from the same faeces(1.84%) and maggots(0.51%) samples using a cultivation method, indicating the wide presence of nongrowing but metabolically active bacterial cells under antibiotic treatment. Conclusively, the cultureindependent D_(2)O-Raman approach detected and quantified a large portion of metabolically active indigenous bacteria to multiple antibiotics in their native environments, illustrating the great potential risks of these active cells to spread antibiotic resistance via food chain.

FAM64A promotes HNSCC tumorigenesis by mediating transcriptional autoregulation of FOXM1

Head and neck squamous cell carcinoma(HNSCC)still lacks effective targeted treatment.Therefore,exploring novel and robust molecular targets is critical for improving the clinical outcome of HNSCC.Here,we reported that the expression levels of family with sequence similarity 64,member A(FAM64A)were significantly higher in HNSCC tissues and cell lines.In addition,FAM64A overexpression was found to be strongly associated with an unfavorable prognosis of HNSCC.Both in vitro and in vivo evidence showed that FAM64A depletion suppressed the malignant activities of HNSCC cells,and vice versa.Moreover,we found that the FAM64A level was progressively increased from normal to dysplastic to cancerous tissues in a carcinogenic 4-nitroquinoline-1-oxide mouse model.Mechanistically,a physical interaction was found between FAM64A and forkhead box protein M1(FOXM1)in HNSCC cells.FAM64A promoted HNSCC tumorigenesis not only by enhancing the transcriptional activity of FOXM1,but also,more importantly,by modulating FOXM1 expression via the autoregulation loop.Furthermore,a positive correlation between FAM64A and FOXM1 was found in multiple independent cohorts.Taken together,our findings reveal a previously unknown mechanism behind the activation of FOXM1 in HNSCC,and FAM64A might be a promising molecular therapeutic target for treating HNSCC.

SWVFS： a saliency weighted visual feature similarity metric for image quality assessment

In this paper, a saliency weighted visual feature similarity （SWVFS） metric is proposed for full reference im- age quality assessment （IQA）. Instead of traditional spatial pooling strategies, a visual saliency-based approach is em- ployed for better compliance with properties of the human visual system, where the saliency allocation is closely related to the activity of posterior parietal cortex and the pluvial nu- clei of the thalamus. Assuming that the saliency map actually represents the contribution of locally computed visual distor- tions to the overall image quality, the gradient similarity and the textural congruency are merged into the final image qual- ity indicator. The gradient and texture comparison play com- plementary roles in characterizing the local image distortion. Extensive experiments conducted on seven publicly available image databases show that the performance of SWVFS is competitive with the state-of-the-art IQA algorithms.

UNCONDITIONALLY OPTIMAL ERROR ANALYSIS OF THE SECOND-ORDER BDF FINITE ELEMENT METHOD FOR THE KURAMOTO-TSUZUKI EQUATION

This paper aims to study a second-order semi-implicit BDF finite element scheme for the Kuramoto-Tsuzuki equations in two dimensional and three dimensional spaces.The proposed scheme is stable and the nonlinear term is linearized by the extrapolation technique.Moreover,we prove that the error estimate in L^(2)-norm is unconditionally optimal which means that there has not any restriction on the time step and the mesh size.Finally,numerical results are displayed to illustrate our theoretical analysis.

A New Classifier for Imbalanced Data Based on a Generalized Density Ratio Model

Achieving higher true positive rate when decreasing false positive rate is always a great challenge to the imbalance learning community.This work combines penalized empirical likelihood method,lower bound algorithm and Nyströmmethod and applies these techniques along with kernel method to density ratio model.The resulting classifier,density ratio classifier(DRC),is a combination of kernelization,regularization,efficient implementation and threshold moving,all of which are critical to enable DRC to be an effective and powerful method for solving difficult imbalance problems.Compared with other methods,DRC is competitive in that it is widely applicable and it is simple and easy to use without additional imbalance handling skills.In addition,the convergence rate of the estimate of log density ratio is discussed as well.And the results of numerical analysis also show that DRC outperforms other methods in AUC and G-mean score.

Single-cell Raman and functional gene analyses reveal microbial P solubilization in agriculture waste-modified soils

Application of agricultural waste such as rapeseed meal(RM)is regarded as a sustainable way to improve soil phosphorus(P)availability by direct nutrient supply and stimulation of native phosphate‐solubilizing microorganisms(PSMs)in soils.However,exploration of the in situ microbial P solubilizing function in soils remains a challenge.Here,by applying both phenotype‐based single‐cell Raman with D_(2)O labeling(Raman‐D_(2)O)and genotype‐based high‐throughput chips targeting carbon,nitrogen and P(CNP)functional genes,the effect of RM application on microbial P solubilization in three typical farmland soils was investigated.The abundances of PSMs increased in two alkaline soils after RM application identified by single‐cell Raman D_(2)O.RM application reduced the diversity of bacterial communities and increased the abundance of a few bacteria with reported P solubilization function.Genotypic analysis indicated that RM addition generally increased the relative abundance of CNP functional genes.A correlation analysis of the abundance of active PSMs with the abundance of soil microbes or functional genes was carried out to decipher the linkage between the phenotype and genotype of PSMs.Myxococcota and C degradation genes were found to potentially contribute to the enhanced microbial P release following RM application.This work provides important new insights into the in situ function of soil PSMs.It will lead to better harnessing of agricultural waste to mobilize soil legacy P and mitigate the P crisis.

Omicron,the new mutant coronavirus,has spread rapidly globally,attracting close attention from different stakeholders worldwide.The complex and constantly changing epidemic situation has had a new impact on the world.Therefore,this paper focuses on the characteristics of the rapid spread of the COVID-19 variant strain.Generally,epidemic prevention experts conduct preliminary screening as part of the existing epidemic plan database according to the current local situation,after which they sort the alternatives deemed more suitable for the situation.Then the decision-makers identify the most divergent expert group,plan for consultation and adjustments,and finally obtain the plan with the smallest divergence.This article aims to integrate the experts'opinions with the method of minimizing the differences,which can maximize the expert consensus and help organize the schemes that best meet the epidemic situation.The experts'negotiation and iteration of the differences in the initial plan align with the current complex and dynamic epidemic situation and are of great significance to the rapid formulation of plans to achieve effective prevention and control.

High linearity U-band power amplifier design:a novel intermodulation point analysis method

A power amplifier’s linearity determines the emission signal’s quality and the efficiency of the system.Nonlinear distortion can result in system bit error,out-of-band radiation,and interference with other channels,which severely influence communication system’s quality and reliability.Starting from the third-order intermodulation point of the milimeter wave(mm-Wave)power amplifiers,the circuit’s nonlinearity is compensated for.The analysis,design,and implementation of linear class AB mm-Wave power amplifiers based on GlobalFoundries 45 nm CMOS silicon-on-insulator(SOI)technology are presented.Three single-ended and differential stacked power amplifiers have been implemented based on cascode cells and triple cascode cells operating in U-band frequencies.According to nonlinear analysis and on-wafer measurements,designs based on triple cascode cells outperform those based on cascode cells.Using single-ended measurements,the differential power amplifier achieves a measured peak power-added efficiency(PAE)of 47.2%and a saturated output power(P_(sat))of 25.2 dBm at 44 GHz.The amplifier achieves a P_(sat)higher than 23 dBm and a maximum PAE higher than 25%in the measured bandwidth from 44 GHz to 50 GHz.

A new global land productivity dynamic product based on the consistency of various vegetation biophysical indicators

Changes in land productivity have been endorsed by the Inter Agency Expert Group on Sustainable Development Goals(IAEGSDGs)as key indicators for monitoring SDG 15.3.1.Multiple vegeta-tion parameters from optical remote sensing techniques have been widely utilized across different land productivity decline processes and scales.However,there is no consensus on indicator selection and their effectiveness at representing land productivity declining at different scales.This study proposes a fusion framework that incorporates the trends and consistencies within the four com-monly used remote sensing-based vegetation indicators.We ana-lyzed the differences among the four vegetation parameters in different land cover and climate zones,finally producing a new global land productivity dynamics(LPD)product with confidence level degrees.The LPD classes indicated by the four vegetation indicators(VIs)showed that all three levels(low,medium,and high confidence)of increasing area account for 23.99%of the global vegetated area and declining area account for 7.00%.The Increase high-confidence(HC)area accounted for 2.77%of the total area,and the Decline-HC accounted for 0.35%of the total area.This study demonstrates the accuracy of the high-confidence(HC)area for the evaluation of land productivity decline and increase.The“forest”landcover type and“humid”climate zone had the largest increasing and declining area but had the lowest high-confidence proportion.The data product provides an important and optional reference for the assessment of SDG 15.3.1 at global and regional scales according to the specific application target.
